Expression of Survivin and Caspase 3 in Oral Squamous Cell Carcinoma and Peritumoral Tissue

  • This study was conducted to investigate the expression of survivin and caspase 3 in oral squamous cell carcinoma and peritumoral tissue, and possible pathogenesis mechanisms. We used ELISA and western blotting to detect the protein expression levels of survivin and caspase 3 in tissue. In situ hybridization and real-time PCR were applied to assess mRNA expression levels. In this study, 13 tumor samples and 13 peritumoral tissue samples were collected from oral squamous cell carcinoma patients and 10 normal tissue samples obtained from patients without tumor. The result showed that the protein and mRNA expression of survivin in carcinoma was the highest among three types of tissue; following was that in peritumoral tissue. No difference in caspase 3 zymogen between peritumoral tissue and normal tissue could be found, while it was evidently decreased in carcinoma tissue. Activated caspase 3 was detected in normal tissue but could not be identified in peritumoral or carcinoma tissue. Our results indicate that the expression of survivin is apparently elevated in tumoral and peritumoral tissue. Expression of activated caspase 3 was not detected in tumoral tissue and the expression of caspase 3 zymogen was decreased in tumoral tissue. Our findings suggest that survivin may inhibit both synthesis and activation of caspase 3, hence inhibiting cell apoptosis and facililitating eventual development of oral squamous cell carcinoma.

접착성(接着性) 레진이 가견치수조직(家犬齒髓組織)에 미치는 독성(毒性)에 관(關)한 실험적(實驗的) 연구(硏究) (TOXIC EFFECT OF ADHESIVE RESINS ON THE DOG'S PULP TISSUE)

  • The toxic effect of adhesive resins on the dog's pulp tissue was studied with 70 teeth from 5 dogs. The experimental materials were Clearfil, a mixture of Clearfil with calcium hydroxide powder, Panavia-EX, and a mixture of Panavia-EX with calcium hydroxide powder. As a control group, calcium hydroxide powder was used. Each material was placed on the pulpotomized tissue surface. After 3 days, 1, 2,4, and 6 weeks, the teeth and apical tissue were processed routinly and stained with hematoxylin and eosin. Pathological tissue changes due to the toxicity of adhesive resins were observed by light microscope, and the pH of Panavia-EX and the Bonding agent of Clearfil were measured. Following were the results; 1. In the group of calcium hydroxide powder, slight inflammatory change was observed in the pulpotomized surface and adjacent pulp tissue on 3 day. 1 week case showed incomplete dentin bridge. The remaining pulp tissue was normalized according to the days elapsed. 2. In the group of Clearfil, early inflammatory change revealed in the superificial portion of the remaining pulp tissue on 3 day. The inflammation spreaded over the total pulp tissue and partial necrosis was observed in 1 week and 2 week cases. Total necrosis of pulp tissue and moderate inflammatory change at the apical tissue was noticed in 4 week and 6 week cases. 3. In the group of Panavia-EX, moderate inflammatory change appeared in the superficial pulp tissue on 3 day, and severe inflammatory change over all pulp tissue found in 1 week case. Pulp necrosis was obvious in 2 week case. 4 week and 6 week cases were totally necrotized up to the periapical tissue. 4. In the groups of mixtures with calcium hydroxide powder, the pulp tissue destruction was retarded, compared with the groups of Clearfil and Panavia-EX. 5. Panavia-EX was more destructive than Clearfil. 6. The acidity of freshly mixed Bonding agent of Cleafil was pH 4.0, and that of Panavia-EX was pH 2.0.

New Insights Into Tissue Macrophages: From Their Origin to the Development of Memory

  • Macrophages are the main effector cells of innate immunity and are involved in inflammatory and anti-infective processes. They also have an essential role in maintaining tissue homeostasis, supporting tissue development, and repairing tissue damage. Until few years ago, it was believed that tissue macrophages derived from circulating blood monocytes, which terminally differentiated in the tissue and unable to proliferate. Recent evidence in the biology of tissue macrophages has uncovered a series of immune and ontogenic features that had been neglected for long, despite old observations. These include origin, heterogeneity, proliferative potential (or self-renewal), polarization, and memory. In recent years, the number of publications on tissue resident macrophages has grown rapidly, highlighting the renewed interest of the immunologists for these key players of innate immunity. This minireview aims to summarizing the new current knowledge in macrophage immunobiology, in order to offer a clear and immediate overview of the field.

The Application of Machine Learning Algorithm In The Analysis of Tissue Microarray; for the Prediction of Clinical Status

  • Tissue microarry is one of the high throughput technologies in the post-genomic era. Using tissue microarray, the researchers are able to investigate large amount of gene expressions at the level of DNA, RNA, and protein The important aspect of tissue microarry is its ability to assess a lot of biomarkers which have been used in clinical practice. To manipulate the categorical data of tissue microarray, we applied Bayesian network classifier algorithm. We identified that Bayesian network classifier algorithm could analyze tissue microarray data and integrating prior knowledge about gastric cancer could achieve better performance result. The results showed that relevant integration of prior knowledge promote the prediction accuracy of survival status of the immunohistochemical tissue microarray data of 18 tumor suppressor genes. In conclusion, the application of Bayesian network classifier seemed appropriate for the analysis of the tissue microarray data with clinical information.

Design Parameters of Polymers for Tissue Engineering Applications

  • The loss or failure of an organ or tissue can occur because of accident or disease, for which tissue or organ transplantation is a generally accepted treatment. However, this approach is extremely limited due to donor shortage. Tissue engineering is a new and exciting strategy, in which patients who need a new organ or tissue are supplied with a synthetic organ or tissue. In this approach, tissues are engineered using a combination of the patient's own cells and a polymer scaffold. The polymer scaffold potentially mimics many roles of extracellular matrices in the body. Various polymers have been studied and utilized to date in tissue engineering approaches. However, no single polymer has been considered ideal for all types of tissues and approaches. This paper discusses the design parameters of those polymers potentially useful in tissue regeneration.

  • Both brown and white adipose tissues (BAT/WAT) are innervated by the peripheral nervous system, including efferent sympathetic nerves that communicate from the brain/central nervous system out to the tissue, and afferent sensory nerves that communicate from the tissue back to the brain and locally release neuropeptides to the tissue upon stimulation. This bidirectional neural communication is important for energy balance and metabolic control, as well as maintaining adipose tissue health through processes like browning (development of metabolically healthy brown adipocytes in WAT), thermogenesis, lipolysis, and adipogenesis. Decades of sensory nerve denervation studies have demonstrated the particular importance of adipose sensory nerves for brown adipose tissue and WAT functions, but far less is known about the tissue's sensory innervation compared to the better-studied sympathetic nerves and their neurotransmitter norepinephrine. In this review, we cover what is known and not yet known about sensory nerve activities in adipose, focusing on their effector neuropeptide actions in the tissue.

광 조사에 따른 온도분포를 이용한 생체조직 내 비정상조직 탐지에 대한 연구 (A Study on the Detection of Abnormal Tissues in Biological Tissue Using Temperature Distribution According to Light Irradiation)

  • 본 연구에서는 비정상조직(파라핀)을 가진 생체조직에 광을 조사하고 그에 따른 조직의 표면온도와 비정상조직 주위에서의 온도분포를 실험과 해석적 방법을 통해 분석하였다. 파라핀을 이용하여 비정상 조직을 모사한 후 조사하는 광의 파장과 시간을 변화시키면서 조직 주위에서의 온도를 K형 열전대를 사용하여 측정하였다. 또한 전산열전달 기법을 이용하여 해석적으로 조직에 대한 온도분포를 예측하였다. 정상조직과 비정상조직의 주위에서의 온도는 차이가 있었으며, 비정상조직이 있는 경우 표면과 조직 주위의 온도가 높게 나타났다.

3D 바이오프린팅과 무지지체 조직공학 기술 기반 추간판 복합 조직 제작 (Fabrication of Tissue Engineered Intervertebral Disc Using Enable 3D bio-printing and Scaffod-Free technologies)

  • Intervertebral disc(IVD) mainly consists of Annulus fibrosus(AF) and Nucleus pulposus(NP), playing a role of distributing a mechanical load on vertebral body. IVD tissue engineering has been developed the methods to achieve anatomic morphology and restoration of biological function. The goal of present study is to identify the possibilities for creating a substitute of IVD the morphology and biological functions are the same as undamaged complete IVD. To fabricate the AF and NP combine biphasic IVD tissue, AF tissue scaffolds have been printed by 3D bio-printing system with natural biomaterials and NP tissues have been prepared by scaffold-free culture system. We evaluated whether the combined structure of 3D printed AF scaffold and scaffold-free NP tissue construct could support the architecture and cell functions as IVD tissue. 3D printed AF scaffolds were printed with 60 degree angle stripe patterned lamella structure(the inner-diameter is 5mm, outer-diameter is 10 mm and height is 3 mm). In the cytotoxicity test, the 3D printed AF scaffold showed good cell compatibility. The results of histological and immunohistochemical staining also showed the newly synthesized collagens and glycosaminoglycans, which are specific makers of AF tissue. And scaffold-free NP tissue actively synthesized glycosaminoglycans and type 2 collagen, which are the major components of NP tissue. When we combined two engineered tissues to realize the IVD, combined biphasic tissues showed a good integration between the two tissues. In conclusion, this study describes the fabrication of Engineered biphasic IVD tissue by using enable techniques of tissue engineering. This fabricated biphasic tissue would be used as a model system for the study of the native IVD tissue. In the future, it may have the potential to replace the damaged IVD in the future.

하악골후방이동술 후 골격구조와 연부조직의 변화 (The Change of Bone and Soft Tissue Profile after Sagittal Split Osteotomy of Ramus)

  • Orthognathic surgery for Class III malocclusion requires an elaborate preoperative planning using cephalometries or Mock surgery models which enable the surgeon to anticipate postoperative skeletal changes of maxilla and mandible as well as dentition. After surgery, patient's satisfaction is greatly influenced by appearance of soft tissue change. Therefore, it is imperative to predict a relatively accurate soft tissue change prior to surgery. A 5 year retrospective study was designed to evaluate the soft tissue change after sagittal split osteotomy of ramus(SSRO) for class III malocclusion. Analyses of preoperative and postoperative anthropometric measurements were performed. Patients who were treated only by SSRO for class III malocclusion and could follow up for 6 months were studied. Among them, the patients who had history of cleft palate and lip or hemifacial microsomia were excluded. Soft tissue changes were estimated by using the frontal and lateral photographs. Skeletal changes were observed by measuring amount of set back and angular changes of mandible to the reference line by using cephalometries. Relapses were also measured 6 months after the operation. We could observe skeletal changes were more profound than soft tissue changes concerning amount of set back, but soft tissue changes were also profound in angle. Relapse was more profound in skeleton than soft tissue but the amount was not significant. In spite of the variables which may affect proper assessment of the soft tissue change after skeletal relocation, this study can serve as a guide for exact prediction of the postoperative change of soft tissue and skeleton.
