방사선 치료에서 광학적 생검의 유용성에 관한 연구 (The Study of Optical Biopsy‘s Usefulness in Radiotherapy)

  • 본 논문의 목적은 조직(Tissue)에서의 광학적 특성(Optical Properties)을 이용한 광학적 생검(Optical Biopsy)방법을 소개하고, 방사선 치료에서 치료 반응 결과를 확인하는데 적당한 도구인가를 확인하고자 한다 본 연구는 구강(Oral Cavity) 내부조직을 샘플로, 건강한 사람 4명과 구강 암환자 4명의 자원을 받았다. 연구실에서 제작한 FastEEM(Excitation Emission Matrix) 장비를 이용하여 생체 내(in vivo)상태에서 측정하였다. 건강한 구강의 정상조직(Normal Tissue)과 병이 있는 구강의 비정상조직에서 기존의 생검과 동시에 새로운 광학적 생검을 하였다. 광학적 생검 결과와 기존의 생검 결과를 비교 확인하고, 암 조직으로 진단 받은 환자들에게 2차 광학적 생검을 실시하였다. 암 조직에 대한 1차 광학적 생검과 2차 측정 결과에 대한 형광스펙트럼을 비교 분석하였고, 자료분석은 Gillenwater가 개발한 337nm에 근거한 진단 알고리즘을 이용하였다. 광학적 생검 방법은 암 조직을 정상조직과 확실하게 구분시키는 장비임을 확인하였다. 건강한 구강조직과 악성 종양 조직의 측정 형광세기를 비교하면 정상조직인 경우 암 조직의 형광세기보다 모든 환자에 대해서 크게 나타났다. 암 조직의 구성이 시간에 따라 변하였을 때(7일) 광학적 생검을 하면 측정된 4명의 환자의 형광의 세기에 변화가 있었다. 7일간 시간이 지난 암 조직이 형광세기가 더 작은 값을 갖는다. 광학적 생검은 조직을 인체에서 분리하지 않는 생체 내, 실시간, 비침습성(noninvasive)생검 방법이다. 본 연구에서는 구강의 정상조직과, 암 조직, 그리고 암 조직의 진화에 따른 구성의 변화를 형광스펙트럼으로 확인하였다. 형광분광법을 이용한 FastEEM장치는 암 조직의 변화를 확인함으로 방사선 치료 후 발생하는 암 조직 구성의 화학적, 생물학적, 형태학적 변화를 실시간으로 정확하게 측정이 가능한 장치임을 확인하였다.

Evaluation of Acoustic, Thermal, and Morphological Properties in the Egg White Phantom

  • The egg white phantom is a thermal lesion visualization phantom able to illustrate a thermal lesion. It is often used to evaluate the performance of HIFU and is less expensive than the BSA phantom. This study determined the optimal phantom composition for evaluated therapeutic ultrasound machines by varying the egg white concentration in the egg white phantom and demonstrated its utility as a therapeutic ultrasound phantom. The egg white phantom at varying egg white concentrations (10-40% in 10% intervals) was fabricated, and its thermal properties and acoustic properties were assessed. In addition, the size and shape of the formed lesion were compared between the egg white phantom and bovine liver tissue according to the electrical power. The results showed that 30% egg white phantom was optimal for the performance evaluation due to its thermal and acoustic properties. The generated thermal lesions formed sequentially as a cigar, ellipse, tadpole, and cone shapes according to the electrical power; a similar tendency was observed in the liver tissue. Hence, we conclude that the egg white phantom will prove useful in quantitatively evaluating the thermal effects of therapeutic ultrasound.

  • The average glandular dose (AGD) is determined by the breast entrance skin exposure, x-ray tube target material, beam quality (half-value layer), breast thickness, and breast composition. Almost breast cancer always arises in glandular breast tissue. As a result, the average radiation absorbed dose to glandular tissue is the preferred measure of the radiation risk associated with mammography. If the normalized average glandular dose is known, the average glandular dose can be computed from the product of the normalized average glandular dose and breast entrance skin exposure. In this study, AGD was calculated by the breast thickness and various x-ray energy (HVL) in 50% glandular 50% adipose breast by Mo.-Rh. assembly. AGD is 84 mrad in compressed 5 cm breast. These results show that as increasing the breast thickness, dose also increases. But as increasing the x-ray tube voltage, dose decreases because of high penetrating ratio through the object. But high tube voltage is reducing the subject contrast. From this result, we have to consider the trade-off between subject contrast of image and dose to the patient and choose proper x-ray energy range.

Preparation of Black Soybean Peptide and Its Effect on Weight Reduction in Rats

  • The large-scale preparation method for black soybean peptide (BSP; about 70% less than 10,000 Da; highly enriched with glutamic acid, aspartic acid, and arginine) was developed, and its effect on weight reduction and lipid profiles in rats was investigated. Sprague-Dawleymale rats were assigned to four dietary groups (high-fat diets containing 0, 2, 6, and 10% BSP) and fed four weeks to examine the effects of BSP. During the experiment, food intake was measured every two days and body weight was monitored two times a week. After the supplementation of BSP, liver and adipose tissues (epididymal, retroperitoneal and perirenal adipose tissue) in the rats were weighted and the lipid profiles in serum, liver, and feces were analyzed. At the results of body weight gain, liver and epididymal adipose tissue weight, BSP groups were more decreased than HF group (0% BSP), with greater decreases at higher BSP levels. The same patterns were shown in lipid profiles of serum, as BSP was increased, triglyceride and total cholesterol concentration decreased. The serum HDL-cholesterol level was increased with increasing at BSP levels. Total cholesterol concentration of liver and feces were decreased and increased, respectively, as BSP increased. The results confirm that BSP is involved in reducing the body weight and the improvement of lipid composition in serum and liver of rats and that BSP can be applied in weight reduction in the food products industry.

Carnitine and Calmodulin N-Methylation in Rat Testis; Calmodulin May beInvolved in Carnitine Biosynthesis

  • Rat testis known to contain all of the enzymes required for carnitine biosynthesis also contains high concentration of calmodulin, a protein which may or may not contain trimethyllysine, the major substrate in carnitine biosynthesis. The purpose of this study was to investigate the levels of carnitine and the state of calmodulin N-methylation in rat testes, and to discuss the possibility of the involvement of calmodulin incarnitine biosynthesis. Nonesterified carnitine , acid soluble acyl carnitine, and acid insoluble acyl carnitine of ra tests were 273 nmole, 62nmole, and 4 nmole/g tissue, respectively. Total carnitine level was 339 nmole/g testes tissue. Calmodulin purified from rat tests was assayed for methylation potential using N-methyltransferase from the rat testes. Rat testes calmodulin showed no 3H-methyl incorporation indicating that the calmodulin was trimethylated already by endogenous calmodulin N-methyltransferase. Amino acid composition analysis revealed that the rat testes calmodulin containd one mole of trimethyllysine per mole of calmodulin. These data suggest that testes calmodulin could provide the trimethyllysine needed for the synthesis of carnitine in the rat tests.

Obesity and Metabolic Syndrome in Adults with Prader-Willi Syndrome

  • Body fat distribution in patients with Prader-Willi syndrome (PWS) is characterized by reduce lean body mass (LBM), increased total body fat mass (FM), and lower percentage of visceral adipose tissue (VAT). Individuals with PWS seem to have a lower risk for insulin resistance with high levels of adiponectin, an anti-atherogenic adipocytokine that is decreased in visceral fat hypertrophy subjects compared to simple obese subjects, both in children and in adults. The mechanism of the reduction in visceral adiposity in PWS is still unclear. It might be related to qualitative intrinsic characteristics of adipocyte or novel genetic influences on the control of fat distribution. However, obesity remains a critical problem, and obesity status plays a crucial role in individual metabolic risk clustering and development of metabolic syndrome (Mets) in PWS children and adults. Long-term growth hormone (GH) treatment after cessation of skeletal growth improved body composition, with an increase in lean body mass and a reduction in total body fat and subcutaneous and visceral fat in PWS adults. Thus, the role of GH is important after childhood because it might attenuate obesity and Mets in PWS adult by adipocyte modification.

Effects of Exercise Treatment on Obesity: What Community Nutritionists Need to Know

  • By improving body composition, such as fat, lean body mass and total body weight, an exercise program can be an effective treatment of obesity. The effects of exercise on obesity have been confirmed via various approaches such as type, intensity, duration, frequency, and combination with diet. Combined exercise and diet is the most efficient strategy for weight loss, and exercise alone could improve metabolism irrespective of weight loss. In addition, physical activity, including exercise, is emphasized to avoid a 'yo-yo' phenomenon. Exercise increases lipolysis stimulated by such factors as catecholamine, growth hormone (GH), and hormone sensitive lipase (HSL). Moreover, changes in insulin and cortisol through exercise affect adipose tissue, which is known as not only an energy storage locale, but also as an endocrine organ. Adipocytokines secreted by adipose tissue respond to signals that modulate metabolism and inflammation. Exercise has generally shown positive effects on adipocytokines, and these effects increase in conjunction with a hypocaloric diet. However, a long duration and a high intensity of exercise could induce an inflammatory response. This review summarizes the effects of exercise on obesity treatment, which contributes to the exercise and nutritional fields, particularly of community nutritionists. Nanofabrication of Microbial Polyester by Electrospinning Promotes Cell Attachment

  • The biodegradable and biocompatible poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV), a copolymer of microbial polyester, was fabricated as nanofibrous mats by electrospinning. Image analysis of the electrospun nanofibers fabricated from a 2 wt% 2,2,2-trifluoroethanol solution revealed a unimodal distribution pattern of fiber diameters with an observed average diameter of ca. 185 nm. The fiber diameter of electrospun fabrics could be controlled by adjusting the electro spinning parameters, including the solvent composition, concentration, applied voltage, and tip-to-collector distance. Chondrocytes derived from rabbit ear were cultured on a PHBV cast film and an electrospun PHBV nano-fibrous mat. After incubation for 2 h, the percentages of attached chondrocytes on the surfaces of the flat PHBV film and the PHBV nanofibrous mat were 19.0 and 30.1 %, respectively. On the surface of the electrospun PHBV fabric, more chondrocytes were attached and appeared to have a much greater spreaded morphology than did that of the flat PHBV cast film in the early culture stage. The electro spun PHBV nanofabric provides an attractive structure for the attachment and growth of chondrocytes as cell culture surfaces for tissue engineering.

쑥을 이용한 과산화수소 정량 바이오센서의 전기화학적 성질 (Electrochemical properties of the mugwort-embedded biosensor for the determination of hydrogen peroxide)

  • 탄소반죽에 쑥조직을 혼입시켜 과산화수소 정량 바이오센서를 제작하고 그것의 전기화학적 성질을 조사하였다. 다른 생체조직을 이용하여 제작한 센서와 비교할 때, 생촉매 안정성이 뛰어났으며, 보다 큰 감응신호를 보여주었다. 조직의 함량 변화에 따른 신호의 변화는 넓은 범위에 걸쳐 직선성을 보여 주었고, Lineweaver-Burk 도시의 직선성은 전극 감응이 효소 촉매작용에 의하여 조절되고 있음을 보였다. 또 감응전류의 pH 의존성 변화는 센서가 두 종류의 동위효소를 포함하고 있음을 암시하였다.

Gelatinized Carbohydrates in the Diet of Catla catla Fingerlings: Effect of Levels and Sources on Nutrient Utilization, Body Composition and Tissue Enzyme Activities

  • A Feeding trial was conducted to study the effects of three different sources and two levels of dietary gelatinized carbohydrate (GC) on nutrient utilization, growth, tissue composition and tissue enzyme activities of fingerlings of Catla catla (15.1-15.3 g). Six isocaloric (17.1-17.5 kJ/g) semi-purified diets were prepared either with rice, corn or tapioca at 40 or 50% GC each. The crude protein (CP) level used in the diet was 35% and 25% for 40% and 50% GC level, respectively to study the protein sparing effect of GC. The degree of gelatinization was higher for corn and tapioca than rice under similar cooking conditions. After a 60-d feeding trial, dry matter, carbohydrate, protein and lipid digestibility were higher in tapioca fed groups at both the levels of GC. However, the highest specific growth rate (SGR) and protein efficiency ratio (PER) were observed in the corn fed groups at 50% GC level indicating better utilization of nutrients from gelatinized corn. Feed conversion ratio (FCR) was almost similar in corn and tapioca fed groups between two levels of GC but in rice fed groups, FCR was lower in 40% GC than 50% GC level. The results indicated higher protein-sparing effect in corn and tapioca fed groups than rice fed groups. The order of gelatinized carbohydrate utilization in Catla catla fingerlings at 50% GC level was corn>tapioca>rice. At 40% GC level, corn and tapioca were comparable and more efficiently utilized than rice. In the corn fed groups, 50% GC was comparable with 40% GC level, whereas in rice and tapioca fed groups the 40% GC was better in terms of nutrient utilization. Liver glycogen content and hepatosomatic index were significantly (p<0.05) higher in those groups fed high GC (50%) irrespective of carbohydrate sources. Higher intestinal amylase and glucose-6-phosphate dehydrogenase activities were observed in higher GC fed groups than the lower GC groups. No mortality was found in any groups at any levels of GC.
