Short-Term High Expression of Interferon-Alpha Modulates Progression of Type 1 Diabetes in NOD Mice

  • Type I diabetes (T1D) is an organ-specific autoimmune disease caused by the T cell-mediated destruction of the insulin-producing ${\beta}$ cells in the pancreatic islets. The onset of T1D is the consequence of a progressive destruction of islet ${\beta}$ cells mediated by an imbalance between effector $CD4^+$ T helper (Th)1 and regulatory $CD4^+$ Th2 cell function. Since interferon-alpha (IFN-${\alpha}$) has been known to modulate immune function and autoimmunity, we investigated whether administration of adenoviralmediated IFN-${\alpha}$ gene would inhibit the diabetic process in NOD mice. The development of diabetes was significantly inhibited by a single injection of adenoviral-mediated IFN-${\alpha}$ gene before 8 weeks of age. Next, we examined the hypothesis that Th2-type cytokines are associated with host protection against autoimmune diabetes, whereas Th1-type cytokines are associated with pathogenesis of T1D. The expression of IFN-${\alpha}$ induced increase of serum IL-4 and IL-6 (Th2 cytokines) levels and decrease of serum IL-12 and IFN-${\gamma}$ (Th1 cytokines) levels. Therefore, overexpression of IFN-${\alpha}$ by adenoviralmediated delivery provides modulation of pathogenic progression and protection of NOD mice from T1D.

Ochnaflavone의 면역보조제 효과 (Effect of Ochnaflavone as An Immunoadjuvant)

  • In this present study, we determined whether or not there is an immunoadjuvant effect of ochnaflavone, a biflavone isolated from Lonicera japonica. As an antigenic source, the cell wall (CACW) of Candida albicans, a fungal pathogen, was used. CACW consists of 95% carbohydrate (mannan). In the experiments, BALB/c mice were immunized with emersion forms of CACW combined with or without ochnaflavone (Och) in the presence of IFA containing mineral oil or CACW alone. Then, the amounts of antisera collected from these mice groups were measured by the ELISA method. Data from these experiments showed that CACW combined with Och (CACW/Och/IFA) provoked the production of antisera app. 2.2 or 5 times more than the corresponding CACW/IFA or CACW alone (CACW/DPBS), respectively, in mice (P<0.05). We further examined the immune response type induced by Och. Analysis of the values of the IgG1/IgG2a ratios obtained from IgG isotyping revealed that Och induced Th2-immunity more dominantly than Th1. This finding was confirmed by cytokine profile. CACW/Och/IFA formulation induced IL-4 (Th2-type cytokine) more than IFN${\gamma}$ (Th1-type cytokine) as compared with CACW/IFA and CACW/DPBS formulations (P<0.05). All data combined, Och appears to have an immunoadjuvant activity that may convert Th1 immunity into Th2 immunity.

Impaired Memory in OT-II Transgenic Mice Is Associated with Decreased Adult Hippocampal Neurogenesis Possibly Induced by Alteration in Th2 Cytokine Levels

  • Recently, an increasing number of studies have focused on the effects of CD4+ T cell on cognitive function. However, the changes of Th2 cytokines in restricted CD4+ T cell receptor (TCR) repertoire model and their effects on the adult hippocampal neurogenesis and memory are not fully understood. Here, we investigated whether and how the mice with restricted CD4+ repertoire TCR exhibit learning and memory impairment by using OT-II mice. OT-II mice showed decreased adult neurogenesis in hippocampus and short- and long- term memory impairment. Moreover, Th2 cytokines in OT-II mice are significantly increased in peripheral organs and IL-4 is significantly increased in brain. Finally, IL-4 treatment significantly inhibited the proliferation of cultured adult rat hippocampal neural stem cells. Taken together, abnormal level of Th2 cytokines can lead memory dysfunction via impaired adult neurogenesis in OT-II transgenic.

Polyphenol-rich Sargassum horneri alleviates atopic dermatitis-like skin lesions in NC/Nga mice by suppressing Th2-mediated cytokine IL-13

  • Atopic dermatitis (AD) is one of major skin inflammatory diseases characterized by excessive Th2-mediated immune responses. Recent evidence provides that interlukin-13 (IL-13) plays the role of a key Th2 cytokine that drives the inflammation underlining AD. Due to adverse effects of commercially available synthetic drugs, the need for treatments based on natural products is gaining much attention. Sargassum horneri is an edible brown algae known for beneficial bioactivities including anti-inflammation. We investigated if polyphenol-rich S. horneri extracts (SHE) could suppress AD-like skin lesions in NC/Nga mice and if that involved inhibition of the infiltration of Th2-mediated cytokine IL-13. We observed markedly increased infiltration of IL-13 positive cells in AD-like skin lesions of mice but SHE treatments decreased it. Also, the dermal expression of IL-13 was sufficient to cause inflammatory responses in mice skin resembling human AD. SHE suppressed the dermal infiltration of inflammatory cells where IL-13 plays a crucial role in skin tissues and in the recruitment of inflammatory cells. Furthermore, it was confirmed that SHE reduced T cell, dendritic cell, and macrophage populations in spleen. Moreover, SHE decreased the collagen deposition in skin and ear dermis resulting in reduced fibrosis that occurs in AD due to excessive collagen. Taken together, our results reveal that SHE suppressed the infiltration of inflammatory cells into skin dermis by decreasing the infiltration of IL-13 positive cells. Therefore, SHE could be taken as a useful therapeutic agent to alleviate AD.

원발성 폐암 154례에서의 수술율 및 술후 생존 (An Analysis of the 154 Cases of Lung Cancer)

  • During the period of 10 years from July, 1976 to July, 1986, 154 cases of primary carcinoma of the lung - by the cell type, stage, operability, and survival rate in the resectable cases - are analyzed at the Dept. of Thoracic Surgery, Paik Hospital in Seoul. The results are as follows: 1] Histopathological types are squamous cell carcinoma 49% [76 cases], adenocarcinoma 25% [39 cases], undifferentiated large cell carcinoma 9% [14 cases], undifferentiated small cell carcinoma 6% [9 cases], bronchioloalveolar carcinoma 4% [6 cases] and adenosquamous carcinoma 3% [4 cases]. 2] Peak incidence is observed in the 4th decade of life [33%], then 5th [29%] and 3rd [21%] respectively. Male to female ratio is 4 to 1. 3] Evidence of inoperability is observed in 64% [99 cases] by clinical staging workup. Thirty six percent [55 cases] were operated. Of these, post-surgical stage I was 5% [3 cases], stage II, 64% [35 cases] and stage III, 31% [17 cases]. Among total 17 cases of stage III, 14 cases were unresectable with evidence of T2N2M0, while 3 cases were resectable. Resectability is 27%, [41 cases] from the total number of 154 cases. And the resectability for the ex 55 cases is 75% [41 cases]. 4] By cell type, highest resectabitity is the squamous cell carcinoma, 49% [20 cases]. Adenocarcinoma is 32% [13 cases] and bronchioloalveolar, 12% [5 cases]. 5] Survival rate is evaluated for 38 cases of 41 resectable stage I, II and III. Overall 5 year survival rate is 24%, 3 year 32% and 10 year 8%. Survival rate in stage II for 5 year is 25%. In squamous cell type for, 5 year is 42%. Authors believe when surgeons continuous effort of early detection is met with patients early visit, 5 year survival rate for the stage I K II resectable patients will improve more effectively. As well, When the efforts are added to combined modality with radiotherapy and chemotherapy for the stage III selected cases of non-small cell carcinoma patients, the enhancement in survival rate is expected.

Ovalbumin으로 유도한 천식 생쥐모델에서 lutein의 Th2 면역반응 연구 (Lutein Modulates Th2 Immune Response in Ovalbumin-Induced Airway Inflammation)

  • Lutein은 식물에서 발견되는 carotenoid 계열에 속하는 물질로 항산화 기능을 가지고 있는 것으로 널리 알려져 있지만, 호흡기 질환과 관련하여 아직까지 lutein의 효능과 작용 기작이 잘 알려져 있지 않다. Ovalbumin (OVA)으로 유도한 천식(asthma) 생쥐모델에서 lutein은 기도 과민성을 억제하였고, 기관지 폐포 세척액에서 OVA의 감작에 의하여 증가한 각종 염증성 지표들을 감소시켰다. 또한, OVA의 감작에 의하여 증가한 제2형 협조 T 세포(Th2 cell)의 증가된 반응을 약화시키는 결과를 볼 수 있었다. 본 실험에서 lutein이 ovalbumin (OVA)으로 유도한 천식 생쥐 모델에서 제 2형 협조 T세포의 싸이토카인과 유전자 발현을 조절할 수 있는 면역약리학적 기능을 할 수 있는 물질로서의 가능성이 있음을 확인할 수 있었다.

정신분열병 환자에서 Interleukin-12와 Transforming Growth Factor Beta 1의 치료 전후의 변화 (Changes of Interleukin-12 and Transforming Growth Factor Beta 1 before and after Antipsychotic Treatments in Schizophrenic Patients)

  • 목 적: 많은 연구에서 정신분열병에서 염증반응체계의 활성화와 사이토카인의 변화가 병태생리학적 및 원인적 역할을 하는 것으로 보고되어 왔으며, 여기에는 type 1 Thelper cell(Th1), type 2 T helper cell(Th2), type 3 T helper cell(Th3)의 조절 이상이 제시되고 있다. 본 연구에서는 정신분열병 환자에서 항정신병 약물 치료 전후로 Th1 사이토카인인 interleukin-12(IL-12), Th3 사이토카인인 transforming growth factor-${\beta}1$(TGF-${\beta}1$)의 혈장 농도를 측정하였다. 방 법: 23명의 정신분열병 환자군과 31명의 정상대조군에서 IL-12와 TGF-${\beta}1$ 농도를 측정하였고 정신분열병 환자군에서는 8주간 항정신병 약물로 치료 후 다시 IL-12와 TGF-${\beta}1$의 농도를 측정하였다. 또한 정신분열병 환자군에서 치료전과 8주간 치료 후, 2차례에 걸쳐 Brief psychiatric rating scale(BPRS)를 측정하였다. 결 과: 치료전 IL-12 농도와 TGF-${\beta}1$ 농도 모두 정상대조군보다 환자군에서 유의하게 높게 나타났다. 8주간의 치료 후 TGF-${\beta}1$ 농도는 유의하게 감소하여 정상대조군의 농도와 차이를 보이지 않게 된 반면, IL-12의 농도는 유의하지 않은 감소를 보였다. BPRS 점수의 변화 및 IL-12 및 TGF-${\beta}1$의 농도의 변화 사이에는 유의한 상관관계가 없었다. 결 론: 정신분열병의 병태생리학에 사이토카인의 이상이 관여할 수 있으며, TGF-${\beta}1$이 중요한 역할을 하는 것으로 생각된다.

십전대보탕의 면역활성에 관한 기초 실험 연구 문헌 분석 (Analysis of Biological Experiment on Immunoactivity of Sipjeondabo-tang)

  • To investigate scientific evidence for the use of a Korean Medicine (KM), the papers regarding Sipjeondabo-tang (Shiquandabu-tang) frequently used in Korean medical clinics and hospitals were collected and analyzed. The papers were classified as being from domestic or international journals and by the year of publication. The mechanisms of immuno-activity Sipjeondabo-tang (Shiquandabu-tang) were researched. Among 41 papers, 4 were published as Korean papers whereas 7 were as Chinese papers and 40 were as Japanese papers. Sipjeondabo-tang (Shiquandabu-tang) regulated the productions of Th1 cytokines such as interleukin-2 (IL-2) and interferon-${\gamma}$ (IFN-${\gamma}$), and Th2 cytokines such as IL-4, IL-5, IL-6, IL-12 improving the function of immune organ including bone marrow, spleen, liver, thymus and peyer's patch. It also modulated the releases of immunoglobulin-G (IgG) and IgM. Thus, Sipjeondabo-tang (Shiquandabu-tang) balanced the Th1/Th2 immune responses as well as T/B cell. The regulation of immunoactivity could be speculated as an objective and scientific evidence of Sipjeondabo-tang (Shiquandabu-tang).

식물생장촉진 Bacillus sp. SB19 균주의 케일 처리에 대한 가뭄 스트레스 완화 효과 (Mitigation Effect of Drought Stress by Plant Growth-promoting Bacterium Bacillus sp. SB19 on Kale Seedlings in Greenhouse)

  • 가뭄은 농작물의 생산성을 저해하는 주요 원인 중 하나이며, 잎을 먹는 쌈채류의 경우 물 부족 스트레스에 더 치명적일 수 있다. 본 연구는 케일 유묘에 대한 식물생육촉진 균주(PGPB)의 가뭄 내성 효과를 알아보기 위해 수행되었다. 쌈채류의 토양 및 근권 토양으로부 터 형태학적으로 구분되는 146개의 콜로니를 분리하고 온실 생물검정을 통해 케일 생육촉진이 우수한 균주 SB19를 최종 선발하였다. SB19 균주는 케일 재배 토양으로부터 분리하였으며 16S rRNA 유전자 염기서열 분석 결과 Bacillus sp.로 확인되었다. Bacillus sp. SB19 균주를 처리한 케일에 7일 간 수분 부족 스트레스를 유도하고 7일째에 가뭄 피해 조사 후 모든 처리구에 1회 관수하였다. 이후 다시 7일 간 수분 부족 스트레스를 주어 14일째에 케일의 내건성 증진 여부를 조사하였다. 가뭄 조건 7일째에 $10^6$$10^7cell\;mL^{-1}$ 농도의 SB19 균주를 처리한 케일에서 무처리와 비교하여 가뭄 스트레스 경감 효과를 보였다. 7일째에 모든 처리구에 관수 후 다시 가뭄 스트레스를 주었을 때에도 $10^6$$10^7cell\;mL^{-1}$ 농도의 SB19 균주 처리구에서 무처리구와 비교하여 가뭄 피해 경감 효과가 있었으며, 7일째와 14일째 모두에서 $10^7cell\;mL^{-1}$ 농도의 SB19 균주 처리구에서 가뭄 피해의 완화 정도가 가장 효과적인 것으로 나타났다. $10^6cell\;mL^{-1}$ SB19 균주 처리구에서는 물 부족으로 인한 잎의 노화가 $10^7cell\;mL^{-1}$ 농도 처리구에 비해 빠르게 발생하였다. 본 연구 결과를 바탕으로 유용 미생물과 식물의 상호작용이 식물의 물 이용률을 증진시키는 중요한 역할을 하고 약한 가뭄 조건에서 쌈채류의 품질을 향상시킬 수 있는 방안이 될 수 있을 것이라고 예측한다. 즉, 미생물학적인 환경 스트레스 극복 방법으로서의 가치를 뒷받침하는 것이라 할 수 있다.

T helper cell subsets and related cytokines in infertile women undergoing in vitro fertilization before and after seminal plasma exposure

  • Objective: In vitro fertilization (IVF) is a well-known method for the treatment of infertility. The present study aimed to compare the differences between infertile women with successful and unsuccessful IVF outcomes regarding the expression of T helper (Th) cell transcription factors and a group of related cytokines before and after exposure to their husbands' seminal plasma. Methods: This study was performed on 19 couples with unexplained infertility undergoing IVF treatment. Among the studied group, nine and 10 couples had successful and unsuccessful IVF outcomes, respectively. This study was carried out using real-time polymerase chain reaction. Results: Before seminal plasma exposure, the expression levels of T-bet (p< 0.007), $interferon-{\gamma}$ (p= 0.013), and tumor necrosis factor $(TNF)-{\alpha}$ (p= 0.017) were higher in the infertile women with IVF failure than in those with successful IVF outcomes, while those of GATA3 (p< 0.001), Foxp3 (p= 0.001), and interleukin (IL)-35 (p< 0.003) were lower. After seminal exposure, the expression of T-bet (p= 0.02), Rorc (p< 0.001), $TNF-{\alpha}$ (p= 0.001), Foxp3 (p= 0.02), and $interferon-{\gamma}$ (p= 0.001) increased in the unsuccessful IVF group, while the expression of Foxp3 (p= 0.02), Rorc (p< 0.001), IL-23 (p= 0.04), IL-17 (p= 0.02), IL-6 (p< 0.001), transforming growth $factor-{\beta}$ (p= 0.01), and IL-35 (p< 0.001) increased in the successful IVF group. Conclusion: In summary, IVF failure was associated with imbalanced Th1/Th2/Th17/Treg responses. Moreover, our results show that seminal plasma might have a positive effect on IVF outcomes via changes in peripheral blood T cell subsets.
