추출조건(抽出條件)이 홍삼(紅蔘)엑기스의 무기성분(無機成分) 조성(組成)에 미치는 영향 (Effect of Extracting Conditions on the Mineral Content of Korean Red Ginseng Extract)

  • 홍미삼엑기스를 제조할 때 추출용매와 그 농도 및 추출온도와 추출시간 등의 추출조건이 홍삼엑기스의 조회분 함량과 무기성분의 조성에 미치는 영향을 조사한 결과 무기성분의 용출량과 이행율은 추출용매에탄올의 농도가 증가 될수록 낮아 졌으며 특히 70% 이상 농도에서 현저하였다. 물 추출구(區)의 경우 용출량은 K의 1.55%로 가장 높았고 Cu가 11ppm으로 가장 낮았으며 원료함량대비로는 Ca의 91.41% 용출이행으로 가장 높았고 Mg이 30.14%로 가장 낮았다. 추출온도 상승에 따라서는 큰 차이가 없었으나 소량씩 증가되는 경향이었고 초기용출이행이 커 $1{\sim}3$회 추출에 80%이상이 용출되었다. 조회분의 경우도 같은 경향이었고 몰 추출구(區)의 경우 3회 추출에 92.38%이상의 용출되어 70% 에탄올추출구(區)에 비하여 훨씬 높았다.

${\beta}-Carotene$의 열분해(熱分解)에 의한 휘발성(揮發性) 화합물(化合物)의 생성(生成) (Formation of Volatile Compounds by the Thermal Degradation of ${\beta}-Carotene$)

  • 잎담배의 중요(重要)한 색소성분(色素成分)인 ${\beta}-carotene$$400^{\circ}C,\;600^{\circ}C$$800^{\circ}C$에서 각각 열분해(熱分解)한 다음 생성(生成)된 휘발성(揮發性) 성분(成分)들을 GC 및 GC/MS에 의해 확인하고 분해온도(分解溫度)에 따른 생성물(生成物)의 차이점을 비교하였다. 각 온도별(溫度別) 분해생성물(分解生成物)로부터 36종(種)의 방향족(芳香族) 탄화수소(炭化水素) 10종(種)의 ${\beta}-ionone$ 관련화합물을 포함하여 53종(種)의 휘발성(揮發性) 화합물(化合物)을 확인하였고 이들 중에서 ${\beta}-cyclogeraniol$을 포함한 37종(種)의 화합물(化合物)은 ${\beta}-carotene$의 열분해생성물(熱分解生成物)로서 아직까지 보고되지 않은 성분(成分)들이다. 한편 $400^{\circ}C$$600^{\circ}C$에서 열분해(熱分解)했을 때의 중요(重要) 생성물(生成物)은 $p-xylene,\;{\alpha}-terpinene,\;{\beta}-cyclocitral,\;ionene(1,2,3,4-tetrahydro-1,1,6-trimethylnaphthalene),\;{\beta}-ionone$ 및 dihydroactinidiolide 등이었고, $800^{\circ}C$에서는 위에서의 6종(種)의 성분(成分) 이외에도 toluene이 많이 생성(生成)되었다.

담배 흡연에서 가열담배 사용으로의 단기간 전환에 따른 심장 자율신경 반응 (Short-term Effects of Switching from Cigarette Smoking to Using Heated Tobacco Products on Cardiac Autonomic Regulation)

  • 가열담배(heated tobacco products, HTPs) 에어로졸 내 유해 성분들의 수준은 담배 연기와 비교해 현저히 낮은 것으로 보고되었지만, HTPs 사용이 담배 흡연과 연관된 심혈관 위험을 완화할 수 있는지는 불분명하다. 현재 연구는 습관성 흡연자들을 대상으로 담배 흡연에서 HTPs 사용으로의 단기간 전환이 심장 자율신경조절(cardiac autonomic regulation, CAR)에 미치는 영향을 탐색하기 위한 목적으로 수행되었다. 건강한 20대 남성 흡연자 7명은 5일 동안 담배 흡연(cigaette smoking, CS), 3가지 서로 다른 HTPs 사용(IQOS use, IQ; lil SOLID use, LS; lil HYBID use, LH) 혹은 흡연 중단(non-smoking, NS)으로 구성된 개방표지, 무작위 교차설계 시험을 완료했다. 각 세션은 1주의 세척 기간으로 분리되었으며, 세션당 할당된 제품 사용 전과 사용 후 24, 48, 72, 96 및 120시간에서 생체 이물[CO (carbon monoxide) and COHb (carboxyhemoglobin)], 혈류역학적 변인(systolic and diastolic blood pressure) 및 CAR 조절을 반영하는 HRV (heart rate variability) 지표들의 수준이 평가되었다. 호기 CO 및 COHb 수준은 NS에서만 통계적으로 유의하게 감소했다. 모든 세션 내 혈류역학적 변인에서 통계적 변화는 없었다. 그러나r HRV 스펙트럼 분석에서 lnHF (log-transformed high frequency, lnHF)는 NS를 포함한 IQ와 LS에서 유의하게 증가하였으며, HFnu (normalized HF)는 NS와 LH에서 통계적으로 증가하였다. lnHF와 HFnu는 각각 세션 간 시간에 따른 유의한 상호작용효과를 나타냈다. 현재 연구는 습관성 흡연자들에게서 HTPs 사용으로의 단기간 전환이 담배 흡연에 의한 심장 미주신경 긴장도 저하를 개선하여 CAR에 영향을 미칠 수 있음을 시사한다.

Comparative Investigation of Flavors in Cigarettes by Electronic Nose and GC/MS

  • An Electronic Nose(E-Nose) and Gas Chromatography/Mass Spectroscopy (GC/MS) are meanwhile conventional technique to analyze volatile materials in many industries (e.g., food, medicine, environment) and have broad acceptance in the analysis of tobacco products. In this study, an experiment where tin oxide gas sensor array responses and GC/MS profiles are used to characterize the volatile compounds of different cigarettes at the same time is performed and the measurements of two instruments are compared for cigarette samples with a known chemical information. E-Nose and GC/MS were employed to differentiate and match flavored cigarettes with commercial tobacco flavoring agents (lavender, vanilla, peppermint, orange, star anise). For verifying reliability of two systems, the analyses were conducted in terms of amount of flavors in each cigarettes using partial least squares (PLS) and with the principal components analysis (PCA). Various chemical sensors and GC/MS data was reduced into two principal factors (PC1, PC2) for being distinguished with visualized regions. Both systems provided adequate results for odor characteristics of cigarettes in this study with each instrument having its own advantages and disadvantages.

IDENTIFICATION AND DETERMINATION OF GINSENG SAPONINS, PROSAPOGENINS AND SAPOGENINS FROM CRUDE DRUG PREPARATIONS FOR QUALITY CONTROL

  • Ginseng saponins have been known as main active principles and analyzed as the index components in ginseng and its products for quality control. But it is generally difficult to analyze the saponins in crude drug preparations. Saponins, Prosapogenins and sapogenins of crude drug preparation were identified by TLC and determined quantitatively by HPLC. $Prosapogemins-Rg_3\;-Rg_2\;and\;{\Delta}^{20}-prosapogenin$ were extracted with ethyl acetate from $50\%$ acetic acid hydrolyzates of saponin fractions and identified by TLC with lower phase of $CHCl_3/MeOH/H_2$ O\65:35:10. v/v)on silica gel plate, and quantified by HPLC on $Lichrosorb-NH_2$ column with $CH_3CN/H_2O(90:10,\;v/v).$ Sapogenins. panaxadiol and panaxatriol. were extracted with ethyl ether from $7\%-sulfuric$ acid hydrolyzates of saponin fractions and identified by TLC with chloroform/acetone(1 : 1 v/v) on silica gel plate. and quantified by HPLC on u - Bondapak $C^{18}$ column with $CH_3CN/MeOH/CHCl_3(83:10:7.\;v/v).$ These analyses of prosapogenins and sapogenins are more useful for quality control than those of saponins in crude drug preparations such as So - Shi - Ho - Tang(소시호탕), Sa - Kun - Ja - Tang(사군자탕), Yook - Kun - Ja - Tang(육군자탕), and In - Sam -Tang(인삼탕)

Associations between the Frequency and Quantity of Heated Tobacco Product Use and Smoking Characteristics among Korean Smoking Adolescents

  • Purpose: Although heated tobacco product (HTP) use among adolescents is an emerging public health problem, little is known about the frequency and quantity of HTP use. Thus, we investigated the associations between the frequency and quantity of HTP use and smoking characteristics (i.e., combustible cigarette [CC] and electronic cigarette [EC] use, and attempts to quit smoking) among CC-smoking adolescents. Methods: We analyzed nationally representative data from 2,470 Korean adolescents who were current CC smokers. To investigate our aim, we conducted multinomial logistic and logistic regression analyses. Results: We found that daily and heavier CC users had greater likelihoods of more frequent and heavier HTP use. In addition, dual users of CCs and ECs were more likely to use HTPs more frequently and heavily than CC users who did not use ECs. Moreover, daily EC users had the highest risk of frequent and heavy HTP use. The frequency and quantity of HTP use were not associated with attempts to quit smoking. Compared to CC-only use, dual use of CCs and HTPs was not associated with quitting attempts, and triple use of CCs, ECs, and HTPs was associated with a lower likelihood of quitting attempts. Conclusion: HTP use was less likely to displace CC use and promote attempts to quit smoking. Thus, strict regulations are required to prevent the promotion of HTPs as a substitute for CCs or as a means of quitting smoking. Additionally, health professionals should consider preventive interventions for HTP, as well as CC and EC use among adolescents.

Non-enzymatic Antioxidant Status and Biochemical Parameters in the Consumers of Pan Masala Containing Tobacco

  • Background: Tobacco consumption is one of the leading causes of oral submucous fibrosis, oral cancer and even premature death. The present study was designed to compare the biochemical parameters and non-enzymatic antioxidant status and the lipid peroxidation products in pan masala tobacco users as compared with age-matched non-user controls. Methods: Pan masala and tobacco users of age $33.2{\pm}9.94$ years and age-matched controls ($31.2{\pm}4.73$ years) were enrolled for the study. Plasma levels of vitamin E, vitamin C, albumin, bilirubin, uric acid, glucose, urea, creatinine, aspartate amino transferase (AST), alanine amino transferase (ALT) were measured by standard methods. Serum malondialdehyde (MDA) levels were estimated as a measure of lipid peroxidation. Results: In the pan masala tobacco users, as compared to the controls, the level of vitamin C ($68.5{\pm}5.9$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.05$) vitamin E ($18.4{\pm}5.3$ vs $97.9{\pm}9.03{\mu}mol/L$, $p{\leq}0.001$), albumin ($37.5{\pm}7.01$ vs $44.3{\pm}9.99g/L$, $p{\leq}0.001$), and malondialdehyde ($10.8{\pm}1.29$ vs $1.72{\pm}1.15nmol/ml$, $p{\leq}0.001$) were found to be significantly altered. Malondialdehyde was significantly correlated with vitamin E (r=1.00, p<0.001) and vitamin C (r=1.00, p<0.001) in pan masala tobacco users. Serum levels of AST ($31.0{\pm}16.77$ IU) and ALT ($36.7{\pm}31.3$ IU) in the pan masala tobacco users were significantly raised as compared to the controls (AST, $25.2{\pm}9.51$ IU, p=0.038; ALT, $26.2{\pm}17.9$ IU, p=0.038). Conclusion: These findings suggest that pan masala tobacco users are in a state of oxidative stress promoting cellular damage. Non-enzymatic antioxidants are depleted in pan masala tobacco users with subsequent alteration in the biochemical parameters. Supplementation of antioxidants may prevent oxidative damage in pan masala tobacco users.

고려인삼 유래 Cytochrome P450 유전자의 동정 및 형질전환에 의한 특성검정 (Identification of Korean Ginseng Cytochrome P450 gene and Its Characterization by Transformation System)

  • 인삼으로부터 뽑은 PgCYP 유전자와 표지유전자인 NPTII 유전자를 함유하고 있는 Agrobacterium tumefaciens GV3101균주를 이용하여 담배 잎절편과 공동배양한 후 MS 기본배지에 kanamycin 100 $\mu$g/ml, cefotaxime 500 $\mu$g/l, BA 2 mg/l와 NAA 0.2 mg/l가 첨가된 선발배지에 치상하여 4주 후 항생제가 첨가된 기본배지에서 발근시켰다. 생존한 선발체의 잎을 이용하여 PCR 반응으로 도입유전자의 삽입여부를 확인하였다. 또한 선발된 형질전환체를 이용하여 RT-PCR을 실시하여 PgCYP 유전자가 담배식물에 안정적으로 도입되어 전사되고 있음을 확인하였다.

Systematic Review on International Practices in Controlling Waterpipe Tobacco Smoking

  • Background: Waterpipe tobacco smoking has becoming popular especially among young people worldwide. Smokers are attracted by its sweeter, smoother smoke, social ambience and the misconception of reduced harm. The objective of this study was to systematically review the effects of waterpipe tobacco policies and practices in reducing its prevalence. Materials and Methods: A systematic review was conducted electronically using the PubMed, OVID, Science Direct, Proquest and Embase databases. All possible studies from 1980 to 2013 were initially screened based on titles and abstracts. The selected articles were subjected to data extraction and quality rating. Results: Three studies met the inclusion criteria and were eligible for this review. Almost all of the waterpipe tobacco products and its accessories did not comply with the regulations on health warning labelling practices as stipulated under Article 11 of WHO FCTC. In addition, the grisly new warning labels for cigarettes introduced by Food and Drug Administration did not affect hookah tobacco smoking generally. Indoor air quality in smoking lounges was found to be poor and some hookah lounges were operated without smoke shop certification. Conclusions: Our findings revealed the availability of minimal information on the practices in controlling waterpipe smoking in reducing its prevalence. The lack of comprehensive legislations or practices in controlling waterpipe smoking warrants further research and policy initiatives to curb this burgeoning global epidemic, especially among the vulnerable younger population.

궐련지의 섬유종류가 담배 연기성분에 미치는 영향 (The comparative analysis of smoke components delivered from cigarette papers manufactured by flax and wood pulp)

  • This study was conducted to evaluate the effect of cigarette papers, flax and wood, on the delivery of mainstream smoke. The main components of cigarette papers were cellulose, hemicellulose, lignin, and pectin. Lignin contents, known as precursor of smoke's phenolic compounds, of the flax and wood cigarette papers were 5.8% and 10.6%, respectively. The pyrolysis products of cigarette papers were similar by the profile of total ion chromatogram. But, the area % of some components, such as 1,3-cyclopentanedione, 3,5- dimethyl cyclopentane-1,2-dione, 2-hydroxy-3-methyl-2-cyclopentenone, dihydro-2(3H)-furanone, 3-methyl-2(5H)-fruanone, and 5-methyl-2-furaldehyde delivered through pyrolysis of the flax cigarette paper were higher than that of wood cigarette paper. Otherwise, the area % of some components, such as 2-methyl-cyclopentene-l-one), 2,3-butanedione, 2-cyclopentene-l-one, and 5-hydroxy-2-methyl-furaldehyde, 2-furaldehyde delivered through pyrolvsis of the wood cigarette paper were higher than that of flax cigarette paper. To identify the difference between two cigarette papers, we used the cigarette column filled with the cut cigarette paper instead of the cut tobacco leaf. The amounts of semi-volatile fraction delivered from flax cigarette paper was more than that of wood cigarette paper. But, by using the cut tobacco, there was no big difference of delivery amount between flax and wood cigarette papers. Also, aroma of TPM by collecting from brening cut tobacco wrapped in flax and wood papers showed a different pattern by the electonic nose system. Although the difference between two cigarette papers by using the cut tobacco was smaller than that of cut cigarette paper, this result indicated that the fax and wood had the different effects on the delivery of smoke components as shown in the sensory test results.
