AADT(Annual Average Daily Traffic) can be obtained by using short-term counted traffic data rather than using traffic data collected for 365 days. The process is a very important in estimating AADT using short-term traffic count data. Therefore, There have been many studies about estimating AADT. In this Paper, we tried to improve the process of the AADT estimation based on the former AADT estimation researches. Firstly, we found the factor showing differences among groups. To do so, we examined hourly variables(divided to total hours, weekday hours. Saturday hours, Sunday hours, weekday and Sunday hours, and weekday and Saturday hours) every time changing the number of groups. After all, we selected the hourly variables of Sunday and weekday as the factor showing differences among groups. Secondly, we classified 200 locations into 10 groups through cluster analysis using only monthly variables. The nile of deciding the number of groups is maximizing deviation among hourly variables of each group. Thirdly, we classified 200 locations which had been used in the second step into the 10 groups by applying statistical techniques such as Discriminant analysis and Neural network. This step is for testing the rate of distinguish between the right group including each location and a wrong one. In conclusion, the result of this study's method was closer to real AADT value than that of the former method. and this study significantly contributes to improve the method of AADT estimation.
In the study, we developed and evaluated a uracil N-glycosylase (UNG)-supplemented single-tube nested reverse transcription-polymerase chain reaction (UsnRT-PCR) assay that can carried out first-round RT-PCR and second-round nested PCR in a reaction tube without reaction tube opening and can simultaneously detect EU- and NA-PRRSV. The UsnRT-PCR confirmed to have a preventing ability of mis-amplification by contamination of pre-amplified PRRSV DNA from previous UsnRT-PCR. Primer specificities were evaluated with RNAs extracted from 8 viral strains and our results revealed that the primers had a high specificity for both genotypes of PRRSV. The sensitivity of the UsnRT-PCR was 0.1 $TCID_{50}$/0.1 mL for EU- or NA-PRRSV, respectively, which is comparable to that of previously reported real time RT-PCR (RRT-PCR). Clinical evaluation on 110 field samples (60 sera and 50 lung tissues) by the UsnRT-PCR and the RRT-PCR showed that detection rates of the UsnRT-PCR was 70% (77/110), and was relatively higher than that of the RRT-PCR (69.1%, 76/110). The percent positive or negative agreement of the UsnRT-PCR compared to RRT-PCR was 96.1% (73/76) or 90.9% (30/33), showing that the test results of both assays may be different for some clinical samples. Therefore, it is recommend that diagnostic laboratory workers use the two diagnostic assays for the correct diagnosis for the relevant samples in the swine disease diagnostic laboratories. In conclusion, the UsnRT-PCR assay can be applied for the rapid, and reliable diagnosis of PRRSV without concerns about preamplified DNA carryover contamination that can occurred in PCR process in the swine disease diagnostic laboratories.
The location-based service is emerging again to the public attention. The location recognition environment up-to-now has been studied with its focus only on a person, an object or a moving object. However, this study proposes a location recognition model that serves to recognize and track, in real time, multiple passengers in a moving vehicle. Identifying the locations of passengers can be classified into two classes: one is to use the high price terminal with GPS function, and the other is to use the economic price compact terminal without GPS function. Our model enables the simple compact terminal to provide effective location recognition under the on-boarding situation by transmitting messages through an interface device and sensor networks for a vehicle equipped with GPS. This technology reduces transmission traffic after detecting the condition of a vehicle (being parked or running), because it does not require transmission/receiving of information on the locations of passengers who are confined in a vehicle when the vehicle is running. Also it extends battery life by saving power consumption of the compact terminal. Hence, we carried out experiments to verify its serviceability by materializing the efficient tracking system for passengers with the detection algorithm of a stopping vehicle proposed in this study. Moreover, about 200 experiments using the system designed with this technology proved successful recognition on on-boarding and alighting of passengers with the maximum transmission distance of 12 km. In addition to this, the running recognition tests showed the test with the detection algorithm of a stopping vehicle has reduced transmission traffic by 41.6% compared to the algorithm without our model.
Differential capacity of the parthenogenetic embryonic stem cells (PESCs) is still under controversy and the mechanisms of its neural induction are yet poorly understood. Here we demonstrated neural lineage induction of PESCs by addition of insulin-like growth factor-2 (Igf2), which is an important factor for embryo organ development and a paternally expressed imprinting gene. Murine PESCs were aggregated to embryoid bodies (EBs) by suspension culture under the leukemia inhibitory factor-free condition for 4 days. To test the effect of exogenous Igf2, 30 ng/ml of Igf2 was supplemented to EBs induction medium. Then neural induction was carried out with serum-free medium containing insulin, transferrin, selenium, and fibronectin complex (ITSFn) for 12 days. Normal murine embryonic stem cells derived from fertilized embryos (ESCs) were used as the control group. Neural potential of differentiated PESCs and ESCs were analyzed by immunofluorescent labeling and real-time PCR assay (Nestin, neural progenitor marker; Tuj1, neuronal cell marker; GFAP, glial cell marker). The differentiated cells from both ESC and PESC showed heterogeneous population of Nestin, Tuj1, and GFAP positive cells. In terms of the level of gene expression, PESC showed 4 times higher level of GFAP expression than ESCs. After exposure to Igf2, the expression level of GFAP decreased both in derivatives of PESCs and ESCs. Interestingly, the expression level of $Tuj1$ increased only in ESCs, not in PESCs. The results show that IGF2 is a positive effector for suppressing over-expressed glial differentiation during neural induction of PESCs and for promoting neuronal differentiation of ESCs, while exogenous Igf2 could not accelerate the neuronal differentiation of PESCs. Although exogenous Igf2 promotes neuronal differentiation of normal ESCs, expression of endogenous $Igf2$ may be critical for initiating neuronal differentiation of pluripotent stem cells. The findings may contribute to understanding of the relationship between imprinting mechanism and neural differentiation and its application to neural tissue repair in the future.

To improve the mechanical properties, such as durabilities and antioxidative characteristics, the covalently cross-linked (CL-) SPEEK (sulfonated polyether ether ketone)/Cs-substituted HPA (heteropoly acid) organic-inorganic composite membranes (CL-SPEEK/Cs-HPAs), have been intensively investigated. The composite membrane were prepared by blending cesium-substituted HPAs (Cs-HPAs), including tungstophosphoric acid (TPA), molybdophosphoric acid (MoPA), and tungstosilicic acid (TSiA) with cross-linking agent content of 0.01 mL. And composite electrolytes composed of Cs-HPAs, prepared by immersion (imm.) and titration (titr.) methods to increase the stability of HPAs in water, were applied to polymer electrolyte membrane electrolysis (PEME). As a result, the proton conductivity of Cs-substituted composite membranes increased rapidly over $60^{\circ}C$ but mechanical properties, such as tensile strength, decreased in accordance with added Cs content. The bleeding-out of Cs-TPA membranes by titration method (50 vol.% Cs) decreased steadily to 2.15%. In the oxidative stability test by Fenton solution, the durability of membranes with Cs-HPA significantly increased. In case of CL-SPEEK/ Cs-TPA membrane, duration time increased more than 1200 hours. It is expected that even though CL-SPEEK/Cs-MoPA membrane shows the high proton conductivity, electrocatalytic activity and cell voltage of 1.80 V for water electrolysis, the CL-SPEEK/Cs-TPA (imm.) is more suitable as an alternative membrane in real system with the satisfactory proton conductivity, mechanical properties, anti-oxidative stability and cell voltage of 1.89 V.
This study was designed to verify reliability and feasibility by analyzing elderly drivers' ability test tools for older drivers aged 65 or older, which were improved in 2018 and are currently being conducted by the Korea Highway Traffic Authority. Only those aged 65 or older who voluntarily applied to the elderly driving ability evaluation system implemented by the Seoul branch of the Korea Highway Traffic Authority. The research was conducted for about 50 days until Aug. 31, 2018, starting with the registration and inspection of the first study subjects. The analysis performed a correlation analysis with existing tools and cognitive testing tools (MMSE_K) to determine their feasibility and reliability as an improved tool in 2018. As a result, the first, the speed distance, time-space memory, and dispersionism of each sub-component of the old version showed statistically significant static correlation with the sub-factor of the current version. Persistence, on the other hand, was not statistically significant to the current version. The limitations of this study were as follows. Most of the people in the study were highly educated and residents in the metropolitan area. Therefore, it is likely that the results of MSE_K, which checks cognitive and judgment skills, have been upgraded. Also, cognitive tools that are measured by computers are likely to have real measurement errors for generations who are not familiar with computers. Therefore, it is expected that improvement and development of tools for improving the limit points at the site and assessing actual operation capability will be required.

Background: Korean Red Ginseng (KRG) is known to have antianxiety properties. This study was conducted to investigate the anxiolytic effects of KRG extract (KRGE) during ethanol withdrawal (EW) and the involvement of the mesoamygdaloid dopamine (DA) system in it. Methods: Rats were treated with 3 g/kg/d of ethanol for 28 d, and subjected to 3 d of withdrawal. During EW, KRGE (20 mg/kg/d or 60 mg/kg/d, p.o.) was given to rats once/d for 3 d. Thirty min after the final dose of KRGE, anxiety-like behavior was evaluated in an elevated plus maze (EPM), and plasma corticosterone (CORT) levels were determined by a radioimmunoassay (RIA). In addition, concentrations of DA and 3,4-dihydroxyphenylacetic acid (DOPAC) in the central nucleus of the amygdala (CeA) were also measured by high performance liquid chromatography (HPLC). Results: The EPM test and RIA revealed KRGE inhibited anxiety-like behavior and the over secretion of plasma CORT during EW. Furthermore, the behavioral effect was blocked by a selective DA D2 receptor (D2R) antagonist (eticlopride) but not by a selective DA D1 receptor (D1R) antagonist (SCH23390). HPLC analyses showed KRGE reversed EW-induced decreases of DA and DOPAC in a dose-dependent way. Additionally, Western blotting and real-time polymerase chain reaction (PCR) assays showed that KRGE prevented the EW-induced reductions in tyrosine hydroxylase (TH) protein expression in the CeA and TH mRNA expression in the ventral tegmental area (VTA). Conclusion: These results suggest that KRGE has anxiolytic effects during EW by improving the mesoamygdaloid DA system.
Objectives: The aim of this study was to observe the reduction of lipid accumulation by treatment with Akkermansia muciniphila extract on 3T3-L1 adipocytes. Methods: After treating pasteurized Akk. muciniphila strains in HT-29 colorectal cancer cell, the relative expression of interleukin (IL)-8, tumor necrosis factor-α, IL-6, and IL-1β mRNA was analyzed by real time polymerase chain reaction, respectively. 27 strains of Akk. muciniphila which have anti-inflammatory effects were selected. 3T3-L1 pre-adipocytes were treated with Akk. muciniphila for 24 hr and then measured the toxicity using water soluble tetrazolium salt assay. The cells were incubated for 4 days and then differentiated into adipocytes using the medium including adipogenic reagents for 10 days. The Akk. muciniphila was treated when the medium was exchanged for differentiation medium at 4th day and insulin medium at 6th day. To observe the lipid accumulation, the cells were stained with Oil red O dye and were measured using a spectrophotometer. Results: In the cytotoxicity test, the cell viability of 3T3-L1 pre-adipocytes was significantly increased compared to the control group which untreated with Akk. muciniphila, and there was no cytotoxicity of Akk. muciniphila at 1×107 CFU/mL. The results on Oil red O staining and absorbance measurements were showed a significant decrease in lipid accumulation in the group which was treated with Akk. muciniphila compared to the control group. Conclusions: In our results, Akk. muciniphila has the inhibitory effect of lipid accumulation in 3T3-L1 adipocytes. This suggests that Akk. muciniphila could be help to improve obesity.

Due to the low C/N ratio of domestic wastewater characteristic, addition of external carbon source for the effective N and P removal is necessary. High organic content of food waste can be used for the external carbon source in biological nutrient removal processes, The applicability of condensate of food waste (CFW), which is produced during the high-rate fermentation process, was examined in membrane bioreactor for the nutrient removal. Under the various operating conditions, nutrient removal efficiencies and membrane fouling characteristics were evaluated using synthetic wastewater. From nitrate utilization rate (NUR) test, denitrification rate was 0.19 g $NO_3-N/g$ VSS/day. With the addition of CFW increased, average removal efficiencies of T-N and T-P could be increased up to 64% and 41%, respectively. Also the optimal retention time was 3 hr/5 hr for anoxic/aerobic reactor. When applied to real sewage, membrane fouling resistance was increased up to 60%, which could be reduced from $10.4{\times}10^{12}m^{-1}$ to $5.9{\times}10^{12}m^{-1}$ with the control of influent suspended solid concentration. In summary, it was suggested that CFW could be used as an economical and effective carbon source for membrane assisted biological N and P removal.
Objective: The present study aims to evaluate the visual reactions of users when they are playing games of different flow levels, and to explore the visual variables that can sensitively reflect the different flow levels. Background: The flow is defined as a psychological state where interface users feel their actions in a virtual setting identical to those in real environment. To measure the flow states of users, the questionnaire-based FSS(Flow State Scale) has mostly been used. However, this method is a qualitative test that has limits in terms of the accuracy of users' flow experiences. Therefore, more accurate methods to measure users' flow experiences are required. Method: Ten subjects participated in the experiment, where the independent variables were three games with different flow levels(puzzle games, dot drawing and coloring) and the time frame(the first and last 10 seconds in game playing), whereas the dependent variables included the pupil size and the frequency and duration of eye blinking. This study was a within-subject design. Each participant performed three types of games with different flow levels 3 times for each for 10 minutes, and their visual reactions to each game were measured. Results: The higher the flow cause the bigger pupil size(p<0.01) and the lower eye blinking frequency(p<0.1), indicating that different types of games lead to different flow levels. The pupil size during the last 10 seconds when the flow level was higher was bigger by 2.1% compared with that during the first 10 seconds in game playing(p<0.1), and the eye blinking frequency decreased by 12%(p<0.01). Conclusion: It was found out that the pupil size and the frequency and duration of eye blinking were psychophysiological indices for evaluating users' flow experiences, which could quantify the flow states users go through. The psychophysiological variables capable of measuring diverse aspects of the flow need diversifying to be applicable to precise measurement of the flow. Application: These studies are warranted for both quantitative analysis of flow levels and qualitative improvement of cyber leisure in line with development of healthy games.
