The charge densities of humic acid and humic extract have been determined by back titration using spectrophotometry as the method of end point detection and several cationic polymers of different charge density(copolymers of acylamide and dimethylaminoethylacrylate and polydiallyidimethylammonium chloride, PDDA), and a cationic indicator o-toluidine blue(o-Tb). The charge densities of humic acid and humic extract obtained depend on the charge densities of cationic polymers used. When the polymers of lower charge density are used, lower apparent charge density values are obtained, but for polymers of high charge density(above 2.5 meq/g), the measured values are almost identical, but also are nearly the same as that obtained for PDDA which has a high charge density(about 6 meq/g). For polymers of lower charge density, it is considered that the optimum dosage does not correspond to 1 :1 charge neutralization between anionic and cationic groups. Polymers of high charge density should be used in order to achieve a complete charge neutralization. As humic acid and humic extract are dissociable acids, their ionization is heavily pH dependent and so higher charge density values are obtaines with increasing pH.
Polyelectrolyte titration, which was called colloid titration is based on the stoichiometric reaction between oppositely charged polyelectrolytes, This can be used, for instance, to determine the charge density of a cationic polyelectrolyte, using an anionic polyelectrolyte of known charge density, such as potassium polyvinyl sulfate (PPVS). The technique requires a suitable method of end-point detection and there are several possibilities. In this work, two methods have been investigated: visual titrimetry based on the color change of a cationic dye (o-toluidine blue, o-Tb) and spectrophotometry based on the absorbance change corresponding to the color change of the same dye. These have been applied to several cationic polyelectrolytes with different charge density and molecular weight. In all cases, the cationic charge was due to quaternary nitrogen groups. In the case of cationic dye, it was shown that the sharpness depends on the charge density of cationic polyelectrolyte. With the polyelectrolytes of lower charge density, the binding to PPVS is weaker and binding of the dye to PPVS can occur before all of the polyelectrolyte charge has been neutralized. However, by carrying out titrations at several polyelectrolyte concentrations, good linear relationships were found, from which reliable charge density values could be derived. Effects of pH and ionic strength were also briefly investigated. For cationic polyelectrolytes (copolymers of acrylamide and dimethylaminoethy] acrylate), there was some loss of charge at higher pH values, probably as a result of hydrolysis. Increasing ionic strength causes a less distinct color change of o-Tb, as a result of weaker electrostatic interactions.
많은 연구들에서 hMSC를 얻기 위해 centrifugation, fluoroscence activated cell sorter(FACS), magnetic activated cell sorter(MACS)가 이용되어져 왔다. 그러나 centrifugation만을 이용한 경우 순도가 떨어지며 FACS나 MACS의 경우에는 비용, 시간이 많이 드는 단점이 있다. 따라서 이 연구에서는 antibody cocktail을 이용하여 hMSC를 좀더 쉽게 얻어내는 방법에 대해 알아보았다. 사람의 골반에서 12G의 바늘을 이용하여 골수를 흡입한 후 heparin이 들어있는 시험관에 넣고 처리과정을 시행하기 전에 냉장고에 보관하며 가능한 한 빨리 처리 과정을 실시한다. 얻은 골수에 적당량의 RosetteSep( Stemcell Technologies)을 첨가한 후 실온에서 20분간 반응시킨다. 그 후 적당량의 Ficoll-paque위에 골수와 RosetteSep의 혼합물을 섞이지 않게 올리고 원심분리를 이용하여 원하는 세포층을 얻어낸다. 이 세포층을 따로 분리한 뒤 배양한다. 배양 시 세포가 80%이상 차기 전에 계속 passage를 시행하며 배양한다. 이는 세포가 밀도가 높아져 원치 않는 세포로 분화되는 것을 막기 위함이다. 배양된 세포가 다양한 분화능력을 가지고 있는지 알아보기 위해 세 가지로 분화를 유도하였다. 적절한 배지와 적절한 환경에서 배양함으로써 얻어진 세포를 osteoblast, chondroblast, adipocyte로 분화를 유도하였다. 분화된 세포가 원하는 형질의 세포로 분화되었는지를 확인하기 위하여 osteoblast의 경우 alizarin red staining, alkaline phosphatase activity, chondroblast의 경우 toluidine blue staining, adipocyte의 경우 Oil-Red-O staining으로 염색하여 분화를 확인하였다. 분리해낸 세포는 각각 세 가지 세포로 분화가 되었으며 이는 RosetteSep이 hMSC를 성공적으로 분리해냈다는 것을 보여준다. 그러나 모든 세포가 분화를 보이지는 않았으며 따라서 hMSC의 순도를 높이기 위한 연구가 더 필요하다. RosetteSep을 이용하면 다른 방법들 보다 쉽게 hMSC를 얻을 수 있으나 기존의 방법과 순도의 측면에서 더 비교할 필요가 있다.
Bacterial cellulose (BC) is generated from citrus gel by Gluconacetobacter hansenii TL-2C. BC has good properties such as high-burst pressure, high-water contact and the ultrafine highly nanofibrous structure of mimic natural extracellular matrix (ECM) for tissue engineering. In this study, acrylic acid (AAc) was grafted onto BC surfaces under aqueous conditions using gamma-ray irradiation, and then immobilized gelatin onto AAc-g-BC. The characterization of scaffolds was performed by scanning electron microscopy (SEM), attenuated total reflectance-Fourier transform infrared spectroscopy (ATR-FTIR), toluidine blue O (TBO) assay. Morphology of gelatin and AAc incorporation onto BC nanofibers did not changed. Our study suggests that gelatin-immobilized BC nanofibers scaffold has a potentiality to fabricate 3D nanofibrous scaffolds for tissue engineering.
In tissue engineering application, a fibrous structure of scaffolds has been issued as an alternative system to regulate cell survival and tissue regeneration, and electrospinning technique has been popularly used to generate fibrous meshes or sheets mimicking the structure of native extracellular matrix (ECM). However, recent strategy in the scaffold development is expanded to provide the structural property as well as a biological property of native ECM, a variety of surface modification techniques have been used to introduce biological property. In this study, we developed biomimetic poly(L-lactide-co-${\varepsilon}$-caprolactone) (PLCL) nano- and micro-fibrous scaffolds as a unique platform with structural and biological properties with native ECM using electrospinning method and gamma-ray irradiation. Surface morphology of the scaffolds was observed by scanning electron microscopy, and alteration of surface property was evaluated with toluidine blue O staining, water contact angle measurement and ATR-FTIR analysis.

Background: Ginsenoside Rb1 (G-Rb1), one of the major active compounds in Panax ginseng, has already been shown to reduce inflammation in various diseases. Osteoarthritis (OA) has traditionally been considered a degenerative disease with degradation of joint articular cartilage. However, recent studies have shown the association of inflammation with OA. In the present study, we investigated whether Rb1 had an antiinflammatory effect on monoiodoacetate (MIA)-induced OA in ovariectomized rats as a model of postmenopausal arthritis. Methods: G-Rb1 at a dosage of 3 and 10 ㎍/kg body weight was administered every 3 days intraarticularly for a period of 4 weeks to observe antiarthritic effects. Diclofenac (10 mg/kg) served as a positive control. Results: The administration of Rb1 significantly ameliorated OA inflammatory symptoms and reduced serum levels of inflammatory cytokines. Furthermore, G-Rb1 administration considerably enhanced the expression of bone morphogenetic protein-2 and collagen 2A and reduced the levels of matrix metalloproteinase-13 genes, indicating a chondroprotective effect of G-Rb1. G-Rb1 also significantly reduced the expression of several inflammatory cytokines/chemokines (interferon gamma (IFN-γ), monocyte chemoattractant protein-1 (MCP-1)/CCL-2, interleukin [IL]-1β, and IL-6). Histological analysis demonstrated that G-Rb1 significantly attenuated the pathological changes in MIA-induced OA in ovariectomized rats. Safranin O and toluidine blue staining further demonstrated that G-Rb1 effectively prevented the degradation of cartilage and glycosaminoglycans, respectively. Conclusion: Overall, our results suggest that G-Rb1 exerts cartilage protective effect on MIA-induced ovariectomized OA rats, by inhibiting inflammatory mediators such as IL-6, IL-1β, MCP-1/CCL-2, cyclooxygenase-2 (COX-2), and prostaglandin E2 (PGE2). These results shed a light on possible therapeutic application of G-Rb1 in OA.
실험 동물의 면역기능을 인위적으로 억제시킴과 동시에 몇 가지 항생 물질을 투여하여 Pneumocystis carin힐 또는 다른 원인에 의한 폐염을 발현시킨 다음 각 원인별 치사율 및 평균 생 존 기간(mean survival day) 등을 비교함으로써 간질성 폐염의 원인충으로서 흑 carinil히 중요성 을 알아보고자 이 연구를 실시하였다. 실험 동물로 Sprague-Dawley계 흰쥐 90마리를 사용하였고 각각 15마리씩 1개 대조군과 다음 5개 투약군으로 분류하였다. 1군 prednisolone(25mg/kg twice weekly), 2군 prednisolone과 tetracycline (75mg/kg/day) , 3군 prednisolone, tetracycline 및 trimethoprim·sulfamethoBazole (50∼250mg/kg/day), 4군 prednisolone과 trimethoprim-sulfamethozazole, 5군 prednisolone과 griseofulvin(300 mg/kg/day). 상기 약제를 횐쥐가 죽을 때까지 투여하고 각 흰쥐의 생존 기간을 측정하였다. 죽은 흰쥐는 즉시 부검하여 폐장을 적출한 후 접촉 도말 표본과 조직 절편 표본을 만들고 toluidine blue O, Giemsa, Gomori's methenamine-silver, hematoxylin-eosin, Brown & Brenn 염색 등을 하여 관찰하였다. 각 군별 평균 생존 기간은 1군 $19.3{\pm}5.2$일, 2군 $41.1{\pm}14.0$일, 3군 $50.5{\pm}18.4$일, 4군 $43.0{\pm}22.9$일, 5군 $21.8{\pm}5.1$일이었다. 카 군별 생존 기간의 차이를 비교하면 1군과 2군, 1군과 3군 및 1 군과 4군이 각각 유의한 차이(p<0.01)를 보여 세균 감염 방지시 생존 기간이 연장되는 것으로 나타났다. 또 1군과 5군의 생존 기간은 거의 차이가 없었고, 2군과 5군, 3군과 5군 및 4군과 5군은 유의한 차이(p<0.01)를 보여 진균 감염보다는 세균이나 P. carinii 감염이 흰쥐 사인에 직접적인 영향을 주는 것으로 나타났다. P. carinii 폐염에 의한 사망은 실험 17일에 최초로 나타났으며 각 군별 비율은 3군이 92.9%로 가장 높고, 2군 및 5군이 80.0%, 4군이 78.6%, 1군이 33.3%를 보였다. P. carinii 폐염으로 죽은 53마리를 분류한 바 stage 1은 11.3%, stage 2는 28.3%, stage 3은 60.4%이었다. 죽은 흰쥐의 사인을 기간별로 분류했을 때 실험 전기(1일∼28일)는 세균 감염에 의한 사망이 많 고, 실험 중기(29∼56일)는 주로 P. carinii 폐염에 의한 사망이었으며 진균과의 혼합감염이 이 시 기에 나타났다. 실험 후기 (57∼82일)에는 세균, 진균 감염 없이 모두 P. carinii 폐염으로 사망하 였다. 이상의 결과로 볼 때 면역억제제 투여 1개월 이후 흰쥐의 사망 원인으로는 세균이나 진균성 폐염 보다 P. carinii 폐염이 더욱 중요하다는 것을 알 수 있었다.
치주염이나 구취발생과 관련된 구강미생물에 대한 저출력레이저의 살균효과를 관찰하기 위하여, Porphyromonas gingivalis 2561 (P. gingivalis 2561), Prevotella intermedia (Pr. intermedia)에 광감수성증가물질을 처리함으로써 실험균주의 광민감성을 증가시킨 후 저출력레이저 650nm의 파장을 연속적으로 1분, 2분, 3분, 5분간, 각각 1cm, 2cm, 3cm 거리에서 조사하여 다음과 같은 결과를 얻었다. 1. P. gingivalis 2561의 경우 Toluidine blue O(TBO) 처리 후 650nm의 파장으로 3cm의 거리에서 1분간 조사한 실험군은 TBO만 처리한 실험대조군에 비해 99.99% 이상의 항균효과를 나타내었다. 2. Pr. intermedia의 경우 TBO 처리 후 650 nm의 파장으로 3 cm의 거리에서 1분간 조사한 실험군은 TBO만 처리한 실험대조군에 비해 99.8% 이상의 항균효과를 나타내었다. 3. P. gingivalis 2561의 경우 TBO 처리 후 650 nm의 파장을 연속으로 각각 1분, 2분, 3분간 조사한 실험군의 경우 시간이 길어질수록 살균효과가 점차 증가되었다. 4. P. gingivalis 2561의 경우 조사거리가 1 cm, 2 cm, 3 cm으로 증가할수록 살균효과가 약간 감소되었다. 5. Pr. intermedia의 경우 조사거리가 1 cm, 2 cm, 3 cm으로 증가할수록 살균효과가 약간 감소되었다. 위와 같은 결과를 바탕으로 저출력레이저의 살균효과는 조사시간이 길어질수록, 조사거리가 짧을수록 살균효과가 증가되었는데, 살균효과가 가장 적게 나타난 3 cm의 거리에서 1분간 조사한 실험군에서도 TBO만 처리한 경우보다 99.8% 이상의 살균효과를 나타내었으므로 저출력레이저의 구강미생물에 대한 살균효과는 명백하다고 생각된다. 따라서 photosensitizer의 부작용이 없는 한계 내에서 사용한다면 저출력레이저가 치주염, 임플란트주위염 등에 대한 보조적 치료와 구취조절 등에 매우 유용하게 사용되리라고 생각된다.
교배효율의 증대를 위해서는 정상화분의 확보가 선행되어야하며 화분의 활성을 파악할 필요가 있다. 본 연구에서는 화분의 활성을 확인하기 위해 여러 가지 염색액을 사용하여 광학현미경하에서 관찰하였다. Aceto-carmine과 Alexander's 염색액을 사용한 실험에서 비슷한 정상화분율을 얻을 수 있었고 FCR염색으로 관찰한 결과 살아있는 화분만 정확히 선별해 주기에 가장 낮은 정상화분율을 얻을 수 있었다. TB는 화분의 상태에 따라 달랐으나 Aceto-carmine과 Alexander's 염색액을 사용한 실험에서의 결과와 비슷한 양상을 보였다. 화분 관찰의 경험이 별로 없는 실험자도 쉽게 접근할 수 있는 염색액은 Alexander's 이었다. 정확한 화분의 활성을 조사하는 데에는 FCR이 필수이다. 형광현미경 장치 없이 정상과 비정상화분의 구별이 용이한 Alexander's 염색액으로 관찰한 자료를 바탕으로 살아있는 정상화분율의 예견이 가능하나 정확한 임성을 확인하려면 추가로 FCR test를 해야 한다.
PURPOSE: The purpose of this study was to evaluate whether light-emitting diodes (LED) irradiation could be effective in a noninvasive, therapeutic device for the treatment of osteoarthritis(OA). METHODS: Twenty-four male Sprague-Dawley rats were divided into four groups: Vehicle control (saline); monosodium iodoacetate-injection (MIA); LED irradiation after MIA injection (MIA-LED); indomethacin-treatment after MIA injection (MIA-IMT). OA was induced by intra-articular injection of 3 mg MIA through the patellar ligament of the right knee. Vehicle control rats were injected with an equivalent volume of saline. The LED was irradiated for 15 min/day for a week after 7 days of MIA treatment. To compare with the effect of LED irradiation, the indomethacin was administrated 20 mg/kg twice a week orally after 7 days of MIA treatment. Knee joints were removed and fixed overnight in 10% neutral buffered formalin and decalcified by EDTA for 2 week before being embedded in paraffin. The assessment of OA induction were monitored by knee movement and radiographic finding. Histologic analysis were performed following staining with hematoxylin and eosin, safranin O-fast green, or toluidine blue, picrosirius red, and histologic changes were scored according to a modified Mankin system. Apoptotic cell in tissue sections was detected using TUNEL method. RESULTS: Radiographic examination could not show the differences between the MIA-treated and the MIA-LED-treated rats. In the histologic analysis, however, LED irradiation prevented cartilage damage and subchondral bone destruction, and significantly reduced mononuclear inflammatory cell infiltration and pannus formation. LED irradiation also reduced apoptosis of cartilage cells, but it prevented apoptosis of infiltrated inflammatory cells in synovium. In addition, LED irradiation showed an increase of collagen production in the meniscus. CONCLUSION: These results suggest that the 840 nm LED irradiation would be a suitable non-thermal phototherapy for the treatment of OA, as a cartilage protection and anti-inflammatory modality.

회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
