Temperature dependent hydrogen exchange study of DNA duplexes containing binding sites for Arabidopsis TCP transcription factors

  • The TCP domain is a DNA-binding domain present in plant transcription factors and plays important roles in various biological functions. The hydrogen exchange rate constants of the imino protons were determined for the three DNA duplexes containing the DNA-binding sites for the TCP11, TCP15, and TCP20 transcription factors using NMR spectroscopy. The M11 duplex displays unique hydrogen exchange property of the five base pairs in the first binding site (5'-GTGGG-3'). However, the M15 and M20 duplexes lead to clear changes in thermal stabilities of these five base pairs. The unique dynamic features of the five base pairs in the first binding site might play crucial roles in the sequence-specific DNA binding of the class I TCP transcription factors.

Pervaporation Characteristics of Water/Ethanol Mixture through Hydrophilic Zeolite Membranes (친수성 제올라이트 분리막의 물/에탄올 투과증발 특성)

  • Zeolite membranes show better thermal, mechanical, chemical stabilities than polymer membranes. Water was separated from water/ethanol mixtures by pervaporation using the NaA and the NaY zeolite membranes synthesized in our laboratory. The effects of concentration of ethanol at the feed side and temperature were studied on the permeation flux and separation factor of water. The separation factors obtained with the NaA zeolite membrane was found to be 1000 times higher than those obtained with the NaY. However, the water flux through the NaA zeolite membrane was observed to be lower than 1/2 of that through the NaY zeolite membrane.

Cure Behaviors and Mechanical Interfacial Properties of Epoxy/Polyurethane Blends (에폭시/우레탄 블렌드의 경화거동과 기계적 계면특성에 관한 연구)

  • In this work, the blend of diglycidylether of bisphenol A (DGEBA) and modified polyurethane (PU) was prepared and characterized in the cure behaviors and mechanical interfacial properties. The N-benzylpyrazinium hexafluoroantimonate was used as a cationic initiator for cure, and the content of PU was varied within 0-20 phr. The cure behaviors and mechanical interfacial properties were studied by DSC, near­IR, and the critical stress intensity actor $(K_{IC})$ measurements. Also thermal stabilities were carried out by TMA and TGA analyses. As a result, the cure activation energy $(E_a)$ and the conversion $(\alpha)$ were slightly increased with increasing the PU content, and a maximum value was found at 10 phr PU. The mechanical interfacial properties measured from $K_{IC}$ showed a similar behaviors with the results of conversion. These results were probably due to the increase of the hydrogen bonding between the hydroxyl groups of DGEBA and isocyanate groups in PU.

Copolymerization of 2-Hydroxyethyl Methacrylate with Functional Silane Monomers (2-히드록시에틸 메타크릴레이트와 기능성 사이레인 단량체와의 공중합에 관한 연구)

  • 2-Hydroxyethyl methacrylate(HEMA) was copolymerized with allyltrimethylsilane- (ATMS) and allylt.imethoxysilane(ATMOS) at 70。C in N, N'dimethylformamide- (DMV) using $\alpha$ , $\alpha$'-azobisisobutyronitrile(AIBN). The compositions of unreacted monomers were determined by gas chromatographic analysis. The monomer reactivity ratios were determined by Fireman-Ross, Helen-Tiidus and intersection methods. The average values are as follows : $$r_1(HEMA) : 6.62 $\pm$ 0.07, r_2(ATMS) : 0.07 $\pm$ 0.01 for HEMA-ATMS system.$$ $$r_l(HEMA) : 4.09 $\pm$ 0.14, r_2(ATMOS) : 0.06 $\pm$ 0.01 for HEMA-ATMOS toys tem.$$ The lower rl(HEMA) In the HEMA-ATMOS system as compared to HEMA ATMS system may be contributed to higher relative reactivity of ATMOS toward the poly(HEMA) radical. The compositions of synthesized copolymers were determined from silicon con tents estimated gravimetrically. The thermal stabilities of the copolymers were investigated by thermogravimetry(TG). The enthalpic changes associated with the endothermic transition were evaluated by differential scanning calorimetry(DSC). The swelling properties of the copolymers in water were also investigated.

  • 3-Chromonealdehyde(2,2-disubstituted)hydrazone derivatives were synthesized by dehydration condensation. They are green-emitting materials for organic light emitting device (OLED) composed of electron acceptor of 3-chromonealdehydes and electron donor of 2,2-disubstituted hydrazones by a conjugated structure. The structural properties of reaction products were analyzed FT-IR and $^1H-NMR$ spectroscopy. The thermal stabilities and reactivities were measured by melting points and yields. The UV-visibles and PL properties can be determined by excitation spectra and emission spectra, respectively.

  • Biocomposites were fabricated with biodegradable polymers and natural fibers. Biocomposites have benefits of low cost, low density, and biodegradability over inorganic fiber composite, and give comparable strength properties. Hydrophobic polymer used for sizing in paper industry, AKD (Akenyl Keten Dimer), was applied to natural fibers, red algae fibers (RAF) in this study, to make fiber surfaces more compatible to hydrophobic nature of matrix polymers. Composites with RAF, kenaf, glass fibers, and carbon fibers have been fabricated by a compression molding method and their thermo-mechanical properties have been studied. Also, the thermal dimensional stability test was done from at 30 to $100^{\circ}C$. The storage moduli and the thermo-mechanical stabilities of polypropylene and poly lactic acid based biocomposites were improved by reinforcing with the RAF and much more with AKD treated fibers. Dimensional stability of biocomposite was also markedly improved by AKD pretrement on RAF.

  • In this study, analysis of the disposal tunnel spacing and disposal pit pitch was carried out, as a factor of the design to estimate the scale and layout of the repository To do this, based on the reference repository concept and the engineered barrier concept, the cross section of the disposal tunnel and disposal pit are established and the mechanical and thermal stabilities of the tunnels are analyzed. Also, the optimized disposal tunnel spacing and the disposal pit pitch which minimize the excavation volume was proposed. The detailed analyses by the exact site characteristics data are needed to reduce the uncertainty of the site in the future.

  • 4-(Dicyanomethylene)-2-methyl-6-(4-dimethylaminostyryl)-4H-pyran (DCM) derivatives were synthesized by Knoevenagel condensation. They are red-emitting materials for OLED (Organic Light-Emitting Diode) composed of electron donor of aminostyryl groups and electron acceptor of two cyano(nitrile)groups in a conjugated structure. The structural properties of reaction products were analyzed by FT-IR and $^1H-NMR$ spectroscopy. The thermal stabilities and reactivities were measured by melting points and yields. The UV-visible and PL properties can be determined by exitation and emission spectra, respectively.

Cyclopolymerization of 1,1-Dipropargyl-1-silacyclohexane by Transition Metal Catalysts

  • A conjugated spirocyclic polymer was synthesized via the cyclopolymerization of 1,1-dipropargyl-1- silacyclohexane with various transition metal catalysts. The monomer, 1,1-dipropargyl-1-silacyclohexane was synthesized by Grignard reaction of 1,1-dichloro-1-silacyclohexane with propargyl magnesium bromide. This polymerization proceeded well to give the corresponding poly(1,1-dipropargyl-1-silacyclohexane). The catalytic activity of WCl6 was found to be similar with that of MoCl5. The structure of polymer having the conjugated backbone with silacyclohexane moieties was characterized by such instrumental methods as NMR (1H-, 13C-), IR, and UV-visible spectroscopies. The resulting polymers were mostly yellow or light-brown powders, depending on the catalyst systems used. This polymer was completely soluble in halogenated and aromatic hydrocarbons such as chloroform, 1,2-dichloromethane, benzene, toluene, and chlorobenzene, etc. The thermal and oxidative stabilities of polymer were also studied and discussed.

  • An agar-degrading Agarivorans sp. AG17 strain was isolated from the red seaweed Grateloupia filicina collected from Jeju Island. A beta-agarase gene from Agarivorans sp. AG17 was cloned and designated as agrA. agrA has a 2,985 bp coding region encoding 995 amino acids and was classified into the glycoside hydrolase family (GHF)-50. Predicted molecular mass of the mature protein was 105 kDa. His-tagged agrA was overexpressed in Escherichia coli and purified as a fusion protein. The enzyme showed 158.8 unit/mg specific activity (optimum temperature at $65^{\circ}C$ and pH 5.5 in acetate buffer) with unique biochemical properties (high thermal and pH stabilities). Enzyme produced neoagarohexaose, neoagarotetraose and neoagarobiose by degrading agar, and hydrolyzed neoagaro-oligosaccharides were biologically active. Hence the purified enzyme has potential for use in industrial applications such as the development of cosmetics and pharmaceuticals.
