  • In total, 131 Escherichia coli isolates from surface seawater of the Gomso Bay, of Korea, were analyzed for their susceptibility to 22 different antimicrobials and for genes associated with antimicrobial resistance and virulence. According to the disk diffusion susceptibility test, the resistance to tetracycline was most prevalent (33.6%), followed by that to ampicillin (22.1%), ticarcillin (22.1%), and trimethoprim (16.8%). More than 46.6% of the isolates were resistant to at least one antimicrobial, and 22.9% were resistant to three or more classes of antimicrobials; these were consequently defined as multidrug resistant. We further found that 29 ampicillin-resistant isolates possessed genes encoding TEM-type (93.1%) and SHV-type (6.9%) ${\beta}$-lactamases. Among the 44 tetracycline-resistant isolates, tetA and tetC were found in 35 (79.5%) and 19 (43.2%), respectively, whereas tetB was detected in only three isolates (6.8%). With regard to virulence genes, merely 0.8% (n = 1) and 2.3% (n = 3) of the isolates were positive for the enteroaggregative E. coli-associated plasmid (pCVD432) gene and the enteropathogenic E. coli-specific attaching and effacing (eae) gene, respectively. Overall, these results not only provide novel insight into the necessity for seawater sanitation in Gomso Bay, but they help reduce the risk of contamination of antimicrobial-resistant bacteria.

  • Bacterial pathogens were isolated from 36 dogs with respiratory signs, that were submitted to Veterinary Clinics in Jeju, including Veterinary Medical Teaching Hospital in Cheju National University. Of 36 isolates, 16 (44.4%) bacterial pathogens were Gram-positive and 20 (55.6%) were Gram-negative. Gram-positive bacteria identified with API Staph were 12 S. intermedius (33.3%), 2 S. aureus (5.6%), 1 S. haemolyticum (2.8%), and 1 S. xylosus (2.8%). Gram-negative organisms identified with API 20E or API NE included 8 Bordetella bronchiseptica (22.2%), 6 Escherichia coli (16.7%), 4 Pasteurella spp. (11.1%), 1 Enterobacter intermedius (2.8%), and 1 Oligella ureolytica (2.8%). Both Staphylococcus spp. isolates and Gram-negative pathogens were resistant to one or more antibiotics, including ampicillin (AM), amoxicillin/clavulanic acid (AMC), chloramphenicol (C), cefazolin (CZ), erythromycin (E), gentamicin (GM), kanamycin (K), lincomycin (L), oxacillin (OX), trimethoprim/sulfamethoxazole (SXT), and tetracycline (TE). All Staphylococcus spp. were susceptible to AMC, OX and VA, while many isolates were highly resistant to L (87.5%), E (68.8%), P (62.5%), and AM (56.3%). Antibiotic-resistant patterns of staphylococcal isolates were shown ranges from single to 9-resistant patterns. Resistant rates to antibiotics of Gram-negative bacteria were usually higher than those of Staphylococcus spp. in this study. Most Gram-negative bacteria were highly resistant to L (90.0%), AM (85.0%), E (85.0%), P (85.0%), OX (80.0%), and CZ (75.0%). B. bronchiseptica isolates showed 5 to 8 antibiotics-resistant patterns and Pasteurella spp., 2 to 8-resistant patterns. In particular, all 6 E. coli isolates were resistant to more than 9 different kinds of antibiotics, including one strain resistant to all antibiotics tested.

  • Objectives: This study aims to understand the concentration, diversity, and antibiotic characteristics of staphylococci present in the indoor air of child-care facilities. Methods: Air sampling was performed from October 2012 to January 2013 in 120 child-care facilities in Seoul, Korea. Methicillin-resistant bacteria were selected from the total obtained airborne bacteria and subjected to 16S rRNA analysis for methicillin-resistant staphylococcal species determination. Identified staphylococcal strains were tested for resistance to a range of antibiotics. Results: Average total airborne bacterial concentration was $508.9{\pm}246.3CFU/m^3$. Indoor concentration of total airborne bacteria had a significant positive correlation with the $CO_2$ concentration in the child-care facilities. Methicillin-resistant staphylococci were present in 13.3% of the child-care facilities studied. A total of four species (S. epidermidis, S. cohnii, S. saprophyticus, S. sp.) and 55 strains were identified from the indoor air of the child-care facilities. Staphylococcus cohnii was the most common species (54.5%), followed by S. epidermidis (38.2%). All of the isolated staphylococcal strains exhibited high resistance to oxacillin, erythromycin, mupirocin, and ceftizoxime. Especially, S. saprophyticus strains showed more multidrug resistance to oxacillin, vancomycin, clindamycin, erythromycin, lincomycin, ceftizoxime, mupirocin, and tetracycline than did other species. Conclusion: The results of this study showed that a monitoring system for multidrug-resistant bacteria is needed in facilities for children, as the community-associated infections of these bacteria are increasing.

  • Causative agents of mastitis were isolated from glandular tissues of 101 culled Holstein cow udders and tested with antimicrobials by standardized disc diffusion method. Pathogens most commonly isolated were coagulase-negative staphylococci (43.9%) and streptococci (21.4%). Most of the udders (88.5%) showed mixed infections with more than two species of bacteria. Antimicrobial sensitivity tests revealed that 90.0% and 84.5% of the isolated organisms were susceptible to amoxicillin and gentamicin, while most organisms were resistant to erythromycin. Coliforms were resistant to ampicillin, cephalothin, cefazolin, erythromycin, kanamycin, neomycin, penicillin and tetracycline and streptococci to erythromycin, kanamycin, neomycin, trimethoprim/sulfamethoxazole and tetracycline.

  • The use of bioluminescence as a sensitive marker for the detection of Pseudomnas sp. in the rhizosphere was investigated. Transposon Tn4431 which contains a promoterless luciferase operon and tetracycline resistant gene was used. This transposon, present on a suicide vector (pUCD623) in E. coli HB101, was mated with spontaneous rifampicin mutant of Pseudomonas fluorescens B16, a plant growth promoting rhizobacteria (PGPR), and then rifampicin and tetracycline resistant survivors were isolated. Twenty tow mutants wer isolated from the conjugants between E. coli HB101 and P. fluorescens B16. One of these, B16::Tn4431 (L22) recombinant which glowed brightly in the dark was selected for analysis. The cucumber seeds inoculated with L22 were grown in moisten two layers of filter paper and nonsterile soil contained in half cut PVC pipe. The roots were removed from the filter paper and PVC pipe, then placed on the 1/2 LB media plates. The plates were incubated at room temperature for 16 hr. L22 could successfully be detected in the rhizoplane by using the ordinary negative camera film (ASA100-400) with 30 minutes exposure under dark condition. The root colonizing ability and the plant growth promoting effect of L22 were not reduced compared to the untreated bacteria and wild type. L22 was superior to will type.

  • Microorganisms causing bovine mastitis, swine diarrhea and swine pneumonia were isolated from farms in Chungbuk and Kyunggi Province and the isolates were identified using microbial identification system(MIS). The most common isolates from bovine mastitis were Staphylococcus sp.,Streptococcus sp., and Corynebacterium sp., those from swine diarrhea were Escherichia coli, Salmonella sp., and Clostridium perfringens, those from swine pneumonia were Bordetella bronchiseptica and Pasteurella multocida. Examination of 16 antibiotics against these pathogens revealed that the incidence of antibiotic-resistant microoganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. These results suggest that the use of antibiotics in a farm should be controlled in order to decrease the number of antibiotic-resistant microorganisms were very high and that many of these isolates had multiple resistance to various commercially available antibiotics such as penicillin, ampicillin, erythromycin, streptomycin, norfloxacin, and tetracycline. • This study was conducted to monitor the antibiotics resistance of human-harmful bacteria isolated in the agricultural environment for hot peppers (Capsicum annuum) and tomato (Lycopersicon esculentum). As a result, we isolated 120 bacterial species (34 on fruits, 48 in soil, 21 in water, and 17 in manure), identified them with the 16S rRNA sequence, analyzed minimum inhibitory concentration (MIC) for 26 antibiotics using Sensititre ARIS Hi-Q system and then evaluated whether each bacterial genus acquired resistance for the tested antibiotics or not, according to the CLSI criteria. From difference in MIC between eco-friendly (EFM) and practical (PFM) cultivation farms, Klebsiella spp. isolated from EFM was resistant to ampicillin (AMP) and nalidixic acid (NAL), and that isolated from PFM was resistant to streptomycin (STR) and tetracycline (TET). Enterobacter spp. isolated from EFM was resistant to AMP and azithromycin (AZI), and that isolated from PFM was resistant to AMP, AZI, and STR. Meanwhile, Pseudomonas spp. isolated from EFM and PFM were all resistant to AMP, AZI, cefotaxime (FOT), cefoxitin (FOX), ceftriaxone (AXO), CHL, NAL, and STR. Staphylococcus spp. isolated from EFM and PFM were resistant to gentamycin (GEN), STR, and kanamycin (KAN), and in particular, that from EFM showed resistance for erythromycin (ERY). In conclusion, our study suggested that EFM lead STR antibiotics resistance for human-harmful bacteria to decrease, because only the bacteria isolated from hot pepper and tomato crop with PFM have showed resistance against STR antibiotics, regardless of bacterial genus.

  • The antimicrobial drug susceptibility of 439 isolates of animal pathogens recovered from various clinical cases during 1978-79 has been investigated by the use of disk diffusion technique. The majority of 308 strains of Eschericihia coli were highly resistant to bacitracin, erythromycin, penicillin, streptomycin and tetracyclinon while only 0.3 per cent of them were resistant to gentamicin and 3.2 per cent to colistin. The percentages of strains resistant to ampicillin, carbenicillin, cephalothin, chloramphenicol and neomycin were 30.5%, 24.7%, 11:4%, 28.2% and 26.2% and repectively. However, none of E. coli cultures of ovine origin were resistant to ampicillin, carbenicillin, chloramphenicol, colistin, gentamicin, kanamycin, and neomycin. A total of 39 patterns of multipe drug1 resistance of 308 strains E. coli against 9 drugs in general use such as ampicillin, cephalothin, chloramphenicol, colistin, gentamicin, kanamycin, neomycin, streptomycin and tetracycline were observed and the most common multiple resistance patterns were SM, TC pattern (20.5%) and AM, CP, KM, NM, SM, TC pattern (9.7%). None of the 43 cultures of salmonella organism from pigs and chickens were resistant to ampicillin, carbenicillin, cephalothin, colistin, gentamicin and kanamycin; and the majority of the cultures were susceptible to chloramphenicol (90.0%), neomycin (97.7%) and tetracycline (93.0%). All the cultures were found to be resistant to bacitracin and penicillin and the rate of resistant strains to erythromycin and s treptomycin being 79.1% and 41.9% respectively. It was found that the majority of 63 cultures of staphylococcal isolates were resistant to lincomycin, penicillin, streptomycin and tetracycline. The percentages of 63 staphylococcal isolates susceptible to gentamicin, nitrofurantoin, cephalothin, ampicillin, methicillin, bacitracin and chloramphenicol were 98.4%, 98.4%, 95.2%, 93.7%, 93.7%, 92.1% and 92.1% respectively. The 25 cultures of streptococcal isolates were resistant in order of prevalence to streptomycin(88.0%), kanamycin(68.0%), gentamicin (44.0%), tetracycline (44.0%) and methicillin (40.0%) wihle the majority of them were sensitive to ampicillin, bacitracin, chloramphenicol and penicillin.

  • BACKGROUND: Large amount of veterinary antibiotics have been used in the livestock industry to prevent diseases and promote growth. These antibiotics are excreted through feces and urine in unchanged form and reach to agricultural fields via application of the livestock manure based composts. The purpose of this study was to evaluate the occurrence of tetracyclines-resistant bacteria in the soil received livestock manure compost for a long term. METHODS AND RESULTS: Tetracyclines (tetracycline TC, chlortetracycline CTC, and oxytetracycline OTC) resistance bacteria in the soil of rice-onion field applied pig manure compost (PM), in the soil of grass-rye field received cow manure compost (CM), and in the soil of rice field applied inorganic fertilizer (NPK) were determined. The soil received livestock manure composts clearly showed higher number of TC, CTC, and OTC resistance bacteria compared with the soil treated with inorganic fertilizer. The antibiotic resistant bacteria recovered appeared at 80 mg/L of tetracyclines was identified 1 specie, 6 genera 7 species, and 6 genera 7species in the soils received CM, PM, and NPK, respectively. The dominant resistant bacteria with the CM and PM application were Ochrobactrum and Rhodococcus. CONCLUSION: The application of livestock manure compost in the agricultural field is likely to contribute the occurrence of antibiotic resistance bacteria in the agricultural environment.
