Isolation of Marine Bacteria Killing Red Tide Microalgae II. Isolation and Algicidal Properties of Pseudomonas sp. LG-2 Possessing Killing Activity for Dinoflagellate, Prorocentrum micans (적조생물 살조세균 탐색 II. 적조생물 Prorocentrum micans 살조세균 Pseudomonas sp. LG-2의 분리와 살조특성)

  • We have isolated a bacterial strain that tends to kill P. micans from the mixed culture of p. minns plus seawater filtrate (poresize, 0.8 $\mu$m) collected at Masan bay in July 1996, in which the mixed culture grown in the f/2 medium. According to the experimental results of the isolated bacterium such as fatty acids analysis, morphological and biochemical characteristic tests, the strain was supposed to be a Pseudomonas and then it was named as Pseudomonas sp. LG-2. The killing effect of Pseudomonas sp. LG-2 against P. micans was proportionally increased with the concentrations of culture filtrate (pore size, 0.8 $\mu$m) is well as with the number of bacterium inoculated. In the mixed culture inoculated with $1.3\times10^6$ cells/ml of Pseudomonas sp. LG-2, the number of P. micans (2,000 cells/ml) was gradually decreased and then killed below 100 cells/ml within 7 days. In addition, the culture filtrate with $30\%$ of final concentration revealed a significant killing effect against P. micans around 3 days after culture. In the relationship between killing effects and growth stage of Pseudomonas sp. LG-2, the culture filtrate at lag phase has little effects on P. micans. In constant, the culture filtrate at mid-log phase showed the killing effect by decreasing P. micans to 112 in number within 5 days. In particular, the culture filtrate at stationary phase showed a significant killing effect against P. micans in which the majority of it was killed after 3 day culture. The species specificity of killing effects of Pseudomonas sp. LG-2 against 5 species of dinoflagellate was only found in P. micans and Scrippsiella trochoidea.

Characteristics of Algicide Produced by Micrococcus luteus SY-13 Inhibiting Cochlodinium polykrikoides and the Effects on Marine Organisms (적조생물 Cochlodinium polykrikoides를 저해하는 Micrococcus luteus SY-13이 생산하는 살조물질의 특성과 해양생물에 미치는 영향)

  • Algicidal bacterium was isolated from sea water during the declining period of Cochlodinium polykrikoides blooms and this bacterium had a significant algicidal activity against C. polykrikoides. In this study, algicidal bacterium was identified on the basis of biochemical and chemotaxonomic characteristics, and analysis of 16S rDNA sequences. The algicidal bacterium showed 98.6% homology with Micrococcus luteus ATCC $381^T$. Therefore, this bacterium was designated Micrococcus luteus SY-13. The optimal culture conditions of the algicidal bacterium was $25^{\circ}C$, initial pH 8.0, and 3.0% NaCl concentration. M. luteus SY-13 is assumed to produce secondary metabolites which have algicidal activity. When 10% culture filtrate of this strain was applied to C. polykrikoides ($1.0\;{\times}\;10^4\;cells/ml$) cultures, over 98% of C, polykrikoides cells were destroyed within 6 hours. The culture filtrate of M. luteus SY-13 exhibited similar algicidal activity after heat-treatment at $121^{\circ}C$ for 15 min. While algicidal activity remained in filtrates with pH adjusted to 8.0, loss of algicidal activity occurred when the pHs of filtrates were adjusted to over 9.0 or heat-treated at $121{\times}180^{\circ}C$ for 1 hour. M. luteus SY-13 showed significant algicidal activities against C. polykrikoides (98.9%) and a wide algicidal range against various harmful algal bloom (HAB) species. However, there was no algicidal effect on diatom and marine livefood organisms except Isocrysis galbana. These results suggest that M. luteus SY-13 could be a candidate for use in the control of HABs.

Temporal Variations of Stratification-Destratification in the Deukryang Bay, Korea (하계 득량만의 연직성층해양의 시간적 변동 특성)

  • The Quantitative estimations of the stratification - destratification(SD) phenomena in Deukryang Bay, Korea have been carried out based on the data of wind speed, heat flux through the sea surface and tidal current amplitude. To find out the main factors causing SD, wv introduce the rate of energy balance of the surface heat flux, tidal and wind stirring proposed by Simpson and Hunter(1974). The calculated potential energy of three terms are compared, from which the energy of wind stirring effect was one order smaller than the heat flux and the tidal stirring. Using the results, we complement time integration of the potential energy with the several s values of 0.010~0.014 at interval 0.001 and with wind speeds of 1.5 and 2.0 times larger than observation values at land. It shows that the variation of SD phenomena in the bay mainly depended on tidal stirring and sea surface heating in summer if there is no exceptionally strong wind event like Typhoon. The stratification become to be foamed from about 5 July although the stratification a little decreases during the second spring tidal period of middle of July.

Variation of Density Stratification due to Fresh Water Discharge in the Kwangyang Bay and Jinju Bay (광양만과 진주만 해역에서 담수 유입으로 인한 밀도성층 변화)

  • This study simulate three-dimensional ocean circulation patterns using the EFDC model in the Kwangyang Bay and Jinju Bay, considering tide, water temperature and salinity. The numerical model results were verified using observed data. The model results well reproduced the observed data. As a result, ocean circulation patterns in the study area show convergence and divergence in the middle area of Noryang waterway and Daebang waterway, the residual flow patterns show typical two-layer circulation. According to the change of the density stratification in the Kwangyang Bay and Jinju Bay, the effect of fresh water is dominant in study area. In the case of Jinju Bay, although it is strongly influenced by the Namgang fresh water, also it is affected by Seomjin River when there is no discharge by Namgang Dam. On the other hand, the stratification of the Kwangyang Bay is relatively enhanced by the discharge of Namgang Dam.

A Study of Distribution of Jellyfish by Particle Numerical Experiment in Masan Bay (마산만에서 입자수치실험에 의한 해파리 분포연구)

  • The spatio-temporal distribution of jellyfish in Masan Bay was investigated in this study using a numerical model. First, a three-dimensional hydrodynamic model (POM) was constructed,taking into account residual flows, tides, temperature, salinity, and wind effects. A particle tracking model based on residual flow was then used to investigate the jellyfish present in Masan Port, referred to as the Heavy Industry and Gapo New Port in Masan. Jellyfish distribution was concentrated with maximum (2,533 individual) in the North Sea near Machang Bridge. Itcan be concluded that this concentration was due to multi-directional residual flows and topography effects. Residual flow currents are a dominant factor in understanding the aggregation of jellyfish, and this study used a numerical simulation of tide-induced residual currents, wind-driven currents and density currents in distinct cases to thoroughly address the topic. As a result, wind-driven currents (effect of the wind) was found to be superior to other components as an influence on the distribution of jellyfish near Machang Bridge and Modo in Masan Bay.

  • This research was attempted to study on the effects of desalinization by the depth and interval of open conduit in Kang-Hwa polder where is located at the Kil-sang Myun, Kang-Hwa Gun, Kyung-gi Do, and it has been continued for the three years from 1967 to 1969. The results obtained are as follows; 1. The depths of saline expulsion by supplying of irrigation water are approximately 30cm to 50cm under the ground surface, but saline expulsion is hardly done in case of the depth which is deeper than the above mentioned, because the moisture and saline content hardly change in such a condition. 2. The speed of vertical percolation gradually decreases below the 30cm depth, but it is noticed that there is a tendency to make the percolation of the horizental direction from its layer in Kang-Hwa reclaimed tidal land. 3. Comparing experimental treatments-varing depths and intervals of open conduits, the interval of open conduit has a more effect upon the promotion of desalinization and increasing of the rice yields than the depth of it. Therefore, according to the results of experimental data, the optimum depth of open conduit is about 0.9m, the effective interval of it is about 18m. 4. Considering the loss of arable area by the layout of open conduit, the reasonable interval of it could extend to 36m.

Composition of Fatty Acid and the Effect of Environmental Factors on the Population Growth of Scrippsiella trochoidea a Dinoflagellate Responsible for a Red Tide (적조와편모조 Scrippsiella trochoidea 군증식에 미치는 환경요인과 지방산 조성)

  • The cyst of Scrippsiella trochoidea from the surface mud in Masan Bay was germinated in the incubator for the culture. This species was one of dinoflagellates responsible for the early spring bloom in the southern coastal water of Korea. The culture experiments were carried out under the various gradients of environmental factors to know their effects on the population growth of this dinoflagellate. With respect to the effects of environmental factors on the growth, it was proved that the maximum cell growth was occurred at 4,000 lux of light intensity, salinity $30\%0$ and temperature $20^{\circ}C$. When 0.25ml/ml of the filtrates of Skeletonema costatum culture medium and the supernatants of soil extracts were added to growth medium as organic growth stimulants, both materials enhanced the population growth. In the fatty acid composition of S. trochoidea, $C_{16:0}$ was the major component, and $C_{18:0},\;C_{18:1},\;C_{22:0}\;and\;C_{22:1}$ were a minor components.

The Variation of Water Quality due to Sulice Gate Operation in Shiwha Lake (시화호의 배수갑문 운용에 따른 수질변화)

  • To evaluate the change for water quality after the water gate operation in Shiwha lake, in situ survey were conducted on september in 2000 and January, march, jane in 2001. Chemical characteristics and eutrophication level was estimated from the survey data. The water quality of the Shihwa lake was greatly affected by pollutant load from rainfall, and formation of stratification in summer and winter was increased to effect on nutrient release from sediment. Especially, high concentration of chlorophyll-a was occurred in autumn, due to increased nutrient, high water temperature and low salinity after rainfall runoff. The mean concentration of DIN, DIP were 0.346mg/L, 0.0217mg/L in surface water and 0.826mg/L, 0.0415mg/L in bottom water, respectively, which were over III grade of seawater standard. Also high percentage of ammonia nitrogen to DIN in bottom water for autumn and winter was affected by released nutrient from sediment. Correlation analysis of chlorophyll-a versus TSS was shown that organic matter was affected by autochthonous organic matter stem from the algae, these factor showed reverse correlation about salinity. Closely correlations among to the water quality constituent in continuity survey was appeared. The results of eutrophication index estimation showed the high potentiality of red tide occurrence in Shiwha lake, particularity in summer or fall. Overall water quality was greatly improve to compared with measuring data during 1997~1998 at the beginning water gate operation, which reported by KORDI. Therefore, to improve of water quality in Shiwha lake, we need to establish of management plan about nutrient release from sediment, rainfall runoff, maximum of seawater exchange.

Analysis of Tidal Effect in Hangang Bridge by Automatic Discharge Measurement (자동유량측정에 의한 한강대교 조석영향 분석)

  • The measuring point of the Hangang Bridge affected by tide has some special topographic characteristics due to Nodle Island. Furthermore the submerged weirs located on the upstream and downstream. Therefore flow is separated and joined by Nodle Island. Discharge measurement at the point of the Hangang Bridge is very important, because Hangang Bridge is key station in managing the discharge and flood forecasting. In the past, it was too difficult to measure discharge in tidal conditions. HRFCO(Han River Flood Control Office) installed automatic discharge measurement facilities for solving this problem. Measuring equipments operates and measures discharge every 10 minutes at 2 points(southern and northern section close to Nodle Island), and calculates flow discharge using Chiu's velocity law(Chiu, 1988). In order to verify the results of automatic discharge measurements, manual discharge measurements were carried out by ADCP. In addition, the monthly discharge were also compared.

Calculating Expected Damage of Breakwater Using Artificial Neural Network for Wave Height Calculation (파고계산 인공신경망을 이용한 방파제 기대피해도 산정)

  • An approach to calculating expected damage of breakwater assisted by artificial neural network was developed. Wave height in front of a breakwater was predicted by a trained artificial neural network with inputs of wave height in deep ocean and tidal level. Prediction results by the neural network can be comparable to that by professional numerical model for wave transformation. Using the wave prediction neural network, it was very easy and fast to obtain a number of significant waves at breakwater and finally analysis time for expected damage can be shortened. In addition, the effect of considering tidal level in the calculation of expected damage was revealed by comparing the expected damages with and without tidal variation. Therefore, it was pointed out that tidal variation should be considered to improve prediction accuracy.
