Adipose Tissue-Derived Mesenchymal Stromal Cells from Ex-Morbidly Obese Individuals Instruct Macrophages towards a M2-Like Profile In Vitro

  • Obesity, which continues to increase worldwide, was shown to irreversibly impair the differentiation potential and angiogenic properties of adipose tissue mesenchymal stromal cells (ADSCs). Because these cells are intended for regenerative medicine, especially for the treatment of inflammatory conditions, and the effects of obesity on the immunomodulatory properties of ADSCs are not yet clear, here we investigated how ADSCs isolated from former obese subjects (Ex-Ob) would influence macrophage differentiation and polarization, since these cells are the main instructors of inflammatory responses. Analysis of the subcutaneous adipose tissue (SAT) of overweight (OW) and Ex-Ob subjects showed the maintenance of approximately twice as many macrophages in Ex-Ob SAT, contained within the CD68+/FXIII-A- inflammatory pool. Despite it, in vitro, coculture experiments revealed that Ex-Ob ADSCs instructed monocyte differentiation into a M2-like profile, and under inflammatory conditions induced by LPS treatment, inhibited HLA-DR upregulation by resting M0 macrophages, originated a similar percentage of TNF-α+ cells, and inhibited IL-10 secretion, similar to OW-ADSCs and BMSCs, which were used for comparison, as these are the main alternative cell types available for therapeutic purposes. Our results showed that Ex-Ob ADSCs mirrored OW-ADSCs in macrophage education, favoring the M2 immunophenotype and a mixed (M1/M2) secretory response. These results have translational potential, since they provide evidence that ADSCs from both Ex-Ob and OW subjects can be used in regenerative medicine in eligible therapies. Further in vivo studies will be fundamental to validate these observations.

  • Recent findings, notably on adipokines and adipose tissue inflammation, have revised the concept of adipose tissues being a mere storage depot for body energy. Instead, adipose tissues are emerging as endocrine and immunologically active organs with multiple effects on the regulation of systemic energy homeostasis. Notably, compared with other metabolic organs such as liver and muscle, various inflammatory responses are dynamically regulated in adipose tissues and most of the immune cells in adipose tissues are involved in obesity-mediated metabolic complications, including insulin resistance. Here, we summarize recent findings on the key roles of innate (neutrophils, macrophages, mast cells, eosinophils) and adaptive (regulatory T cells, type 1 helper T cells, CD8 T cells, B cells) immune cells in adipose tissue inflammation and metabolic dysregulation in obesity. In particular, the roles of natural killer T cells, one type of innate lymphocyte, in adipose tissue inflammation will be discussed. Finally, a new role of adipocytes as antigen presenting cells to modulate T cell activity and subsequent adipose tissue inflammation will be proposed.

옥병풍산(玉屛風散)이 생쥐의 선천성 및 특이적 면역반응에 미치는 영향 (Effects of Okbyungpoongsan Administration on Innate and Specific Immune Response in the Mouse)

  • Okbyungpoongsan(OBPS) has long been known to have anti-allergic effect. In order to evaluate the influence on innate and specific immune response, the effects of OBPS on vascular permeability. hypersensitivities and phagocytic functions were measured. As the results, OBPS increased phagocytic activity of peritoneal macrophages in vitro and in vivo. But OBPS depressed formation of reactive oxygen intermediates(ROI) in vitro and in vivo, while the drug enhanced generation macrophages. Foot pad swelling in the mouse and contact hypersensitivity against dinitroflouorobenzene were decreased. OBPS had no effect on NK cells. But OBPS decreased vascular permeability induced by histamine without statistical significance. These results demonstrate that OBPS suppresses hypersensitivity reactions without affecting phagocytic functions and formation of ROI from macrophages. It also means that OBPS acts as a effective inducer to synthesis of nitric oxide which is effective for the infectious disease while it does damage to tissue less as it suppresses ROI, So we can conclude that OBPS could be used for the treatment of the disease related with immune function.

가토 창상치유에 미치는 Yeast Glucan의 효과 (Effect of Yeast Glucan on Wound Healing in Rabbits)

  • To investigate the effect of yeast glucan on wound healing in rabbits, histopathological findings and wound contraction after topical application of glucan on the open wound were observed. Formalin fixation and embedding of tissues at wound were prepared for histopathological findings at the other day after wounding. Wound size was measured for contraction. Macrophages appeared in the deeper site under the scab at day 1 after wounding in the glucan treated group, but not in the control. A number of macrophages, fibroblasts and a small number of neutrophils were observed at day 3 after wounding In the glucan treated group as compared with the control. Histopathological findings at day 5 after wounding in control were similar with those at day 3 in the glucan treated group. Glucan like granules were scattered in the cytoplasm of macrophages at day 5 in the glucan treated group. A great portion of wound reepithelialized at day 5 in the glucan treated group but a small portion of wound in the control. The Present study indicated that the treatment of 5,000$\mu\textrm{g}$/$m\ell$ of glucan has an effect on wound heal. ins in respect to migration of macrophage into the wound site, granulation tissue formation and epithelialization.

Development and Functions of Alveolar Macrophages

  • Macrophages residing in various tissue types are unique in terms of their anatomical locations, ontogenies, developmental pathways, gene expression patterns, and immunological functions. Alveolar macrophages (AMs) reside in the alveolar lumen of the lungs and serve as the first line of defense for the respiratory tract. The immunological functions of AMs are implicated in the pathogenesis of various pulmonary diseases such as allergic asthma, chronic obstructive pulmonary disorder (COPD), pulmonary alveolar proteinosis (PAP), viral infection, and bacterial infection. Thus, the molecular mechanisms driving the development and function of AMs have been extensively investigated. In this review article, we discuss the roles of granulocyte-macrophage colony-stimulating factor (GM-CSF) and transforming growth factor (TGF)-β in AM development, and provide an overview of the anti-inflammatory and pro-inflammatory functions of AMs in various contexts. Notably, we examine the relationships between the metabolic status of AMs and their development processes and functions. We hope that this review will provide new information and insight into AM development and function.

  • 폐흡충(Paragonimus Tuestermani) 피낭유충을 흰쥐(Wistar) 및 고양이에 감염시키고 감염 힐청이나 분회분리된 IgG또는 보체가 정상 또는 감염 흰쥐 복강 대식세포의 폐흡충 유충 살충에 어떠한 영향을 미치는지 부착 실험 (adherence assay) 및 세포독성을 통하여 관찰하였다. 폐흡충 감염은 복강 대식세포를 비특이적으로 활성화시퍼 대식세포의 유충에 대한 부착률 및 세포독성을 증가시켰으며, 감염 혈청을 첨가하였을 때 항체-의존 세포매개성 세포독성에 의해 배양 6시간 후에 세포 부착률 및 세포독성이 가장 강하였다. 감염 혈청을 56℃에서 30분간 가열하였을 때 IgG 항체 변성에 의해 세포독성이 저하되었다. IgG 및 보체를 첨가한 경우 세포 부착률은 낮았으나 24시간 후에는 유충이 사멸하였다. 그러나 보체의 단독적인 역할은 이 실험에서 알 수 없었다.

  • 인체 중간엽 줄기세포는 지방세포, 골세포, 연골세포, 근육세포 등 여러 형태의 세포로의 분화되는 것이 특징이다. 특히, 최근 연구 결과를 살펴 보면, 중간엽 줄기세포는 생체 내에서 조직 특이적인 세포 형태로 분화 된다. 본 연구에서는 염증 상태에 존재하는 중간엽 줄기 세포가 혈관 형성에 관여하는지 알아보고, 염증 상태에 존재하는 줄기세포의 역할을 규명하고자 본 연구를 진행하였다. 생체 내 염증 상태와 유사한 환경을 만들고자, 강력한 염증 유발 물질인 LPS를 대식세포에 처리하여 그 배양액을 중간엽 줄기세포에 처리하여 변화를 관찰하였다. LPS 배양액을 처리한 중간엽 줄기세포는 평활근 세포로 분화가 촉진되는 것을 확인하였으며, LPS 배양액에 존재하는 분화 유도 물질이 $PGF2{\alpha}$임을 확인하였다. 이에 본 연구결과를 통해 염증 상태에서 존재하는 중간엽 줄기세포는 평활근 세포로의 분화가 촉진되는 것을 확인하였다. 본 연구는 염증성 미세환경 내에 존재하는 중간엽 줄기세포가 평활근 세포로 분화가 유도됨을 확인하였고, 혈관 형성을 촉진하는데 영향을 미칠 수 있음을 제시한다.

Pulsed Nd : YAG 레이저를 조사한 임플란트를 가토복강내에 이식시 주위조직에 미치는 영향 (SOFT TISSUE RESPONSE TO THE PULSED Nd : YAG LASED DENTAL IMPLANTS IN THE RABBIT ABDOMINAL WALL)

  • Pulsed Nd : YAG laser has been used to treat various soft tissue lesions. However, there have been warnings of using laser to treat peri-implantitis because the laser can alter the surface of implant and can rise its temperature so that it might be harmful to implant itself and its surrounding tissue. However, the study in vivo has not been done yet. The purpose of this study was to evaluate the biocompatibility to the pulsed Nd:YAG lased dental implant. 16 HA coated implants and 16 titanium plasma sprayed implants were used. They were divided 4 groups as unlased control, 0.3 watt, 1.0 watt and 2.0 watt lased groups after lasing. And they were implanted in the peritonial walls of 12 rabbits. The rabbits were sacrificed 6 weeks after implantation and the LM and EM specimems were made. 1. In the unlased implant group, both HA coated and titanium plasma sprayed implants were surrounded by fibroblasts and collagen fibers. No macrophages and inflammatory cells were seen. 2. In HA coated implants 0.3 watt, 1.0 watt lased group had a little inflammation, but in 2.0 watt lased group had scattered macrophages, a significantly larger number of chronic imflammatory cells were seen. 3. In titanium plasma sprayed implant 0.3 watt, 1.0 watt lased group had a little inflammation, but in 2.0 watt group had severe inflammation as in the conditions of HA coated group. 4. The inflammatory reaction of both lased HA coated and titanium plasma sprayed implant groups increased as the increase of the power of the laser.

  • Objective: Endometriosis is a chronic debilitating inflammatory condition characterized by the presence of endometrial tissues outside the uterine cavity. Pelvic soreness and infertility are the usual association. Due to the poor effectiveness of the hormone therapy and the high incidence of recurrence following surgical excision, there is no single effective option for management of endometriosis. Mesenchymal stem cells (MSCs) are multipotent stromal cells studied for their broad immunoregulatory and anti-inflammatory properties; however, their efficiency in endometriosis cases is still a controversial issue. Our study aim was to evaluate whether adipose tissue-derived MSCs (AD-MSCs) could help with endometriosis through their studied anti-inflammatory role. Methods: Female Wistar rats weighting 180 to 250 g were randomly divided into two groups: group 1, endometriosis group; established by transplanting autologous uterine tissue into rats' peritoneal cavities and group 2, stem cell treated group; treated with AD-MSCs on the 5th day after induction of endometriosis. The proliferative activity of the endometriosis lesions was evaluated through Ki67 staining. Quantitative estimation of interferon γ, tumor necrosis factor-α, interleukin (IL)-6, IL-1β, IL-10, and transforming growth factor β expression, as well as immunohistochemical detection of CD68 positive macrophages, were used to assess the inflammatory status. Results: The size and proliferative activity of endometriosis lesions were significantly reduced in the stem cell treated group. Stem cells efficiently mitigated endometriosis associated chronic inflammatory reactions estimated through reduction of CD68 positive macrophages and the expression of the proinflammatory cytokines. Conclusion: Stem cell therapy can be considered a novel remedy in endometriosis possibly through its anti-inflammatory and antiproliferative properties.

개에서 거세 후 발생한 봉합사 육아종의 병리조직학적인 연구 (A Histopathological Study on Suture Granuloma in Neutralized Dogs)

  • Postoperative suture granuloma have rarely been reported in animals. Eight biopsy masses from testes areas of neutralized dogs were diagnosed as suture granuloma. The suture granuloma occurred at any time from several weeks to a few years after surgery and appeared to be testicular tumors by macroscopic examination. The granulomas were classified into three types based on the histopathological findings. The first type of pyogranuloma was mainly composed of neutrophils, macrophages, and suture fragments. The second type was chronic necrotizing granuloma which was well demarcated by fibrous connective tissues and was composed of a few suture fragments, macrophages and central fibrinoid necrosis. The third type of granuloma had a poorly defined margin with scarcely observed suture fragments and central necrosis in the tissue. These histopathological findings suggested that various types of suture granuloma may be caused by suture material and could even appear long after surgery.
