Studies on the Correlation between SPF index and Primary Irritation Index of Octylmethoxycinnamte (Octylmethoxycinnamate의 SPF 지수와 피부자극지수와의 상관성 연구)

  • In recent years, the safety of sunscreens have been challenged based on the reports of its adverse effects on users; dermatitis, allergic contact dermatitis, photoallergic contact dermatitis. In the toxicological aspect, the need to investigate the safety of sunscreens is steadily increasing. However, there were few studies on the correlation between sun protection factor (SPF) and the safety of sunscreens. The objective of this study was to assess whether there was a correlation between SPF and the skin irritation index of sunscreens. Octyl methoxycinnamate (OMC) was selected, which was the most frequently used (about 92% in korea and about 90% in the world) in sunscreens. In vitro SPF test was performed using the standard samples recommended by KFDA guideline. We examined primary and cumulative skin irritation and eye irritation of OMC standard sample as well as homosalate. As in vitro SPF was increased, skin irritation index was increased in primary skin irritation test ($r^2$=0.986) and cumulative skin irritation test ($r^2$=0.939). In addition, OMC was non-irritating and non-sensitizing. In this results, there was a good correlation between SPF index and skin irritation index.

  • In vitro cell culture system has been proposed as a promising alternative model to in vivo skin irritation test. These studies were performed to screen the cytotoxicity effects of surfactants using normal human skin fibroblasts. Cell membrane integrity assessed by the leakage of lactate dehydrogenase (LDH) and mitochondrial integrity by MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromides reduction test were affected in a dose dependent manner. The irritation potential of surfactants to human skin patch test, and the changes of capillary permeability by rabbit intradermal safety test were assessed as in vivo methods. Our results suggest that LDH leakage assay and MTT reduction test using cultured human fibroblasts could be predictive for the irritancy of various surfactants in human, and LDH assay is superior correlated with in vivo test (r=0.886) to MTT test with in vivotest (r=0.757).

  • In-vivo and in-vitro MR Microscopy has been performed to investigate any differences in-between the normal and the abnormal skin layers. Noninvasively acquired images could provide the skin histology. Also, it was found that the epidermis of the abnormal skin was thickened more than that of normal skin. MR Microscopy utilized for the present study may be applicable to the noninvasive investigation of skin pathology and this technique may be of use in the development of cosmetics.

Safety Evaluation and Anti-wrinkle Effects of Retinoids on Skin

  • Retinoids have many beneficial effects on dermatological applications. But, retinoids cause skin irritation. In this study, the safety of retinoids was clarified via both primary skin irritation test in rabbits and sensitization study using an integrated model for the differentiation of chemical-induced allergic and irritant skin reaction (IMDS), an alternative method to sensitization test. The effects of retinoids on the change of ultraviolet A (UVA)-induced matrix metalloproteinase-1 (MMP-1) in human skin fibroblasts and the modulation of type-1 pN collagen synthesis in hairless mice were examined to clarify the anti-wrinkle effects. Alltrans retinol (t-ROL) and its derivative, all-trans retinoic acid (t-RA), showed mild skin irritation but did not induce the sensitization. t-ROL and t-RA exerted anti-wrinkle effects by inhibiting the UVA-induced MMP-1 in human skin fibroblasts and increasing the type-1 pN collagen synthesis in hairless mice. These findings suggest that retinoids do not induce the allergy, and show anti-wrinkle effects by decreasing MMP-1 activation and increasing collagen synthesis.

  • The marine leisure activities are specially increasing among the other leisure activities due to the improving of people's life quality and formation of people's values which pursuit the various life styles with the development of economy and society and operation the system of five working days per week. Among the various marine leisure activities, skin scuba diving is the mast prevalent activity, even though it doesn't have proper legal system in relation to the safety of the activity. Thus, this paper studies the various legal issues in relation to skin scuba diving to improve the safety and activation of the activity, and identifies the problems and proposes the ways of improving it.

  • Hyaluronic acid(HA) is a naturally occurring mucopolysaccharide composed of the repeating disaccharide unit of D-glucuronic acid and N-acetyl-D-glucosamine. HA exists as a high molecular weight polymer in the extracellular matrix of many tissues in the body. HA has been widely used not only for osteoarthritis and ophthalmology but also for cosmetics for skin care. Recently HA has drawin much attention for use in health foods. Bioland Corporation started production of HA t1y biotechnological process in 1990 for cosmetical use. At present Bioland produces HA for cosmetic and food use. We have done many researches to evaluate the helpful effect for skin and safety of HA as food. In order to evaluate the effect of HA on the skin after its oral intake, we measured its bioavailability, skin-care effects on anti-wrinkle, moisturization elasticity and desquamation by clinical trial. The results showed that HA is very safe and could bs helpful to improve anti-wrinkling, moisturization, elasticity, and desquamation on the skin.

  • Chemicals for cosmetics, including skin, the skin absorbs some of the research in the field of science or pharmacy recently, about the environment and the health of the heightened interest in skin absorption, and many other human attributes and absorption evaluation studies are underway in various areas. In this study, The effects of commercial permanent wave products to skin which are composed with cysteine and bases using rat. Results are as follows; the content of penetration 4 hours later with steady state and no significant changeable after 20 hours later. In cysteine groups lag time and permeability coefficient of young skin is 3.32hr and $0.102{\mu}g/cm^2{\cdot}hr$, lag time and permeability coefficient of old skin is 4.04hr and $0.106{\mu}g/cm^2{\cdot}hr$. In conclusion of study lag time and permeability coefficient in old skin and wounded skin are faster than healthy skin. We notified that fine rinkle and rash of skin were changeable in the case of treating with permanent wave drugs than normal skin.

NMR Microscopy for the Relaxation Phenomena of Skin Layers

  • In-vivo and in-vitro NMR Microscopy has been performed to investigate the histological and/or pathological difference between the normal and the abnormal skin layers. The acquired images in the present study were able to provide the information of both the relaxation changes and the histological changes due to skin abnormality. The present study is also intended to show the possibility of NMR Microscopy application to both the noninvasive investigation of skin pathology and the helpful use in the development of cosmetics.

  • Carthami tinctorii Fructus is known for its good effect on diseases such as fracture, menorrhalgia, menoschesis, puerperium aneilema and so on. The study of irritation and toxicity of Carthami oil aquapuncture solution applied topically to the skin and the eye mucous membrane were carried out to prove the safety of Carthami oil aquapuncture solution in clinical use. Animal for the research was the rabbit and the solution for the test was made from Carthami semen. 6 animals were used for the skin test and 9 animals were used for the eye mucous membrane test. In results, both tests proved that Carthami oil aquapuncture solution makes no irritable reaction on skin and eye mucous membrane of rabbit. We consider that this result is helpful for saying about the safety of Carthami oil aquapuncture solution in clinical use.

The application of alternative methods for skin irritation evaluation on pesticides (농약에 대한 피부자극성 대체시험법 적용연구)

  • It is common to use many experiment animals to evaluate the toxicity of chemicals including pesticides. For protecting animal, the concepts of 3R (Reduction, Replacement, Refinement) were introduced and in vitro alternatives methods actively have been developed all over the world. Many experimental animals for toxicological tests have been used, so that it is important to establish the alternative methods. In this study, the alternative method using reconstituted human skin model (Keraskin$^{TM}$) was conducted for classification of skin irritation on pesticides. Sixteen formulations selected on the basis of the degree of irritation were treated by Keraskin$^{TM}$ test. The percent of cell viability was measured into the culture medium collected after treatment of the pesticides for 24-72 hrs. The skin irritations of formulations were evaluated by the cell viability. In this study, The 4 formulations with mild irritation in rabbits were evaluated as nonirritant, the 6 formulations with moderate and severe irritation were evaluated as irritant in human skin model test. We suggest that the alternative test using Keraskin$^{TM}$ model could be used as toxicity evaluation for primary irritation index (P.I.I.) score of greater than or equal to 2.1 of pesticides. The further studies should be required to apply for hazardous assessment of pesticides on alternative skin irritation methods because of the interindividual variability of the sensitivity of skin irritation on pesticides.
