콜라겐은 포유동물의 가장 풍부한 동물성 단백짙로, 전체 단백질의 약 30 %를 차지하며, 결합 조직에 존재하며 대부분의 장기의 구조적 지지에 기여한다. Tripeptide (glycine-proline-hydroxyproline: INCI name Tripeptide-29) 는 collagen type 1의 주성분이며, palmitoyl tripeptide (palmitoyl-glycine-proline-hydroxyproline: INCI name Palmitoyl Tripeptide-29) 는 콜라겐의 합성을 촉진하는 항노화 불질로서 디자인된 합성소재이다 합성된 웹타이드 유도체는 HPLC를 이용하여 분식하였다. in vitro test를 통하여 콜라겐합성과 섬유아세포 증식 효능을 확인하였고, 비침습적 기기를 사용하여 피검자에 대한 8주간의 적용결과 피부주름과 탄력의 상당한 개선을 확인하였다. Palmitoyl tripeptide는 우수한 항노화 효능을 갖는 화장품 소재로 사료된다.

Background: Topical hemostatic agents are used when ligation, electrocauterization, or other conventional hemostatic methods are impractical. Because a hemostatic agent is a foreign body, it can cause foreign body reactions, inflammation, and infections that can interfere with the wound healing process. Therefore, we should select hemostatic agents after considering their effects on wound healing. Here, we compared the effects of hemostatic agents on wound healing in a rectus abdominis muscle defect in rats. Methods: Twelve Sprague Dawley rats were subjected to creation of a $6{\times}6mm$ defect in the rectus abdominis muscle and divided into four groups: control group; group A, Tachosil fibrin sealant patch; group B, Surgicel Fibrillar oxidized regenerated cellulose; and group C, Surgicel Snow oxidized regenerated cellulose. For the histologic analysis, biopsies were performed on the 3rd, 7th, and 27th days. Results: The foreign body reaction was the weakest in group A and most significant in group C. The inflammatory cell infiltration was the weakest in group A and similar in groups B and C. Muscle regeneration differed among periods. The rats in group A were the most active initially, while those in group C showed prolonged activity. Conclusion: Tachosil and Surgicel administration increased inflammation via foreign body reactions, but the overall wound healing process was not significantly affected. The increased inflammation in the Surgicel groups was due to a low pH. We recommend using Tachosil, because it results in less intense foreign body reactions than Surgicel and faster wound healing due to the fibrin action.
Shea butter (Vitellaria paradoxa) is a fat extracted from shea tree nuts and contains relatively high levels of non-glycerides. Triterpenes, the main non-glyceride component, exhibit a variety of biological activities such as antitumor, antibacterial, and anti-inflammatory. Shea butter extract (SBE) has been used to treat various skin problems such as burns, eczema, and rash in human medicine, but little is known about the activity of SBE on canine skin. This study evaluated the cytotoxicity and anti-inflammatory effect of SBE in canine keratinocytes. Cytotoxicity of lipopolysaccharide (LPS, 5-50 ng/mL) and SBE (50-200 ㎍/mL) was evaluated using the CCK-8 assay. Non-cytotoxic concentrations of LPS and SBE were administered to canine cell cultures to evaluate anti-inflammatory effects. To evaluate the anti-inflammatory activity of SBE, the levels of IL-1β, IL-8, IL-12, and TNF-α were measured using ELISA kits. The concentration of each cytokine was quantified in control, LPS-treated, LPS + SBE-treated groups. Increased levels of IL-1β, IL-8, and IL-12 were found in LPS-treated groups relative to control groups. LPS + SBE-treated groups showed a lower level of IL-1β, IL-8, and IL-12 than LPS-treated groups. These results suggest that SBE may have application as a topical agent for canine inflammatory skin diseases. However, further in vivo study is needed to evaluate the safety and efficacy of SBE in dogs.
Background: A wide range of environmental factors, such as diseases, nutritional deficiencies, ageing, hormonal imbalances, stress, and ultraviolet (UV) radiation, may affect the structure and function of the skin that covers the entire surface of the human body. In this study, we investigated roles of red ginseng oil (RGO) in enhancing skin functions, including hair growth and skin protection, using mouse models. Methods: For hair growth experiment, shaved dorsal skins of C57BL/6 mice were topically applied with vehicle, RGO, RGO's major compounds, or minoxidil for consecutive 21 days and skin tissues were examined the hair growth promoting capacity. For skin protection experiment, SKH-1 hairless mice were topically applied with vehicle or RGO twice a day for three days prior to exposure to UVC radiation at 20 kJ/cm2. Skin tissues were collected to evaluate skin protective effects of RGO. Results: Topical application of RGO to C57BL/6 mice effectively promoted hair regeneration by inducing early telogen-to-anagen transition and significantly increasing the density and bulb diameter of hair follicles. Major compounds, including linoleic acids and β-sitosterol, contributed to RGO-promoted hair growth. Treatment with RGO as well as its major components upregulated expression of hair growth-related proteins. Furthermore, in SKH-1 hairless mice, RGO had a protective effect against UVC-induced skin damage by inhibiting inflammation and apoptosis, as well as inducing cytoprotective systems. Conclusion: These data suggest that RGO may be a potent agent for improving skin health and thereby preventing and/or treating hair loss and protecting skin against UV radiation.

Fibrin glue is a topical agent widely used for hemostasis, wound healing, and surgical adhesion. Complications of fibrin glue itself are extremely rare because it is absorbed over time, but can occur as a result of inappropriate application. We report a case of a postoperative complication caused by inappropriate application of fibrin glue in blow-out fracture surgery. A 65-year-old male patient presented with periorbital swelling and an open wound on the right infraorbital area. Computed tomography showed a right orbital floor fracture. After reduction of the herniated tissue into the orbit, an implant was inserted and fibrin glue was applied to stabilize the implant. This procedure was performed without difficulty, but the patient complained of persistent diplopia and limited eyeball movement after surgery. An imaging study showed a mass-like lesion, which was not a hematoma, in the orbital cavity. In a second operation, the mass was identified as clotted fibrin glue that had not been applied properly. After removal, the patient's symptoms were relieved without further complications. Appropriate and careful application of fibrin glue is necessary to avoid unnecessary complications.

Free flap transplantation with microvascular anastomosis has been successfully performed by development of surgical technique, materials and postoperative monitoring equipments of flap. But success rate of microvascular anastomosis is influenced by various factors, and failure rate is about 5-10%. The most influential factor for success rate is surgical technique and other factors that influence failure of microvascular anastomosis are ischemic time of free flap, thrombus formation of anastomosis region and vascular spasm. Many studies has been published in microvascular anastomosis with histologic effect for irrigating solution. But local irrigation solution has been used clinically in microvascular anastomosis, the comparison with each solution, microhistological study for endothelial cell repair and vascular patency has not been reported. The heparin which is anti-thrombotic agent, and urokinase which is fibrinolytic agent are used for this study. Vascular patency and thrombus formation in experimental micro-arterial anastomosis, and endothelial repair were observed with histologic analysis, scanning electron microscopy, transmission electron microscopic examination. The results were obtained as follows: 1. In vascular patency test in 30 minute and 7 days after micro-arterial anastomosis, equal effects of good vascular patency were obtained in group of local irrigation with heparin and urokinase. 2. In thrombus formation in 7 days after micro-arterial anastomosis, equal effects of minimal thrombus formation were obtained in group of local irrigation with heparin and urokinase. 3. In toluidin blue staining in 7 days after micro-arterial anastomosis, local destruction of endothelial cell and inner elastic lamina were seen and endothelial repair was not seen. 4. In scanning electron microscope examination in 7 days after micro-arterial anastomosis, endothelial cell was not seen in peripheral to suture materials, thrombus associated fibrin network was observed. 5. In transmission electron microscope examination in 7 days after micro-arterial anastomosis, inflammatory cell was seen within smooth muscle cells in site of endothelial cell destruction, smooth muscle cell around suture material were arranged irregularly, some collagenous change were seen. From the results obtained in this study, same results of good vascular patency and anti-thrombotic effect of heparin and urokinase were obtained as a local irrigation solution, and repair of endothelial cell was not seen in 7 days after micro-arterial anastomosis.
스테로이드 함유 표시가 없는 화장품에서 항염증 효과가 있는 글루코코티코스테로이드인 베타메타손프로피오네이트 성분이 검출되었다. 이 화장품은 외용스프레이 및 샴푸로서 주성분으로 아연피리치온을 함유하는 것으로 표기되어 있었다. 화장품에서 스테로이드 구조와 활성을 갖는 물질의 존재를 확인하기 위하여 실리카겔 박층판을 이용한 박층크로마토그래프를 사용하였으며 이 성분을 분리하기 위해 high-performance liquid chromatography (HPLC)를 이용하여 확인 및 정량을 수행하였다. 분취용 HPLC를 이용하여 스테로이드를 함유한 것으로 판단되는 분획을 모은 다음 nuclear magnetic resonance (NMR) 및 mass spectrometry (MS)를 이용하여 스테로이드 성분을 확인하였다. 스테로이드 표준물질로 베타메타손 17-프로피오네이트 및 베타메타손 21-프로피오네이트를 합성하여 사용하였고 이 표준물질과 HPLC 크로마토그램을 비교하여 스테로이드 성분의 함량을 분석하였다. 이 방법으로 아연피리치온 제제와 같은 일부 시판 화장품에서 스테로이드 성분을 확인하였고 reversed-phase high-performance liquid chromatography (RP HPLC) 상의 유지시간 비교를 통하여 스테로이드 성분을 정량한 결과 시험한 총 8종의 화장품 시료 중 2개 제품에서 0.005 ${\sim}$ 0.02%의 베타메타손프로피오네이트가 검출되었다.
본 실험에서는 피부질환을 위한 새로 구성된 HC001의 처방이 외용제로서 아토피성 피부질환에서의 항염증 효능을 확인하였다. H&E염색을 통한 조직학적 검사에서 DNCB로 유도된 진피 및 표피의 과각화증이 HC001 국소도포로 인해 호전되는 것을 관찰되었으며, 염증과정에서 중요한 지표인 NF-${\kappa}B$의 활성이 억제됨을 확인하였다. 이를 통해 HC001이 아토피성 피부질환의 외용제 사용에 있어 일정한 효과를 나타낼 수 있을 것으로 기대 된다.
최근 연구에서 잠재적 피부 색소침착 경감제로서 파라-쿠마린산(PCA)의 주목되는 특성이 발견되었다. 본 연구의 목적은 이 물질의 자외선 차단 효과를 탐구하는 것이다. 자외선에 노출된 HaCaT 세포의 생존율에 대한 PCA의 영향을 in vitro에서 조사하고, 자외선 흡수 스펙트럼이 유사한 방향성 아미노산 대사물들의 작용과 비교하였다. In vivo시험으로는 PCA 크림(1.5 %)과 크림 베이스를 SKH-1 무모 쥐의 등 피부에 도포하고 UVB에 의한 염증 반응으로 나타나는 피부색(홍반) 및 두께 변화(부종)를 측정하였다. 크림 도포-자외선 조사는 2일 간격으로 총 12회 반복하였다. HaCaT 세포를 UVB에 노출시켰을 때 광량 의존적으로 세포 생존율이 감소하였다. 자외선 노출(10 mJ $cm^{-2}$)에 의한 세포 생존율감소는 100 ${\mu}M$의 PCA, cinnamic acid, urocanic acid, 그리고 indole acrylic acid에 의해 각각 39, 27, 39, 31 %가 억제되었다. 무모 쥐의 등 부위에 도포된 PCA크림(10 ${\mu}g\;cm^{-2}$)은 자외선(150 mJ $cm^{-2}$)-노출 피부의 색 지수, 즉 $L^*$, $a^*$ 및 $b^*$ 값, 그리고 두께의 변화를 각각 59, 50, 58, 53 %씩 억제하였다. 본 연구의 결과는 PCA의 멜라닌 생성 억제 작용을 밝힌 선행 연구와 함께 PCA가 자외선에 노출된 피부의 색소 이상 침착과 염증 반응을 막아줄 수 있음을 시사하였다.
C57BL/6 마우스에서 자외선(Ultraviolet, UV) B 조사에 의한 표피 멜라닌세포의 변화에 대한 보중익기탕(Bu-Zhong-Yi-Qi-Tang, BZYQT)의 효과를 관찰하였다. 마우스에 UVB를 매일 $80\;mJ/cm^2$ (0.5 mW/sec)씩 7일간 조사하고, BZYQT를 UV 조사전 또는 조사 후에 복강내주사 또는 피부에 도포하여, 멜라닌세포 형성 억제효과 및 형성된 멜라닌세포에 대한 미백 효과를 dihydroxyphenylalanine (DOPA) 염색으로 관찰하였다. 마우스의 귀등쪽 표피를 분리하여 관찰한바, 정상대조군에서는 $mm^2$ 당 11-16개의 멜라닌세포가 관찰되었으며, UV 조사 일주일 후 발달된 가지를 가진 DOPA양성 멜라닌세포는 급격히 증가하였다. 멜라닌세포 형성 억제 실험에서는 평균치를 기준으로 복강 내 주사군에서 16.3%, 피부도포군에서 26.6%(p<0.01)의 효과가 관찰되었으며, 형성된 멜라닌세포의 감소 효과 실험에서는 평균치를 기준으로 복강 내 주사군의 경우 3주에 24.0%(p<0.01), 6주에 26.0%(p<0.01)의 효과가 관찰되었고, 피부도포군의 경우 3주에 5.2%, 6주에 12.5%(p<0.05)의 효과를 보였다. 이상의 결과는 BZYQT가 UV에 의한 멜라닌세포 형성 억제제 및 미백제로서의 적용 가능성을 제시하였다.
