Construction of genetic linkage maps of Allium cepa using genotyping-by-sequencing

  • The onion (Allium cepa L.) is the most widely cultivated species of the genus Allium, especially it has been valued because of the pungent flavor and aroma. Allium species including onion has very large genome sizes ranging from approximately 10 to 20 Gbp, which have complicated genomic studies and precluded genome sequencing until recently. A population of 186 F2 individuals derived from a cross of 'Umjinara' ${\times}$ 'Sinsunhwang' and the two parental lines were used for this study. For the development of framework map, various types of markers including SSRs, RAPD, SNPs, and CAPS makers have been used for polymorphism test. Especially, a lot of SNP and CAPS loci were developed from the onion transcriptome sequence by RNASEQ of two parental lines. The GBS libraries have been constructed based on a modified protocol from Poland Lab using a two-enzyme system. We have been developing markers showing polymorphism between two parental lines, and genotyping for all F2 individuals were finished for a number of polymorphic markers. For the construction of GBS libraries, a set of 192 barcoded adapters were generated from complementary oligonucleotides with XhoI overhang sequence and unique barcodes of length 4-8 bp and they have been tested using two parental linesto determine the optimum conditions for GBS analysis.

Numerical simulation of an adobe wall under in-plane loading

  • Adobe is one of the oldest construction materials that is still used in many seismic countries, and different construction techniques are found around the world. The adobe material is characterized as a brittle material; it has acceptable compression strength but it has poor performance under tensile and shear loading conditions. Numerical modelling is an alternative approach for studying the nonlinear behaviour of masonry structures such as adobe. The lack of a comprehensive experimental database on the adobe material properties motivated the study developed here. A set of a reference material parameters for the adobe were obtained from a calibration of numerical models based on a quasi-static cyclic in-plane test on full-scale adobe wall representative of the typical Peruvian adobe constructions. The numerical modelling, within the micro and macro modelling approach, lead to a good prediction of the in-plane seismic capacity and of the damage evolution in the adobe wall considered.

  • The study examined fire resistance of concrete followed by change of mixed rate in PP and NY composite fiber and the results were as follows. In the event of fluidity in concrete not set, plane satisfied 600±100, its target slump flow, and fluidity was reduced as organic fiber's mixed rate was increased. Air amount satisfied 3.0±1.0, its target air amount, and didn't have distinct differences in reduction and increase according to organic fiber's kind and change of its mixed rate. However, it had a tendency that fluidity was reduced as the mixed rate was increased. In characteristics of hardening concrete, the 28th day compressive strength followed by organic fiber's kind and change of its mixed rate didn't have a lot of differences and satisfied high strength scope as about 70MPa. In spalling characteristics after fire resistance test, spalling was happened in non-mixture, plane combination, and P1N0. In other combinations, spalling resistance was happened. The relic compressive strength rate was 56%, the best condition, in P3N1(PP0.03%, NY0.01% compositeness) mixing PP fiber with NY fiber at once.

Properties of concrete incorporating sand and cement with waste marble powder

  • Marble is a metamorphic rock used widely in construction which increases amount of marble powder obtained from it. Marble powder is a waste product obtained from marble during its processing. Marble waste is high in calcium oxide content which is cementing property but it creates many environmental hazards too if left in environment or in water. In this research, partial replacement of cement and sand by waste marble powder (WMP) has been investigated. Seven concrete mixtures were prepared for this investigation by partially replacing cement, sand with WMP at proportions of 0%, 10% and 15% by weight separately and in combined form. To determine compressive strength, flexural strength and split tensile strength of concrete made with waste marble powder, the samples at the curing ages of 7, 28 and 90 days was recorded. Different tests of durability were applied on samples like ultrasonic pulse wave test, absorption and sorptivity. For further investigation all the results were compared and noticed that WMP has shown good results and enhancing mechanical properties of concrete mix on partially replacing with sand and cement in set proportions. Moreover, it will solve the problem of environmental health hazard.

Laboratory evaluation of roller compacted concrete containing RAP

  • This paper investigates mechanical properties of roller compacted concrete (RCC) involving reclaimed asphalt pavement (RAP). In this way, a set of 276 cylindrical RCC specimens were prepared with different RAP sizes (i.e., fine, coarse & total) at various ratios (i.e., 10%, 20%, and 40%). Results reveal that incorporation of RAP decreases unconfined compressive strength (UCS), modulus of elasticity (E), and indirect tensile (IDT) strength of RCC. For each RAP size, a regression model was used to maximize RAP content while satisfying the UCS lower limit (27.6 Mpa) mentioned by ACI as a minimum requirement for RCC used in pavement construction. Moreover, UCS of RAP incorporated mixes, dissimilar to that of control mixes, was found to be sensitive and insensitive to the testing temperature and curing time after 7 days, respectively. The results also demonstrate that the higher amounts of RAP, the more flexibility in RCC is. This issue was also proved by the results of modulus of elasticity test. In addition, the toughness index (TI) shows that increase in RAP content leads to up to 43% increase in energy absorbance capacity of RCC.

3차원을 이용한 중년 비만 여성용 스커트 설계 방법론 연구 (Development of Skirt Pattern for the Middle Aged Women of Obese using the 3-Dimension Technology)

  • The purpose of this paper is to find how to do the width of skirt and the girth of the waist in the adequate pattern making for the obese women's skirt. Appearance test of the five experimental skirts was evaluated by the four experts in clothing construction. At the same time, 3D clothing air volume was observed for the five types of experimental skirt with different size specifications. The results from the appearance test were as follows; when the width of skirt pattern is set for(the shell girth/2), it was suggested w/4+1(front), w/4(back) for girth of the waist. On one hand, in case of (the shell girth of front)/2+(the shell girth of back)/2, it was suggested(the waist girth of front)/2 and(waist girth of back)/2 for obese women's skirt with the best appearance. As results, it was found that the width of skirt pattern for the obese women should be the greatest shell girth instead of hip girth. In the case of the hip girth, the amount of ease on hip was suggested 6cm. It was found that pattern with the wrinkle of ease was full of the gaps between body and skirt in 3D clothing air volume. In spreading out to 2D flat pattern from 3D scan data, when the width of skirt pattern was set for(the shell girth of front)/2+(the shell girth of back)/2, it was suggested(the waist girth of front/2)+(the waist girth of back/2) than the shell girth/2 in girth of the waist for the best appearance. And the conversion of 3D scan data into 2D flat pattern in curve shape of crosswise had to spread out of the plane in straight line. The obese women's clothing should be manufactured with systematical consideration of the diversity and scarcity of the obese women's body shape.

벤치발파 설계에서 발파계수 설정에 관한 연구 (Blast Coefficient for Bench Blasting)

  • 본 연구에서는 국내 77개소의 벤치발파 현장을 조사하여 암석의 종류 및 산업별로 발파계수 C값을 설정하였고, 대표적인 산업용 화약류에 대한 실험데이터와 화약류 제조사의 데이터를 이용하여 폭약위력계수 e값을 설정하였다. 그 결과로 발파계수 C값은 화강암 등 대표적으로 조사한 암석 5종에 대한 평균적인 값의 범위가 0.21~0.30이었다. 그리고 석산, 광업 및 건설현장의 산업별로 산출한 발파계수 C값은 각각 평균 0.22, 0.13 및 0.26이었다. 한편, 폭약위력계수 e값은 화약류의 반응에너지, 탄동구포비, 폭발속도 및 Langefors 단위 중량당 강도 등 4가지로 산출하였고, 대표적인 화약류인 일반에멀젼은 1, 고성능에멀젼은 0.9 그리고 ANFO는 1로 산출되었다.

  • For Pneumatic system control, we need a data transmission system with high speed and high realibility for information interchange between main computer and solenoid valves and I/O devices. This paper presents a set of design techniques for a data communication system that is mainly used for pneumatic system control. For this purpose, we first designed hardware modules for an interface between central control module and local node that handles the operation of solenoid valves. In addition we developed a communication protocol for construction of RS-485 based multidrop network, and this protocol is basically designed with a kind of polling technique. Finally we evaluated performance of the developed system. The field test results show that, even under high noise environment, the data transmission of 375Kbps rate is possible up to 1,000m without using repeater. In addition, the system developed in this research is proved to be used easily for extension of a communication network because of its module structure.

  • Because of simplicity and easiness, dynamic pile driving formulae have long been used by most of the field engineers for pile quality control purposes. Yet their reliability have been repeatedly reported unsuitable and the results can lead to significant errors. According to the research results by the authors, the two most important sources of unreliability of dynamic pile driving formulae are uncertainty in the estimation of hammer efficiency and time dependent characteristics of pile bearing capacity. Based on this finding a new method is proposed. By using the actual value of hammer efficiency the pile bearing capacity at the time of driving could be reasonably estimated. By performing restrike test sometime after pile installation, time effect coefficient could be determined. The effectiveness of the proposed method was proven in the actual construction project.

Study on the Fluidity and Strength Properties of High Performance Concrete Utilizing Crushed Sand

  • Recently, it has been difficult to get natural sand for concrete due to an insufficient supply in Korea. Crushed sand was thought as a substitute and previous research has been focused on low fluidity and normal compressive strength (24-30 MPa). Study on high performance concrete using crushed sand is hardly found in Korea. In this study it was investigated that the effect of the crushed sand on fluidity and compressive strength properties of high performance concrete. Blending crushed sand (FM: 3.98) produced in Namyangju, Kyunggido and sea sand (FM: 2.80) produced in Asan bay in Chungnam. The final FMs of fine aggregate were 3.50, 3.23, and 3.08. W/B was set as 0.25 to get high performance. With the test results an analysis of relationship was performed using a statistical program. It was shown that strength property of concrete using crushed aggregate at the very early age or after specific time was mainly affected by strength development properties of binders instead of the crushed sand.
