Ultrasonic Tissue Characterization by Digital Spectrum Analysis Technique (Digital Spectrum 분석방법을 이용한 조직특성 변수에 관한 연구)

  • A digital spectrum analysis technique was used to estimate the tissue characteristic parameters (transmission velocity and attenuation coefficient) in the phantom study and the human liver's ultrasound scanning. The soft tissue equivalent phantom was made with the combination materials of agar, water, powdered graphite, and n-propyl alcohol. In the human study, twenty five normal subjects and three patients with liver diseases were studied using the ultrasonic reflection signals and the spectrum analysis method The following results were obtained; 1. The soft tissue-equivalent materical could be produced with various acoustic parameters by changing the composition amount of the powdered graphite and n-propyl alcohol. 2. Attenuation coefficients of normal human liver tissue were estimated to be 0. 36 dB/cm MHz$\pm$0.11. In patients with liver disese, tile attenuation coefficients were shown to be different from the above normal values.

  • In this paper, we present the software on the development of the computerized imaging system for diagnosis and treatment plannig of orthodontics and orthognathic surgery. Soft tissue changes followed by orthognathic surgery mainly depends on surgical movements of hard tissue. Then, the stepwise multiple regression method was used to investigate the soft tissue changes followed by hard tissue changes. As a result of this research, we were able to develop a system which diagnoses automatic X-ray images and predicts soft tissue changes after othognathic surgery.

  • A laser moxibustion therapy device having effect similar to that of traditional moxibustion is being developed using 1064 nm infrared laser. The therapy device allows direct interaction of laser light with the tissue rendering temperature distribution both on the skin surface and deep under the skin. We made a device that could measure temperature of deep under the surface of agar gel tissue phantom using thermocouples. A thermal imaging camera was used to verify results from the temperature measurement device. We compared the characteristics of heat transfer inside the tissue phantom during moxibustion and laser irradiation. The temperature distribution measured by thermocouples was found to be similar to that of distribution given by thermal imaging camera.

  • Hypoxia (decreased tissue oxygen tension) is a component of many diseases such as tumors, cerebrovascular diseases and ischemic heart diseases. Although hypoxia can be secondary to a low inspired $pO_2$ or a variety of lung disorders, the most common cause is ischemia due to an oxygen demand greater than the local oxygen supply. In the heart tissue, hypoxia is often observed in persistent low-flow states, such as hibernating myocardium. Direct "hot spot" imaging of myocardial tissue hypoxia is potentially of great clinical importance because it may provide a means of identifying dysfunctional chronically ischemic but viable hibernating myocardium. A series of radiopharmaceuticals that incorporate nitroimidazole moieties have been synthesized to detect decreased local tissue pO2. In contrast to agents that localize in proportion to perfusion, these agents concentrate in hypoxic tissue. However, the ideal agents are not developed yet and the progress is very slow. Furthermore, the research focus is on tumor hypoxia nowadays. This review introduces the myocardial hypoxia imaging with summarizing the development of radiopharmaceuticals.

Expression of Extracellular Superoxide Dismutase Protein in Diabetes

  • Background Diabetes is characterized by chronic hyperglycemia, which can increase reactive oxygen species (ROS) production by the mitochondrial electron transport chain. The formation of ROS induces oxidative stress and activates oxidative damage-inducing genes in cells. No research has been published on oxidative damage-related extracellular superoxide dismutase (EC-SOD) protein levels in human diabetic skin. We investigated the expression of EC-SOD in diabetic skin compared with normal skin tissue in vivo. Methods The expression of EC-SOD protein was evaluated by western blotting in 6 diabetic skin tissue samples and 6 normal skin samples. Immunohistochemical staining was also carried out to confirm the EC-SOD expression level in the 6 diabetic skin tissue samples. Results The western blotting showed significantly lower EC-SOD protein expression in the diabetic skin tissue than in the normal tissue. Immunohistochemical examination of EC-SOD protein expression supported the western blotting analysis. Conclusions Diabetic skin tissues express a relatively small amount of EC-SOD protein and may not be protected against oxidative stress. We believe that EC-SOD is related to the altered metabolic state in diabetic skin, which elevates ROS production.

  • Deformation of soft tissue is known inhomogeneous and non-linear in general. In this study, we propose a measurement methodology of local/global strain during soft tissue elongation precisely using laser extensometer which has high accuracy, resolution and is possible to measure global/local strain. The mechanical tensile test are performed on tibialis cranialis, flexor hallucis longus, extensor digitorum longus of swine hindlimb. In order to measure target displacement, reflective marker is attached to detect elongation on specimen using surgical adhesive. The result of this study is to show that laser extensometer is valid to measure longitudinal elongation which is inhomogeneous and non-linear fur soft tissue.

  • There were many difficulties in the treatment of extensive, massive, and composite defect in the lower extremity until early 1980's. Recently, microscopic reconstruction of wide soft tissue defect is popularized. But, the combined flap, which requires wide coverage of lower extremity after soft tissue sarcoma excision or traffic accident, is still challenging to the orthopaedic surgeons. We experienced 12 cases of combined scapular and latissimus dorsi flap from 1983 to 1997 in the lower extremity reconstruction of soft tissue defect with satisfactory result. There were no serious donor site complications such as functional disturbance of shoulder joint.

Analysis of Tissue Plasminogen Activator Expression using Pollen Culture in vitro (기내 화분배양을 이용한 Tissue Plasminogen Activator 발현분석)

  • In an effort to use plant biotechnology for the production of biopharmaceuticals, pollens collected from lily (Lilium longiflorum) were grown in vitro and transformed with a PCR-amplified 1.7 kb cDNA encoding human tissue plasminogen activator (tPA) using Agrobacterium via a vacuum infiltration process. Western blotting showed that transgenic lily pollen tubes selected on kanamycin for 16 hrs expressed a tPA protein with a size similar to the human standard, suggesting their possible use as a disposable host for rapid foreign protein production.

  • Complex cell-to-cell communication underlies the basic processes essential for homeostasis in the given tissue architecture. Obtaining quantitative gene-expression of cells in their native context has significantly advanced through single-cell RNA sequencing technologies along with mechanical and enzymatic tissue manipulation. This approach, however, is largely reliant on the physical dissociation of individual cells from the tissue, thus, resulting in a library with unaccounted positional information. To overcome this, positional information can be obtained by integrating imaging and positional barcoding. Collectively, spatial transcriptomics strategies provide tissue architecture-dependent as well as position-dependent cellular functions. This review discusses the current technologies for spatial transcriptomics ranging from the methods combining mechanical dissociation and single-cell RNA sequencing to computational spatial re-mapping.

SOFT HARD TISSUE CHANGES FOLLOWING ANTERIOR SEGMENTAL SURGERY IN BIMAXILLARY PROTRUSION (상하악 치조전돌증의 양악 분절골절단 수술후 경, 연조직 변화에 관한 연구)

  • The purpose of this study was to evaluate the interrelationship of the soft and hard tissue changes after both maxillary and mandibular anterior segmental surgery in bimaxillary protrusion patients. 11 patients had received both maxillary and mandibular anterior segmental surgery and were investigated radiologically with lateral cephalogram. The results were as follows : 1. The correlation of maxillary hard and soft tissue horizontal changes were relative high. : Coefficient between UIE and Stms was 0.89 (p<0.001). 2. The correlation of mandibular hard and soft tissue horizontal changes were very high, especially at the chin. : Coefficients were over 0.90 (p<0.001) 3. All points were moved superiorly except SLS, LS, Stms. 4. Upper and lower lip convexity to the E-Line were decreased (p<0.001) and postsurgical facial profiles were changed very esthetically.
