Antioxidative Activity of Korean Wild Leaf Vegetables : Pleurospemum Kamtschaticum, Aderophora Remotiflor and Aster Glheni

  • The purpose of this study was to evaluate the antioxidative potential of Korean wild leaf vegetables in vivo as well as in vitro. The antloxidative activities of Pleurospemum kamtschaticum, Aderophora remotiflor and Aster gfheni were evaluated as a reference for Spinacia oleracea. Fifty rats were find either a control diet or one of several vegetable diets for 4 weeks. The level of thiobarbituric acid reactive substance (TBARS) and the activity of catalase and superoxide dismutase (SOD) in the liver were compared. The levels of total cholesterol (TC), high density lipoprotein cholesterol (HDL-C) and triglyceride (TG) in plasma were also compared. Also, the contents of $\beta$-carotene, vitamin C, vitamin E and total phenolic compound, including flavonoid and thiobarbituric acid (TBA) value using linoleic acid model system, were measured in Korean wind leaf vegetables. The TBARS values of Aderophora remotiflor and Aster giheni tended to be lower than that of Spinacia olerncea. The SOD activity of Artrr glheffi was significantly higher than that of Spinacia oleracea and reached 265% that of the control animals, whereas there was no difference between the control animals and the vegetable diet animals in terms of catalase activity. Compared to the control animals, TG and TC levels were significantly solver and showed a lower artheriosclerotic index. TBA values of Pleurospemum Kamschaticum and Aderophora remptiflor were only 18% of control value. TBA value of Spinacia olerucra was 41% of control value. These results suggest that Aderophora remotinor and Aster glheni could have antioxidative potency in vivo as well as in vitro and potential value far functional flood to improve the plasma lipid profile. flavonoid and phenolic compounds could be the major contributing factor in the antioxidative potential of Aderophora remotiflor and Aster glheni.

Meat Color, TBARS and VBN Changes of Vacuum Packaged Korean Pork Loins far Export during Cold Storage (수출용 진공포장 냉장 돈육 등심의 육색, TBARS 및 VBN 변화)

  • The Korean fresh pork loins in vacuum packaged were obtained from three different Korean export companies and investigated for meat color, TBARS(thiobarbituric acid reactive substance) and VBN(volatile basic nitrogen). The fresh pork loins were stored at 2$^{\circ}C$ for 50 days and analyzed with an interval of 5~10 days. The L* values of meat from the company II and III were relatively higher than those from the company I. The L* values of loins from the companies II and III were increased until 35 days of storage and decreased after that period. The TBARS values were increased as the storage time increased for meat from all companies. Although the TBARS values of all loins were similar at the beginning stage of storage, those from the companies II and III were higher than loins from the company I after 40 days of storage. There were no significant difference in VBN among loins from three companies during the storage.

Effects of Dietary Supplemental Activated Carbon and Sardine Oil on the VBN, TBARS and Fatty Acid of Chicken Meat (활성탄과 정어리유의 급여가 계육의 VBN, TBARS 및 지방산 조성에 미치는 영향)

  • This study was conducted to investigate the influence of dietary activated carbon(0.9%) and sardine oil (0, 1, 2, 4%) on volatile basic nitrogen(VBN), thiobarbituric acid reactive substance(TBARS), and fatty acid in meat sample of chicken. Broilers were randomly assigned to one of the file dietary treatments: 1) Control(commercial feed) 2) T1(commercial feed supplemented with 0.9% activated carbon) 3) T2(commercial feed with 0.9% activated carbon and 1% sardine oil) 4) T3(commercial feed with 0.9% activated carbon and 2% sardine oil) 4) T4(commercial feed with 0.9% activated carbon and 4% sardine oil). They were fed one of the experimental diets for five weeks and slaughtered. After that, the meat samples were stored over a period of 0, 1, 3, 7 and 10 days at 4${\pm}$1$^{\circ}C$. The VBN of all treatments significantly increased during the storage periods(p<0.05). Also, the VBN was not significantly between control and treatment group. In VBN of breast was higher compared with that of thigh. The TBARS of all treatments were significantly increased as storage period extended (p<0.05). The TBARS of control and thigh tended to be higher than that of treatments and breast. Oleic acid, palmitic acid, linoleic acid, stearic acid were major fatty acid in chicken meat. Saturated fatty acid decreased and unsaturated fatty acid increased of all treatments during storage. Oleic acid, EPA, DHA contents was higher in treatment group than the control.

Antioxidant Activity of Soybean Extracts by Mixed Solvents Ethanol-Water and Methanol-Water (에탄올-물과 메탄올-물 혼합 용매를 이용한 대두 추출물의 항산화 효과)

  • The effective extraction of antioxidative substances from soybeans was studied by the mixed solvents of methanol or ethanol with water at 30$^{\circ}C$ and 85$^{\circ}C$. At accelerated peroxidation condition, POV (peroxide value) of soybean oil and TBARS (thiobarbituric acid reactive substances) value of egg yolk liposome with extracts added, were applied for the evaluation of antioxidative activity of the extracts. In the peroxidation of soybean oil, antioxidative substance of soybeans was extracted more effectively by methanol and at higher temperature 85$^{\circ}C$, and the deffated and roasted soybean extract (DRS) showed higher antioxidative activity. But in the Peroxidation of egg yolk liposome, methanol extract from the roasted soybeans (ROS) showed higher activity.

Effects of Medicinal Plant Extracts on Antioxidant System in Ethanol-intoxicated Rats

  • Four medicinal plants selected from preliminary screening study were evaluated in the aspects of their antioxidant activities in alcohol-intoxicated rats. Rats fed 1% ${\alpha}-tocopherol-supplemented$ diet as positive control and ones done ${\alpha}-tocopherol-deficient$ diet as negative control were compared with ones done the plant extract-supplemented diet (n=8). After the administration of the experimental diets for 4 weeks, typical increments in activities of manganese-superoxide dismutase (Mn-SOD) and glutathione peroxidase (GSH-px) indicated in alcohol-intoxicated rats, were not observed in ones fed Lagerstroemia and Ulmus extract-supplemented diet. The content of thiobarbituric acid reactive substance (TBARS), the product of lipid peroxidation, did not increased in rats fed plant extracts-supplemented diet except for Terminalia. From the results, it is concluded that Lagerstroemia and Ulmus have physiologically efficient antioxidant activities.

In vivo Antioxidative Characteristics of Extracts from the Aromatic Herb Elsholtzia splendens

  • The antioxidative properties of an Elsholtzia splendens ethanol extract (ESE) were examined in vivo. Oral administration of 10 or 50 mg ESE/kg BW in mice for 50 days resulted in a dose-dependent decrease in several biomarkers of oxidative stress including thiobarbituric acid reactive substance (TBARS), protein carbonyls, and serum 8-hydroxy-2'-deoxy guanosine (8-OH-dG). Moreover, the level of activity and mRNA expression of catalase and superoxide dismutase (SOD) were significantly increased by ESE treatment. Taken together, these results indicate that ESE may be beneficial to human health via its antioxidative properties.

Microbiological and Oxidative Stability of Low Fat Ground Beef during Refrigeration (취반 재고미를 첨가하여 제조한 저지방 분쇄우육의 냉중중 안정성)

  • Four low fat ground beef groups containing 10% fat plus 0, 5, 10 and 20% additional cooked lod rice and a control ground beef containing 30% fat were prepared and the analysis for microbiological and oxidative stability were conducted. During 6 days of storage at 4$^{\circ}C$ microbial analysis including total plate count and coliform groups were performed and 2-thiobarbituric acid reactive substance (TBARS) absorbances were measured. The growth rate of total aerobic bacteria and coliform groups tended to increase with the increase in fat content and the amount of added cooked lod rice. Development of oxidative rancidity were not significantly different between 10 and 30% fat ground beef but among the 10% ground beef the rancidity development significantly(p<05) decreased with the in-crease in the amount of added cooked old rice. Low fat ground beef groups were not stable over 3 days during storage at 4$^{\circ}C$.

Inhibitory Effect of Euscaphic Acid and Tormentic Acid from the Roots of Rosa rugosa on High Fat Diet-Induced Obesity in the Rat (고지방 식이로 유도된 비만 흰쥐에서 해당근에서 분리된 Euscaphic Acid 및 Tormentic Acid의 효과)

  • The roots of Rosa rugosa have been used to treat diabetes mellitus in the folkloric society of Korea. To demonstrate the active component for the rat obesity induced by high fat diet for 6 weeks, the phytochemical fractionation and the pharmacological activity test were performed on this crude drug. It was shown that the methanolic extract and its EtOAc fraction inhibited the weight increase of the rat body, abdominal fat pad and hyperlipidemia at 200 mg/kg dose. Further, the triterpenoids, euscaphic acid and tormentic acid, isolated from R. rugosa roots were active at 30 mg/kg in the same assay. The two components shifted serum total-, HDL, and LDL-cholesterol levels toward the values of the unteated group, suggesting that the active compounds has hypolipidemic effects. The rats fad euscaphic acid and tormentic acid also reduced thiobarbituric acid-reactive substance (TBARS) and hydroxyl radical in the rat blood and increased superoxide dismutase activity compared to the control. TBARS values and carbonyl contest of the hepatic protein were reduced by treatment with the two triterpenoids. Antioxidative enzyme (SOD, glutathione peroxidase, and catalase) activities in hepatic were increased by treatment of rats with the triterpenoids, which suggests that triterpenoids inhibited the reduction of hepatic antioxidative activity caused by high fat diet. Taken together, these results support that euscaphic acid and tormentic acid improve a high fat diet-induced hyperlipidemia via the activation of antioxidative mechanism.

Effect of Stem Bark Extract from Cudrania tricuspidata on the Concentrations of Lipid and Lipid Peroxidation in Rats Fed a Cholesterol Diet. (꾸지뽕나무 수피 추출물의 콜레스테롤 급여 흰쥐의 지질 농도 및 과산화지질 농도에 미치는 영향)

  • Male Sprageu-Dawely rats fed a cholesterol diet (Control group) or cholesterol diet supplemented with the water-soluble extract of stem bark from Cudrania tricuspidata(CTSB group) at the level of 0.5% for 2 weeks. Concentration of triglyceride in serum of CTSB group was significantly lower than that of control group. However, the other lipids concentrations of liver and serum, body weight gain, and food intake did not show significant difference between CTSB group and control group. Antioxidative activities of water-soluble extract from stem bark of Cudrania tricuspidata on the lipid peroxidation in rat were also studied in vivo by measuring the formation of thiobarbituric acid reactive substance(TBARS). Concentration of TBATS in the liver homogenates of CTSB group was significantly higher than that in control group. Nonheme iron concentration was significantly increased in the liver of CTSB group compared to control group. suggested that enhanced nonheme irom was associated with enhanced peroxidation of liver in CTSB group. These results suggested that water soluble extract from stem bark of Cudrania tricuspidata exert the hypotriglycerolemic effect and might amplify the lipid peroxidation of tissues in cholesterol fed rats.

Effect of Dried Powder and Juice of Aster Scaber on Lipid Metabolism and Antioxidative Capacity in Rats (참취의 건분 및 녹즙이 흰쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder.
