Effects of season and breed on the reproductive performance of sheep

  • The aim of the current study was to compare the effects of season and breed on the reproductive performance of male and female sheep using 12 rams and 318 ewes of Assaf and Awassi breeds under the seasonal environmental condition of United Arab Emirates for two years. The blood level of testosterone hormone was measured monthly. Semen was collected twice a month from each male using artificial vagina and evaluated for volume, motility, livability, abnormality and concentration. The scrotal circumference and thickness as well as the left testicular length, width, height and volume were measured at one-month intervals. The level of testosterone in Assaf breed was significantly higher in autumn than winter and summer. The scrotal circumference and thickness as well as the left testicular length were significantly higher in Assaf breed than Awassi breed. While, left testicular width and volume were significantly higher in Awassi breed than Assaf breed. Scrotal circumference which was higher in spring and summer than in autumn and winter season in both breeds. The SCC of semen was significantly higher in autumn than in other seasons in both breeds. The sperm abnormality was significantly higher in summer than other seasons in both breeds. The livability was significantly lower in summer in both breeds. Fecundity and prolificacy were significantly higher in Assaf than Awassi breed during autumn season. Assaf breed showed the superior reproductive performance in the autumn season when compared with Awassi breed in the same season and other seasons. The Assaf breed tolerated the climatic conditions in UAE and kept the litter size of 1.72 in comparison to Awassi breed which showed litter size of 1.09. in conclusion, the results showed the superiority of Assaf over Awassi breed and offer a good model of breeding with increased fecundity and prolificacy specially in autumn season.

  • The ultrastructures of spermatogenesis and sperm in Xiphophorus maculatus, ovoviviparous fish were investigated by electronmicroscopy The testis of Xiphophorus maculatus contained numerous testicular sacs, and spermatogenesis was synchronized in these testicular sac. In the case of spermatogonium, the nucleus was comparatively large ellipsoidal, and the nucleolus and mitochondria showed a marked development. The size of primary spermatocyte was smaller than that of spermatogonia, and that of secondary spermatocyte was smaller than that of primary spermatocyte. The chromatin of spermatocyte was highly condensed according to their development. The nucleus with electron-dense was round shape. In spermiogenesis, flagella started to be formed and chromatin was more condensed. The mitochondria were rearranged along the tail. The sperm was formed by loss of cytoplasm. The head of mature sperm was long cone shape and had not acrosome. The microtubules of flagella were arranged 9+2 structure. Also, the sperm has a loop-like structure at the end of a tail.

  • Cytochrome P450 aromatase (P450arom) catalyzes the conversion of androgens to estrogens and plays an important role in reproduction and development in vertebrates. We investigated the expression patterns of ovarian P450arom (P450aromA) and brain P450arom (P450aromB) mRNA during sex change in black porgy. Maturity was divided into seven stages from male to female (immature testis, mature testis, testicular portion of mostly testis, ovarian portion of mostly testis, testicular portion of mostly ovary, ovarian portion of mostly ovary, and mature ovary). P450aromA expression was significantly higher in the ovarian portion of mostly-ovarian stage fish, and P450aromB expression was highest in the brain of black porgy with mostly-ovarian gonads. Histology showed that testicular tissues were disintegrated with the development of ovarian tissue associated with an increase in the expression of the two P450arom mRNAs during sex change. Interestingly, among various tissues, P450aromA was only expressed in the ovary, and P450aromB was only expressed in the brain. To understand the role of gonadotropin-releasing hormone (GnRH) and estradiol ($E_2$), we injected exogenous hormone (GnRH analogue [GnRHa] and $E_2$) into immature black porgy. In the GnRHa group, expression of the two P450arom genes decreased 12 h after injection, and expression of the two P450arom genes were significantly higher at 6 dafter $E_2$ injection. These results provide useful baseline knowledge on the mechanism of natural sex change in black porgy.

Effect of Anionic Alkali Mineral Complex Barodon$^{(R)}$ on Body Growth and Testicular Development in Rats

  • Effects of Barodon$^{(R)}$ on body growth, histological changes in seminiferous tubules of testes and serum level of testosterone and FSH were examined in juvenile rats from 5 to 38 wks of age. All rats supplied feed and Barodon-added water(0, 0.1, 0.15, 0.3 and 1.0%) available ad libitum. DNA distribution of testicular cells from control rats obtained by flow cytometry was characterized by 4 peaks representing I : elongated, II round spermatids(1N), III: a variety of 2N cells and IV: 4N cells. Frequencies of 4 testicular cell types were calculated using cumulative DNA frequency distributions. Body growth from 18wk of age was significantly accelerated in rats supplied water with 0.15% Barodon compared to other groups. Testes weights tended to be greater in 0.15% Barodon-treated rats than in control, without significant difference. Diameter of seminiferous tubules advanced in 0.15-0.3% Barodon till 26 wk of age, but there was not significant different. Proportion of spermatids in seminiferous tubules was 48.4% at 6wk of age(round: 81.2% and elongated: 18.8% as proportion of total spermatids) and frequency of spermatids was higher in 0.3% Barodon group(57.1%) than in control(44.0%), but there was no significant difference. Serum testosterone of all groups significantly elevated at 18wk of age and level in 0.15% Barodon group was greatly higher than in others. Serum FSH at 10wks was greatly higher in 0.1-0.3% Barodon groups compared to control, but there were no significant differences. It is concluded that 0.15-0.3% Barodon treatment tended to induce precocious puberty in rats.

  • The spermatogenesis of pale chub (Zacco platypus) was investigated morphologically. The testis of pale chub contained numerous testicular sacs. These testicular sacs were bounded on neighboring sacs by single layer of squamous cells. Also, differentiated sperms were filled in the sacs. In the stage of spermatogonium, the germ cells had a large nucleus and a distinct nucleolus, and mitochondrial development was prominent. In the primary and secondary spermatogonia, these cells had a round electron-dense nucleus, reduced cytoplasm, and mitochondria were congregated in the side of cytoplasm. The highly condensed chromatin of sperms was electron-dense, the acrosome was not found in the head of the sperm and a motile flagellum consisted of an axoneme with a typical 9+2 pattern of microtubules.

  • In ultrastructure study of testis, Sertoli cells start to differentiate at 16 days of gestation. Transcripts of FSH receptor, IGF-I receptor, ER $\alphareceptor, $ $TGF-\beta$ receptor and androgen receptor were highly and initially expressed at 16 day of gestation. As results of in situ PCR at 16 day of gestation, transcripts of FSH, IGF-I receptor were detected in Sertoli cells and spermatogonia, whereas the receptors of $ER\alpha, $ $TGF-\beta$ and androgen were detected in Sertoli cells. Therefore, expression of FSH and estrogen $\alpha, $ androgen, IGF-I and $TGF-\alpha$ could play an important role during fetal and prepubertal testicular development by stage specific manner in mouse.

  • Simazine (6-chloro-N,N'-diethyl-1,3,5-triazine-2,4-diamine) is a triazine herbicide that has been applied worldwide including Korea for agricultural purposes. Simazine is the second most commonly detected pesticide in surfaceand ground-water in the United States, Europe and Australia. It has been shown that simazine is a potent endocrine disruptor in wildlife and laboratory animals. Although many endocrine disruptors can induce apoptosis in various types of cells, the effects of simazine on apoptosis and on the expression of Bcl-2 family genes are not known. Also it is unknown the effect of simazine on the expression of steroidogenesis-regulating genes in testicular cells. In this study, we investigated the effect of simazine on the expression levels of apoptosis- and steroidogenesis-regulating genes in testicular cells. We found that a low concentration of simazine can alter the mRNA expression levels of steroidogenesis-related genes and Bcl-2 family genes in mouse Sertoli cells and rat Leydig cells. Thus, our results suggest that simazine can disturb normal testicular development and reproductive function by altering the expression of genes that are critical for the regulation of apoptosis and steroidogenesis.

Histological Studies on Gonad and Germ Cell Development of Diploid and Triploid Mud Loach (Misgurnus mizolepis) (2배체와 3배체 미꾸라지(Misgurnus mizolepis)의 생식소 발달)

  • A Study was conducted by using a histological method to examine gonadal development in diploid and triploid mud loach, Misgurnus mizolepis. Shape of ovaries from both groups looked like a single mass however, size of triploid ovary was significantly smaller than that of diploid ovary. Testis of triploid was also similar that of diploid in shape, but it was smaller than that of diploid. Ovarian development and oogenesis in diploid were significantly more rapid than those process in triploid, and first matured eggs were observed 100-days after hatching (SL, 5.68cm). Triploid ovary from hatching to 9-month-old had external appearance of undeveloped gonad and a few of oocytes of perinucleolus stage. However, normal matured eggs like those of female diploid were observed in one 5year-old triploid fish ovary examined. Although testicular development and sperrnatogenesis of diploid were prosperous sexually, testicular development of triploid were rather retarded compare to their male diploid counterpart in spermatogenesis.

  • PURPOSE: Gene expression and apoptosis in testicular germ cells has been demonstrated in many transgenic animals. However, little is known about the transgenic pig and rates of apoptosis during spermatogenesis. METHODS : Morphological and biochemical features of apoptosis reported in other species were used to confirm that the TdT-mediated dUTP Nick end labeling (TUNEL) assay is an acceptable mothos for idendtification and quantification of apoptotic transgenic germ cells in histological tissue section from transgenic pig testis. Gonadal Abnormality and Intersexuality of Oplegnathus fasciatus (Teleostei: Oplegnathidae) Collected from the Southern Coast of Korea: A Case Report

  • We report on the gonadal structure abnormality and intersexuality found from a survey regarding the reproductive potential of the rock bream, Oplegnathus fasciatus, along the southern coast of Korea. In total, 448 samples were collected for histological analysis (total length 27.5±6.8 cm; total weight 522.9±385.6 g). The sex ratio (F:M) was 1:0.46 (n=307:141), with 68.5% being females. The frequency of oocyte atresia and ovarian neoplasm in the females was 85.3% and 21.5%, respectively, and the frequency of testicular lobule and cyst deformation and testicular neoplasm in the males was 73.1% and 37.6%, respectively. The scale formation in the ovary and testis was approximately 2.0% for both sexes. The intersexuality was 42.2%, with the intersexuality among females being 49.5%, which was significantly higher than that of males (26.2%). Such findings indicate a negative impact on the reproductive output of the rock bream along the southern coast of Korea; however, a more detailed study is required to ascertain accurate causes.
