Studies on the Growth and Nutrient Uptake of Flag Leaf and Chaff of Rice Plant in Cold Injury Location I. Difference of Some Inorganic Elements of Grain Chaff Having Different Rice Variety and Elevation (냉해지대의 수도생육과 임, 불임 인각의 양분흡수에 관한 연구 제1보 지대별 수도품종에 따른 인각의 무기성분조성차)

  • In 1980, rice was considerably damaged by abnormal low temperature. In this paper, to determine the effect of low temperature on the growth and nutrition of rice, the experiment were carried out: varietal response to low temperature in the region with different elevations. Regional differences of heading response to low temperature were observed among varieties. The difference of days between regions were bigger in tongil lines than Japonica lines. Especially, Milyang 42 and Hangangchalbeo might belong the cold suceptible group, since the varieties were severely delayed their heading in the high mountainous region as comparred to plain region. The delay of heading with low temperature was brought out grain sterility, and fertility and ripening ratio is influenced cold tolerance and elevations, and it's decreased yield. Varieties with higher grain sterility by low temperature have higher total nitrogen content, but tended to have lower potassium and phosphate contents in the flag leaf. High content of total nitrogen, low contents of potassium and silicate were observed in the sterilized grain chaff an the opposite result were noted in the ferilized grain chaff at the ripening stage. The results reveal that the balance of these mineral element may play an important role in ripening and possibly cold tolerance.

Metallothionein Induction and Its Protective Effect in Liver and Kidney of Rats Exposed to Cadmium Chloride (Cadmium에 의한 흰쥐의 간장 및 신장의 Metallothionein 변화와 방어효과)

  • Tolerance to several toxic effects of cadmium, including lethality has been shown following pretreatment with cadmium and zinc. This study was designed to determine if tolerance also develops to Cd-induced hepatotoxicityandrenaltoxicity. Three groups of rats (A, B, C), each consisting of 16 rats, were studied and each group was divided into four subgroups (1, 2, 3, 4), 4 rats for each subgroup. Rats were subcutaneously pretreated with saline (A), $CdCl_2$ (0.5 mg/kg, B), and $ZnCl_2$ (13.0 mg/kg, C) during time periods of $1{\sim}6$ weeks. At the end of the period, rats were challenged with $CdCl_2$ (3.0, 6.0 and 9.0 mg/kg, ip). After giving the challenge dose, cadmium and metallothionein (MT) concentrations were determined and also observed the histologic change in liver and kidney. The concentration of cadmium in liver and kidney increased dose-dependently to the challenge dosage. These da indicate the kidney is a major target organ of chronic cadmium poisoning, and suggest that cadmium induced hepatic injury, via release of Cd-MT, may play an important role in the nephrotoxicity observed in response to long-term exposure to cadmium. In addition, histologic examination of group $A_2,\;A_3\;and\;A_4$ revealed moderate to severe cadmium toxicity, evidenced by infiltration of inflammatory cells, cell swelling, pyknosis, enlarged sinusoids and necrosis in liver, and tubule cell necrosis and degeneration in kidney. However, MT concentrations in liver and kidney were increased by the pretreatment of $CdCl_2$ and $ZnCl_2$, and their morphological findings were not significantly changed, comparing with control group. Higher MT concentration in liver and kidney observed in the pretreated groups constitutes a plausible explanation of the protective effects of pretreatment against the cadmium toxicity after challenge dosing.

Effect of Allopurinol on Ultrastructural Changes in Ischemia Reperfusion Injury to Skeletal Muscle of Rats After Graded Periods of Complete Ischemia (흰쥐에서 허혈시간에 따라 재관류후 나타나는 근조직의 미세구조 변화에 allopurinol이 미치는 영향)

  • It has been well known that ischemia and reperfusion injury to skeletal muscle following an acute arterial occlusion causes significant morbidity and mortality. The skeletal muscle, which contains high energy phosphate compounds, has ischemic tolerance. During the ischemia, the ATP is catalyzed to hypoxanthine anaerobically and hypoxanthine dehydrogenase is converted to xanthine oxidase. During reperfusion, the hypoxanthine is catalyzed to xanthine by xanthine oxidase under $O_2$, presence and that results in production of cytotoxic oxygen free radicals. These cytotoxic free radicals, $O_2^-,\;H_{2}O_2,\;OH^-$, are toxic and make lesions in skeletal muscle during reperfusion. The authors perform the present study to investigate the effects of allopurinol, the inhibitor of xanthine oxidase, on reperfused ischemic skeletal muscles by observing the ultrastructural changes of the muscle fibers. A total of 48 healthy Sprague-Dawley rats weighing from 200 g to 250 g were used as experimental animals. Under urethane(3.0mg/kg., IP) anesthesia, lower abdominal incision was done and the left common iliac artery were ligated by using vascular clamp for 1, 2 and 6 hours. The left rectus femoris muscles were obtained at 6 hours after the removal of vascular clamp. In the allopurinol pretreated group, 50mg/kg of allopurinol was administered once a day for 2 days and before 2 hours of ischemia. The specimens were sliced into $1mm^3$ and prepared by routine methods for electron microscopic observations. All preparations were stained with uranyl acetate and lead citrate, and then observed with Hitachi -600 transmission electron microscope. The results were as follows: 1. In 1 hour ischemia/6 hours reperfused rectus femoris muscles of rats, decreased glycogen particles and electron density of mitochondrial matrix and dilated terminal cisternae are seen. In 2 hours ischemia/6 hours repersed rectus femoris muscles of rats, mitochondria with electron lucent matrix, irregularly dilated triad and spheromembranous bodies are observed. In 6 hours ischemia/6 hours reperfused rectus femoris muscles of rats, irregularly arranged myofibrils, and many spheromembranous bodies, fat droplets and lysosome are seen. 2. In 1 hour ischemia/6 hours reperfused rectus femoris muscles of rats pretreated with allopurinol, decreased glycogen particle and dilated cisternae of sarcoplasmic reticulum and triad are observed. In 2 hours ischemia/6 hours reperfused rectus femoris muscles of rats pretreated with allopurinol decreased electron density of mitochondrial matrix and spheromembranous bodies are seen. In 6 hours ischemia/6 hours reperfused rectus femoris muscles of rats pretreated with allopurinol, mitochondria with electron lucent matrix, spheromembranous bodies and dilated cisternae of sarcoplasmic reticulum and terminal cistern are observed. The results suggest that the allopurinol attenuates the damages of the skeletal muscles of rats during ischemia and reperfusion.

Study on the Potential of Phytoremediation using Wild Plants for Heavy Metal Pollution (중금속 오염에 대한 Phytoremediation 용 야생식물 연구)

  • The potentials of some Korean wild plants as a phytoremediator for cleaning heavy metal pollution were measured. Several plant species, Ambrosia trifida, Brassica juncea, Rumex crispus, and Abutilon theophrasti screened previously for phytoremediator were treated with cadmium and copper solution. In order to know the growth response to heavy metal stress, the plants were cultivated in hydroponic system containing heavy metals with different concentration. To know the effects of heavy metals on emergence and seedling growth, seeds of 4 species were sown in the pot and watered with heavy metal solution adjusted pH to 6.5, 5.5, and 4.5. A proposed species as potential phytoremediator, A. trifida, showed tolerance to $20{\mu}mol/L$ Cd and $80{\mu}mol/L$ Cu in nutrient solution without apparent growth reduction, and up to $100{\mu}mol/L$ Cd and $400{\mu}mol/L$ Cu without critical visual injury. Up to 311mg/kg of Cd and 369mg/kg were accumulated in dried aerial part in A. trifida. In contrast, A. theophrasti showed injury at $400{\mu}mol/L$ Cu. Significant differences were shown in Cu accumulation among the four species. A. trifida had much higher concentrations of Cd in the shoot, whereas R, crispus accumulated higher concentrations of Cd in the shoot. Testing plant species showed reduced emergence rate with heavy metal treatment. When pH was lowered, the emergence and seedling growth were affected severely with heavy metal. We can suggested that A. trifida was the most proper species for phytoremediation in heavy metal-polluted regions.

  • Mode of safening action of N-(4-chlorophenyl)maleimide (CPMI) on metolachlor [2-chloro-N-(2-ethyl-6-methylphenyl)-N-(2-methoxy-l-methylethyl) acetamide] was investigated in sorghum(Sorghum bicolor L.). CPMI was synthesized by dehydration of N-(4-chlorophenyl)maleamic acid (CPMA) which was obtained from amination with maleic anhydride and 4-chloroaniline. Melting points of CPMA and CPMI (>95% purity) were $200-202^{\circ}C$ and $116-118^{\circ}C$, respectively. Growth response study indicated that seed treatment of CPMI increased tolerance of sorghum shoot to metolachlor approximately threefold. Sorghum shoot was more sensitive to injury caused by metolachlor and CPMI activity than the root. Metolachlor was initially absorbed by sorghum shoot and metabolized to the metolachlor-glutathione conjugate in CPMI-untreated and treated shoots. However, CPMI treatment significantly accelerated metabolism of $[^{14}C]$metolachlor in sorghum shoot, resulting in decrease in metolachlor content and increase in formation of the glutathione conjugate. It was concluded that the protection against metolachlor injury conferred by CPMI appeared to be correlated to detoxification of metolachlor in sorghum shoot tissue.

Effects of vitamin C as antioxidant on recovery of left ventricular function after ischemia and reperfusion in isolated rat heart (항산화제로서 비타민 C가 적출된 쥐심장에서 허혈 및 재관류후 좌심실 기능회복에 미치는 영향)

  • The large number of past investigation on extended myocardial protection clearly indicates that cold potassium cardioplegia and topical cooling have limited capabilities. Accordingly, more recent experimen- tal approaches have focused on the modalities of reperfusion and their implication on postischemic myo- cardial recovery. Oxygen may play a crucial role in the development of ischemic and reperfusion injury. Reactive oxygen radicals may be produced during ischemia or reperfusion after incomplete reduction of molecular oxygen or from other pathway and then induce fatal injury of the heart. The important obser- vation of oxygen-induced myocardial damage during reperfusion has led to the concept of applying oxy- gen free radical scavengers. So, this study is on dietary vitamin C supplementation as antioxidant in rats to determine whether or not they have a higher tolerance against cardiac ischemia-reperf'usion injury under Langendorff system. Male or female Sprague-Dawley rats (190-33Og) were randomly separated into two groups. Group A was not treated(n=10). Group B received vitamin C supplement (n=10). Experiment was performed 24 hours after vitamin C 200mg fed orally as injectable ascorbic acid. There were significant differences in contractile parameters between control and vitamin C-treated group. The RLVP (r te of post/preischemic left ventricular pressure) and Rdp/dt (rate of post/preischemic dp/dt) were significant statistically between two groups (p<0.05). But, RHR (rate of post/preischemic heart rate), time to first beat and sta'utilization were not significant. In conclusion, pretreatment with the antioxidant, ascorbic acid, was found to preserve left ventricular contractile function. But the precise mechanism of action of ascorbic acid has not as yet been determined, so further study will be required.

  • This study was carried out to investigate the specific activity fo peroxidase(POD) in conjunction with the phytotoxic effect of the tolerant soybean(Glycine max Merr.) and the susceptible crabgrass(Digitaria adscendens(L.) Scopol.) to acifluorfen. The POD activity increased during the growth of soybean and crabgrass seedlings. But after the foliar application of acifluorfen, the POD activity in the treated leaves was considerably higher than in controlled leaves, suggesting that soybean had higher tolerance to acifluorfen with growth. All the treated soybeans recovered from herbicidal injury in 14 days after treatment, while all the treated crabgrasses were dead with extreme necrosis and leaf burning. It was measured that the POD activity of crabgrass increased abruptly in 1 to 2 days after treatment and then significantly decreased in 5 days.

Involvement of Adenosine in Cardioprotective Effect of Catecholamine Preconditioning in Ischemia-Reperfused Heart of Rat

  • Preconditioning of a heart with small doses of catecholamines induces a tolerance against the subsequent lethal ischemia. The present study was performed to find a specific receptor pathway involved with the catecholamine preconditioning and to test if adenosine plays a role in this cardioprotective effect. Isolated rat hearts, pretreated with small doses of ${\alpha}-\;or\;{\beta}-adrenergic$ agonists/antagonists, were subjected to 20 minutes ischemia and 20 minutes reperfusion by Langendorff perfusion method. Cardiac mechanical functions, lactate dehydrogenase and adenosine release from the hearts were measured before and after the drug treatments and ischemia. In another series of experiments, adenosine $A_1\;or\;A_2$ receptor blockers were treated prior to administration of adrenergic agonists. Pretreatments of a ${\beta}-agonist,\;isoproterenol(10^{-9}{\sim}10^{-7}\;M)$ markedly improved the post-ischemic mechanical function and reduced the lactate dehydrogenase release. Similar cardioprotective effect was observed with an ?-agonist, phenylephrine pretreatment, but much higher $concentration(10^{-4}\;M)$ was needed to achieve the same degree of cardioprotection. The cardioprotective effects of isoproterenol and phenylephrine pretreatments were blocked by a ${\beta}_1-adrenergic$ receptor antagonist, atenolol, but not by an ${\alpha}_1-antagonist,$ prazosin. Adenosine release from the heart was increased by isoproterenol, and the increase was also blocked by atenolol, but not by prazosin. A selective $A_1-adenosine$ receptor antagonist, 1,3-dipropyl-8-cyclopentyl xanthine (DPCPX) blocked the cardioprotection by isoproterenol pretreatment. These results suggest that catecholamine pretreatment protects rat myocardium against ischemia and reperfusion injury by mediation of ${\beta}_1-adrenergic$ receptor pathway, and that adenosine is involved in this cardioprotective effect.

Comparison of Summer Turf Performance, Color, and Green Color Retention among Cool-Season Grasses Grown under USGA Soil System (USGA 지반구조에서 한지형 잔디의 여름 고온기 적응력, 색상 및 연중 녹색 유지기간 비교)

  • Research was initiated to investigate turf performance under USGA soil system. USGA system 45 centimeters deep was built with rootzone layer, intermediate layer, and drainage layer. Six turfgrass entries were comprised of 3 blends and 3 mixtures from cool-season grasses (CSG). Turfgrass color and quality ratings were best in spring and fall, especially early May to early July and late August to early November. Kentucky bluegrass (KB) consistently produced the greatest performance, while perennial ryegrass (PR) the poorest. Intermediate turf performance between KB and PR was observed with tall fescue (TF). Among CSG mixtures it increased with KB but decreased with PR. There were considerable variations in summer turf Performance. No summer drought injury was found in KB and TF. However, PR showed poor performance through summer as compared with others. Among mixtures, it decreased with PR. It was suggested that PR mix in less than $20\%$ in the mixtures to have an acceptable quality in summer. Cultural intensity also affected it. With lowering mowing height, KB of rhizomatous-type in growth habit kept good quality, while PR and TF with bunch-type in growth habit poor quality. Mowing quality was greatly different among CSG. KB produced clean-cut surface, but PR unclean one. If had an intermediate mowing quality between KB and PR. A great difference in green color retention was observed among CSG. The longest CSG was PR that kept green for 339 days, while the shortest one TF for 267 days. KB continued to keep green for 290 days. The mixtures kept green in color for 292 to 315 days, depending on turfgrass mixing intensity. The greater the PR in content, the longer the green color duration. These results demonstrate that KB was the best and PR the worst among CSG grown in USGA system under a domestic climate, in regards of turf quality, color, mowing quality, summer turf performance and green color duration. KB and TF are most adequate for high-maintenance and low-maintenance area, respectively. In case of mixtures for high-quality turf, it was desirable to use KB-based mixture with PR of below $20\%$ in seeding rate.

The Effect of Extracellular Glutamate Release on Repetitive Transient Ischemic Injury in Global Ischemia Model

  • During operations, neurosurgeons usually perform multiple temporary occlusions of parental artery, possibly resulting in the neuronal damage. It is generally thought that neuronal damage by cerebral ischemia is associated with extracellular concentrations of the excitatory amino acids. In this study, we measured the dynamics of extracellular glutamate release in 11 vessel occlusion(VO) model to compare between single occlusion and repeated transient occlusions within short interval. Changes in cerebral blood flow were monitored by laser-Doppler flowmetry simultaneously with cortical glutamate level measured by amperometric biosensor. From real time monitoring of glutamate release in 11 VO model, the change of extracellular glutamate level in repeated transient occlusion group was smaller than that of single occlusion group, and the onset time of glutamate release in the second ischemic episode of repeated occlusion group was delayed compared to the first ischemic episode which was similar to that of single 10 min ischemic episode. These results suggested that repeated transient occlusion induces less glutamate release from neuronal cell than single occlusion, and the delayed onset time of glutamate release is attributed to endogeneous protective mechanism of ischemic tolerance.
