SLC35B2 Expression is Associated with a Poor Prognosis of Invasive Ductal Breast Carcinoma

  • Background: Breast cancer is the most common malignancy in women worldwide, including Thailand, and is a major cause of mortality and morbidity, despite advances in diagnosis and treatment. Novel gene expression in breast cancer is a focus in searches for prognostic biomarkers and new therapeutic targets. Materials and Methods: The mRNA expression of novel B4GALT4, SLC35B2, and WDHD1 genes in breast cancer were examined in invasive ductal breast carcinoma (IDC) patients using quantitative real-time reverse transcription polymerase chain reaction (QRT-PCR). Results: Among these genes, increased expression of SLC35B2 mRNA was significantly associated with TNM stage III + IV of IDC (p<0.001). Hence, up-regulation of SLC35B2 may serve as a prognostic biomarker for poor prognosis, and is also a potential therapeutic target in breast cancer.

3-Deazaneplanocin A is a Promising Therapeutic Agent for Ovarian Cancer Cells

  • Background: Recent studies have shown that 3-deazaneplanocin A (DZNep), a well-known histone methyltransferase inhibitor, disrupts polycomb-repressive complex 2 (PRC2), and induces apoptosis, while inhibiting proliferation and metastasis, in cancer cells, including acute myeloid leukemia, breast cancer and glioblastoma. However, little is known about effects of DZNep on ovarian cancer cells. Materials and Methods: We here therefore studied DZNep-treated A2780 ovarian cancer cells in vitro. Proliferation of ovarian cancer cells under treatment of DZNep was assessed by MTT and apoptosis by flow cytometry. Cell wound healing was applied to detect the migration. Finally, we used q-PCR to assess the migration-related gene, E-cadherin. Results: DZNep could inhibit the proliferation of A2780 and induce apoptosis Furthermore, it inhibited migration and increased the expression of E-cadherin (P<0.05). Conclusion: DZNep is a promising therapeutic agent for ovarian cancer cells, with potential to inhibite proliferation, induce apoptosis and decrease migration.

Clinical Effects of Chuongsangboha-tang in Asthmatic Patients (청상보하탕의 기관지 천식 환자에 대한 임상적 효과)

  • Background: Nowadays asthma is considered to be an inflammatory disease characterized by airway hyperresponsiveness and pulmonary eosinophilia. Production of cytokines by bronchial epithelial cells may contribution to the local accumulation of inflammatory cells in patients with bronchial asthma. In many recent studies molecular biological methods have been used to investigate the role of cytokines in pathogenesis and new therapeutic targets of asthma. Objectives: We aimed to identify the clinical effects of Chuongsangboha-tang and the effects of Chuongsangboha-tang on serum cytokines in asthmatic patients. Materials and Methods: The subjects consisted of 36 patients with asthma who had been treated with Chuongsangboha-tang for four weeks. Chuongsangboha-tang is an herbal decoction which has been used of the traditional therapeutic agent of asthma. PFT, Quality of Life Questionnaire for Adult Korean Asthmatics (QLQAKA), blood eosinophil, serum IgE, Serum IL-4, IL-5, IFN- were checked before and 4 weeks after treatments. Results: Treatment with Chuongsangboha-tang for four weeks resulted in significant increase in FEV1.0%, PEFR%, QLQAKA. The blood eosinophil, serum IgE, IL-4 and IL-5 in asthmatic patients increased significantly compared to the normal control group, while the serum IFN-decreased significantly. Conclusions: This study shows that Chuongsangboha-tang has effects on improvement of pulmonary function and quality of life in asthmatic patients. Obviously, further research concerning this is still necessary.

Therapeutic Effects of Eucalyptus Globulus Essential Oil on an Animal Model with Allergic Rhinitis (안엽유의 흡입이 알레르기성 비염 동물모델의 비염치료에 미치는 영향)

  • Background and Objectives: Aroma therapy using Eucalyptus Globulus essential oil for curing allergic rhinitis is an inhalation treatment which has been applied as an aid to oriental clinics which use a fundamental treatment such as acupuncture and herbs. There have been no studies so far performed on the effect of this oils' use, only . We aimed to find out the therapeutic effect of its exclusive use only on an Animal Model with Allergic Rhinitis. Materials and Methods: Sprague-Dawley rats were induced the allergic rhinitis by ovalbumin solution. And then, rats in the treated group were treated by diffusing Eucalyptus essential oil in a flask for 7 days. Compared with the treated group, rats in the control group were treated by diffusing normal saline in a flask for 7 days. We observed changes in glandular cells and mast cells in nasal mucosa and submucosa; also changes in the number of eosinophils and neutrophils in blood and the succus of rhinenchysis. Results: The number of neutrophils in the succus of rhinenchysis was significantly decreased in treated group compared with control group(p<0.05). The number of neutrophils in blood and mast cells in nasal mucosa were decreased in the treated group but, these were not significant statistically(p>0.05). Histologic changes showed edema; congestion and expansion of grandular cells in nasal submucosa and hypertrophy of epithelium in nasal mucosa were decreased in treated group compared with control group. Conclusions: The results may suggest that the inhalation treatment using Eucalyptus essential oil decreases the inflammatory response on an Animal Model with Allergic Rhinitis.

A Retrospective Review of Iatrogenic Skin and Soft Tissue Injuries

  • Background Even though the quality of medical and surgical care has improved remarkably over time, iatrogenic injuries that require surgical treatment including injuries caused by cast and elastic bandage pressure, extravasation, and dopamine-induced ischemia still frequently occur. The goal of this study was to estimate the incidence and analyze the distribution of iatrogenic injuries referred to our department. Methods A retrospective clinical review was performed from April 2006 to November 2010. In total, 196 patients (116 females and 80 males) were referred to the plastic surgery department for the treatment of iatrogenic injuries. We analyzed the types and anatomic locations of iatrogenic complications, along with therapeutic results. Results An extravasation injury (65 cases, 37.4%) was the most common iatrogenic complication in our study sample, followed by splint-induced skin ulceration, dopamine-induced necrosis, prefabricated pneumatic walking brace-related wounds and elastic bandage-induced wounds. Among these, prefabricated pneumatic walking brace-related complication incidence increased the most during the 5-year study period. Conclusions The awareness of the very common iatrogenic complications and its causes may allow physicians to reduce their occurrence and allow for earlier detection and referral to a plastic surgeon. We believe this is the first study to analyze iatrogenic complications referred to a plastic surgery department in a hospital unit.

Immunohistochemical and Electron Microscopic Studies on Intermediate Filament of Epithelial Cell and Non-Epithelial Cells (상피 및 비상피세포들의 Intermediate Filament에 대한 면역조직화학적 및 전자현미경적 연구)

  • The intermediate filament is one of the most important constituents of the intracytoplasmic cytoskeleton microtubule, actin, myosin and intermediate filament. It is composed of keratin, desmin, vimentin, neurofilament and glial filament, and has important role as a cellular marker, epithelial or mesenchymal origin. So it will be important to differentiated from some poorly or undifferentiated neoplasm to provide adequate therapeutic modalities. This study was performed by using immunohistochemical staining and electron microscopic observation to find out intermediate filaments of epithelial and non-epithelial tumor cells evaluate the degree of differentiation in tumors and therefore to provide some diagnostic and therapeutic modalities. The materials consisted of 83 epithelial and non-epithelial elements bearing 23 normal control, 28 epithelial tumors, and 32 non-epithelial tumors, that are resected for definite treatment at Chosun University Hospital from June, 1988 to June, 1990. Immunohistochemical stain for keratin, desmin and vimentin, and electron microscopic study were performed in all cases. The results obtained were as follows. 1. Immunohistochemical stain for intermediate filament were very useful diagnostic aid for differentiated epithelial tumor to non-epithelial tumor in diagnostic neoplasia. 2. In the electron microscopic finding, the size of intermediate filaments were possible differentiated to cell components of epithelial tumor and non-epithelial tumors.

Protein-Coating Evaluation Method of Colloidal Gold Nanoparticles (콜로이드 골드 나노입자의 단백질 수송성 평가법)

  • Colloidal gold nanoparticles might be of use as nano scale delivery systems of various therapeutic materials in the future. Recent studies have reported the feasibility of colloidal gold nanoparticles as gene delivery systems or protein delivery systems. In this study, we aimed to develop a short-step method useful for screening the optimal coating conditions of colloidal gold nanoparticles with proteins. We observed that colloidal gold nanoparticles have properties of changing its unique color when they were exposed to NaCl solution. Taking advantage of the color changing properties of colloidal gold nanoparticles, we applied the color testing method of colloidal gold nanoparticles solutions for evaluating the protein coating nature. Using bovine serum albumin as a model protein, we tested the protein coating of colloidal gold nanoparticles via the color change upon NaCl addition. The optimal coating concentration and coating conditions of colloidal gold nanoparticles with bovine serum albumin were fixed using the color testing methods. We suggest that the color testing method might be applied to optimize the coating condition of colloidal gold nanoparticles with other therapeutic proteins.

Experimental Evaluation of Scattered X-Ray Spectra due to X-Ray Therapeutic and Diagnosis Equipment for Eye Lens Dosimetry of Medical Staff

  • Background: For proper monitoring of the eye lens dose, an appropriate calibration factor of a dosimeter and information about the mean energies of X-rays are indispensable. The scattered X-ray energy spectra should be well characterized in medical practices where eye lenses of medical staffs might be high. Materials and Methods: Scattered X-ray energy spectra were experimentally derived for three different types of X-ray diagnostic and therapeutic equipment, i.e., the computed tomography (CT) scan, the angiography and the fluoroscopy. A commercially available CdZnTe (CZT) spectrometer with a lead collimator was employed for the measurement of scattered X-rays, which was performed in the usual manner. Results and Discussion: From the obtained energy spectra, the mean energies of the scattered X-rays lied between 40 and 60 keV. This also agreed with that obtained by the conventional half value layer method. Conclusion: The scattered X-rays to which medical workers may be exposed in the region around the eyes were characterized by means of spectrometry. The obtained mean energies of the scattered X-rays were found to match the flat region of the dosimeter response.

Skin Permeation Characteristics of Antihyperlipoproteinemic Agent using Natural Polymer Bases in Rats (천연고분자 기재에 의한 수용성 항고지단백혈증제의 흰쥐 피부투과 특성)

  • Transdermal therapeutic system(TTS) is often used as the method of drug dosage into the epidermic skin. Natural polymer were selected as ointment material of TTS. We investigated the permeation of natural polymer ointment containing drug in rat skin using horizontal membrane cell model. Permeation properties of materials were investigated for water-soluble drug such as oxiniacic acid in vitro. These results showed that skin permeation rate of drug across the composite was mainly dependent on the property of ointment base and drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate. This result suggests a possible use of natural polymer ointment base as TTS of antihyperlipoproteinemic agent.

  • Herbal medicines are increasingly being utilized to treat a wide variety of disease processes. We previously reported that aqueous extract from the roots of Platycodon grandiflorum A. DC (Campanulaceae), Changkil (CK), had hepatoprotective effects against acetaminophen induced liver injury. In the present study, we assayed the preventive and therapeutic effects of CK on experimental hepatic fibrosis induced by dimethylnitrogamine or carbon tetrachloride in rats. (omitted)
