반고형 이유식의 개발을 위한 변형 쌀가루 제조 및 이화학적 특성 I - 열-효소 처리한 쌀가루 (Physicochemical Properties of Modified Rice Powder for Rice-Based Infant Foods - Thermal-enzymatic Treatment on Rice Powder)

  • The establishment of optimal producing condition for rice-based infant foods using modified rice powder was attempted. The modification of rice powder was prepared by microwave heating as well as partial enzymatic hydrolysis. Not only thermal treatment but also enzymatic hydrolysis on rice powder increased D.E. value from 1.25 to 3.81. The water binding capacity, swelling power, solubility, digestibility by ${\alpha}-amylase$, light transparency, and paste clarity of the modified rice powder compared to unmodified rice powder were increased from 107% to 249%, from 7.80 to 42.52, from 0.04% to 0.89ft, from 9.19% to 23.01, from 33% to 42%, and from 2.2% to 3.9%, respectively. On the other hand, gelatinization temperature, apparent viscosity, and degree of retrogradation of modified rice powder showed negative correlation with D.E. value. The 「results suggested that the thermal and enzymatic treatment on rice powder improved the physicochemical properties of rice based infant food by enhancing carbohydrate absorptionability and lowering the viscosity and opacity.

Gelatinization and retrogradation characteristics of Korean rice cake in the presence of citric acid

  • The effect of citric acid on rice starch gelatinization and low-temperature (4 ℃) storage was studied in order to produce rice cake with a lower retrogradation rate. A citric acid solution in the ratio of 0, 0.5, 1.0, and 1.5% (w/w) of the water used during production was utilized. The gelatinization properties, gel strength, thermal properties, and texture analysis were evaluated to determine the retrogradation rate. The result showed that acid hydrolysis occurred in samples treated with citric acid. Thus, increasing citric acid decreased gelatinization temperature (58.63±1.98 to 45.84±1.24 ℃). The moduli of elasticity increased with increasing citric acid concentration, indicating an increased gel strength. Thermal analysis of starch showed that the onset, peak, and conclusion temperatures of retrogradation were increased significantly with the storage period and decreased with citric acid concentration. After 72 h of low-temperature storage (4 ℃), the retrogradation rate was lowest in the rice cake with 1.5% citric acid solution, with an increased ratio of 12.01 to 13.60% compared to the control sample, with a ratio of 12.99 to 43.54%. This shows a high retrogradation rate in the control sample. Additionally, sensory properties and retrogradation ratio suggest that the addition of 1.0% citric acid solution during rice cake production is efficient in retarding the retrogradation without an adverse effect on the rice cake modeling and acceptance.

  • Hydrotalcite, layered double hydroxides (LDH), with hexagonal morphology has been rapidly synthesized by microwave reaction within 1 hour by urea hydrolysis from homogeneous solution. Different synthesis parameters, Mg/Al molar ratio, microwave reaction temperature and microwave power were systematically investigated. Pure hydrotalcite phase was obtained for Mg/Al ratios of 2:1 and 3:1, and higher reaction temperature gave higher crystallinity. The hydrotalcite synthesized at 600W power shows the highest crystallinity and more homogeneous crystal size distribution. The hydrotalcite samples were characterized by powder X-ray diffraction (XRD), simultaneous thermogravimetric/differential thermal analysis (TG/DTA), Fourier Transform Infrared (FT-IR) and Scanning electron micrograph (SEM).

Biochemical and Thermal Stabilization Parameters of Polygalacturonase from Erwinia carotovora subsp. carotovora BR1

  • With an emphasis on its thermal behavior with different pHs and salts, the kinetic and thermodynamic parameters of the purified polygalacturonase (PG) from E. carotovora subsp. carotovora (Ecc) BR1 were studied, as the characterization of an enzyme is significant in the context of burgeoning biotechnological applications. The thermodynamic parameters for polygalacturonic acid hydrolysis by the purified PG were ${\Delta}H^*$=7.98 kJ/mol, ${\Delta}G^*$=68.86 kJ/mol, ${\Delta}S^*$=-194.48 J/mol/K, ${\Delta}G_{E-S}$=-1.04 kJ/mol, and ${\Delta}G_{E-T}$=-8.96 kJ/mol. In addition, its turnover number ($k_{cat}$) was 21/sec. The purified PG was stable within a temperature range of $20-50^{\circ}C$ and was deactivated at $60^{\circ}C$ and $70^{\circ}C$. The thermodynamic parameters (${\Delta}H^*$, ${\Delta}G^*$, ${\Delta}S^*$) for the irreversible inactivation of the PG at different temperatures ($30-60^{\circ}C$) were determined, where the effectiveness of various salts and different pHs (4-8) for the thermal stability of the PG were also characterized. The efficacy of various salts for the thermal stability of the PG was in the following order: $MgCl_2$ > $BaCl_2$ > KCl > $CaCl_2$ >NaCl. Therefore, the present work presents the biochemical, substrate hydrolysis thermodynamics and the thermal stabilization parameters of the PG from Ecc.

폐식용유로 제조된 재생비누의 생분해 속도 (Biodegradation Rate of Recycling Soap Prepared from Non-Cooking Oils)

  • A recycling soap was prepared from non-cooking oils. The effects of physlcal and chemical properties of the recycling soap on biodegradation are expected to be different due to the thermal histories of the non-cooking oils. Therefore, the biodegradation rate of the recycling soap was studied by using Klebssella Pneumoniae(K. pneumoniae), and the growth rate of K. pnewoniae in soap solution was observed. The biodegradation rate of the recycling soap appeared to be slower as the thermal histories of the non-cooking oils became larger. This might be resulted from hydrolysis, in which the ester bonds in the oils are broken to produce hydroxyl group. It was also observed that the growth rate of the microorganism decreased with the increase in the thermal histories of the oils. As a result, it is desired that recycling soap should be produced from the non-cooking oils with the prober ranges of thermal histories to reduce water contamination. The non-cooking oils with larger thermal histories are considered to be recycling through the cracking process before used. 화염 가수분해 증착공정에서 온도 및 OH 분포측정 (Measurements of Temperature and OH Radical Distributions in Flame Hydrolysis Deposition Process)

  • The effects of SiCl$_4$addition on flame structures have been studied in flame hydrolysis deposition (FHD) processes using Coherent anti-Stokes Raman spectroscopy (CARS) and planar laser induced fluorescence (PLIF) to measure temperatures and OH concentrations, respectively. The results demonstrate that even a small amount of SiCl$_4$ addition can change thermal and chemical structures of H$_2$/O$_2$ diffusion flames. When SiCl$_4$ is added to a flame temperature decreases in non-reacting zone due to the increases in both specific heat and density of the gas mixture, while flame temperature increase in particle formation zone due to the heat release through hydrolysis and oxidation reactions of SiCl$_4$. It is also found that OH concentration decreases dramatically in particle formation zone where temperatures increase. This can be attributed to consumption of oxidative species and generation of HCl during silica formation.

열가수분해 전처리가 양돈 슬러지의 메탄생산퍼텐셜에 미치는 영향 (Effect of the Pretreatment by Thermal Hydrolysis on Biochemical Methane Potential of Piggery Sludge)

  • 본 연구는 양돈슬러리의 혐기소화 효율 증진을 위하여 양돈슬러리를 고액분리 하고 이때 발생하는 슬러지케이크를 200, 220, 250, $270^{\circ}C$에서 각각 열가수분해 전처리하여 열가수분해 온도별 유기물의 가용화 효율과 혐기적 메탄생산 퍼텐셜을 분석하였다. 최종메탄생산퍼텐셜 ($B_u$)은 서로 다른 S/I 비율 (1:9, 3:7, 5:5, 7:3의 부피비)에서 73일간 혐기배양하여 구하였다. 양돈슬러리의 유기물 가용화율 ($S_{COD}$)은 $200{\sim}270^{\circ}C$ 열가수분해 반응에서 98.4~98.9%를 보였으며, 열가수분해액의 이론적 메탄생산퍼텐셜 ($B_{th}$)은 반응온도의 증가와 함께 증가하여 $200^{\circ}C$, $220^{\circ}C$, $250^{\circ}C$, $270^{\circ}C$에서 각각 0.631, 0.634, 0.705, $0.748Nm^3\;kg^{-1}-VS_{added}$로 나타났다. 열가수분해액의 최종메탄생산퍼텐셜 ($B_u$)은 $200^{\circ}C$의 열가수분해액에서 S/I 비율이 1:9에서 7:3으로 증가할수록 $0.197Nm^3\;kg^{-1}-VS_{added}$에서 $0.111Nm^3\;kg^{-1}-VS_{added}$로 감소하는 경향이 나타났으며, 다른 열가수분해 반응 온도 ($220^{\circ}C$, $250^{\circ}C$, $270^{\circ}C$)에서도 $200^{\circ}C$의 열가수분해액과 동일한 경향의 최종메탄생산퍼텐셜을 나타내었다. 유기물의 혐기적 분해율 ($B_u/B_{th}$)을 보면, $200^{\circ}C$ 열가수분해액은 S/I비율이 증가함에 따라 31.2%에서 17.6%까지 감소하였으며, $220^{\circ}C$, $250^{\circ}C$, $270^{\circ}C$의 열가수분해액에서 각각 36.4%에서 9.6%, 31.3%에서 0.8%, 26.6%에서 0.8%로 감소하는 것으로 나타나, 열가수분해 온도의 상승에 따라 유기물의 혐기적 분해능이 낮아졌다. 이러한 결과는 98% 대의 유기물 가용화율 ($S_{COD}$)을 보인 것과는 반대로 $250{\sim}270^{\circ}C$의 열가수분해액은 혐기소화에 분해저항성을 지니는 것으로 나타났다.

Optimization of Subcritical Water Hydrolysis of Rutin into Isoquercetin and Quercetin

  • Maximum production of isoquercetin and quercetin simultaneously from rutin by subcritical water hydrolysis (SWH) was optimized using the response surface methodology. Hydrolysis parameters such as temperature, time, and $CO_2$ pressure were selected as independent variables, and isoquercetin and quercetin yields were selected as dependent variables. The regression models of the yield of isoquercetin and quercetin were valid due to the high F-value and low P-value. Furthermore, the high regression coefficient indicated that the polynomial model equation provides a good approximation of experimental results. In maximum production of isoquercetin from rutin, the hydrolysis temperature was the major factor, and the temperature or time can be lower if the $CO_2$ pressure was increased high enough, thereby preventing the degradation of isoquercetin into quercetin. The yield of quercetin was considerably influenced by temperature instead of time and $CO_2$ pressure. The optimal condition for maximum production of isoquercetin and quercetin simultaneously was temperature of $171.4^{\circ}C$, time of 10.0 min, and $CO_2$ pressure of 11.0 MPa, where the predicted maximum yields of isoquercetin and quercetin were 13.7% and 53.3%, respectively. Hydrolysis temperature, time, and $CO_2$ pressure for maximum production of isoquercetin were lower than those of quercetin. Thermal degradation products such as protocatechuic acid and 2,5-dihydroxyacetophenone were observed due to pyrolysis at high temperature. It was concluded that rutin can be easily converted into isoquercetin and quercetin by SWH under $CO_2$ pressure, and this result can be applied for SWH of rutin-rich foodstuffs.

산 가수분해를 이용하여 microcrystalline cellulose로부터 추출 된 cellulose nano-whisker의 특성분석 (Preparation and Characterization of Cellulose Nano-Whiskers Extracted from Microcrystalline Cellulose by Acid Hydrolysis)

  • 본 연구에서는 친환경적인 특징을 가지면서도 우수한 기계적 특성을 가지고 있어 고분자 복합재료의 보강제로 주목 받고 있는 Cellulose nanowhisker (CNW) 를 염산 혹은 황산을 사용한 산가수분해 방법을 이용하여 Microcrystalline cellulose (MCC) 로 부터 추출하였다. 염산 혹은 황산을 사용하여 추출된 CNW는 직경이 20 에서 30nm 정도였고, 길이가 200 에서 300 nm 로써, 형상학적 측면에서 유사한 특성을 가지는 것을 확인하였다. 또한 전해질 용액의 전기전도도를 이용한 적정 (conductometric titration) 결과 황산을 이용하여 제조되어진 CNW의 경우, 셀룰로오스 표면의 sulfate group 에 의해 나타나는 표면전하 값이 각각 140, 197.78 mmol/kg으로 나타났으며, 염산을 이용하여 제조되어진 CNW 의 경우 셀룰로오스 표면에 약한 전하 값을 가져 표면전하 값을 구할 수 없었다. 황산을 이용하여 추출된 CNW의 열중량 분석 결과 염산을 이용하여 추출된 CNW와 비교하여 열분해 온도가 급격히 감소하는 것을 확인할 수 있었으며, Broido's method를 이용하여 정의 되어진 열분해 거동에 대한 활성화 에너지 역시 상대적으로 낮음을 확인할 수 있었다.

가열용융 방법에 의한 Ge-BPSG 마이크로렌즈 어레이 제작 (Ge-doped Boro-Phospho-Silicate Glass Micro-lens Array Produced by Thermal Reflow)

  • 화염가수분해 증착법(FHD : Flame Hydrolysis Deposition)으로 제작된 Ge이 첨가된 BPSG(Boro-Phospho-Silicate-Glass)막의 표면을 절단톱(dicing saw)을 이용하여 일정한 깊이로 절단함으로써 각 단위 마이크로렌즈 셀들을 형성시켰다. 또한 절단된 각 단위 마이크로렌즈 셀들을 가열용융(thermal reflow) 방법을 이용하여 $1200^{\circ}C$에서 가열용응시킴으로써 직경이 $53.4{\mu}m$인 마이크로렌즈 어레이를 제작할수 있었다. 이 때 렌즈간 간격은 $70{\mu}m,$ 렌즈 두께는 약 $28.4{\mu}m$이었다. 제작된 마이크로렌즈 어레이의 형상을 이미지-프로세스로 분석하였으며. 초점거리는 $62.2{\mu}m$이었다. 본 제작방법은 일반적인 사진식각 공정을 이용한 마이크로렌즈 제작보다 간단하면서도 저렴한 비용으로 제작이 가능하다. 또한 곡률반경의 조절이 용이하고, 보다 정밀하며 다양한 마이크로렌즈 어레이를 구현할 수 있다.
