Cadmium (Cd) generates reactive oxygen species (ROS), which in turn cause the apoptosis of various cell types including developing germ cells in rodent testis. Ascorbic acids (AA), one of the ROS scavengers, had been reported to protect against Cd-induced apoptosis. N-Acetyl-L-cysteine (NAC), another ROS scavenger, is known to remove ROS and alleviate the Cd-induced apoptosis in various cell types. In this study we tried to elucidate how NAC affected on Cd-induced cell apoptosis in rat testis. Rats were administered with NAC before and after Cd treatment and then testicular cell apoptosis was examined. NAC treatment resulted in the reduction of Cd-induced chromosomal DNA fragmentation in agarose gel electrophoresis. Terminal deoxynucleotidyl transferase dUTP nick end-labeling (TUNEL) assay showed that treatment of NAC reduced the Cd-induced apoptosis of germ cells. The administration of NAC showed that the translocation of apoptosis inducing factor (AIF) from mitochondria to nucleus was prevented, which indicated that the mechanism of Cd-induced testicular apoptosis is mediated through the release of AIF in caspase-independent manner. Taken together, the NAC may remove Cd-induced ROS and protect ROS-induced cell apoptosis in rat testis.
Cyclic AMP-response element binding protein zhangfei (CREBZF), a member of ATF/CREB (activating transcription factor/ cAMP response element binding protein) family, regulates numerous cellular functions and development of cells by interacting transcription factors. This study discovered the expression pattern of CREBZF in seminiferous tubule of testes during the postnatal development of mice. In testis, CREBZF mRNA expression was the highest among other organs. Immunofluorescence analyses showed that the CREBZF was specifically expressed on spermatocyte but not in spermatogonia and Sertoli cells in seminiferous epithelium of mouse testis. Semi-quantitative polymerase chain reaction (PCR) analysis showed that CREBZF transcript level was significantly elevated during postnatal development of mouse testis. Confocal imaging analysis indicated that the protein expression of CREBZF in seminiferous tubule remained low until postnatal day (PD) 14, and was dramatically increased in PD 21. Interestingly, only one type of the spermatocyte expressed CREBZF specifically among SCP3-positive spermatocytes. Taken together, these results suggest that CREBZF may be novel putative marker of the spermatocyte and regulate meiosis during postnatal development of mice.

Kisspeptins, products of KISS1 gene, are ligands of the G-protein coupled receptor (GPR54), and the kisspeptin-GPR54 signaling has an important role as an upstream regulator of gonadotropin releasing hormone (GnRH) neurons. Interestingly, extrahypothalamic expressions of kisspeptin/GPR-54 in gonads have been found in primates and experimental rodents such as rats and mice. Hamsters, another potent experimental rodent, also have a kisspeptin-GPR54 system in their ovaries. The presence of testicular kisspeptin-GPR54 system, however, remains to be solved. The present study was undertaken to determine whether the kisspeptin is expressed in hamster testis. To do this, reverse transcription-polymerase chain reactions (RT-PCRs) and immunohistochemistry (IHC) were employed. After the nest PCR, two cDNA products (320 and 280 bp, respectively) were detected by 3% agarose gel electrophoresis, and sequencing analysis revealed that the 320 bp product was correctly amplified from hamster kisspeptin cDNA. Modest immunoreactive (IR) kisspeptins were detected in Leydig-interstitial cells, and the weak signals were detected in germ cells, mostly in round spermatids and residual bodies of elongated spermatids. In the present study, we found the kisspeptin expression in the testis of Syrian hamster. Further studies on the local role(s) of testicular kisspeptin are expected for a better understanding the physiology of hamster testis, including photoperiodic gonadal regression specifically occurred in hamster gonads.

The maturation changes in morphology were studied with the spermatozoa collected from the testis and three successive parts of the epididymis in Korean native Cattle. Acrosomal granules were observed in the testis. Avoiding the cap and acrosome phases, the tail base and the striated column of the neck were formed in spermatides. The volume of the acrosome was decreased during transit from the testis to the epididymis. The cell membranes were also separated from the acrosome or damage during the spermatozoan passage through successive parts of the reproductive tract. Cytoplasmic droplets were observed in the spermatozoa collected from various parts of the reproductive tract.

Studies comparing the relative effects of water soluble extract of Ganoderma lucidum kale juice and sodium dextrothyroxine on lipid component in liver brain and testis of hypercholeste-rolemic rats for 4 weeks. Therefore the aim of this work was to investigate for prevention in cardiovascular disease, Total cholesterol concentration in liver were significantly lower kale juice and sodium dextrothyroxine(1.25mg/kg diet) fed groups than the other groups and trigly-ceride concentration was significantly lower Ganodrma lucidum fed group than in control group;. Total cholesterol concentration in testis were lower other groups than in control group and triglyceride concentration were significantly lower cholesterol Ganoderma lucidum and kale juice groups in control group. Phospholipid concentration in testis was higher sodium dextroth-yroxine fed group thanin control group.
SRY gene is normally responsible for testis induction, yet testis development can occur in the absence of SRY. In here, we analyzed the SRY-negative sex reversal in cocker spaniel, at 1.5 year-old. The attacked dog was suffered from enlarged clitoris, and resulted in disorder of urination. By surgically approach, enlarged clitoris and one testis, which are apparently seen, are removed. Additionally, thorough the abdomen surgery, uterus and ovary-like mass were removed. The dog had XX, chromosome, showed negative for SRY-gene, and the mass had the ovary-testis structure. In other words, based on the macroscopic, cytogenic, and histological study, we can diagnose the cocker spaniel as SRY-negative sex reversal.
Environmental endocrine disruptors are very toxic to the animals including humans. They are hormone-like acting chemicals which can be found in our normal daily life. We have examined the toxic effect of bisphenol A and if lactic acid bacteria could suppress this toxic effect. Thirty rats were divided into three groups (control, bisphenol A treated, bisphenol A and lactic acid bacteria treated). Treatments were carried out at an interval of 12 hours for each group. Control group showed normal and clear morphology of tissues. Cells were fine in their shape and color, and density was high enough for the normal function. However, bisphenol A treated group was abnormally destructed in cell morphology. In the testis, sperms were totally destructed. When treated with lactic acid bacteria together, the toxic effect of bisphenol A was clearly decreased. This study indicated that bisphenol A was toxic in any concentration especially for the liver and testis, however, lactic acid bacteria could suppress the toxic effects of bisphenol A.
The testis is major male gonad responsible for spermatogenesis and steroidogenesis. Much knowledge is still remained to be learned about the control of these events. In this study, we performed a comprehensive bioinformatics analysis on 1,196 mouse testis proteins screened from public protein database. Integrated function and pathway analysis were performed through Database for Annotation, Visualization and Integrated Discovery (DAVID) and ingenuity Pathway Analysis (IPA), and significant features were clustered. Protein membrane organization and gene density on chromosomes were analyzed and discussed. The enriched bioinformatics analysis could provide clues and basis to the development of diagnostic markers and therapeutic targets for infertility and male contraception.
Monocrotophos was administered orally to adult male albino mice at dose level of 3.0 mg/kg body weight/day/mice for 50 days. The treatment has found to affect spermatogenesis as well as the endocrine functions of the testis as indicated by gravimetric, histopathological and biochemical changes. The treatment has caused degenerative changes in the seminiferous tubules and Leydig cells of the testis and regression of the epididymis, seminal vesicle, vas deferens, prostate, Cowper's gland and levator ani. Similarly, cauda epididymal sperm count and sperm motility have shown significant reduction. There was a significant reduction in the protein, glycogen, sialic acid, acid and alkaline phosphatase and increase in cholesterol in the testis of monocrotophos treated mice compared with the control. The causative factors for these changes due to monocrotophos administration were discussed.
