Characterization of Tetracycline-Resistant Genes of Staphylococci isolates from Human and Marine Environment (사람과 해양환경에서 분리된 Staphylococci의 Tetracycline 내성 유전자의 특성 비교)

  • For comparison of tetracycline-resistant ($Tc^R$) genes, we obtained 21 and 14 $Tc^R$ Staphylococcus spp. from marine environment and human patient, respectively. Although all isolates from human were identified as Staphylococcus aureus, higher proportion of $Tc^R$ isolates (12 out of 14) from human were utilizing tet(M) gene compared to that of $Tc^R$ isolates (6 out of 21) from marine environment. Additionally, collaborated utilization of tet(M) and erm(A) in $Tc^R-Em^R$ S. aureus in human patient, but not in $Tc^R$ Staphylococcus spp. isolates from marine environment was also characterized. Based on the nucleotide sequence of transposon related to $Tc^R$ gene, we confirmed the origin of tet(M) gene in $Tc^R$ Staphylococci isolated from marine environments and human are derived from Tn916/1545-like and Tn5801 transposon, respectively. It is the first report showing the presence of Tn5801 in all $Tc^R$ S. aureus carrying tet(M) in human patient. Alignment of the fully sequenced tet(M) from marine environmental isolates was also agreed with the determined transposons by showing the genomic mosaic structure composed with three genomic parts from Tn916/1545 and unknown transposons. Genetic characteristics of these tet(M) in environmental isolates were similar to each other but different from those in isolates from human showing only tet(M) from Tn916/5801 type. It may imply the presence of less dramatic communication of antibiotic resistant genes between Staphylococci isolated from marine environment and human.

  • The location of R-determinants, $Ap^{r}$ and $Tc^{r}$, and replication origin in pKU41 determined using the construction of miniplasmid by the BamHI and the HindIII restriction fragment from pKU41 and the cloning of the restriction fragments from pKU41 into pSY343. The gene encoding resistance to ampicillin (Ap) as well as replication origin in pKU41 were located on the region overlapping BamHI B fragment and HindIII A fragment. The gene encoding resistance to tetracycline (Tc) was located on the region of the HindIII C fragment, which was cleaved by BamHI as well.

Analysis of Antimicrobial Resistance Pattern and Distribution of Multi-drug Efflux Pump Genes and Virulence Genes in Enterococcus faecalis Isolated from Retail Meat in Seoul (서울시내 시판 식육에서 분리한 Enterococcus faecalis의 항생제 내성 유형, 다중약물 유출 펌프 유전자 및 병독성 유전자의 분포도 분석)

  • The aim of this study was to investigate the distribution of genes that encode multi-drug efflux pumps and virulence factors in Enterococcus faecalis isolated from retail meat and antibiotic resistance patterns of these strains. Of the 277 retail meat samples, 93 Enterococcus faecalis were isolated. The strains exhibited resistance to ampicillin (35.5%), chloramphenicol (6.4%), ciprofloxacin (4.3%), erythromycin (18.3%), levofloxacin (0%), quinupristin-dalfopristin (76.3%), tetracycline (45.2%), teicoplanin (0%) and vancomycin (0%). The strains were positive for MFS type eme(A) (100%), ABC type efr(A) (100%), ABC type efr(B) (98.9%) and ABC type lsa (91.4%) efflux pump gene. The strains were positive for gelE (68.8%), ace (90.3%), asa1 (47.3%), efaA (91.4%) and esp (12.9%) virulence gene. This research will help to assess the hazards associated with the occurrence of drug resistance among enterococci from retail meat. Therefore, it is necessary to monitor enterococcus spp. isolated from retail meat continuously.

  • The properties and DNA structures of R plasmids differ depending on the species of the fish-pathogens Aeromonas hydrophila, A. salmonicida, Edwardsiella tarda, Enterococcus seriolicida, Pasteurella piscicida and Vibrio anguillarum. However, some R plasmids with the same resistance markers in similar DNA structures were found in A. hydrophila and E. tarda, as well as in A. hydrophila and A. salmonicida. R plasmids from V. anguillarum were classified into three groups according to their DNA structures. The first group was detected before 1977, the second from 1980 to 1983, and the third from 1989 to 1991. R plasmids have been retained within P. piscieida having the same DNA structures and detected at various locations and times. E. seriolicida strains carrying the same R plasmids, which were encoded with resistance to macrolide antibiotics(MLs), lincomycin(LIM), and TC, and to MLs, LIM, and CP. were distributed in yellowtail farms in various districts. The chloramphenicol-resistance(cat) gene of the R plasmids of P. piscicida was classified as CAT type I. The cat of the R plasmids of E, tarda. A. salmonicida was classified as type II. The cat of R plasmids of V. anguillarum was classified into two types. One type detected before 1977, was classified as CAT IV and the other type, detected after 1980, was classified as CAT II. Tetracycline-resistance (tet) V. anguillarum, isolated before 1977 and after 1981, was classified as Tet B and Tet G, respectively. The class D tet gene was widely distributed in R plasmids from fish-pathogens A. hydrophila, E. tarda, P. piscicida, and also V. anguillarum isolated after 1989.

Safety Assessment of Coagulase-Negative Staphylococci from Jeotgal, a Korean High-Salt-Fermented Seafood (젓갈 유래 Coagulase-Negative Staphylococci의 안전성 평가)

  • Seventeen ampicillin-sensitive coagulase-negative staphylococci (CNS) isolates identified in jeotgal were subjected to assessments for antibiotic susceptibility and safety hazards. Fifteen of the 17 CNS strains exhibited phenotypic resistances to at least one antibiotic, and their prevailing resistance was to penicillin G. The dfrA gene for trimethoprim and tetK for tetracycline were amplified by PCR from the two strains, respectively. α-Hemolytic activity was not detected from the 17 strains, while five strains presented δ-hemolytic activity. Among the five strains, two strains exhibited β-hemolytic activity. Biofilm was formed from twelve strains. All of the tested phenotypic characteristics were expressed in a strain-specific manner.

Prevalence and Antimicrobial Resistance of Enterococus faecalis and Enterococcus faecium Isolated from Beef, Pork, Chicken and Sashimi (시판 축산물 및 수산물에서 Enterococcus faecalis와 Enterococcus faecium 분포 및 항생제 감수성에 관한 연구)

  • In this study, a total of 256 samples of retail raw meats (beef, pork and chicken) and sashimi were investigated for the presence of Enterococcus faecalis and Enterococcus faecium. We isolated a total of 117 E. faecalis and E. faecium from the samples, with contamination rates ranging from 18.8% for sashimi samples to 68.8% of chicken samples. E. faecalis was the predominant species recovered from all of the retail raw meats beef (42.2%), pork (42.2%), chicken (65.6%) and sashimi (12.5%). Among 117 isolates, 61 isolates (52.1%) were resistant to tetracycline, 32 isolates (27.4%) were resistant to erythromycin, 23 isolates (19.7%) were resistant to chloramphenicol, 16 isolates (13.7%) were resistant to ripampin, 10 isolates (8.5%) were resistant to gentamycin, 9 isolates (7.7%) were resistant to ciprofloxacin and 1 isolate (0.9%) was resistant to ampicillin and penicillin G. No resistance to amoxicillin + clavulanic acid and vancomycin was observed. Although no strain was resistant to vancomycin, the vanB gene was observed in 9 of 117 of Enterococcus (7.7%) demonstrating potential risk of vancomycin-resistant Enterococcus (VRE). Our results indicate that E. faecalis and E. faecium were highly prevalent in retail raw meats, but most strains were sensitive to tested antibiotics.

Construction of Plasmid Vectors for Zymomonas mobilis (Zymomonas mobilis의 Plasmid Vector 제조에 관한 연구)

  • In order to develop useful plasmid vectors for Zymomonas cells, attempts were made to isolate natural plasmids from Z. mobilis ATCC10988. Among a few plasmids isolated, a small plasmid of 3.9 Kb size was chosen and designated as pZM3. By introducing the replication origin of pZM3 into pBR325, a hybrid plasmid vector of 8.4 Kb size, pHZ22, was constructed. This vector contained chloramphenicol resistant gene as a selectable marker and proved to be conjugally transmissible and stably maintained in Z. mobilis. Tetracycline resistant gene was isolated from RP4 and introduced into pHZ22 to make a new vector called pHZT224 of 10.7 Kb size. Through n series of experiments, it was evident that these plasmid vectors containing selectable markers of chloramphenicol and tetracycline resistance were shuttle vectors functional in Z. mobilis as well as E. coli.

Antimicrobial Resistance and Implicated Genes of E. coli Isolated from Commercial and Cooked Foods in Seoul (서울시내 유통식품에서 분리한 대장균의 항생제 내성 및 내성유전자)

  • Distribution of foodborne E. coli strains, antimicrobial resistant genes and antimicrobial susceptibility have been carried out on E. coli isolated from commercial and cooked foods distributed food in Seoul. Of total 1,313 samples, fifty samples(3.8%) were found E. coli that included one of the ETEC and EPEC, respectively. The serotype of ETEC in seasoning raw meat was E. coli O26 and produced Verotoxin 2. Fifty percentage of total isolates were susceptible to all antimicrobial agents. Specially, there were ampicillin(36%), amoxicillin/clavulanic acid(32%) and tetracycline(22%) etc. Resistant gene (tetB) were found in four tetracycline resistant E. coli strains, and TEM gene was found in one ampicillin resistant E. coli isolate.

Isolation and Characterization of Antibiotic Resistant Vibrio Strains from Japanese Eel (Anguilla Japonica) Cultured in Korea (국내산 양식 뱀장어에서 항생제 내성 비브리오 세균 분리 및 특성)

  • Continuous mortality in commercially cultured Japanese eel (Anguilla japonica), showing symptoms of dermal ulcerations and focal hemorrhages on the body, occurred on a private farm in November, 2019 in Korea. A series of mortality had been described in one local eel culture farm from November to December in 2019. From the three cases, three isolates of Vibrio spp. were recovered from the blood, ascitic fluid, and kidney of the dead fish, respectively. Based on the 16S rRNA sequence comparisons, the Vibrio isolates from the 1st and 3rd cases (strain named 1E1-2 and 3K1-2) were identified as V. fluvialis and the isolate from the 2nd case was identified as V. plantisponsor (strain named 2A3-1). Moreover, the 16S rRNA-based phylogenetic analysis revealed that strain 1E1-2 and 3K1-2 were most similar to V. fluvialis NBRC 103150T, and strain 2A3-1 was most similar to V. plantisponsor NBRC103148T. According to the results of the antibiotic resistance determination, V. fluvialis 1E1-2 showed intermediate resistance to tetracycline and chloramphenicol, and was resistant to trimethoprim-sulfamethoxazole. V. plantisponsor 2A3-1 showed intermediate resistance to ciprofloxacin and levofloxacin, and was resistant to trimethoprim-sulfamethoxazole. V. fluvialis 3K1-2 showed intermediate resistance to tetracycline, and was resistant to ampicillin and trimethoprim-sulfamethoxazole. These results have provided the evidences on the occurrence of antibiotic-resistant Vibrio infection in commercially cultured Japanese eels are present in Korea.

Isoaltion and characterization of petroleum degrading bacteria (원유분해세균의 분리 및 특성)

  • From several sites of petroleum storage basement in South Coasts in Korea, various petroleum degrading bacteria have been isolated and characterized as Pseudomonas fluorescens, Acinetobacter baumanii, Pseudomonas maltophila and Pseudomonas aeruginosa, respectively. They show the ability of petroleum degradation on minimal media which contains petroleum as sole carbon source and loose the ability at high concentration of NaCl as increasing the concentration of NaCl from 0.5% to 6%. It has been confirmed that such bacteria have utilized the simple saturate hydrocarbon; n-decane, n-hexane, n-octane and n-decane because petroleum consists of various kinds of organic compounds. It has been also identified that petroleum degrading bacteria habor the plasmid and show the antibiotic resistance against ampicillin, tetracycline and chloramphenicol. These results strongly suggest that the petroleum degrading gene and antibiotic resistance gene might be located on the high molecular weight plasmid.
