Factors Affecting the Motility and Fertility of Frozen-thawed Stallion Semen (말 동결융해 정자의 생존성 및 수정능에 영향을 미치는 인자)

  • The techniques for the collection, cooling and freezing of semen and artificial insemination of horses are not fully understood in Korea. We investigated the percentages of total motile (TM) and progressively motile (PM) sperms after the collection, cooling and freezing of stallion semen. The average volume of semen was 167 ml in Thoroughbred and 68 ml in Arab. The average numbers of spermatozoa in Thoroughbred and Arab were $104\times10^6/ml$ and $86\times10^6/ml$ respectively. The average percentages of TM and PM were 82.3% and 88.6% in Thoroughbred, and 61.4% and 82.6% in Arab, respectively. The average percentage of TM at 4 hr after cooling at $5^{\circ}C$ was significantly lower than that at 0 hr ($30.0\pm4.1%\;vs.\;78.0\pm2.5%,\;p<0.05$), but the percentage of PM was similar between 66.5 and 73.2% at 0, 1, and 4hr. The average percentage of frozen-thawed Thoroughbred semen frozen in MFR5 extender was 56.2%, which was significantly higher than that of the semen frozen in LE extender (average 32.9%, p<0.05). The percentage of TM in Arab was similar for semen frozen in MFR5 extender and LE extender (18.2% and 21.2%, respectively), but the percentage of PM was significantly higher in sperm frozen in MFR5 extender than in sperm frozen in LE extender (69.0% vs. 36.4%, p<0.05). Four mares were artificially inseminated by Thoroughbred frozen-thawed semen and one of them fertilized at 11 day after artificial insemination. In this study, the collection, cooling and freezing of equine semen were possible under domestic conditions.

Therapeutic Shoeing for Spontaneous Quarter Cracks Induced by Sheared Heel in Thoroughbred Race Horse (더러브렛 경주말에서 부등제종 기인 특발성 제측열제 처치술)

  • A 3-year-old colt Thoroughbred horse was referred with obvious lameness (3/5G) and mild heat and pain on left hoof of forelimb. He was diagnosed with quarter cracks that have a typical conformation of sheared heel, which are a different length and height between medial/lateral heels. Various materials and techniques were carried out to repair quarter cracks by using many different kinds of clips, bar shoes on fifth times for about 10 months. It worked on him without pain and heat on the cracked hoof, and then he could retrain for racing from the 36th weeks beginning of therapeutic shoeing. We suggested that especially fiberglass reinforced plastic (FRP) material and therapeutic shoeing were a great help to treat quarter cracks induced by sheared heel.

Characteristics and antimicrobial susceptibility of Escherichia coli isolated from horse (말에서 분리한 Escherichia coli의 특성 및 항생제 감수성)

  • This study was conducted to investigate characteristics and antimicrobial susceptibility of Escherichia (E.) coli isolates isolated from vaginal mucosa and clitorial fossa of 105 Thoroughbred mares suspicious of the genital disease in Korea during the period from March 2006 to July 2007. Ninety six E. coli isolates were identified as standard biochemical properties and using BIOLOG system. Fifty three isolates (55.2%) could be classified into a total of 21 O serotypes and forty three isolates (44.8%) were non-typeable with 51 O antisera used in this study. The verotoxin 1 (VT 1) and verotoxin 2 genes were analyzed by multiplex PCR. Among them, one isolate was detected VT 1 gene (130 bp). Most of isolates showed a high susceptibility in ciprofloxacin (100%), enrofloxacin (100%), norfloxacin (100%), cefoxitin (96.9%), gentamicin (96.9%), sulphamethoxazole (96.9%), nitrofurantoin (94.8%), amikacin (93.8%), nalidixic acid (92.7%) and tetracycline (90.6%). These results may provide the basic information to establish strategies for the treatment and prevention of reproductive disease in Thoroughbred mares in Korea.

  • A 11-year-old gelding Thoroughbred horse was presented with a history of unilateral nasal discharge of right side. The horse was diagnosed as maxillary sinusitis based on blood examination and radiography. The horse was treated with bone flap of right maxillary sinuses. Abscess of maxillary sinus was drained and irrigated with antibiotic-saline and povidone iodine solution. After surgical procedure, nebulization and medication applied by the result of antibiotic sensitivity test. After treatment for two weeks, the patient was recovered completely.

  • The present study demonstrates a new approach that enables effective horse parentage testing using 22 ISAG microsatellite markers involving 6 heads of Thoroughbred horse(TB) and non-TB. In the comparison allele distribution between these horses, the alleles found in the TB were numerously detected in the non-TB. As results, we confirmed that these ISAG microsatellite markers might apply the pedigree registration of Korean native horse(Jeju horse).

Outbreaks of Strangles in Thoroughbred Horses of Jeju (제주지역의 더러브렛 말에서 선역의 발병)

  • This work describes an outbreak of strangles due to Streptococcus equi subsp. equi in 1 to 2 years old Thoroughbred horses. A total of 7 samples were collected from 5 horses in two different horse farms during 2003. Six ${\beta}$-hemolytic colonies selected from each sample were identified by biochemical tests using API STREPTO followed by PCR amplification which is able to recognize unique region of SeM gene of S. equi subsp. equi. All colonies selected from the ruptured lymphadenitis of 2 horses in farm A were identified as S. dysgalactiae subsp. equisimillis. It seems to be secondary infection because the sampling sites have been already opened and the horses have been treated with antibiotics for a week. All colonies from 2 submandibular lymphadenitis samples in Farm B were S. equi subsp. equi while the isolates from 3 nasal discharges in this farm were mixed with S. equi subsp. zooepidemicus.

Intussusception of the uterine horn associated with dystocia in a Thoroughbred broodmare (Thoroughbred 씨암말에서 난산과 연관된 자궁각 중첩 예)

  • Horses that have been retired from racing or imported from abroad are retired as broodmares. Whether at private farms or government institutes they are bred to stallions with the aim of improving fertility and enhancing the breed. Accidental deaths as a result of surgical or obstetrical complications lead to decreased productivity and economic losses to the horse breeding industry. Intussusception of the uterine horn is a frequent complication of the equine and bovine species, but rarely seen in other species. The most common causes are thought to be tearing of the placenta which is suspended from the uterus and ovaries and the weight of the placenta. Expulsion of the embryonic membranes is also thought to be a contributing factor. If symptoms are minor and treatment affected quickly, the problem will likely resolve itself. In more severe cases the uterus will prolapse completely. This case study was observed in a Thoroughbred broodmare. After treatment of dystocia, the broodmare could not stand up. She became dehydrated, collapsed, suffered colic and was euthanised. In this case was observed serious swelling vulva and perineal region by morphology, and there was seldom ascites and unusual view in thoracic and abdominal cavity after opening abdomen. However, intussusception of uterine horn involuted left uterine horn into the uterine body was observed.

Evaluation on Blood Chemistry in a Thoroughbred Neonate (1-day old) with Congenital Torticollis (선천성 사경을 동반한 더러브렛 신생망아지의 혈액화학 평가)

  • There have been no reports on the laboratory screening of congenital torticollis in equine medicine. The purpose of this study was to evaluate the blood chemistry of a newborn female foal with congenital torticollis. The filly was born after a normal parturition period, and the clinical pathology results were as follows: biochemistry of the plasma showed significantly higher levels of CK, LDH, and Na than those in a normal foal. However, Cl level was remarkably lower than that of the control. In conclusion, the cells in the neck muscles influenced the clinical pathological value in the neonate. A disorder of the muscles as well as the bone problem, such as the cervical vertebrae, may have caused congenital torticollis. To our knowledge, this is the first report detailing the clinical pathology results in a newborn foal with torticollis. These laboratory results can be used as a reference for interpretation of pathology results in foals.

Development of Sequence Characterized Amplified Regions (SCAR) Showing for Cheju Native Horse (품종 특이성을 이용한 제주마 판별 표지인자 재발)

  • This study was conducted to analyze genetic characteristics and to develop the specific marker for Cheju native horse (Coo) at the level of sequence characterized amplified regions (SCARs). We collected blood samples from Cheju native horse and Thoroughbred horse (Th) and obtained genomic DNA from the blood of 50 individuals randomly selected within the breeds. Seven hundred primers were chosen randomly and were used to examin the polymorphism and 40 kinds of primers showed polymorphic RAPD band patterns between two breeds. Thirty primers of them showed horse specific bands. With the primer MG 30, amplified band of 2.0 kb showed the specificity to Cheju native horse (Cnh). Additionally MG 53 detected the thoroughbred horse (Th) specific markers at size of 2.3 kb. As the next, 2.3 kb band from MG 53 was checked with the all individuals from all the breeds of this study, and it maintained the reproducible breed specificity to thoroughbred horse (Th). With this results, 2.3 kb band was cloned into plasmid vector and sequenced bidirectionally from both ends of the cloned fragment. With the obtained sequences 10 nucleotide extended primers including the original arbitray primer were designed as a SCARs primer. Finally, the primer with extended sequence showed the reproducible breed differentiation pattern and it was possible to identify Cheju native horse (Cnh) from other breeds. The SCARs marker 2.3 kb from MG 53 could be used to identify Cheju native horse (Cnh) for not only registration but also horse breeding programe.

Investigation of Antibody Titers after Inoculation with Commercial Equine Influenza Vaccines in Thoroughbred Yearlings (Thoroughbred 1세말에서 상업용 말 인플루엔자 백신접종 후 항체역가 추적)

  • The object of this study was to evaluate the change of antibody titers on virus strains after inoculation with commercial killed equine influenza (EI) vaccines in horses. Serum antibodies of 20 Thoroughbred yearlings were detected using hemagglutination inhibition test for 41 weeks. Second vaccination is inoculated 4 weeks after the initial vaccination. Most of antibody titers were not increased until 4 weeks after first vaccination. The highest titers were detected 6-10 weeks after vaccination. The titers were decreased slowly and maintained for 16 weeks after inoculation. We could barely detect the antibody 41 weeks after vaccination in most cases. Vaccine anergia were appeared in 3 horses (15%) but it depended on virus strains. A/Equine/La Plata/93(H3N8) strain that induce high and durable antibody responses was the most effective among three strains. This study presents the first comprehensive data on the endurance of antibody titers against EI. Our data also suggests that yearlings should be inoculated three times in order to maintaining optimal antibody titers against EI. We speculate the causes of anergia were vaccine break down or individual specificity. Further research is needed to investigate immunological unresponsiveness. This was the first study on strain of equine vaccine in Korea.
