The objectives of this study were to examine the physiological responses to ozone and to measure ozone uptake rates of Betula species exposed to relatively high concentration of pollutants. At the end of the growing season, photosynthesis, pigments contents, antioxidants (SOD and GR) and ozone uptake rates were measured or estimated at the leaves of five Betula species (Betula costata, B. davurica, B. platyphylla var. japonica, B. schmidtii and B. ermanii) exposed to 100ppb ozone concentration. On the termination of the experiment, growth effects were determined by measuring leaf area and dry weights of leaf, stem and root. Ozone treatment showed the significant reduction the leaf area and dry weight of four Betula species, except for B. ermanii. Shoot / root (SR) ratio of five species represented two different types. SR ratio of B. costata and B. davurica were lower than control, in contrast, SR ratio of B. platyphylla var. japonica, B. schimidtii and B. emani, were higher than that of control. The photosynthetic responses of five species were different in responses to ozone exposure. Four species, except for B. emanii, maintained or increased the stomatal conductance, but B. emanii decreased both stomatal conductance and photosynthesis. SOD activities of five species decreased by the ozone exposure, especially B. ermanii showed the largest reduction, GR activities of B. platyphylla var. japonica and B. schmidtii increased, B. costata and B. emanii decreased. Instantaneous ozone uptake rate was the highest at the leaves of B. ermanii and B. costata, ozone uptake per seedling was the highest at the leaf of B. schmidtii and B. emanii. It was concluded that B. costata, B. davurica and B. platyphylla var. japonica, appeared the growth reduction and visible ozone injury, were sensitive species to ozone, and B. schmidtii with the increased antioxidant activity and B. ermanii without the growth reduction were relatively resistant species to high ozone concentration at the early growing stage.

The rate of oxygen consumption of the silkworm eggs was measured to set up the barometer for measuring the effect of egg chilling on diapause termination. In diapause eggs, $O_2$ uptake showed a maximum of 79.2 ${mu}ell$/mg eggs/hr, at one day after oviposition and then gradually decreased to 2.2${mu}ell$/mg eggs/hr at 9days. The rates of oxygen uptake of eggs raised immediately after HCl-treatment and reached to a maximal level of 484.5 ${mu}ell$/mg eggs/hr in 9-day-old eggs, which corresponds to 220 fold that of diapause eggs, and the hatching ability was also over 50%. In order to break diapause, eggs incubated at $25^{\circ}C$ for 30 days after oviposition had to be kept at least for 45 days at 5$^{\circ}C$, but chilled eggs for 90 days or longer hatched at 10 days as normal hatching periods. We also investigate periods which the value of $O_2$ uptake of eggs chilled during 15 to 120 days at 5$^{\circ}C$ reached at 200 ${mu}ell$/mg eggs/hr. As a result of that, $O_2$ uptake of eggs chilled during 15 to 45days were not reached at 200 ${mu}ell$/mg eggs/hr and longer the chilling durations are, the higher the rate of $O_2$ uptake is and the longer the chilling durations are, the shorter the periods up to hatching is. And also, hatch-ability increased rapidly as $O_2$ uptake reaches over 20/eggs/hr.
Objectives: The purpose of this study was to examine the efficacy of Picrorrhizae Rhizoma (PR) aqueous extracts on the formalin-induced paw chronic inflammation in mice. Methods: PR extracts (500, 250 and 125mg/kg) or distilled water (DW) were orally administered once a day for 10 days to formalin-injected chronic inflammatory mice. The paw thicknesses and volumes were measured daily and the paw wet-weight and histological profiles were conducted at termination with paw tumor necrosis factor (TNF)-${\alpha}$ contents measurement. The anti-inflammatory effects of PR extracts were compared with dexamethasone. Results: In DW treated control group, the paw thickness, paw wet-weights and paw TNF-${\alpha}$ contents were markedly increased. Severe chronic inflammation signs such as severe fibrosis, the formation of necrotic debris, and infiltration of inflammatory cells were detected in histopathological observations. However, these formalin-induced changes were dramatically decreased by treatment of dexamethasone and all three different dosages of PR extracts. The anti-inflammatory effects of PR at highest dose were slighter than that of dexamethasone, but it did not show any harmful effects on the body weight contrary to dexamethasone. Conclusion: These results suggest that PR extracts have safe and favorable effects on formalin-induced chronic inflammation.

The effects of pentylenetetrazol (PTZ), a GABA receptor antagonist, were studied on passive avoidance learning and expression of heat shock protein 70 (hsp70), neuroglobin, and fatty acid binding protein-7 (fabp-7) genes. Zebrafish were trained to stay in a dark compartment to avoid a weight dropping in an acryl shuttle box with a central sliding door. In two training sessions of 2 h interval, each consisting of 3 trials, the crossing time was significantly increased from $43.2{\pm}14.4s$ to $149.3{\pm}38.5s$ in the first training session and remained $116.1{\pm}36.0s$ s in the first trial of the second training session in the control. In zebrafish treated with PTZ before the first training session, the crossing time was significantly increased neither in the first nor in the second training session. However, the increased crossing time was maintained in the second training session when 10 mM PTZ was treated three times for 10 min at 30 min intervals between the first and second training session. Quantitative real-time PCR showed that expression level of hsp70 mRNA increased two to eight fold over that of control in the brain at 0-24 h after termination of PTZ treatment. No change in expression of neuroglobin and fabp-7 mRNA was shown in PTZ-treated zebrafish. Our studies suggest that PTZ impairs learning ability in avoidance response and also modifies expression of genes related to the neuroprotection.
A indigenous freshwater bivalve Unio douglasiae was introduced to compare the differences in the efficacy of algal bloom control and the appearances of mussel-mediated adverse effects between two different reservoirs such as mesotrophic (Shingu r.) and hypertrophic (Seokmun r.). We constructed the study mesocosm in the shore of each reservoir, stocked the mussel at density of $30indiv./m^3$ for 7 days, and measured daily the phytoplankton density and water quality. In mesotrophic reservoir, even though approximately 38% of suspended solids and chlorophyll-a was reduced by stocked bivalves for the first 3 days, algal density, ammonia and soluble reactive phosphorus gradually increased with increasing mussel death. In hypertrophic reservoir, mussels strongly inhibited suspended solids and chlorophyll-a by the termination of study with no increase of mussel death and nutrient, especially ammonia concentration. In both reservoirs, a strong selectivity showed mussels preferred to diatom rather than cyanobacteria and green algae without algal density and nutrient level. Our results indicate that an introduction of freshwater bivalve U. douglasiae is more strategic to improve water quality of hypertrophic than mesotrophic reservoir, but many preliminary studies on the treatment method and the selection of target water system are required.
This study investigated the effects of temperature on the shoot growth and flowering of potted kumquat [Fortunella margarita ( Lour.) Swingle] trees grown in subtropical conditions of I-Lan County in Taiwan. Temperature treatments included T 25-32, T 17-25, T 22, and T 18. The T 25-32 treatment trees were to the day/night temperatures of $25/18^{\circ}C$ for 2 weeks, followed by 28 weeks at $32/25^{\circ}C$. T 17-25 was exposed for 4 weeks to $17/10^{\circ}C$ followed by 26 weeks at $25/18^{\circ}C$. T 22 and T 18 were exposed at $22/18^{\circ}C$ and $18/13^{\circ}C$, respectively, for the entire duration of the experiment. Control trees were placed in a plastic greenhouse under conditions similar to the natural environment. The kumquat trees exposed to high-temperature environment of $32/25^{\circ}C$ showed more frequent and speedy sprouting of new buds, but induced the earlier termination of shoot elongation growth, resulting in decreased vegetative growth. The temperature treatments lower than $22^{\circ}C$ suppressed the new shoot production but increased the shoot growth period, resulting in increased shoot length and diameter. Temperatures higher than $25/18^{\circ}C$ readily induced flowering, with flowering being advanced under the higher temperature conditions such as $32/25^{\circ}C$. However, flowering was substantially inhibited under temperature conditions lower than $22/18^{\circ}C$, indicating the negative role of relatively lower temperatures on flowering of kumquat trees.
Background: Anticoagulant therapy can be required during pregnancy with prosthetic heart valves. Warfarin and heparin provide real protection against thromboembolic phenomena, but they also carry serious risks for the fetus and the mother. In an attempt to identify the best treatment for pregnant women with cardiac valve prostheses who are receiving anticoagulant, we studied 19 pregnancies, the warfarin was discontinued and heparin was administered every 12 hours by subcutaneous injection in doses adjusted to keep the midinterval aPTT in the therapeutic range(at least 2-2.5 control) from the conception to the 12th week of gestation and oral antiocagulant was then administered until the middle of the third trimester in the therapeutic range(at least 2 INR), and heparin therapy was restared until delivery. Also in order to avoid an anticoagulant effect during delivery, it has been our practice to instruct women to either discontinue their heparin injections with the onset of labur or to stop heparin injections 12 hours prior to the elective induction of labour. Result: The outcome of 19 pregnancies managed with above protocol was spontaneous abortion in 3 cases, voluntary termination in 2 cases, premature delivery at 35 weeks in 1 case and delivery at full-term in 14 cases. There was no maternal morbidity and moratality and fetopathy. Conclusion: We conclude that in the second and third trimester of pregnancy, warfarin provide effective protection against thromboembolism, Oral antiocagulant therapy should be avoided in 2 weeks before delivery because of the risk of serious perinatal bleeding caused by the trauma of delivery to the anticoagulated fetus. However, the substitution of heparin at first trimester and 2 weeks before delivery reduce the incidence of complications.
Effects of repeated physical exercise on the carbon tetrachloride ($CCl_4$) hepatotoxicity were examined in adult female rats. Rats were introduced into a cylindrical rotating cage and forced to exercise for 1 hr each day, 6days/week, for 5 consecutive weeks at a speed starting from 10m/min, increased by 1m/min per day until the speed reached 27m/min. Significantly less body weight gain was observed in the exercise group suggesting that physical fitness had been induced in these animals. Eighteen hours following termination of the last exercise bout rats were treated with $CCl_4$(2 mmol/kg.ip). The $CCl_4$-induced heptotoxicity was significantly potentiated in the repeated exercise group compared to the resting sedentary animals as determined by changes in serum sorbitol dehydrogenase (SDH), glutamic oxaloacetic transaminase(GOT), glutamic pyruvic transaminase (GPT), and glucose-6-phosphatase(G-6-Pase) activities when measured 24hrs following the $CCl_4$ treatment. Hepatic drug metabolizing activity was determined in order to elucidate the underlying mechanism of potentiating action of the $CCl_4$ hepatotoxicity induced by repeated physical exercise. Repeated exercise increased the hepatic microsomal cytochrome P-450 contents and aminopyrine N-demethylase activity. The results suggest that the potentiation of $CCl_4$ hepatotoxicity by repeated exercise is associated with induction of the mixed function oxidase (MFO) enzyme system mediating the metabolism of $CCl_4$ to its active metabolite(s).
Various kinds of genetically modified organisms (GMO) and processed foods have been developed during recent years. Genetically modified organisms can be classified into several groups as their development methods. Generally, GMO has three foreign DNA regions such as gene expression adjustment region(Promoter), termination region (terminator) and structure gene. Detection of these regions can be done particularly by polymerase chain reaction (PCR). PCR-based detection can virtually be performed for any GMO within short of time. The most important prerequisite for the application of PCR-based detection is to decide abstraction method of efficient nucleic acids. Specially, in the case of processed food, because nucleic acids of foodstuffs are damaged by heat treatment (sterilization), pressure and fermentation, DNA must be extracted ken the samples prior to PCR analysis. Although many DNA extraction protocols are available, they have rarely been compared in a comprehensive method. In this study low widely used commercial and non-commercial DNA extraction methods-DNeasy$^{TM}$, Wizard$^{TM}$, CTAB, phenol/chloroform system-were compared with respect to the quality and yield of nucleic acids and insertion genes.nes.
To verify anti-diabetic effect of oligopeptide with His-Pro repeats (mHP peptide), the oligopeptide was first secreted and optimized using the secretion vector, pRBAS with alkaline protease gene promoter and the signal sequence in Bacillus subtilis and directly the anti-diabetic effect of the mHP peptide was investigated in insulinoma cell, RINm5F cell line. The oligopeptide gene was obtained by annealing oligonucleotides with repeated His-Pro sequence and finally was constructed as 18 dipeptides (108 bp and 4.0 kDa) coding gene, named oligopeptide with His-Pro repeats (mHP peptide) to make cyclo(His-Pro) known to be anti-diabetic effects. The region encoding the oligopeptide gene was subcloned into the pRBAS secretion vector (E.coli-Bacillus shuttle vector) after PCR amplification using the designed primers including initiation and termination codons and His tag, named pRBAS-mHP (6.56 kb). To optimize secretion of the oligopeptide, various culture conditions were investigated in Bacillus subtilis LKS. As a result, the secreted oligopeptide was maximally measured (approximately $59.6{\mu}g/mL$) in 3 L batch culture and the highest secretion was achieved at $30^{\circ}C$, PY medium, and carbon sources (particularly barley and glycerol). In the RINm5F cells treated with 2 mM STZ, the oligopeptide treatment (0.1 mg/mL) restored the cell viability (10%) and reduced the nitric oxide (NO) generation (35%) and DNA fragmentation (90%). And also, insulin secretion level was increased to 17% higher than in STZ-treated RINm5F cells. These results suggest that the oligopeptide with His-Pro repeats could be a candidate material for anti-diabetic agent against STZ-induced diabetes.
