Two New Antiinflammatory Triterpene Saponins from the Egyptian edicinal Food Black Cumin (Seeds of Nigella sativa)

  • An extensive phytochemical investigation of the polar fractions of a methanolic extract of Egyptian medicinal food, black cumin (seeds of Nigella sativa L.) led to the isolation of two new triterpene saponins, named sativosides A and B (1-2), along with four known saponins (3-6). Sativoside A (1) is the first example of saponins containing 18-ene triterpene aglycon not only in this Nigella genus but also in the family Ranunculaceae. The structure of the new saponins was elucidated mainly by a combination of 1D and 2D NMR data, together with HRFABMS and acid hydrolysis. Three compounds (1-3) showed the significant inhibition effect of phorbol 12-myristate 13-acetate (PMA) plus calcium ionophore A23187-induced production of IL-6 in a human mast cell (HMC-1) line.

Anti-allergic Effect of Aqueous Extract of Stachys riederi var.japonica Miq.in vivo and in vitro

  • The effect of aqueous extract of Stachys riederi var. japonica Miq. (Labiatae) (SRAE) on the immediate-type allergic reactions was investigated. SRAE was found to exhibit a inhibitory activity on the compound 48/80-induced systemic anaphylaxis in mice. SRAE inhibited the plasma histamine release induced by compound 48/80 in mice. In addition, SRAE also inhibited the passive cutaneous anaphylaxis reaction induced by IgE/anti-IgE in mice. The effect of SRAE on the histamine release from rat peritoneal mast cells (RPMC) was studied. SRAE inhibited the histamine release induced by compound 48/80 in RPMC. To clarify the mechanism of these inhibiting reactions, we investigated the effects of SRAE on cyclic AMP (cAMP). The level of cAMP in human leukemia cell line, HMC-1, when SRAE (1 mg/ml) was added, significantly increased compared with that of basal cells. These results indicate that SRAE may be beneficial in the treatment of immediate-type allergic reaction.

봉약침액(蜂藥鍼液)의 NO 소거 및 Chemokine 유전자 발현에 대한 효과 (Nitric Oxide Scavenging Effect and Expression of Chemokine Genes in Bee Venom)

  • Although the effect of Bee Venom has been reported, its mechanism has not been fully elucidated. Nitric Oxide(NO) is one of the free radicals and mediates in inflammation diseases. Chemokines contribute to the pathogenesis of several disorders such as allergic rhinitis and rheumatoid arthritis and so on. The objective of this study was to investigate the scavenging effect of Bee Venom on NO and on expression of chemokine genes. There was no significant NO scavenging effect in Crude Bee Venom, Apamin, Melittin, and MCD-peptide. The expression of chemokines was examined by RT-PCR using the human mast cell line(HMC-1), which is known to secrete and express chemokines. In order to investigate the protective effect of Bee Venom, HMC-1 cells were incubated with pretreatment of Bee Venom for 24 hrs and stimulated with 1 uM calcium ionophore A23178 for 2 hrs. RT-PCR analyses of chemokine genes showed that expressions of RANTES and MCP-1 were increased compared to the calcium ionophore-only treated group. But IL-8 and MCP-3 did not express increasing effect compared to control group. This study may provide important basic data on the possibility of the clinical treatment of Bee Venom in inflammation diseases.

곰취 열수 추출물의 항산화 및 항염증 활성 (Antioxidant and anti-inflammatory activities of hot water extracts of Ligularia fischeri)

  • 본 연구는 대표적인 산채지역으로 알려져 있는 영양군 곰취 열수추출물의 총 폴리페놀 함량과 DPPH 라디컬 소거능을 측정하였으며 사람 비만세포주인 HMC-1 세포를 이용하여 PMACI을 처리하여 비만세포를 활성화시켜 세포 생존률을 확인하였고, 염증성 사이토카인인 $TNF-{\alpha}$, IL-6, IL-8의 변화량을 측정하여 곰취의 기능성 식품 소재로서의 이용 가능성을 알아보고자 실험을 실시하였다. 영양군 곰취 열수 추출물의 추출수율은 원물 건조량 대비 $24.43{\pm}1.82%$(w/w), 총 폴리페놀 함량은 $198.07{\pm}0.24mg/g$으로 다른 곰취 추출물의 연구결과와 유사한 값을 나타내고, 영양군 곰취 열수 추출물의 DPPH radical 소거 활성을 측정한 결과, $IC_{50}$ 값이 $28.2{\pm}0.04{\mu}g/mL$로 양성대조군인 ascorbic acid의 $IC_{50}$$3.5{\pm}0.01{\mu}g/mL$보다 낮은 항산화활성을 보였으나 일반 천연물 유래 추출물보다 비교적 높은 항산화활성을 나타내었다. 영양군 곰취 열수 추출물을 HMC-1 세포에 처리했을 때 MTT assay법으로 분석한 결과, 세포 생존률에는 아무런 영향을 미치지 않고, $TNF-{\alpha}$, IL-6, IL-8 생성에 대한 억제효과는 유의성 있게 나타났다. 이상의 결과를 종합해 보면 항산화력 및 세포를 이용한 in vitro 실험을 실시한 결과 영양군 곰취 열수 추출물은 항산화력이 매우 우수할 뿐만 아니라 항염증효과도 뛰어나 향후 기능성 소재로서의 이용 가능성이 높은 것으로 판단되나 보다 다양하고 진보된 연구로 곰취의 염증성 질환 치료에 대한 정확한 기전이 설명되어야 할 것이다.

Hydrolysis by Alcalase Improves Hypoallergenic Properties of Goat Milk Protein

  • Goat milk is highly nutritious and is consumed in many countries, but the development of functional foods from goat milk has been slow compared to that for other types of milk. The aim of this study was to develop a goat milk protein hydrolysate (GMPH) with enhanced digestibility and better hypoallergenic properties in comparison with other protein sources such as ovalbumin and soy protein. Goat milk protein was digested with four commercial food-grade proteases (separately) under various conditions to achieve the best hydrolysis of αs -casein and β-lactoglobulin. It was shown that treatment with alcalase (0.4%, 60℃ for 30 min) effectively degraded these two proteins, as determined by SDS-PAGE, measurement of nonprotein nitrogen content, and reverse-phase high-performance liquid chromatography. Hydrolysis with alcalase resulted in a significant decrease in β-lactoglobulin concentration (almost to nil) and a ~40% reduction in the level of αs-casein. Quantification of histamine and TNF-α released from HMC-1 cells (human mast cell line) showed that the GMPH did not induce an allergic response when compared to the control. Hence, the GMPH may be useful for development of novel foods for infants, the elderly, and convalescent patients, to replace cow milk.

Anti-allergic effects of Perilla frutescens var. acuta Kudo 30% ethanol extract powder

  • Perilla frutescens var. acuta Kudo (PF) is a traditional Korean medicinal herb for allergic reaction regulation. In the present study, we investigated the effect of 30% ethanol extract powder of PF (EPPF) and rosmarinic acid (RA), the active compound of EPPF on various allergic reactions using in vivo and in vitro models. EPPF and RA significantly inhibited compound 48/48-induced systemic anaphylactic reaction and histamine release (P < 0.05). In addition, EPPF and RA significantly inhibited passive cutaneous anaphylaxis (PCA) in a dose-dependent manner (P < 0.05). These effects were stronger than those of disodium cromoglycate, the reference drug tested. EPPF and RA also significantly inhibited production of inflammatory cytokines, tumor necrosis factor-a interleukin (IL)-6, and vascular endothelial growth factor on the PCA reaction and phorbol 12-myristate 13-acetate and calcium ionophore A23187-stimulated human mast cell line, HMC-1 cells (P < 0.05). Moreover, EPPF and RA showed an inhibitory effect on lipopolysaccharide (LPS)-induced IL-4 production from whole spleen cells. Finally, EPPF and RA significantly decreased IL-4-dependent IgE production by LPS-stimulated whole spleen cells (P < 0.05). In conclusion, these results indicate that EPPF has potent anti-allergic activities.
