Cytotoxic Activity from Curcuma zedoaria Through Mitochondrial Activation on Ovarian Cancer Cells

  • ${\alpha}$-Curcumene is one of the physiologically active components of Curcuma zedoaria, which is believed to perform anti-tumor activities, the mechanisms of which are poorly understood. In the present study, we investigated the mechanism of the apoptotic effect of ${\alpha}$-curcumene on the growth of human overian cancer, SiHa cells. Upon treatment with ${\alpha}$-curcumene, cell viability of SiHa cells was inhibited > 73% for 48 h incubation. ${\alpha}$-Curcumene treatment showed a characteristic nucleosomal DNA fragmentation pattern and the percentage of sub-diploid cells was increased in a concentration-dependent manner, hallmark features of apoptosis. Mitochondrial cytochrome c activation and an in vitro caspase-3 activity assay demonstrated that the activation of caspases accompanies the apoptotic effect of ${\alpha}$-curcumene, which mediates cell death. These results suggest that the apoptotic effect of ${\alpha}$-curcumene on SiHa cells may converge caspase-3 activation through the release of mitochondrial cytochrome c.

Hypoglycemic Effect of Padina arborescens Extract in Streptozotocin-induced Diabetic Mice

  • This study investigated the hypoglycemic effect of the Padina arborescens extract in STZ-induced diabetic mice. Freeze-dried Padina arborescens were extracted with 80% methanol and concentrated for use in this study. The hypoglycemic effect was determined by inhibitory activities against ${\alpha}$-glucosidase and ${\alpha}$-amylase as well as the alleviation of postprandial blood glucose level. Padina arborescens extracts showed higher inhibitory activities than acarbose, a positive control against ${\alpha}$-glucosidase and ${\alpha}$-amylase. The $IC_{50}$ values of Padina arborescens extracts against ${\alpha}$-glucosidase and ${\alpha}$-amylase were 0.26 and 0.23 mg/mL, respectively, which evidenced as more effective than observed with acarbose. The increase of postprandial blood glucose levels were significantly suppressed in the Padina arborescens extract administered group than the control group in the streptozotocin induced diabetic mice. Furthermore, the area under the curve (AUC) was significantly lowered via Padina arborescens extract administration in diabetic mice (p 0.05). These results indicated that the Padina arborescens extract might be used as an inhibitor of ${\alpha}$-glucosidase and ${\alpha}$-amylase and delay absorption of dietary carbohydrates.

Anti-inflammatory and utero-relaxant effect of α-bisabolol on the pregnant human uterus

  • The aim of this study was to evaluate the in vitro anti-inflammatory and utero-relaxant effect of ${\alpha}$-bisabolol on the pregnant human myometrium. Samples from the pregnant human myometrium were used in functional tests to evaluate the inhibitory effect of ${\alpha}$-bisabolol (560, 860, 1,200 and $1,860{\mu}M$) on spontaneous myometrial contractions. The intracellular cyclic adenosine monophosphate (cAMP) levels generated in response to ${\alpha}$-bisabolol in human myometrial homogenates were measured by ELISA. The anti-inflammatory effect of ${\alpha}$-bisabolol was determined through the measurement of two pro-inflammatory cytokines, tumor necrosis factor-${\alpha}$ ($TNF{\alpha}$) and interleukin $(IL)-1{\beta}$, and the anti-inflammatory cytokine IL-10, in pregnant human myometrial explants stimulated with lipopolysaccharide (LPS). Forskolin was used as a positive control to evaluate the cAMP and cytokine levels. ${\alpha}$-Bisabolol was found to induce a significant inhibition of spontaneous myometrial contractions at the highest concentration level (p<0.05). ${\alpha}$-Bisabolol caused a concentration-dependent decrease in myometrial cAMP levels (p<0.05) and a concentration-dependent decrease in LPS-induced $TNF{\alpha}$ and $IL-1{\beta}$ production, while IL-10 production did not increase significantly (p>0.05). The anti-inflammatory and utero-relaxant effects induced by ${\alpha}$-bisabolol were not associated with an increase in cAMP levels in pregnant human myometrial samples. These properties place ${\alpha}$-bisabolol as a potentially safe and effective adjuvant agent in cases of preterm birth, an area of pharmacological treatment that requires urgent improvement.

토코페롤류의 항산화작용과 Linoleic Acid Methylester에서 생성된 cis/trans-, trans/trans-Hydroperoxide Isomer (Antioxidant Effect of Tocopherols and Tocotrienols and cis/trans-, trans/trans-Hydroperoxide Isomer from Linoleic Acid Methylester)

  • Linoleic acid methylester와 토코페놀류$({\alpha}-,\;{\beta}-,\;{\gamma}-,\;{\delta}-tocopherol$${\alpha}-,\;{\gamma}-,\;{\delta}-tocotrienol)$를 사용한 model system에서의 자동산화과정에서 온도$(40,\;60,\;80^{\circ}C)$와 산소의 양 $(0,\;10,\;20%\;O_2)$에 따르는 항산화효과를 고찰하였다. Linoleic acid methylester에서 생성된 주요 산화생성물인 13-cis/trans-, 13-trans/trans-, 9-trans/cis-, 9-trans/trans-hydroperoxide를 HPLC를 이용하여 분석하였으며, 그 양적 변화로 항산화정도를 비교하였다. 모든 토코페롤류$({\alpha}-tocotrienol$제외)는 $60^{\circ}C$ 범위에서 항산화효과가 있었다. ${\alpha}-tocopherol$, ${\alpha}-tocotrienol$ and ${\delta}-tocotrienol$의 경우 $80^{\circ}C$ 범위에서 산화촉진효과가 관찰되었다. $40^{\circ}C$ 범위에서는 주로 cis/trans-hydroperoxide가 많이 생성되었고 $80^{\circ}C$ 범위에서는 trans/trans-hydroperoxide가 많이 생성되었다. $80^{\circ}C$ 범위의 hydroperoxide 생성량간의 오차가 심한 구간을 제외한 $40^{\circ}C/10%,\;O_2$에서부터 $60^{\circ}C/20%,\;O_2$까지의 산화구간에서 cis/trans-: trans/trans-hydroperoxides의 비율은 ${\alpha}-T>{\alpha}-T_3>{\gamma}-T>{\beta}-T>{\gamma}-T_3>{\delta}-T>{\delta}-T_3$의 순으로 고찰되어, 토코페롤류 중 ${\alpha}-tocopherol$이 가장 낮은 항산화효과가 있는 것으로 나타났다.

Spinal Noradrenergic Modulation and the Role of the Alpha-2 Receptor in the Antinociceptive Effect of Intrathecal Nefopam in the Formalin Test

  • Background: Nefopam has shown an analgesic effect on acute pain including postoperative pain. The reuptake of monoamines including serotonin and noradrenaline has been proposed as the mechanism of the analgesic action of nefopam, but it remains unclear. Although alpha-adrenergic agents are being widely used in the perioperative period, the role of noradrenergic modulation in the analgesic effect of nefopam has not been fully addressed. Methods: Changes in the antinociceptive effect of intrathecal (i.t.) nefopam against formalin-elicited flinching responses were explored in Sprague-Dawley rats pretreated with i.t. 6-hydroxydopamine (6-OHDA), which depletes spinal noradrenaline. In addition, antagonism to the effect of nefopam by prazosin and yohimbine was evaluated to further elucidate the antinociceptive mechanism of i.t. nefopam. Results: Pretreatment with i.t. 6-OHDA alone did not alter the flinching responses in either phase of the formalin test, while it attenuated the antinociceptive effect of i.t. nefopam significantly during phase 1, but not phase 2. The antagonist of the alpha-2 receptor, but not the alpha-1 receptor, reduced partially, but significantly, the antinociceptive effect of i.t. nefopam during phase 1, but not during phase 2. Conclusions: This study demonstrates that spinal noradrenergic modulation plays an important role in the antinociceptive effect of i.t. nefopam against formalin-elicited acute initial pain, but not facilitated pain, and this action involves the spinal alpha-2 but not the alpha-1 receptor.

인지질 Loposome 에 미치는 대두 Saponin의 항산화효과 (Antioxidant Effect of Soyasaponin on the Liposomal Phospholipid Membrane)

  • Egg 인지질 liposome에 미치는 대두 saponin의 항산화 작용에 대한 영향을 흡광분석법으로 검토하였다. ${\alpha}-tocopherol$이 함유된 egg 인지질 liposomed의 산화지수와 산화속도가 순수 eff 인지질 liposome의 산화지수와 산화속도에 비하여 현저하게 감소되었다. Liposome내 함유된 ${\alpha}-tocopherol$은 순수 egg 인지질 liposome의 산화를 지연시켰다. 특히 대두 saponine은 egg 인지질 liposome에 대한 ${\alpha}-tocopherol$의 항산화 효과를 촉진시켰다. 이러한 결과로 보아 대두 saponine이 egg 인지질 liposome에 대한 ${\alpha}-tocopherol$의 항산화 작용에 영향을 미치는 것으로 사료된다.

Effect of the Volatile Oil of Nigella sativa Seeds and Its Components on Body Temperature of Mice: Elucidation of the Mechanisms of Action

  • The effect(s) of the volatile oil (VO) of Nigella sativa and its two components, ${\alpha}-pinene$ and ${\rho}-cymene$ on body temperature of male and female conscious mice were studied. Further investigations to delineate the mechanism(s) of action of the observed effect(s) by using various blockers involved in the central regulation of body temperature were made. VO and ${\alpha}-pinene$ caused significant reductions in rectal body temperature at is and 30 minute after treatment. ${\rho}-cymene$ had negligible effect on body temperature of mice. Cyproheptadine inhibited VO and ${\alpha}-pinene-induced$ hypothermia significantly. Nalbuphine inhibited ${\alpha}-pinene-induced$ hypothermia significantly but did not affect VO-induced hypothermia. Droperidol potentiated VO and ${\alpha}-pinene-induced$ hypothermia to a non-significant level; whereas atropine potentiated VO-induced hypothermia non-significantly. The study confirms further the role of serotoninergic receptors in the mechanism(s) of the observed pharmacological effects of the VO of Nigella sativa. It also indicated a possible role of opioid receptors in ${\alpha}-pinene-induced$ hypothermia.

  • The effect of salts on the formation of ${\alpha}$-hemihydrated gypsum at boiling temperature under atmospheric pressure was studied by the solubility measurement method, and the formation of ${\alpha}$-hemihydrated gypsum from by-product gypsum of phosphoric acid process in the salts solution were investigated. The order of catalytic effect of salt on the formation of ${\alpha}$-hemihydrated gypsum are as follows: NH4Cl>NaCl>NaNO3. In the salts solution of sodium nitrate, sodium chloride, and ammonium chloride, prismatic ${\alpha}$-hemihydrated gypsum was obtained and the crystal form was converted to needle form in complex solution with sulfuric acid. The P2O5 content in gypsum was largely decreased in this atmosphoric solution process.

  • When the reaction rate constant k of phospholipase D on liposome was measured by the ANS fluorometry, k of phospholipase D on DMPC liposome which was made of L-$\alpha$-PI, cholesterol and L-$\alpha$-PS decreased than that of phospholipase D on DMPC liposome with cholesterol or with PI and cholesterol. Optimal $Ca^{2+}$ concentration, the most important factor on effect of phospholipase D, also decreased to 1mM, as compared with 10mM and 60mM respectively when cholesterol and PI were added, and cholesterol only was added. The change of cholesterol Mol% had a great influence on k value of phospholipase D. But in case of addition of L-$\alpha$-PS to cholesterol, the influence was relatively diminished.

한국산 겨우살이 추출물 M11C (렉틴 구성물질)가 단구세포의 $TNF-\alpha$ 유전자 발현유도 및 분비에 미치는 효과 (Effect of Korean Mistletoe Extract M11C (Non-lectin Components) on the $TNF-\alpha$ Expression and Secretion from Human Peripheral Blood Monocytes)

  • 한국산 겨우살이 (Viscum album)는 면역 활성효과와 항암효과가 있다는 것으로 잘 알려져 있다. 본 연구에서는 한국산 겨우살이 추출물 M11C (비렉틴 구성물질)가 사람의 말초혈액 단구를 활성화시켜 tumor necrosis factor-alpha $(TNF-\alpha)$를 생산 분비하게 하는지를 규명하기 위해 실험에 이용되었다. 단구로부터 $TNF-\alpha$의 생산에 있어 M11C의 효과를 알기 위해 단구를 여러 농도 $(0-2000{\mu}g/m\ell)$의 M11C로 0.5-24시간 동안 자극한 후 배양액 MCM을 수거했다. 수거한 배양액 MCM을 $TNF-\alpha$에 민감한 L929 세포에 첨가한 후 L929 세포의 독성 정도를 MTT 기법으로 검사하였는데, 배양액 MCM이 L929 독성효과를 가졌으며, 이 MCM의 L929 세포 독성효과는 $TNF-\alpha$ 항체에 의해 거의 완전하게 억제되었다. 이러한 결과는 배양액 MCM에 $TNF-\alpha$가 존재함을 지적했으며, 또한 M11C가 단구를 자극해 $TNF-\alpha$를 생산하리라고 암시했었다. 이러한 암시를 확인하기 위해 immunoblotting 기법을 사용해 배양액 MCM에 존재하는 $TNF-\alpha$을 검사를 수행했다. 검사결과 배양액 MCM에 많은 량의 $TNF-\alpha$가 검출되었다. 이러한 $TNF-\alpha$ 검출량은 단구를 자극한 M11C의 농도와 자극시간에 비례해서 증가했다. 즉, 단구로부터 $TNF-\alpha$의 생산 분비량은 M11C의 농도와 자극시간에 의존적이었다. 이러한 M11C의 농도와 자극시간의 의존적 효과는 단구로부터 $TNF-\alpha$ mRNA 발현에도 같은 경향을 보였다. 결론적으로 한국산 겨우살이로부터 열탕 추출한 M11C가 면역활성제로 작용할 수 있음을 시사함과 동시에 한방탕제로 사용되는 한국산 겨우살이가 면역학적 효능을 가지고 있음을 말해주고 있다.
