Molecular Biological Studies on the Stress Protein HSP90$\beta$ Gene from Flounder (Paralichthys olivaceus) (넙치(Paralichthys olivaceus) HSP90$\beta$ 유전자의 분자생물학적 연구)

  • Heat shock proteins (HSPs) were induced in cells in the thermal stress, and the HSP90 family is one of the major classes of HSPs. Gene encoding HSPs have been characterized from various mammals and piscine. We have cloned and sequenced the HSP90 cDNA from a brain cDNA library constructed from flounder (Paralichthys oliThe result of sequence analysis shows it to be the HSP90~. The nucleotide sequence of the HSP90$\beta$ was composed of 2791 long, encoding 726 amino acid residues. The flounder hsp90$\beta$ gene showed very high sequence homology with hsp90f3 of European sea bass (96.6%), zebrafish (92.9%), Atlantic salmon (92.0%) and human (89.5%). We also constructed a phylogenetic tree based on HSP90 amino acid sequences from vertebrate species. Gene-specific primers were selected and used in RT-PCR reactions to measure the basal hsp90$\beta$ mRNA. The hsp90f3 gene is constitutively expressed at a fairly high level in all examined tissues (brain, liver, kidney, muscle, and spleen). In order to express protein of flounder hsp90$\beta$ in E. coli, we used the His-tagged pETvector. Then, the expression of flounder HSP90$\beta$ was confirmed by Western blot analysis.

Postbuckling response and failure of symmetric laminated plates with rectangular cutouts under in-plane shear

  • This paper deals with the buckling and postbuckling responses, and the progressive failure of square laminates of symmetric lay-up with a central rectangular cutout under in-plane shear load. A detailed investigation is made to show the effects of cutout size and cutout aspect ratio on the buckling and postbuckling responses, failure loads and failure characteristics of $(+45/-45/0/90)_{2s}$, $(+45/-45)_{4s}$ and $(0/90)_{4s}$ laminates. The 3-D Tsai-Hill criterion is used to predict the failure of a lamina while the onset of delamination is predicted by the interlaminar failure criterion. In addition, the effects of boundary conditions on buckling loads, failure loads, failure modes, and maximum transverse deflection for a $(+45/-45/0/90)_{2s}$ laminate with and without a square cutout have been presented. It is concluded that because of early onset of delamination at the net section of cutouts before first-ply failure, total strength of the laminate with very small cutouts can not be utilized.

  • The genres of fantastic of Korean films created in the 90s are diverse. The fantastics of these works are not manifested only in 'Ghosts' movies. For example, If the film Mystery Of The Cube, which is based on the mystery of Lee Sang, has a story of structuralist fantasy, we can say that a movie like Tell Me Something has a psychologically conventional fantastic. This study examines the fantastic of contemporary Korean films through 'completion of allegory based on realism'. For this, we borrows the fantastic concept of Etienne Souriau. In some films, fantastics can be found directly in the developing nature. In this case, the human being who justifies their domination of nature by upgrading their position as the objectifying subject of nature becomes 'alienated' in return. The notion of this alienation in film narratives is often revealed through allegories, particularly in the manifestation of fantasy by Étienne Souriau in the early 'personal experience'. Kim kiduk's film is a representative example of allegorizing elements of society through individual experience. Focusing on Kim Giduk's Bad Guy, this essay analyzes the process in which fantastic films of Korea in the 90s build up aesthetic fantastic through imitation of society.

In vitro Evaluation of Antimicrobial Agents Susceptibility Against Several Clinical Isolates (임상 분리 균주의 항생제 감수성 유형)

  • In vitro activity of commonly used antimicrobial agents against several clinical isolates were studied. In the case of E. coli, the MICs at which 90% of the bacteria are inhibited of ampicillin, Unasyn, cefazoline, cefotaxim, carbenicillin, gentamicin and ofloxacin were 100<, 100, 25, 0.2, 100<, 3.13, and $12.5{\;}\mu\textrm{g}/m$ , respectively. In the case of K . pneumoniae, the MICs at which 90% of the bacteria are inhibited of ampicillin, Unasyn, crfazoline, cefotaxim, carbenicillin, gentamicin and ofloxacin were 100<, 12.5, 100<, 0.1, 100<, 1.6, and $0.4{\;}\mu\textrm{g}/m$ , respectively. In the case of Enterobacter sp, the MICs at which 90% of the bacteria are inhibited of ampicillin, Unasyn, cefazoline, cefotaxim, carbenicillin, gentamicin and ofloxacin were 100<, 100, 100<, 6.25, 100<, 100 and $1.57{\;}\mu\textrm{g}/m$ , respectively. In the case of Acinetobacter sp, the MICs at which 90% of the bacteria are inhibited of ampicillin, Unasyn, cefazoline, cefotaxim, carbenicillin, gentamicin and ofloxacin were 100<, 100<, 100<, 100<, 100< 100< and $50{\;}\mu\textrm{g}/m$ , respectively. In the case of Pseudomonas sp, the MICs at which 90% of the bacteria are inhibited of ampicillin, Unasyn, cefazoline, cefotaxim, carbenicillin, gentamicin and ofloxacin were 100<, 100<, 100<, 50, 100<, 25 and $25{\;}\mu\textrm{g}/m$, respectively. In the case of S. aureus, the MICs at which 90% of the bacteria are inhibited of ampicillin, Unasyn, cefazoline, cefotaxim, carbenicillin, gentamicin and ofloxacin were 50, 50, 100<, 100<, 50, 50, and $100{\;}\mu\textrm{g}/m$, respectively.

  • Ornithine decarboxylase(ODC) is an indicate enzyme in carcinogenesis. We divided Sprague Dawley rats into six groups: control, electric field exposure, X-ray only irradiation, X-ray irradiation with electric field exposure group, Sr-90 injected group and Sr-90 and electric field complex exposure group. The ODC activity was measured in rat's bone marrow cell every week. The results were summarized as follows: The ODC activitied was increased in X-ray irradiated, Sr-90 injected and Sr-90 and electric field complex exposed group as compared with that of control(p<0.05). The ODC activity was increased comparing that of control neither in X-ray and electric field complex exposed group nor electric field only exposed group. This result suggests that the electric field doesn't have myeloid carcinogenicity and myeloid cancer incidence caused by ionized radiation is suppressed by electric field exposure.

  • The purpose of this study is to investigate the content and changes of modern Korean men's wear advertisement from the 1960s to the 1990s by reviewing fashion magazines and pictorials. Research problems were as follows.; First, analyze the types and their units of measurement in men's fashion advertisements from 1962 to 1998. Second, examine the differences of quantitative change in men's fashion advertisements according to each period. The total 857 advertisements were selected from Shin Dong-A, Joongang Magazine, and Bokjangwolbo from 1972 to 1998. For analyze the result, frequency analysis and $x^2$-test were used. Results were as follows. First, through the review of literatures and pre-tests, 6 units of measurement were identified. They were product type(formal wear, casual wear, sports wear, dress shirts, accessory, inner wear, fabric and the others), brand type(national brand, licensed brand, imported brand, others), appeal type(image, product, others), representation type(photograph, illustration, others), medium type(man, product, others), and color type of advertisement(black & white picture, color picture). Second, for the types of advertisements, formal wear, national brand, image appeal, figure medium, photograph for presentation and color picture are the most frequently shown in men's fashion advertisements from 1962 to 1998. Third, for product, more accessory advertisements were found than the clothing in the 60s and the 70s, and casual wear advertisements in the 80s and formal wear advertisements in the 90s were dominated. National brand type was dominant through all four period. Product appeal type in the 60s and the 70s and image appeal type in the 80s and the 90s were prevailed. For medium, product type in the 60s and the 70s and figure type in the 80s and the 90s were dominated. Black & white picture in the 60s and the 70s and color picture in the 80s and the 90s were prevailed. Lastly, photograph was the most frequently used for presentation type through all periods.

Test Anxiety Investigation of Students in Oriental Medicine College by Using Korea's version-Test Anxiey Inventory, State-Trait Anxiety Inventory, Symptom Check List-90-Revision (시험불안척도, 상태-특성불안 척도, 간이정신진단검사를 이용한 한의학과 대학생의 시험불안 특성)

  • Objective : The aim of this study is to investigate and measure the Test-anxiety of college students in OO Oriental Medicine by using Korea's version-Test Anxiey Inventory(K-TAI-K), State-Trait Anxiety Inventory(STAI), Symptom Check List-90-Revision(SCL-90-R), etc. Methods : We studied 181 students who being at OO College of Oriental Medicine in second semester, 2007 We used some questionaries like K-TAI-K, STAI, SCL-90-R , College Life Satisfacaion, Life Event Questionnaire. Results and Conclusions : 1.141 persons(83,43%) of students feel the largest test anxiety before test. 18 persons(9.94%) of students feel the largest test anxiety after test. 2.The 1st grade students of preparatory course and 4th grade students of regular course have the lowest score in State Anxiety Inventory and obsessive-compulsive of SCL-90-R. The 2nd grade students of regular course have the lowest score in State Anxiety Inventory and obsessive-compulsive of SCL-90-R 3. Students who failed in test one time have the lowest score , and who failed in test more than two times have the highest score in K-TAl-K Students who failed one time in grade promotion more than two times showed high score in second K-TAI-K. 4. The higher grade, the lower score in College Life Satisfaction. The scores of College Life Satisfaction have negative correlations with those of K-TAI-K, STAI SCL-90-R except phobic anxiety, 5. The scores of Life Event Questionnaire have positive correlations with those of K-TAI-K, STAI and SCL-90-R except phobic anxiety. 6. The scores of K-TAl-K have positive correlations with those of STAI, SCL-90-R, College Life Satisfaction and Life Event Questionnaire. A correlation coefficient of obsessive-compulsive scale is highest, anxiety scale is second, interpersonal sensitivity scale is third, depression scale is fourth high among the scales of SCL-90-R.

Characteristics of α-Amylase and Protease Produced from Bacillus amyloliquefacies CNL-90 Isolated from Malt Grain (맥아에서 분리한 Bacillus amyloliquefacies CNL-90이 생산하는 α-amylase와 Protease의 특성)

  • A bacterium, identified as $Bacillus$ $amyloliquefacies$, CNL-90 using 16S rDNA analysis, was isolated from malt grain. The optimal activities of its ${\alpha}$-amylase and protease were observed at pH 6 and $60^{\circ}C$, and at pH 6 and $50^{\circ}C$, respectively although their activities remained stable at pH 7 and $40^{\circ}C$for ${\alpha}$-amylase and at pH 7 and $50^{\circ}C$ for protease. After solid-state fermentation of $B.$ $amyloliquefacies$, CNL-90 on wheat bran for 72hr or 144hr, the ${\alpha}$-amylase and protease activities were 170,000 and 290,000 units/kg, and 290,000 and 310,000 units/kg, respectively. The viable bacterial cell counts were $1.5{\times}10^9$ CFU/g and $2.2{\times}10^9$ CFU/g at 72hr and 144hr of the solid-state fermentation, respectively. A feeding trial with a total of 127 piglets was also conducted. The animals were divided into two groups: an experimental group fed with the fermented product (63 piglets) and a control group (64 piglets). The growth rate of the experimental group was 6.66% higher than that of the control group (P<0.05). The results of this study indicate that the ${\alpha}$-amylase and protease from $B.$ $amyloliquefacies$, CNL-90 can be used for industrial applications due to their activity in production of carbohydrate hydrolysates.

STRAIN CHANGES OF ACRYLIC RESIN SPECIMENS CURED BY THREE CURING CYCLES

  • The acrylic resin was first introduced as denture base materials in 1937 and it is commonly used for denture base fabrication nowadays. Three different curing cycles (Conventional curing cycle, short curing cycle and long curing cycle) and three commercially available heat-activated acrylic resins (Vertex RS, Lucitone 199 and ProBase Hot) were investigated to find the curing cycle and material that showed the minimum shrinkage of the resin during polymerization process. A brass master mold was fabricated and duplicated by additional silicone impression material. Stone molds were made by pouring of type III dental stone (SILKY-ROCK YELLOW, Whip-Mix, Louisville, Kentucky). It was embedded in the flask. Strain gauge and thermocouple were embedded in the specimen. Strain gauge and thermocouple were connected to signal conditioning amplifier and data was recorded by pre-programmed software. The parameters ESmax (Maximum expansion strain), Sb (Strain measured just before deflasking procedure), Sa (Strain measured just after deflasking procedure) and Sf (Strain measured at the end of the experiment) were measured. ${\Delta}$S was calculated from Sb and Sa (${\Delta}$S=Sb-Sa). In the experiment concerned about materials, the parameters 90-ESmax (Maximum expansion strain measured during early 90 minutes of curing procedure), 180-ESmax (Maximum expansion strain measured from 90 minutes to 180 minutes), Sb, Sa, ${\Delta}$S and Sf were measured and the following conclusions were made. 1. The ESmax value of conventional curing cycle showed the largest value and the 180-ESmax value of Lucitone 199 showed the smallest value. 90-ESmax values showed no significant difference (p<0.05). 2. ${\Delta}$S values of conventional curing cycle showed the positive values. ${\Delta}$S values of short curing cycle and long curing cycle showed the negative values. All three materials cured by conventional curing cycle showed the positive values. 3. The Sf values of long curing cycle and ProBase Hot (cured by conventional curing cycle) showed the smallest values.

The Neutrophil-to-Lymphocyte Ratio as a Predictor of Postoperative Outcomes in Patients Undergoing Coronary Artery Bypass Grafting

  • Background: The neutrophil-to-lymphocyte ratio (NLR) has been suggested as a novel predictive marker of cardiovascular disease. However, its prognostic role in patients under-going coronary artery bypass grafting (CABG) is unclear. This study aimed to determine the association between the preoperative NLR and early mortality in patients undergoing CABG. Methods: Cardiac surgery was performed in 2,504 patients at Seoul St. Mary's Hospital from January 2010 to December 2021. This study retrospectively reviewed 920 patients who underwent isolated CABG, excluding those for whom the preoperative NLR was unavailable. The primary endpoints were the 30- and 90-day mortality after isolated CABG. Risk factor analysis was performed using logistic regression analysis. Based on the optimal cut-off value of preoperative NLR on the receiver operating characteristic curve, high and low NLR groups were compared. Results: The 30- and 90-day mortality rates were 3.8% (n=35) and 7.0% (n=64), respectively. In the multivariable analysis, preoperative NLR was significantly associated with 30-day mortality (odds ratio [OR], 1.28; 95% confidence interval [CI], 1.17-1.39; p<0.001) and 90-day mortality (OR, 1.17; 95% CI, 1.07-1.28; p<0.001). The optimal cut-off value of the preoperative NLR was 3.4. Compared to the low NLR group (<3.4), the high NLR group (≥3.4) showed higher 30- and 90-day mortality rates (1.4% vs. 12.1%, p<0.001; 2.8% vs. 21.3%, p<0.001, respectively). Conclusion: Preoperative NLR was strongly associated with early mortality after isolated CABG, especially in patients with a high preoperative NLR (≥3.4). Further studies with larger cohorts are necessary to validate these results.
