Germination Characteristics and Seed Dormancy of Iris dichotoma Pall., an Endangered Species Native to Korea

  • Iris dichotoma Pall. found on Daechung Island in Korea has been designated as an endangered species. To aid in conservation efforts of this species, this study investigated its germination characteristics and seed dormancy type. Four sets of seeds were incubated at different temperatures (4/1℃, 15/6℃, 20/10℃, and 25/15℃). One set of seeds was cold stratified (4 weeks at 4/1℃). The final germination rate and mean germination time showed that the optimal germination temperature was 25/15℃. Final germination rates were ~70%, showing no significant difference among temperature treatments. However, mean germination time were significantly different among all temperature treatments except for 4/1℃. Mean germination time for seeds with temperature treatments of 15/6℃, 20/10℃, and 25/15℃ were 3.2, 2.1, and 1.5 weeks, respectively. At 25/15℃, the mean germination time was half of that at 15/6℃. Seeds of I. dichotoma had fully developed embryos at the time of dispersal. No additional growth of the embryo was observed. Cold stratification did not affect the final germination rate or the mean germination time. This study shows that seeds of I. dichotoma have no physiological or morphological dormancy, unlike other members of the Iris genus known to have seed dormancy that needs a relatively high incubation temperature (≥25/15℃) for mass propagation to occur. These results will be useful for understanding ecophysiological mechanisms related to the species' habitat. They are also useful for mass propagation of I. dichotoma for the purpose of conserving this endangered species.

The Variation of Water Temperature and the Mass Mortalities of Sea Squirt, Halocynthia roretzi along Gyeongbuk Coasts of the East Sea in Summer, 2006. (2006년 하계 경북 연안의 수온변화와 우렁쉥이 폐사)

  • Mass mortalities of the farmed sea squirt, Halocynthia roretzi, occurred in August, 2006 along the Gyeongbuk coasts of the East Sea without any pathological symptom. Suspecting the No. 10 typhoon, Wukong, which had passed over the relevant locations in the middle of August, 2006 as a culprit of the mortalities, we compared water temperature stratifications of the affected coasts before and after the typhoon. Just prior to the typhoon, cold waters lower than $15^{\circ}C$ were stratified at depths deeper than 10m probably by a persisting upwelling of cold bottom waters. The typhoon enforced water off-coast waters into coasts for 5 days, replacing the cold waters at depths $10{\sim}20m$ which can be justified by Ekman Spiral Effect. The results were an increment of water temperature by $10^{\circ}C$ and more at depths where sea squirts were suspended. The sudden elevation of water temperature might drive the sea squirts under abnormal stress to mass mortality.

Species Composition and Spatial Distribution of Euphausiids of the Yellow Sea and Relationships with Environmental Factors

  • We investigated species composition and spatial distribution of the euphausiid community in the Yellow Sea and identified the relationship with environmental factors (temperature, salinity, chlorophyll $\alpha$, nitrate, phosphate, and silicate) using bimonthly data from June, 1997 to April, 1998. The environment varied during the sampling period. In warm seasons, thermocline was well developed rendering lower temperature and higher salinity and nutrient concentrations in the bottom layer. During cold seasons the water column was well mixed and no such vertical stratification was noted. Horizontal distribution of temperature, however, differed slightly between near-coast and offshore areas because of the shallow depth of the Yellow Sea, and between southern and northern areas because of the intrusion of water masses such as Yellow Sea Warm Current and Changjiang River Diluted Water. Four euphausiid species were identified: Euphausia pacifica, E. sanzoi, Pseudeuphausia sp. and Stylocheron affine. E. sanzoi and S. affine were collected, just one juvenile each, from the southern area in June and December, respectively. Pseudeuphausia sp. were collected in the eastern area all the year round except June. E. pacifica occurred at the whole study area and were the predominant species, representing at least 97.6% of the euphausiid abundance. Further, the distribution pattern of the species was varied in regards to developmental stages (adult, furcilia, calyptopis, egg). From spring to fall, E. pacifica adults were abundant in the central area where the Yellow Sea Bottom Cold Water prevailed. Furcilia and calyptopis extended their distribution into nearly all the study area during the same period. From late fall to winter, adults were found at the near-coastal are a with similar pattern for furcilia and calyptopis. The distribution pattern of E. pacifica was consistent regarding temperature, salinity, and three nutrients during the sampling period, whereas chlorophyll $\alpha$ showed a different pattern according to the developmental stages. The nutrients should indirectly affect via chlorophyll $\alpha$ and phytoplankton concentration. With respect to these results, we presented a scenario about how the environmental factors along with the water current affect the distribution of E. pacifica in the Yellow Sea.

Simulations of Temporal and Spatial Distributions of Rainfall-Induced Turbidity Flow in a Reservoir Using CE-QUAL-W2 (CE-QUAL-W2 모형을 이용한 저수지 탁수의 시공간분포 모의)

  • A real-time monitoring and modeling system (RTMMS) for rainfall-induced turbidity flow, which is one of the major obstacles for sustainable use of reservoir water resources, is under development. As a prediction model for the RTMMS, a laterally integrated two-dimensional hydrodynamic and water quality model, CE-QUAL-W2 was tested by simulating the temperature stratification, density flow regimes, and temporal and spatial distributions of turbidity in a reservoir. The inflow water temperature and turbidity measured every hour during the flood season of 2004 were used as the boundary conditions. The monitoring data showed that inflow water temperature drop by 5 to $10^{\circ}C$ during rainfall events in summer, and consequently resulted in the development of density flow regimes such as plunge flow and interflow in the reservoir. The model showed relatively satisfactory performance in replicating the water temperature profiles and turbidity distributions, although considerable discrepancies were partially detected between observed and simulated results. The model was either very efficient in computation as the CPU run time to simulate the whole flood season took only 4 minutes with a Pentium 4(CPU 2.0GHz) desktop computer, which is essentially requited for real-time modeling of turbidity plume.

Effects of Stratification and Gibberellins on Germination of Chelidonium majus L. subsp. asiaticum H.Hara Seed (애기똥풀 종자의 휴면타파를 위한 층적과 GAs의 효과)

  • 애기똥풀(Chelidonium majus L. subsp. asiaticum H.Hara)의 지상부는 백굴채라고 불리며 항암, 항균 등의 효능을 가지고 있어 예로부터 약용으로 이용되었다. 그러나 천연 약용소재로 개발가치가 높음에도 불구하고 실생묘의 대량 확보를 위한 종자번식 관련 연구가 전무한 실정이다. 본 연구에서는 애기똥풀의 발아 및 휴면 특성을 분석함으로써 추후 종자번식을 위한 기초자료를 제공하기 위해 수행되었다. 실험재료로 사용된 애기똥풀 종자는 2019년 6월 경상북도 봉화군에서 채종하였으며 정선 후 건조상태로 실온(25℃)에 보관하였다. 종자의 1000립중은 0.57±0.01g이며, 배종비는 0.21±0.12로 조사되어 형태적으로 미성숙한 배를 가지고 있다. 종자의 활력은 테트라졸륨 검정을 통해 79%로 확인되었다. 종자의 기본 함수율은 7.64±0.25%였으며, 침지 2일 차에 최대함수율(32.6±0.34%)에 도달하였다. 초기 발아검정은 25/15℃ (light/dark, 16/8h)챔버에서 명조건과 암조건으로 구분하여 30일간 진행하였으며, 암조건은 알루미늄 호일로 광을 완전히 차단하고 최종 발아율만 조사하였다. 발아율을 조사한 결과, 명조건은 0.75±0.80%, 암조건은 5.50±1.55%로 조사되어 형태적인 휴면과 생리적인 휴면을 모두 갖는 것으로 판단하였다. 애기똥풀의 생태적인 환경을 고려하여 12주의 Summer temperature(S12, 25/15℃, 16/8h)로 층적하고 연속하여 Autumn temperature(A, 15/10℃, 16/8h)를 4주(A4)와 8주(A8)로 달리하여 종자의 휴면타파를 유도하였다. 층적 후 발아율은 모두 증가되었으며, S12-A4(54%)는 S12-A8(24.5%)에 비해서 휴면타파에 효과적이었다. 휴면타파 효과의 증진을 위해 층적처리한 종자를 500ppm의 GA3와 GA4+7에 각 24시간동안 4℃에서 침지시킨 뒤 동일한 조건에 파종하였다. GA3가 처리된 S12-A4는 발아율이 82.0%, S12-A8는 30.8%로 유의적으로 증가하였으나 GA4+7 처리는 S12-A4(48.5%)와 S12-A8(21.3%)에서 발아율 향상 효과가 없었다. 결론적으로 애기똥풀의 휴면타파를 위해서 12주의 Summer temperature와 4주의 Autumn temperature 변온층적과 추가 GA3 처리가 휴면타파를 위한 효과적인 처리방법으로 판단된다.

Development and Assessment of LSTM Model for Correcting Underestimation of Water Temperature in Korean Marine Heatwave Prediction System (한반도 고수온 예측 시스템의 수온 과소모의 보정을 위한 LSTM 모델 구축 및 예측성 평가)

  • The ocean heatwave is emerging as a major issue due to global warming, posing a direct threat to marine ecosystems and humanity through decreased food resources and reduced carbon absorption capacity of the oceans. Consequently, the prediction of ocean heatwaves in the vicinity of the Korean Peninsula is becoming increasingly important for marine environmental monitoring and management. In this study, an LSTM model was developed to improve the underestimated prediction of ocean heatwaves caused by the coarse vertical grid system of the Korean Peninsula Ocean Prediction System. Based on the results of ocean heatwave predictions for the Korean Peninsula conducted in 2023, as well as those generated by the LSTM model, the performance of heatwave predictions in the East Sea, Yellow Sea, and South Sea areas surrounding the Korean Peninsula was evaluated. The LSTM model developed in this study significantly improved the prediction performance of sea surface temperatures during periods of temperature increase in all three regions. However, its effectiveness in improving prediction performance during periods of temperature decrease or before temperature rise initiation was limited. This demonstrates the potential of the LSTM model to address the underestimated prediction of ocean heatwaves caused by the coarse vertical grid system during periods of enhanced stratification. It is anticipated that the utility of data-driven artificial intelligence models will expand in the future to improve the prediction performance of dynamical models or even replace them.

The Effect of Hydrology on Phytoplankton Assemblages and Its Adaptive Strategies in Lake Hwaseong, Estuarine Reservoir with Seawater Exchange, Korea (해수유통 중인 간척담수호 화성호에서 식물플랑크톤의 군집과 적응전략에 대한 수문학적 영향)

  • A survey was carried out to understand the influence of hydrology on the composition, abundance and adaptive strategies of phytoplankton in artificial Lake Hwaseong, an estuarine reservoir with seawater exchange through a sluice. Samples were collected seven times from May to October 2012. Hydrological events (seawater exchange, rainfall) resulted in a wide variation in salinity along with nutrients and turbidity. Shifts in the dominant phytoplankton composition occurred on every survey. Chlorophyll-a ranged from 9.7 to $104.1{\mu}g\;L^{-1}$. Multivariate analysis allowed us to identify the four phases on phytoplankton community change. Phase I (May~June) was characterized by small-sized Gymnodinium sp. and Heterosigma akashiwo dominated in warm temperature and high salinity derived from seawater exchange, and followed by Cylindrotheca closterium blooms due to rainfall and winds during phase II (July and September). During phase III (August), the dominance of Oscillatoria spp. was correlated with high temperature and low salinity. Abundant cryptomonads were associated with lower temperature during phase IV (October). Adaptive strategies were identified in the phytoplankton as morphological and physiological characteristics. These strategies identified small-sized flagellates as CR-strategists, fast-growing opportunistic species, which might favor the weak stratification of lake due to the seawater exchange during phase I and IV. Dominant species during phase II and III were characterized with R-strategists, medium-sized stress-tolerant species, which might favor turbulence by river flow. The results indicate that stronger stratification following the termination of seawater exchange for the freshening might intensify the predominance of smaller flagellates. In conclusion, this study suggests that hydrology may drive phytoplankton community change and blooms through the controls of salinity, turbulence and nutrients.

Effects of Temperature, Light and Plant Growth Regulators on the Seed Germination of Lavandula angustifolia Mill (잉글리쉬 라벤다의 종자발아(種子發芽)에 대한 온도(溫度), 광(光) 및 생육조절물질(生長調節物質)의 영향(影響))

  • This study was conducted to establish mass propagation system from seeds of Lavandula angustiolia Mill. Only 4% of dry seeds were germinated when they were incubated in $4^{\circ}C$. Germination rate of stratified seeds for 9 weeks was 15% higher in light than darkness. Soaking with 1000mg/l of $GA_3$ was helpful to overcome the effect of darkness. The optimal temperature for germination was $25^{\circ}C$ for the seeds that were treated with$GA_3$, solution. For the improvement of germination rates, pretreatment of $GA_3$ at 500-2000mg/l showed about 75% of germination, and in the combination treatment of $GA_3$, and BA, germination rate increased by about 10% in the treatment of 1000mg/l $GA_3+10mg/l$ BA compared with the 1000mg/l $GA_3$ treatment. Cold stratification treatment was very effective for seed germination, and over 70% of seeds were germinated when they were incubated in $4^{\circ}C$ for 9-12 weeks. In addition, there was synergic effect on the seed germination subject to stratification and 1000mg/l $GA_3$, treatment for 6 months. In the mixture of vermiculite: peatmoss (1 : 1, v/v), emergence rate was 6.7% in control and 65% in 1000mg/l $GA_3$ treatment, respectively.

Seed Germination, Seedling Grwoth and Optimal Seedling Density of Phellodendron amurense Rupr. in Nursery (황벽나무의 종자발아(種子發芽)와 묘목생장(苗木生長) 및 적정생육밀도(適正生育密度)에 관(關)한 연구(硏究))

  • We examined the effect of pretreatments on seed germination, seedling growth, optimal seedling density of Phellodendron amurense. The seeds were carefully purified after collection in late Oct. from natural population at Mt. Chiri. For the evaluation of pretreatment effects, seeds were sowed in an experimental nursery after being stored in open ground($2{\sim}8^{\circ}C$) with soaking in 20% $H_2O_2$, 50-fold pon-pon solution, cold moist stratification in refrigerator($2{\sim}4^{\circ}C$) and dry storage in roam temperature($4{\sim}18^{\circ}C$), respectively, For the effect of growth density of seedlings, seedlings germinated in nursery were precisely controlled with 53, 104, 220, 380 seedlings per $m^2$. Among pretreatments for the promotion of germination rate, pon-pon treatment(68.8%) was highest, the others were higher than that in dry storage(2.3%), and these seed pretreatment methods were significantly different at 1% level. The height growth of seedlings showed the most vigor from Jun.12 to Jul. 8, and from Jul.20 to Aug. 2, and it reached to 72.8% of height growth. In case of 1-0 seedling, although seedling density per $m^2$ was increased, seedling growth was decreased. The optimal seedling density was 104 seedlings per $m^2$. The standard seedlings for plantation were above height 55cm, root collar diameter 8.1mm, and root length 21cm. These results demonstrated that seed of P. amurense requires stratification for the germination promotion.

Seed Dormancy of Jerusalem Artichoke (Helianthus tuberosus L.) and Seed Treatment for Germination Induction (돼지감자 종자의 휴면성과 발아유도를 위한 종자처리)

  • Jerusalem artichoke (Helianthus tuberosus L.) as a biomass potential crop has several distinct advantages such as vigorous growth on poor land and high yield of carbohydrate. In this crop, seed dormancy has hampered the efforts of seed-propagration and to use them in breeding programs for improving jerusalem artichoke. Several seed treatments were tested to determine their effectiveness in overcoming the seed dormancy found in five collected varietes of jerusalem artichoke. The first results showed that the seed fertilities of five collected varieties ranged from 2.4% to 14.7% and the number of seed produced by one plant ranged from 88 to 1058. Germinability of seeds stored for 3 months at room temperature after harvest was almost 0% and it was not improved by addition to the treatments of temperature, light and GA3, while germinability of seeds stored for 27 months at room temperature after harvest increased to 47.5% in germination rate. But the removal and pin-pricking of seedcoat were very effective in breaking the seed dormancy, giving germination of 96.8% and 82.3%, respectively. These results showed that the seed dormancy of jerusalem artichoke was induced by the seedcoat. Besides the treatment of seedcoat removal and seedcoat-pinpricking, the treatment of low and wet stratification was also effective in breaking the seed dormancy of jerusalem artichoke. Whole dormant seeds incubated for 70 days in low and wet condition germinated over 85%.
