Anti-adherence of Antibacterial Peptides and Oligosaccharides and Promotion of Growth and Disease Resistance in Tilapia

  • Four hundred and fifty tilapias ($6.77{\pm}0.23$ g) were assigned randomly to six groups to evaluate the feasibility of the tested antibacterial peptides (ABPs) and oligosaccharides as substitutes for antibiotics. The control group was fed with a commercial tilapia diet; other five groups were fed with the same commercial diet supplemented with konjac glucomannan (KGLM), cluster bean galactomannan (CBGAM), and three animal intestinal ABPs derived from chicken, pig and rabbit at 100 mg/kg respectively. After 21 days of feeding, growth, disease resistance, and in vivo anti-adherence were determined. Furthermore, the inhibitory effect of tested agents on adhesion of Aeromonas veronii biovar sobria (A.vbs) strain BJCP-5 to tilapia enteric epithelia in vitro was assessed by cell-ELISA system. As a result, the tested agents supplemented at 100 mg/kg show significant benefit to tilapia growth and disease resistance (p<0.05), and the benefit may be correlated with their interfering in the contact of bacteria with host mucosal surface. Although none of the tested agents did inhibit the growth of BJCP-5 in tryptic soy broth at $100{\mu}g/ml$, all of them did inhibit the adhesion of A.vbs to tilapia enteric epithelia in vivo and in vitro. In vitro mimic assays show that three ABPs at low concentrations of $25{\mu}g/ml$ and $2.5{\mu}g/ml$ have the reciprocal dose-dependent anti-adherence effect. The inhibition of ABPs may be correlated with a cation bridging and/or receptor-ligand binding, but not with hydrophobicity. The KGLM and CBGAM inhibited the adherence of BJCP-5 to tilapia enteric epithelia with dose-dependent manner in vitro, and this may be through altering bacterial hydrophobicity and interfering with receptor-ligand binding. Our results indicate that the anti-adherence of the tested ABPs and oligosaccharides may be one of the mechanisms in promoting tilapia growth and resistance to A.vbs.

Nitrogen and Phosphorus Excretion of Nile Tilapia (Oreochromis niloticus) Fed Commercial Diets (시판 사료를 먹인 틸라피아( Oreochromis nijoticus) 의 질소 및 인 부하량)

  • Attempts were made to find out nitrogen and phosphorus loads to aquatic environment resulting from feeding Nile tilapia (Oreochromis niloticus). Two different size groups, small and large, were used. The average sizes of small and large tilapia were 65.2 g and 389.2 g respectively, and three kinds of commercial diets were used for each size. The 3 kinds of commercial diets for tilapia contained in average 33.8% crude protein ($5.4\%$ nitrogen) and $1.4\%$ total phosphorus. The load of nitrogen and phosphorus were measured by subtracting the amounts of nutrients retained in the body of fish from consumed nutrients. Sixty, five percentage of total feces was excreted within 24 hours after feeding at $23^{\circ}C$. Nitrogen content in the feces was higher in large fish than small ones. The apparent digestibility of dietary protein for small and large tilapia was $90.0\%$ and $89.7\%$, respectively. Availability of dietary phosphorus for small and large tilapia was $44.7\%\;and\;51.4\%$, respectively. The total load of nitrogen and phosphorus per 1 metric ton of tilapia production was 49.5kg and 6.3kg, respectively, for small ones with feed conversion ratio (FCR) of 1.4, and 61.3 kg and 13.4kg, respectively, for large ones with the FCR of 1.8. Nitorgen balance appeared that small and large tilapia excreted $7.1\%\;and\;9.9\%$ of consumed nitrogen through fecal-nitrogen and $55.5\%\;and\;62.3\%$ through urine and gills, retaining $37.4\%\;and\;27.8\%$ in the body, respectively. These results show that small fish pollute less than large fish, excreting less and retaining more nutrients in the body.

  • A combined field release of indigenous larvivoroug fish (Aplocheilus latipes) and hybrid herbivore (Tilapia mossambicus miloticus) was conducted to determine the effectiveness of biological control, in particular, against malaria vector (anopheles sinensis), breeding in weedy habitat of rice fields at the University rice paddies at Suwon during the period of June through September in 1988. A combined fish introduction at the release rate of 2.0 fish per $M^{2}$ for Aplocheilus and 1-pair $10M^{2}$ water surface for Tilapia resuited in 70.8% Anopheles larval reducton in a week period, the rates of rontrol increased to 73. 5% and 80.2% in 4th and 5th week respectively after the fish introdurtion, maintaining mosquito suppression in range of 80~82% control after the 5th week against Anopheles and Culex spp. combined. In a single fish treatment with Aplocheilus at 2.0 $fish/M^{2}$ release rate, Anopheles larval reduction ranged on the average 51.4~56.5%in 4 wrek period which was later integratpd with B.t.i. (Rl53.78) treatment at 1.0 kg/ha dosage rate to suppress vector mosquito population down and the the results was compared with that of combined fish introdurtion.

Effect of Calcium Source using Tilapia Mossambica Scales on the Bone Metabolic Biomarkers and Bone Mineral Density in Rats (Tilapia Mossambica 비늘 (어린) 유래 칼슘소재가 흰쥐의 골격대사지표와 골밀도에 미치는 영향)

  • This study was done to evaluate the effect of Ca source using fish (Tilapia mossambica) scales on the bone metabolism. Male Sprague-Dawley rats, 4 weeks of age, were fed low-calcium diet (0.15% Ca) for 2 weeks. The rats on the low-calcium diet were further assigned to one of following three groups for an additional 4 weeks: 1) Ca-depletion group (LoCa) given 0.15% Ca diet ($CaCO_3$), 2) Ca-repletion group (AdCa) given 0.5% Ca diet ($CaCO_3$), 3) Ca-repletion diet (AdFa) received 0.5% Ca diet (Ca source from Tilapia mossambica scales). Serum parathyroid (PTH) and calcitonin showed no differences among experimental groups. Whereas LoCa group elevated the turnover markers, serum ALP and osteocalcin, and urinary deoxypyridinoline (DPD), AdCa and AdFa groups reduced their values. Elevation in the femoral weight, ash and Ca contents was observed in AdCa and AdFa groups. Bone mineral density was increased in AdCa and AdFa groups by 25-26% compared with LoCa group. These data demonstrate that Ca repletion with either Ca source from Tilapia mossambica scales or $CaCO_3$ is similarly effective in the improvement of bone turnover markers and BMD, suggesting the usefulness of Tilapia mossambica scales in the prevention of bone loss compared with $CaCO_3$.

Studies on Polymorphism of Transferrin of Serum Proteins in Tilapia (Oreochromis niloticus) (Tilapia(Oreochromis niloticus)의 혈청과 단백질의 transferrin의 다형현상에 관한 연구)

  • This study was taken to isolate transferrin fractions from the sera of tilapia(Oreochromis niloticus) by physico-chemical analyses such as the rivanol precipitation, iron-staining method, SDS-polyacrylamide gel electrophoresis and $^{59}FeCl_3$ autoradiography, and to calculate gene frequencies by using Hardy-Weinberg Law. The results obtained in this experiment were summarized as follow : 1. The transferrin fraction is composed of several components possessing relative lower electrophoretic mobilities and higher molecular sizes than the albumin components. 2. When different staining method was compared with transferrin in band, it was not found difference. 3. It was concluded that the optimun ratio of rivanol to serum was 2 : 1 and this ratio was used in all further fractionation. 4. The molecular weight of transferrin component was about 70,000 $\pm$ 2,000. 5. Tilapia transferrin fractionations were found to be polymorphic. 6. There transferrin variants(A, B and C) have been found in tilapia(Oreochromis niloticus) and Tf types were assumed to be controlled by three codominant alleles Tf A, Tf B and Tf C. Six different phenotypes can be theoretically expected Tf AA, Tf AB, Tf AC, Tf BB, Tf BC, and Tf CC. Only five types of these were observed and Tf CC types(homozygotes) was not found. 7. The frequencies of the three allele Tf A, Tf B and Tf C were 0.795, 0.15 and 0.055 respectively.

The Proliferative and Apoptotic Properties of Nile Tilapia (Oreochromis niloticus) sIg+ Lymphocytes by Cortisol Treatment

  • The effects of cortisol on proliferation and apoptosis of tilapia surface immunoglobulin positive ($sIg^+$) lymphocytes isolated from different tissues were investigated. $sIg^+$ lymphocytes from the tilapia head kidney (HK) and spleen showed a higher proliferation and lower intracellular calcium ($Ca^{2+}{_i}$) level to Ig-crosslinking compared with peripheral blood $sIg^+$ lymphocytes. Peripheral blood $sIg^+$ lymphocytes stimulated with lipopolysaccharide (LPS) showed high levels of apoptosis in the presence of cortisol. HK and to a lesser extent spleen $sIg^+$ lymphocytes, although less sensitive than their equivalent in peripheral blood, showed cortisol-induced apoptosis irrespective of LPS stimulation of control levels. Compared to plasma values measured during stress conditions, proliferation regardless of LPS stimulation was apparently suppressed by cortisol that is effective in inducing a significant increase in apoptosis in all three different cell populations of $sIg^+$ cells, suggesting the immunoregulatory effect of cortisol in both LPS stimulated and non-stimulated conditions. Different sensitivity of $sIg^+$ cells to the cortisol, in regard to developmental stage and activity, could be related in inhibiting excessive and continuing depletion of $sIg^+$ lymphocytes.

  • The approximate compositions and the taste compounds of tilapia, Oreochromis nitoticus, cultured in freshwater and seawater for six months, were analyzed. As for the approximate compositions, there were little differences in moisture and protein contents of tilapia cultured in seawater and freshwater, but somewhat higher lipids and ash than that cultured in freshwater. Also, no significant differences in the content of free amino acids, nucleoli-des, betaine, TMAO, and total creatinine were observed between both groups.

  • Neurokinin B (NKB) and neurokinin B related peptide (NKBRP) belong to tachykinin peptide family. They act as a neurotransmitter and/or neuromodulator. Mutation of NKB and/or its cognate receptor, NK3R resulted in hypogonadotropic hypogonadism in mammals, implying a strong involvement of NKB/NK3R system in controlling mammalian reproduction. Teleosts possess NKBRP as well as NKB, but their roles in fish reproduction need to be clarified. In this study, NKB and NKBRP coding gene (tac3) was cloned from Nile tilapia and sequenced. Based on the sequence, Nile tilapia NKB and NKBRP peptide were synthesized and their biological potencies were tested in vitro pituitary culture. The synthetic NKBRP showed direct inhibitory effect on the expression of GTH subunits at the pituitary level. This inhibitory effect was confirmed in vivo by means of intraperitoneal (ip) injection of synthetic NKB and NKBRP to mature female tilapia (20 pmol/g body weight [BW]). Both NKB and NKBRP had no effect on the plasma level of sex steroids, E2 and 11-KT. However, NKBRP caused declines of expression level of GnRH I, Kiss2 and tac3 mRNAs in the brain while NKB seemed to have no distinct effect. These results indicate some inhibitory roles of NKBRP in reproduction of mature female Nile tilapia, although their exact functions are not clear at the moment.

Physiological Responses of Grey Mullet (Mugil cephalus) and Nile Tilapia (Oreochromis niloticus) by Rapid Changes in Salinity of Rearing Water (사육수의 급격한 염분변화에 따른 숭어 (Mugil cephalus)와 틸라피아 (Oreochromis niloticus)의 생리적 반응)

  • Physiological responses (cortisol, glucose, GPT, GOT, total protein, hematocrit, sodium, chloride, potassium, total calcium and osmolality), growth and survival rates of grey mullet (Mugil cephalus) and Nile tilapia (Oreochromis niloticus) by the manipulation of salinity were studied. Salinity was increased directly from freshwater to $33\%_{\circ}$ and remained for 15 days. Then, salinity was decreased directly from seawater to freshwater and remained for other 15 days. All tilapias were dead at the third day, but $96.5\%$ of grey mullet were survived at the 30th day. The grey mullet showed no significant differences for cortisol and glucose concentrations in the seawater and freshwater. However, those of the Nile tilapia were increased in seawater. The grey mullet showed no significant differences for sodium, chloride and potassium concentrations during experimental period. However, those of the Nile tilapia were significantly increased when exposed to seawater. The osmolality of grey mullet ranged between 293.5 mOsm/kg and 335.0 mOsm/kg for the whole experimental period.
