The 52 kD Protein Gene of Odontoglossum Ringspot Virus Containing RNA-Dependent RNA Polymerase Motifs and Comparisons with Other Tobamoviruses

  • Complementary DNA of the genomic RNA of odontoglossum ringspot virus Cymbidium strain (ORSV-Cy) was synthesized from polyadenylated viral RNA and cloned. Selected clones containing the viral RNA-dependent RNA polymerase gene of the virus has been sequenced by automated sequencing system. The complete nucleotide sequence of an open reading frame is 1377 base pairs in length, and encodes a protein of 458 amino acids about 52, 334 D. The 52 kD protein of ORSV shares four sequence motifs characteristic of viral RNA-dependent RNA polymerase. Comparison of the ORSV 52 kD protein sequence with that of other five viruses in tobamovirus group showed 76.0 to 60.7% homologies at the amino acid level and the conservation of the four motifs betwen the viruses.

Pseudomonas oleovorans Strain KBPF-004 Culture Supernatants Reduced Seed Transmission of Cucumber green mottle mosaic virus and Pepper mild mottle virus, and Remodeled Aggregation of 126 kDa and Subcellular Localization of Movement Protein of Pepper mild mottle virus

  • Efforts to control viral diseases in crop production include several types of physical or chemical treatments; antiviral extracts of a number of plants have also been examined to inhibit plant viral infection. However, treatments utilizing naturally selected microorganisms with activity against plant viruses are poorly documented. Here we report isolation of a soil inhabiting bacterium, Pseudomonas oleovorans strain KBPF-004 (developmental code KNF2016) which showed antiviral activity against mechanical transmission of tobamoviruses. Antiviral activity was also evaluated in seed transmission of two tobamoviruses, Pepper mild mottle virus (PMMoV) and Cucumber green mottle mosaic virus (CGMMV), by treatment of seed collected from infected pepper and watermelon, respectively. Pepper and watermelon seeds were treated with culture supernatant of P. oleovorans strain KBPF-004 or control strain ATCC 8062 before planting. Seeds germinated after treatment with water or ATCC 8062 yielded about 60% CGMMV or PMMoV positive plants, whereas < 20% of KBPF-004-treated seeds were virus-infected, a significantly reduced seed transmission rate. Furthermore, supernatant of P. oleovorans strain KBPF-004 remodeled aggregation of PMMoV 126 kDa protein and subcellular localization of movement protein in Nicotiana benthamiana, diminishing aggregation of the 126 kDa protein and essentially abolishing association of the movement protein with the microtubule network. In leaves agroinfiltrated with constructs expressing the coat protein (CP) of either PMMoV or CGMMV, less full-size CP was detected in the presence of supernatant of P. oleovorans strain KBPF-004. These changes may contribute to the antiviral effects of P. oleovorans strain KBPF-004.

  • 시판고추 종자로부터 Tobamovirus를 검출하고 Tobamovirus 오염률을 조사하고자 gelatin particle agglutination test (GPAT)의 최적조건을 조사하였다. Gelatin 입자에 감작할 담배 모자이크 바이러스 고추 계통에 대한 ${\gamma}$-globulin G 의 최적농도는 100$\mu\textrm{g}$/$m\ell$이었고, GPAT의 바이러스 검출 감도는 효소면역항체법 및 dot immuno-binding assay와 유사하였으며, Tobamovirus 검출을 위한 종자시료의 최적 희석배수는 5배에서 25배였다. 그리고 최적화된 GPAT에 의한 국내시판 고추종자의 Tobamovirus 오염률은 평균 79.1%인 것으로 판명되었다.

Immunocapture RT-PCR을 이용한 박과작물 종자전염 바이러스의 검출 (Immunocapture RT-PCR for Detection of Seed-borne Viruses on Cucurbitaceae Crops)

  • 박과작물에 발생하는 종자전염 바이러스 3종(CGMMV, KGMMV, ZGMMV)에 대한 검출을 위해 IC-RT-PCR의 적용 가능성을 시험하였다. IC-RT-PCR을 이용하여 감염 종자와 잎으로 부터 바이러스의 특이적인 검출이 가능하였으며, 검출 민감도는 ELISA 보다 100배 이상 높았다. 또한 반응을 마친 ELISA 마이크로플레이트에 남아있는 항원을 IC-RT-PCR의 template로 사용할 경우 ELISA 결과를 곧바로 확인할 수 있었다. 따라서 IC-RT-PCR에 의한 본 검정방법은 박과작물의 대규모 포장검사 및 종자검사에 편리하게 사용될 수 있을 것으로 기대된다.

Evaluation of Resistance to Pepper Mild Mottle Virus (PMMoV) in Pepper Germplasm

  • The pepper mild mottle virus (PMMoV), belonging to the tobamovirus genus, is currently one of the most destructive pathogens in pepper production. Tobamoviruses have been classified in terms of increased pathogenicity as pathotypes P0, P1, P1,2, P1,2,3 and P1,2,3,4, based on their ability to infect systemically Capsicum L0 , L1 , L2 , L3 and L4 resistant plants, respectively. Two hundred eighty pepper germplasms and 5 reference accessions known as resistant L alleles, were analyzed to select the resistance cultivars against PMMoV- P1,2,3 (CV130614-2) using bioassay and genetic markers. The susceptible accessions showed systemic symptom when inoculated with PMMoV- P1,2,3. However, accessions including IT223737, were resistant as they developed necrotic local lesions only on inoculated leaves, whereas no symptoms were observed on the upper leaves. Moreover, RT-PCR results for detecting the presence of virus were also negative. Thus, those accessions will be used as a novel source to facilitate introduction the resistant gene into commercial cultivars of pepper.

시판 고추 종자에서 분리한 Tobamovirus의 병원형 (Pathotype of Tobamovirus Isolates from Commercial Red Pepper Seeds)

  • Nicotiana glutinosa를 이용한 단병반 분리를 통하여 30품종의 시판 고추종자로부터 모두 23개의 Tobamovirus 분이주(分離株)를 얻었다. 병원형(病原型) 판별(判別)고추를 사용하여 조사한 이들 분이주(分離株)의 병원형(病原型)은 P0 또는 P1,2형으로 나타났으며 P1과 P1,2,3형은 관찰되지 않았다. 시판고추 중 한 종류를 제외한 모든 품종이 P0와 P1 병원형에 대해서는 저항성을 보였고 P1,2형에는 모두 감수성이었다. Tobamovirus 병원형(病原型)에 관한 연구결과는 앞으로 우리나라 고추의 Tobamovirus 저항성육종(抵抗性育種)과 Tobamovirus 무감염(無感染) 고추종자의 생산에 널리 이용될 수 있을 것으로 생각된다.

고추냉이에서 분리한 담배 모자이크 바이러스(TMV-W)의 전체 유전자 염기서열 분석 (Complete Nucleotide Sequence of Tobacco Mosaic Virus Isolated from Wasabi(Eutrema wasabi Maxim.))

  • 고추냉이에 모자이크 병징을 나타내는 이병주로부터 고추냉이 모자이크 바이러스를 분리하였다. 고추냉이 모자이크 바이러스의 genomic RNA를 추출하여 전체 유전자 구조를 결정하였다. 유전자 전체길이는 6,298 염기를 가지고 있었으며, 4개 ORF로 구성 되어 있었다. ORF 1은 180KD 단백질, ORF 2는 130KD 단백질 , ORF 3은 30KD 단백질, ORF4는 18KD로 외피단백질로 구성되어 있었다. ORF 유전자간에는 ORF4와 ORF 3 유전자간 130개의 염기, ORF 2와 ORF 3 유전자갈 20개 염기 그리고 ORF 1 과 ORF2 유전자간에는 40개의 염기로 overlaps되어 있었다 3'NCR부분은 238개 염기, 외피단백질은 537개 염기, 30KD 이동단백질은 825개 염기, 130KD 단백질은 1,896개 염기와 180k단백질의 2,958개의 염기로 구성되어 있었다. TMV-WTF전체 염기 서열의 유전자 상동성에서는 비교 유전자에서 미보고된 일본의 TMV-WSF와 러시아의 TMV-crucifer와 각각 98.6%와 82.4%로 매우 높았다.

풋고추 수경재배에서 발생하는 tobamovirus의 특성 (Tobamoviruses of Green Peppers Growing on Hydroponic Systems)

  • 풋고추 수경재배에서 tobamovirus 병의 발병률은 생육 후기 단계에서 100%였다 바이러스 종류별 감염 빈도는 PMMOY 34%, TMGMY 41.5% 및 이들 2종의 바이러스 복합감염이 24.5%로 나타났다. 다클론 항체로 제작된 DAS-ELISA에서 각각의 항체에 대하여 특이적인 반응을 보였다. 수경재배하는 풋고추에서 77개의 tobamovirus를 순수분리하여 tobamovirus의 pathotype을 구분한 결과, $P_{0}$은 61% 및 $P_{1,2}$는 39% 빈도로 분리되었다. 모든 TMGMV 분리주는 pathotype $P_{0}$에 속하였다. PMMOY에서 구분된 pathotype $P_{0}$$P_{1,2}$의 외피단백질유전자에 상응하는 cDNA를 합성하여 제한효소에 대한 분석을 실시한 결과, $P_{1,2}$는 TaqI 위치가 두 곳에 존재하였으나 $P_{0}$은 단지 한 곳에만 존재하였다.존재하였다.

Prevalence of Tobacco mosaic virus in Iran and Evolutionary Analyses of the Coat Protein Gene

  • The incidence and distribution of Tobacco mosaic virus (TMV) and related tobamoviruses was determined using an enzyme-linked immunosorbent assay on 1,926 symptomatic horticultural crops and 107 asymptomatic weed samples collected from 78 highly infected fields in the major horticultural crop-producing areas in 17 provinces throughout Iran. The results were confirmed by host range studies and reverse transcription-polymerase chain reaction. The overall incidence of infection by these viruses in symptomatic plants was 11.3%. The coat protein (CP) gene sequences of a number of isolates were determined and disclosed to be a high identity (up to 100%) among the Iranian isolates. Phylogenetic analysis of all known TMV CP genes showed three clades on the basis of nucleotide sequences with all Iranian isolates distinctly clustered in clade II. Analysis using the complete CP amino acid sequence showed one clade with two subgroups, IA and IB, with Iranian isolates in both subgroups. The nucleotide diversity within each subgroup was very low, but higher between the two clades. No correlation was found between genetic distance and geographical origin or host species of isolation. Statistical analyses suggested a negative selection and demonstrated the occurrence of gene flow from the isolates in other clades to the Iranian population.

First Report of Tobacco mild green mosaic virus Infecting Pepper in Korea

  • A rod-shaped virus was isolated from pepper showing mild mosic during the winter growing seasons of 2001 and 2002 in Korea. Based on its biological reactions, serological relationships, reverse transcription-poly-merase chain reaction (RT-PCR) using specific primers, and nucleotide sequence analysis of coat protein (CP) gene, the isolated virus was identified as Tobacco mild green mosaic virus (TMGMV) and designated as Korean pepper isolate (TMGMV-KP). Crude sap from infected tissue was mechanically transmitted to various indicator plants, which produced characteristic symptoms of tobamovirus infection. However, no symptom was observed in Gomphorena globosa. In RT-PCR assays with specific primers toy respective detection of TMGMV, Tobacco mosaic virus (TMV), Pepper mild mottle virue (PMMoV), and Tomato mosaic virus (ToMV), a single strong band of about 500 bp in length was produced from the sample used only with TMGMV primers. The amplified DNA was cloned and the nucleotide sequence was determined. Sequence comparisons with the CP gene of other tobamoviruses indicated that TMGMV-KP shared 99.3% identity with TMGMV Japanese isolate and only 59.1, 58.6, and 58.1% identity with TMV, PMMoV and ToMV, respectively. This is the first report of TMGMV in Korea.
