Ectopic Thymus Presenting as a Submandibular Neck Mass (경부 종물로 발현된 이소성 흉선 1예)

  • Ectopic cervical thymic tissue is rarely reported in medical literature, but it should be included in the differential diagnosis of neck masses, especially in children. It occurs in the line of descent of the thymus from the angle of the mandible to the superior mediastinum. A preoperative diagnosis is seldom considered and is often misdiagnosed as a possible tumor or a lymph node. We present a case of a 2-month-old infant with an asymptomatic enlarging right neck mass. Patient underwent complete excision of the mass.

  • Experiments were performed on mice to investigate the effect of Panax ginseng petroleum ether fraction on the immunotoxicity of lead acetate. Lead acetate was administered in the drinking water and ginseng p-ether fraction was injected intraperitoneally, Mice were sensitized and challenged with sheep red blood cell. Humoral immune responses were evaluated by antibody production and Arthus reaction. Pathotoxicological influences were measured by serum protein, alkaline phosphatase and total cholesterol. The weight of liver, spleen and thymus were measured. Lead acetate exposure significantly decreased hemagglutination titer, hemolysin titer, Arthus reaction, spleen and thymus weight. Ginseng p-ether fraction administration significantly restored or potentiated reduced humoral immune response, spleen and thymus weight. Reduced serum A/G ratio, total cholesterol and alkaline phosphatase activity were restored or increased by ginseng p-ether fraction administration.

  • Apoptosis is now widely recognized as a common form of cell death and represents mechanism of cell clearance in many physiological situations where deletion of cells is required. In vivo administration of bacterial lipopolysaccharide (LPS) to Balb/c mice induced DNA fragmentation in the thymus. DNA fragmentation in the thymus was roughly dependent on the dose of LPS injected and reached the peak 18 hours after injection. This apoptosis in the thymus might be mediated due to LPS stimulant. DEN (diethylnitrosamine) has been shown to cause liver cancer in experimental animals and humans. The hepatic tumorigenesis was induced by ad libitum feeding of DEN only. It was suggested that DEN induced hepatic tumorgenesis in rat is a good reproducible model for studying biochemical and pathophysiological changes associated with the development of hepatic tumorigenesis and apoptosis.

  • On day 2 after treatment of Lewis rat with 25mg of hydrocortisone, the cell number in the thymus was reduced to less than 10% of the matched control. By day 12 after hydrocortisone treatment the thymic cell population was recovered almost its original value and on day 24 after treatment the number of thymocytes was equivalent to that of normal thymocytes. The density distribution profile of hydrocortisone treated rats as compared with normal rats showed a marked decrease in the denser fraction D while the lighter fractions. (A plus B, and C) showed a considerable proportional increase. The proportion of dead cells in thymus suspension from hydrocortisone treated rats was higher than that from their normal counterparts. On separation, the dead cells accumulated selectively in the pellet and A fraction. The response of the thymocytes from hydrocortisone treated rats to PHA was increased compared to that from normal rats. Among the subpopulations, D fraction, which was relatively unresponsive in normal rats, showed a marked increase in PHA response and C fraction showed some increase in the response.

  • The pathological aspects of purified ricin from the seeds of the castor oil plant, Ricinus communis, were examined, using light and transmission electron microscopy. ICR mice were exposed to ricin by peritoneal injection with 100 ng/1 $m\ell$ PBS(pH 7.0) mouse and histological observations on the liver, spleen, thymus, lung and heart were carried out at intervals up to 48 h after exposures. All the organs examined were damaged by ricin. Among the organs, the spleen and thymus; immune organs were the most sensitive to ricin, whereas the effect delayed in the liver, lung and heart. Furthermore, the immune cells in each organ were the most sensitive to ricin. Accordingly, the effect of ricin on the organs seems to be affected by the immune cells existed in each organ, In each organ, the immune cells showed apoptotic changes, while the capillary endothelial and parenchymal cells showed necrotic changes.

  • Quantitation of N7-methyldeoxyguanosine (N7-MedG) produced in the in vitro N-methly-N-nitrosuourea (NMU) action on thymus DNA has been achieved by enzymatic degradation, liquid chromatoraphic separaphic separation and desorption chemical ionization tandem mass spectrometry. In conjunction with the resolving power HPLC in the separation of isomers, desorprion chemical ionization tandem mass spectrometry has utilized in determining modified nucleosides at low levels using a stable-isotope labled compound as an internal reforence. The quantitative estimation of N7-methyldeoxyguanosine was previously established by an independent HPLC analysis of methylated calf thymus DNA. A sensitive and specific methodogy for the quantitation of N7-MedG at the picomole level using HPLC combined with tandem mass spectrometry without radioisotope labeling process is presented. The potential of the liquid chromatoraphic tandem mass exposure to methlation agents in vitro.

  • For evaluating the cytolytic effects on the mouse thymocytes, four typical antiinflammatory steroids (dexamethasone, triamcinolone acetonide, prednisolone, hydrocortisone) were selected in this study. When steroids were treated to the mouse thymocytes in vitro cytolysis occurred with dose-dependent fashion and the activities were found to be paralle with the known local anti-inflammatory activities. In vivo thymus atrophogenic activities appeared by the treatment of topical and subcutaneous applications of the derivatives were also found to dose-dependent, but not coincided with the thymocyte cytolytic activities in vitro and local anti-inflammaatory activity in the case of triamcinolone acetonide. Triamicinolone acetonide induced potent thymocyte cytolysis in vitro, but showed less thymus atrophy.

In Vitro Inhibitory Activities of Essential Oils from Two Korean Thymus species against Antibiotic-Resistant Pathogens

  • The in vitro inhibitory activities of essential oils from Thymus magnus and T. quinquecostatus as well as their main constituents were evaluated against susceptible and resistant species of Streptococcus pneumoniae, Staphylococcus aureus, Salmonella enteritidis, and Salmonella typhimurium. Notably, the essential oil fraction of T. magnus and its main components displayed significant inhibitory action against both antibiotic-susceptible and resistant strains of S. pneumoniae, S. aureus, and S. typhimurium with minimum inhibitory concentrations (MICs) ranging from 0.125 to 8 mg/mL. The differential MIC values imply that the oil fraction and its main components exhibit distinct patterns of activity against the tested bacterial species. Moreover, the disk diffusion test revealed that the inhibitory activities of oil fraction and components were dose-dependent. Data from the checkerboard titer test confirmed synergism between the antibiotic, norfloxacin, and T. magnus oil or thymol, particularly against the resistant strains of S. aureus.

Effects of Astragali Radix extract on the Cell Mediated Immunotoxicity of Zinc Chloride (염화아연의 세포성 면역독성에 미치는 황기 추출물의 효과)

  • Effects of Astragali Radix extract (AG) on the cell mediated-and nonlpecific immunotoxic responses of zinc chloride (Zn) were studied usign ICR mice. Mice were divided into 4 groups (10 mice/group), and Zn was given to the mice 1 hr after i.p. injection with 0.5g/kg of AG by i.p. injection daily for 10 days at a dose of 25 mg/kg. Immune responses on the responses on the relative weight of thymus, delayed-type hypersensitivity to SRBC (DTH), phagocytic activity and circulating leukocytes were evaluated. Zn treatment decreased body weight gain, the relative weight of thymus, DTH and circulating leukocytes compared with those in controls. AG treatment increased DTH, phagocytic activity and circulating leukocytes compared with those in controls. Combination of AG and Zn increased DTH and circulating leukocytes compared with those in controls, but decreased body weight gain and the relative weight of thymus. These findings indicated that AG decreased immunotoxicity of Zn on the DTH and circulating leukocytes.

  • We have introduced the nonresonant holes in the satellite hole spectrum to examine the interaction of two tautomers of thionin to various polynucleotides, including [d(GC)7]2 , [d(AT)7]2 and calf thymus. The saturated depths of nonresonant holes are much reduced upon interacting with [d(GC)7]2 and calf thymus than with [d(AT)7]2 , implying that the binding affinity of thionin to [d(GC)7]2 and calf thymus is stronger than [d(AT)7]2 . In addition, the less perturbation on the exocyclic amino group of the amino -form of thionin upon interacting with [d(AT)7]2 suggests that a direct hydrogen bond formed between the amino group of thionin and the phosphate of nucleic acids is unlikely.
