New Degenerate Primer for the Cyanobacterial Non-ribosomal Peptides (시아노박테리아 Non-ribosomal Peptides의 효과적인 연구를 위한 New Degenerate Primer의 개발)

  • Cyanobacteria have been identified as one of the most promising group producing novel biochemically active natural products. Cyanobacteria are a very old group of prokaryotic organisms that produce very diverse secondary metabolites, especially non-ribosomal peptide and polyketide structures. Large multienzyme complexes which are responsible for the non-ribosomal biosynthesis of peptides are modular for the addition of a single amino acid. An activation of amino acid substrates results in an amino adenylate occuring via an adenylation domain (A-domain). A-domains are responsible for the recognition of amino acids as substrates within NP synthesis. The A-domain contains ten conserved motifs, A1 to A10. In this study, ten conserved motifs from A1 to A10 were checked regarding their amino acid sequence of the NRPS-module of Microcystis aeruginosa PCC 7806. The part of the amino acid sequence chosen was that which contained as many conserved motives as possible, and then these amino sequence were compared between other cyanobacteria to design a new degenerate primer. A new degenerate primer (A3/A7 primer) was designed to detect any putative NP synthetase region in unkwon cyanobacteria by a reverse translation of the conserved amino acid sequence and a search for cyanobacterial DNA bank.

Tylosin Production by Mutant Resistant to Oleic Acid (Oleic Acid 내성균주로부터 Tylosin 생산)

  • When rapeseed oil as the carbon source was used for tylosin production from Streptomyces fradiae TP-1239 was very sensitive to oleic acid. Cell growth was restrained by adding 0.8 g/l of oleic acid to the culture broth. Mutant strain TM-224-1 resistant to 1.2 g/l of oleic acid was obtained by screening in solid and liquid media containing oleic acid. The uptake rate of oleic acid by TM-224-1 was approximately 3.8 fold higher than the parent strain. For comparing the TM-224-1 and the parent strain, batch cultures were carried out in a jar fermentor. Cell growth of TM-224-1 strain was higher than the parent strain after two days of culturing. However, after four days of culturing, it was similar to that of the parent strain. The amount of rapeseed oil consumed by TM-224-1 and the parent strain were 60.5 and 78.2 g/l, respectively. The production and yield of tylosin was aproximately 2.0 and 3.2 fold higher than the parent strain, respectively. From these results, it was concluded that this mutant, which was resistant to oleic acid, has improved tylosin production.

  • In this study, we fabricated a 1.55um polarization-insensitive semiconductor optical amplifier(SOA) with rectangular buried heterostructure using a InGaAsP/InP double heterostructure wafer. Measured characteristics of the fabricated SOA are that 3dR bandwidth is 35nm and 3dB saturation output power is 4dBm. Maximum gain under the 150mA CW driving condition is 19.4dB. We measured the ASE(amplified spontanouse emission) Power spectrum or n and TM mode in the fabricated SOA using ASE measurement system and knew that distributions of the TE and TM mode about the maxinum region are nearly coincident. this shows the fabricated SOA is a polarization-insensitive.

Asymmetric Reduction of 3-Ketoproline Ethyl Ester by Modified Borohydrides and Various Vegetables

  • Reduction of (${\pm}$)-3-ketoproline ethyl ester (1) by $NaBH_4$ in the presence of $CaCl_2$ and $MgCl_2$ as the chelating agents gave selective products cis-3(R/S)-alcohols, while reduction by $NaBH_4$ alone or chelated with $NiCl_2$ and $AlBr_3$ gave mixtures of cis- and trans-alcohols. The reduction of (${\pm}$)-1 by various vegetables however, gave exclusively the cis-alcohol as the major and trans-alcohol as the minor. On the contrary, reduction of (${\pm}$)-1 by carrot afforded a mixture of cis- and trans-alcohols, in which the trans-alcohol exists as the major product. In addition, we found that this biocatalyst selectively converted S-enantiomer of (${\pm}$)-1 to the cis-alcohol, and R-enantiomer to a mixture of cis- and trans-alcohols with cis-alcohol as the major product. This fact prompted us to use various fresh plant tissues for stereoselective reduction of diverse types of pyrrolidinones, as its stereoselectivity towards racemic mixtures is higher compared to that using chemical reducing agents.

A Study on Ti:LiNbO3 Integrated Optical Wavelength Tunable Polarization Mode Controllers (Ti:LiNbO3 집적광학형 파장가변 편광모드 조절기에 관한 연구)

  • We designed and fabricated integrated-optic tunable polarization controllers based on $LiNbO_3$ with the Ti-indiffused waveguide along the y-axis utilizing the electro-optic effect. The device consists of $TE↔TM$ mode converters and TE/TM phase shifters. We analyzed the operation principles of each device utilizing transfer matrices based on a Jones matrix and simulated shifting of the center wavelength by inducing voltage. We confirmed experimentally that the fabricated devices control the tunability of the center wavelength and the input SOP.

  • 최근 PC 시장은 USB라는 새로운 인터페이스의 보급으로 이용한 장치들이 급속도로 증가하고 있다. 또한 USB를 이용하여 저장 장치를 접근할 수 USB Mass Storage는 디지털 카메라나 MP3 Player 등에서 빠른 전송속도와 편리함 때문에 각광 받고 있다. 본 논문에서는 실시간 운영 체제인 iRTOS$^{TM}$에서 USB 인터페이스를 이용한 USB Mass Storage가 가능하도록 설계하고 이를 구현한 내용을 기술한다.

  • In bitrate control of MPEG-2 TM5, $Q_{j}$, N-act, and mquant, of present macroblock are predicted from those of previous macroblock. It results in poor image quality because of abrups change of them. We proposed a method predicting $Q_{j}$, N-act, and mquant, of present macroblock by averaging those parameters of adjacent previous macroblocks. As a results, it shows improved PSNR compared to bitrate control of MPEG-2 TM5.TM5.

Clinical and Radiologic Assessment for Anterior Cervical Interbody Fusion with Synthetic Cages

  • Objective : This purpose of this study was to determine the clinical efficiency and applicability, and to analyze the radiologic findings of the anterior cervical approach using two synthetic cages for interbody fusion. Methods : A total of 41 patients with cervical diseases underwent anterior discectomy and interbody fusion with the PEEK $Solis^{TM}$ cage in 21 patients and the carbon composite $Osta-Pek^{TM}$ cage in 20 patients. Outcome assessment was done using Odom's criteria. Radiological assessment was performed with respect to subsidence, bony fusion and lordosis. The mean follow-up period was 13 months. Results : There were 34 [92.9%] successful cases. The average height of the disc space 12 months after surgery compared the height just after surgery was decreased over 3mm in 4 cases, indicating severe subsidence. The use of these synthetic cages have provided the increase in postoperative cervical lordosis. Conclusion : There were no significant differences between the $Solis^{TM}$ and $Osta-Pek^{TM}$ cages on clinical and radiologic outcomes. Both $Solis^{TM}$ and $Osta-Pek^{TM}$ cages showed low subsidences and complications associated with hardware with good clinical outcomes, high fusion rates, restored disc heights, and restored cervical lordosis.

  • 현재 건설교통부 한강홍수통제소에서는 96개의(2006 한국수문조사연보 기준) 수위관측소를 설치 운영하고 있으며 현장에서 수집되는 수위자료의 전송방법은 크게 두 가지로 중계소를 통해 실시간으로 전송되는 TM(TeleMetering) 방식과 기록지(Recording) 방식으로 구분된다. 고품질 수위자료의 생산 및 제공은 비단 수자원 관련 연구 분야에서 뿐만 아니라 하천의 효율적인 관리, 각종 국토개발계획 등에 다양하게 이용되고 있으며 특히 TM 자료의 경우는 실시간으로 현장에서 관측되는 수문자료를 수집하여 홍수예보시스템의 가장 중요한 입력자료로서 활용된다. 한강홍수통제소에서 구축 운영하고 있는 TM 수위자료와 일반 수위자료를 대상으로 현황을 검토한 결과 일반적으로 수위관측소의 자료 오류 유형을 관측기기부터 전송단계별로 분류하면 수위계 기기 고장(부자 걸림 등), 전송로 변경 및 통신 장비 고장 등으로 인한 오 결측으로 구분될 수 있다. 과거 오 결측된 자료를 보정하기 위한 방법으로는 2시간, 3시간 전(前)수위 자료를 이용해 이상치를 보정하는 것이 유일하게 활용되고 있었으나 작년에 한강수계를 대상으로 "국가수문자료 품질관리시스템구축(1차)" 연구 용역을 실시하여 시범 구축 결과를 금년부터 활용하고 있으며 본 시스템에 자료보정에 대한 다양한 방법이 탑재되어 있다. 이와 별도로 기왕자료의 보정방법으로 TM과 일반방식이 이중화 되어 있는 관측소의 경우에는 연속적인 자료를 나타내는 기록지 자료를 활용하는 것이 대안으로 제시되고 있다. 하지만, 기록지 자료를 통해 오 결측된 TM 자료를 보완하는 것에 대해서는 아직 연구된 바가 없으며 이와 관련된 다각적인 검토가 국내에서 부족한 실정이므로 본 연구에서는 실제 한강홍수통제소에서 관할하고 있는 이중화 기록방식의 관측소를 선정하여 TM과 기록지 수위관측자료의 비교 분석을 통해 오 결측된 TM 자료를 일반 기록지 자료로 보완에 하는 것에 대한 실효성을 심도 있게 검토하여 수위자료 품질향상의 기반을 마련코자 한다.
