Endophytic Association of Trichoderma asperellum within Theobroma cacao Suppresses Vascular Streak Dieback Incidence and Promotes Side Graft Growth

  • Trichoderma species are able to persist on living sapwood and leaves of cacao (Theobroma cacao) in an endophytic relationship. In this research, we evaluated the ability of Trichodema asperellum introduced at the incision site in the bark for side grafting with the concentration of 4 g/10 mL, 4 g/100 mL, and 4 g/1,000 mL (suspended in water) in suppressing vascular streak dieback (VSD) incidence and promoting growth of side grafts in the field. The incidence of VSD in two local clones of cacao, MCC1 and M04, without application of T. asperellum was 71.2% and 70.1% at 21 wk after grafting, respectively. However, when the two clones were treated with a concentration of 4 g/10 mL T. asperellum, the incidence was 20.6% and 21.7%, respectively, compared to 29.1% and 20.9% at 4 g/100 mL and 18.2% and 15.6% at 4 g/1,000 mL. By comparing to the control, the treatment with the same concentrations of T. asperellum listed above, the total number of stomata in MCC1 decreased by 41.9%, 30.2%, and 14.0% and in M04 by 30.5%, 21.9%, and -2.5% (exception), respectively. Otherwise, the total area of stomata opening increased by 91.4%, 99.7%, and 28.6% in MCC1 and by 203.8%, 253.5%, and 35.9% in M04, respectively. Furthermore, the number of buds and branches treated with a mixture concentration on the the two clones increased by 90.7% and 21.7%, respectively. These data showed that the application of T. asperellum to cacao scions while grafting can decrease VSD incidence in side grafts and increase growth of grafts in addition to decreasing total number of stomata, increasing total area of opened stomata, and increasing number of buds and branches.

카카오(Theobroma cacao L.) 용매 분획물의 항산화 및 항염증 효과 (Anti-Oxidative and Anti-Inflammatory Effects of the Solvent Fraction from Theobroma cacao L. Extract)

  • 본 연구는 카카오를 화장품 소재로 활용하기 위하여 항산화 및 항염증 효과를 검증하여 기능성 화장품 소재로서의 가능성을 검토하였다. 아세톤 추출물을 이용하여 클로로포름층, 에틸아세테이트층, 부탄올층, 물층의 극성별로 분획을 실시하였다. 카카오 분획물의 전자공여능은 100 ${\mu}g/ml$에서 에틸아세테이트 분획물이 76.2%, 부탄올 분획물이 53.9%의 소거능을 나타내었다. 또한, superoxide anion radical 소거능 측정 결과, 에틸아세테이트 분획물과 부탄올 분획물이 50 ${\mu}g/ml$에서 각각 76.09%, 51.4%의 소거능이 나타났고, $Fe^{2+}$, $Cu^{2+}$의 첨가에 따른 지방산패 억제능에서 $Fe^{2+}$를 첨가한 군에서는 에틸아세테이트 분획물이 50 ${\mu}g/ml$에서 64%로 $Cu^{2+}$를 첨가한 군보다 저해능이 높았다. 항염증 효과 측정으로 hyaluronidase 저해 활성을 측정한 결과, 부탄올 분획물의 경우 100 ${\mu}g/ml$에서 53.04%의 저해활성을 나타내었으며, lipoxygenase 저해 활성 측정결과 10 ${\mu}g/ml$에서 51.32%의 저해활성을 나타내었다. 대식세포를 이용한 nitric oxide (NO) 저해활성을 측정한 결과 카카오 에틸아세테이트 분획물에서 가장 높은 NO 저해활성을 나타냄을 확인할 수 있었다. 또한 대식세포 내에서 에틸아세테이트 분획물 100 ${\mu}g/ml$ 농도에서 iNOS, COX-2 단백질 발현을 저해하였다.

Effect of Antioxidant Addition on Milk Beverage Supplemented with Coffee and Shelf-life Prediction

  • This study aimed to extend the shelf-life of coffee-containing milk beverage by adding Theobroma cacao (cacao nibs) extract. To prepare the beverage sample containing cacao nibs extract, 0.8% cacao nibs hydrothermal extract was aseptically injected. Qualitative changes in the beverage samples, including antioxidant effect, peroxide value (POV), caffeine content, and sensory parameters were monitored regularly during storage at 10℃, 20℃, and 30℃ for 4 wk. The inclusion of cacao nibs extract produced higher antioxidant activity compared to the control. As the storage temperature increased, the POV of all samples increased. Samples with cacao nibs extract generally displayed lower POV than the control. The caffeine content of all samples tended to decrease during storage, with the decrease accentuated by higher storage temperatures. In the shelf-life prediction using the Arrhenius model, the kinetic regressions of the cacao nibs extract-added sample and control were YPOV=1.2212X-2.1141 (r2=0.9713) and YPOV=1.8075X-2.0189 (r2=0.9883), respectively. Finally, the predicted shelf-life of cacao nibs-added group and control to reach the quality limit (20 meq/kg POV) were approximately 18.11 and 12.18 wk, respectively. The results collectively indicate that the addition of cacao nibs extract extends the shelf-life of the coffee-containing milk beverage and heightened the antioxidant effect.

  • Cacao bean husk로부터 2종류의 2당체 flavan-3-ol을 분리하였고 $anisaldehyde-H_2SO_4$ 용액, $FeCl_3$ 및 TLC상에서 축합형 탄닌으로 확인되었다. 이들 2종류의 flavan-3-ol 화합물은 procyanidin B-1[epicatechin-$(4{\beta}{\rightarrow}8)$-catechin], procyanidin B-3[catechin-$(4{\alpha}{\rightarrow}8)$-catechin]로 기기분석에 의해 분자구조가 결정되었다. Procyanidin B-1은 0.3 mM에서 50% glucosyltransferase 저해효과가 관찰되었고 비경쟁적 저해작용기작을 보였으며 epicatechin을 함유한 화합물이 저해 효과가 높은 것으로 확인되었다.

카카오 빈 함량에 따른 쌀 압출성형물의 이화학적 특성 및 항산화 활성 (Physicochemical Properties and Antioxidant Activity of Extruded Rice Flour with Various Cacao (Theobroma cacao L.) Bean Content)

  • 본 실험은 카카오 빈의 함량(0, 20, 40, 60%)을 달리하여 쌀가루와 압출성형물을 제조하여 물리화학적인 특성을 관찰하고자 하였다. 공정 조건은 수분 함량 25%, 스크루 회전 속도 200 rpm, 사출구온도 $130^{\circ}C$로 조절하였다. 팽화율은 카카오 빈의 함량이 증가할수록 감소하였다. 밀도와 비길이, 겉보기 탄성계수는 카카오 빈의 함량이 증가함에 따라 증가하였고, 파괴력은 카카오 빈의 함량이 증가할수록 감소하였다. 수분용해지수와 수분흡착지수는 압출성형 공정 후 모두 증가하였다. 카카오 빈의 첨가량이 증가할수록 명도는 감소하였고 적색도는 증가하였으며, 황색도는 무처리구(RAW)에서 카카오 빈이 증가함에 따라 증가하였지만, 압출성형 공정 후 카카오 빈이 증가함에 따라 감소하였다. 색도차는 카카오 빈의 함량이 증가할수록 증가하였다. 페이스트 점도는 카카오 빈의 함량이 증가할수록 모든 조건에서 감소하였으며, 압출성형을 한 모든 실험군과 RAW의 roasted 카카오빈(RC) 첨가군에서 저온최고점도(CPV)를 나타내어 낮은 온도에서도 점성을 나타내는 특성을 보였다. 총 페놀 함량은 non-roasted 카카오 빈(NRC) 첨가군이 RC 첨가군보다 높았으며, DPPH 라디칼 소거능은 압출성형한 NRC 첨가군에서 가장 높게 나타났다. 결론적으로 카카오 빈 첨가가 쌀 카카오의 팽화 특성에 영향을 미치며, 카카오 빈의 총 페놀 함량은 로스팅과 압출성형에 의해 감소하였지만, DPPH 라디칼 소거능은 로스팅을 하지 않고 압출성형 하였을 때 증가하였다.

  • Cacao bean외피로부터 분리된 물질은 anisaldehyde-$H_2SO_4$$FeCl_3$에 각각 갈색 및 청색반응을 하였고 TLC상에서 monomer 및 dimer의 fIavan-3-ol 물질로 확인되었다. 물질의 구조를 NMR로 검정한 결과 compound I 과 II 는 각각 (-)-epicatechin과 epicatechin-(4${\alpha}$$\rightarrow$8)-epicatechin의 결합 화합물인 procyanidin B-2이었다. 분리된 두 화합물은 GTase에 대해 높은 저해활성이 있었고 특히 procyanidin B-2는 1.0mM에서 완전히 저해시켰다. 이것은 f1avan-3-ol 구조내에 OH가의 증가에 따라 GTase 저해활성도 높아지는 것으로 추정되었다.

Improving Nutritional Quality of Cocoa Pod (Theobroma cacao) through Chemical and Biological Treatments for Ruminant Feeding: In vitro and In vivo Evaluation

  • Cocoa pod is among the by-products of cocoa (Theobroma cacao) plantations. The aim of this study was to apply a number of treatments in order to improve nutritional quality of cocoa pod for feeding of ruminants. Cocoa pod was subjected to different treatments, i.e. C (cocoa pod without any treatment or control), CAm (cocoa pod+1.5% urea), CMo (cocoa pod+3% molasses), CRu (cocoa pod+3% rumen content) and CPh (cocoa pod+3% molasses+Phanerochaete chrysosporium inoculum). Analysis of proximate and Van Soest's fiber fraction were performed on the respective treatments. The pods were then subjected to an in vitro digestibility evaluation by incubation in rumen fluid-buffer medium, employing a randomized complete block design (n = 3 replicates). Further, an in vivo evaluation of the pods (35% inclusion level in total mixed ration) was conducted by feeding to young Holstein steers (average body weight of $145{\pm}3.6kg$) with a $5{\times}5$ latin square design arrangement (n = 5 replicates). Each experimental period lasted for 30 d; the first 20 d was for feed adaptation, the next 3 d was for sampling of rumen liquid, and the last 7 d was for measurements of digestibility and N balance. Results revealed that lignin content was reduced significantly when cocoa pod was treated with urea, molasses, rumen content or P. chrysosporium (p<0.01) with the following order of effectiveness: CPh>CAm>CRu>CMo. Among all treatments, CAm and CPh treatments significantly improved the in vitro dry matter and organic matter digestibility (p<0.05) of cocoa pod. Average daily gain of steers receiving CAm or CPh treatment was significantly higher than that of control (p<0.01) with an increase of 105% and 92%, respectively. Such higher daily gain was concomitant with higher N retention and proportion of N retention to N intake in CAm and CPh treatments than those of control (p<0.05). It can be concluded from this study that treatment with either urea or P. chrysosporium is effective in improving the nutritive value of cocoa pod.

Relationship between some Phenological Parameters and Somatic Embryogenesis in Theobroma cacao L.

  • The relationship between three phenological parameters and somatic embryogenesis was investigated during a two-year period. Staminodes and petals from six hybrids and two clones as controls were sowed on three distinct primary callus growth media. Flowering level, fructification level, and leaf thrusts rhythm as phenological parameters were measured simultaneously during the weekly harvest of flower buds. Mean and coefficient of variation of the measured parameters highlighted stable phenological phases. The relationship between phenological parameters and somatic embryogenesis was investigated first by comparing the variation of somatic embryogenesis and that of the phenological parameters, and second by using Pearson's linear correlation. Except for the fructification level in both control clones the first year, the other parameters recorded stable phenological phases, regardless of the genotype and year. Favorable and unfavorable phases for the somatic embryogenesis were identified. In hybrids, favorable phases included February, August, September, and October. In both control clones, time interval propitious to embryogenesis stretched from February to December. The significance of the coefficient of correlation seemed to establish a relationship between somatic embryogenesis and phenology. However, a causal link could not be established. Leaf thrusts rhythm was revealed to be the phenological parameter most linked to somatic embryogenesis. Attempts to optimize embryogenesis during unfavorable phases, showed that a correction of 2.4 D/TDZ concentration is not the solution.

한방복합추출물의 항산화 및 항비만 효과 (Antioxidant and Anti-obesity Effects of Herbal Complex Extract)

  • 본 연구에서는 한방복합추출물(마황, 창출, 석고, 카카오닙스)을 제조하여, 추출물에 대한 항산화 활성 및 지방세포형성억제 활성을 확인하고자 하였다. 한방복합물의 항산화 활성을 확인하기 위하여 DPPH, ABTS+ radical 소거능 및 xanthine oxidase 저해 활성을 측정한 결과 추출물 1,000 ㎍/ml 농도에서 각각 75.0, 100.8 및 79.5%로 높은 소거능을 나타내었다. 3T3-L1 지방전구세포에서 한방복합물에 의한 지방형성 및 지방세포 분화 억제를 조사한 결과 0~50 ㎍/ml 농도에서 3T3-L1 세포 생존율에 영향을 미치지 않았다. 3T3-L1 지방전구세포에서 25, 50 및 75 ㎍/ml의 한방복합물 처리는 농도 의존적으로 지질 축적을 억제하였다. Western blot 실험 결과 한방복합물은 MDI 유도 분화 3T3-L1 세포에서 분화 전사인자인 PPARγ, C/EBPα 을 75 ㎍/ml 농도에서 각 44.2, 76.6% 억제함을 나타내었다. 또한, 지방산 합성 조절 인자인 FAS의 발현도 억제하였다. 이와 같은 결과를 바탕으로 본 연구는 항비만 기능성 소재로서 한방복합추출물의 활용 가능성을 제시해 줄 수 있을 것으로 사료 된다.
