Pharmacokinetics of Propentofylline and the Quantitation of Its Metaolite Hydroxypropentofylline in Human Volunteers

  • Propentofylline (PPF, 3-methyl-1-(5-oxohexyl)-7-propylxanthine) has been reported to be effective for the treatment of both vascular dementia and dementia of the Alzheimer type. The pharmacological effects of PPF may be exerted via the stimulation of nerve growth factor, increased cerebral blood flow, and inhibition of adenosine uptake. The objectives of this experiment are to determine the kinetic behavior of PPF, to identify, and to quantify its metabolite in human. Blood samples were obtained from human volunteers following oral administration of 200mg of PPF tablets. For the identification and quantification of the metabolite, 3-methyl-1-(5-hydroxyhexyl)-7-propylxanthine (PPFOH), PPFOH was synthesized and identified by gas chromatography/mass spectroscopy (GC/MS) and $^1H$-nuclear magnetic resonance spectroscopy. The molecular weight of synthesized metabolite is 308 dalton. The PPF and PPFOH in plasma were extracted with diethyl ether and identified by electron impact GC/MS. The plasma concentrations of PPF and PPFOH were determined by gas chromatography/nitrogen phosphorus detector in plasma and their pharmacokinetic parameters were determined. The mean half-life of PPF was 0.74 hr. The areas under the curve (AUCs) of PPF and PPFOH were 508 and 460ng.hr/ml, respectively. $C_{max}$ of PPF was about 828.4ng/ml and the peak concentration was achieved at about 2.2 hr ($T_{max}$). These results indicate that PPF is rapidly disappeared from blood due to extensive metabolism into PPFOH.

Isolation and Characterization of Thermophilic Bacillus licheniformis YJ09 from Spent Mushroom (Pleurotus ostreatus) Substrates (느타리 수확후배지로부터 분리된 고온성 Bacillus licheniformis YJ09의 특성)

  • In order to isolate thermophilic bacteria with high activity of CMCase and xylanase, spent mushroom substrates was collected from an oyster mushroom cultivation farm in Jinju, Gyeongnam, Korea. Among the isolates, one strain designated as YJ09 was selected by agar diffusion method. The isolate YJ09 was identified as a member of Bacillus licheniformis based on biochemical characteristics using Bacillus ID kit and MicroLog system. Comparative 16S rDNA sequence analysis showed that isolate YJ09 formed a distinct phylogenetic tree within the genus Bacillus and was most closely related to Bacillus licheniformis with sequence similarity of 98.9%. Based on its physiological properties, biochemical characteristics and phylogenetic distinctiveness, the isolate YJ09 was classified as Bacillus licheniformis. The CMCase and xylanase activity of B. licheniformis YJ09 was slightly increased corresponding to the bacterial population from exponential phase to stationary phase in the growth curve of B. licheniformis YJ09.

An Study of Demand Forecasting Methodology Based on Hype Cycle: The Case Study on Hybrid Cars (기대주기 분석을 활용한 수요예측 연구: 하이브리드 자동차의 사례를 중심으로)

  • This paper proposes a model for demand forecasting that will require less effort in the process of utilizing the new product diffusion model while also allowing for more objective and timely application. Drawing upon the theoretical foundation provided by the hype cycle model and the consumer adoption model, this proposed model makes it possible to estimate the maximum market potential based solely on bibliometrics and the scale of the early market, thereby presenting a method for supplying the major parameters required for the Bass model. Upon analyzing the forecasting ability of this model by applying it to the case of the hybrid car market, the model was confirmed to be capable of successfully forecasting results similar in scale to the market potential deduced through various other objective sources of information, thus underscoring the potentials of utilizing this model. Moreover, even the hype cycle or the life cycle can be estimated through direct linkage with bibliometrics and the Bass model. In cases where the hype cycles of other models have been observed, the forecasting ability of this model was demonstrated through simple case studies. Since this proposed model yields a maximum market potential that can also be applied directly to other growth curve models, the model presented in the following paper provides new directions in the endeavor to forecast technology diffusion and identify promising technologies through bibliometrics.

Dudleya brittonii extract promotes survival rate and M2-like metabolic change in porcine 3D4/31 alveolar macrophages

  • Objective: Although alveolar macrophages play a key role in the respiratory immunity of livestock, studies on the mechanism of differentiation and survival of alveolar macrophages are lacking. Therefore, we undertook to investigate changes in the lipid metabolism and survival rate, using 3D4/31 macrophages and Dudleya brittonii which has been used as a traditional asthma treatment. Methods: 3D4/31 macrophages were used as the in vitro porcine alveolar macrophages model. The cells were activated by exposure to phorbol 12-myristate 13-acetate (PMA). Dudleya brittonii extraction was performed with distilled water. For evaluating the cell survival rate, we performed the water-soluble tetrazolium salt cell viability assay and growth curve analysis. To confirm cell death, cell cycle and intracellular reactive oxygen species (ROS) levels were measured using flow cytometric analysis by applying fluorescence dye dichlorofluorescein diacetate and propidium iodide. Furthermore, we also evaluated cellular lipid accumulation with oil red O staining, and fatty acid synthesis related genes expression levels using quantitative polymerase chain reaction (qPCR) with SYBR green dye. Glycolysis, fatty acid oxidation, and tricarboxylic acid (TCA) cycle related gene expression levels were measured using qPCR after exposure to Dudleya brittonii extract (DB) for 12 h. Results: The ROS production and cell death were induced by PMA treatment, and exposure to DB reduced the PMA induced downregulation of cell survival. The PMA and DB treatments upregulated the lipid accumulation, with corresponding increase in the acetyl-CoA carboxylase alpha, fatty acid synthase mRNA expressions. DB-PMA co-treatment reduced the glycolysis genes expression, but increased the expressions of fatty acid oxidation and TCA cycle genes. Conclusion: This study provides new insights and directions for further research relating to the immunity of porcine respiratory system, by employing a model based on alveolar macrophages and natural materials.

Effects of Medroxy Progesterone Acetate (MPA) Treatment on Growth of Velvet Antler in Sika Deer and Elk Deer (MPA 투여가 사슴뿔의 성장에 미치는 영향)

  • The study was conducted to determine the effects of hormone injection on casting day of antler, velvet antler yield, and blood hormone concentration in elk deer and Sika deer. The study revealed that the casting day of Elk and Sika deer at medroxy progesterone acetate(MPA) injection averaged 21 days after MPA injection, which was earlier 38 and 24 days, respectively, compared control(P<0.01). The regrowth of antler in both Sika deer and Elk occurred in the MPA injection and the duration of antler growth was 2 times longer than control. The total yield of velvet antler of Elk in the control and MPA injection was 7.31 and 10.11kg and the that of sika deer was 1.00 and 1.41kg, respectively. Blood testosterone concentration of Sika deer and Elk was less than 4.0ng/ml for both at the casting and during the antler growing. Blood IGF-1 concentrations of Sika deer and Elk during the antler growing tended to increase with the same as growth curve of antler.

Effect of Wheat Flour Brew with Bifidobacterium bifidum on Rheological Properties of Wheat Flour Dough (Bifidobacterium bifidum을 이용한 밀가루 brew가 반죽의 이화학적 성질에 미치는 영향)

  • In order to economically utilize flour brew with Bifidobacterium bifidum as a bread improver, the effect of flour brew on the rheological properties of dough, growth curve and acid production, and symbiosis with yeast were investigated. Growth of bifidobacteria was not increased more than initial seed volume but was consistent during 24 hours of incubation. pH was decreased and T.T.A was increased up to 12 hours of incubation. Symbiosis between bifidobacteria and yeast was little. Bifidobacteria produced more lactic acid than acetic acid in flour brew and the opposite in skim milk broth. This result was inferred from Lactobacillus sp. inherent in flour. On rheological properties of dough, farinograms of flour showed progressively decreasing baking absorption, mixing time and stability as the amount of flour brew increased. The validation of extensograms showed that R/E ratio linearly increased with increment of flour brew, and nearly doubled in all treatments comparing to that of control, which suggest the reduction of actual fermentation time. On visco/amylograms, malt index increased with addition of flour brew, accordingly showing the decrease in viscosity. Break down and set back value decreased with increment of flour brew, suggesting that staling rate of bread can be delayed.

Developmental Ecology and Temperature-dependent Development Model of Scotinophara lurida (Heteroptera: Pentatomidae) (먹노린재(Scotinophara lurida)의 발생생태와 온도의존 발육모형)

  • The developmental ecology and temperature-dependent growth model were calculated to develop the Scotinophara lurida control technology, which is mainly affected by environmentally friendly rice cultivation. The survival rate of S. lurida after overwintering in 2019 showed that 167 of 224 survived and the survival rate was 72.8%. Overwintering adult of S. lurida occur in rice fields in mid-June, spawn in early July, and first-generation adults develop in mid-August. In order to determine the temperature-dependent growth model, the growth periods by temperature and development stage were investigated in a incubator at 18, 21, 24, 27, 30℃ and 14L: 10D. The period from egg to adult at the temperature of 18, 21, 24, 27, 30℃ was 119.8, 73.1, 53.5, 39.4, and 82.0 days, respectively. The best development temperature was at 27℃. The regression curve was obtained by analyzing the relationship between temperature and growth rate using the Excell program, and the base temperature threshold and effective cumulative temperature for each development stage were calculated. From eggs to 5 nymphs of S. lurida the base temperature threshold was 17.9℃ and the effective cumulative temperature was 380.2 DD.

  • Demand for optical glass device used for lighting could increase rapidly because of LED lighting market growth. The optical glass devices that have been formed by hot press molding process the desired optical performance without being subjected to mechanical processing such as curve generation or grinding. EL-Max material has been used for many engineering applications because of their high wear resistance, high compressive strength, corrosion resistant and very good dimensional stability. EL-Max is very useful for a glass lens mold especially at high temperature and pressure. The performance and reliability of optical components are strongly influenced by the surface damage of EL-Max during grinding process. Therefore, the severe process condition optimization shall be necessary for the highly qualified EL-Max glass lens mold. To get the required qualified surface of EL-Max, the selection of type of the diamond wheel is also important. In this paper, we report best grinding conditions of ultra-precision grinding machining. The grinding machining results of the form accuracy and surface roughness have been analyzed by using Form Talysurf and NanoScan.

Histological Observations and Regeneration of Barbels in Juveniles of the Chinese Longsnout Catfish Leiocassis longirostris

  • Barbel structure and regenerated barbel length in the juvenile Chinese longsnout catfish Leiocassis longirostris (G$\ddot{u}$nther), were evaluated. The barbles consisted of an epidermis, a dermis, and a central rod. The epidermis harbored taste buds, granular cells and epidermal cells. The taste buds were basophilic and situated along the distal portion of the epidermis. The dermis was composed of loose connective tissue containing blood vessels pigment cells. The innermost central region was cartilage enclosed within layers of muscle layers. After 30 days, the regenerated barbel length measured $0.92{\pm}0.404mm$ at $15^{\circ}C$ (regenerated growth curve: y = 0.5085x + 4.0678, $r^2$ = 0.9654, where y is regenerated length and x is experimental period in days), $1.88{\pm}0.521mm$ at $20^{\circ}C$ (y = 0.1806x + 4.808, $r^2$ = 0.9822), and $6.44{\pm}0.751mm$ at $25^{\circ}C$ (y = 0.0914x + 4.9918, $r^2$ = 0.9944). Fifteen days after amputation, the regenerated length was significantly longer at $25^{\circ}C$ than at 15 or $20^{\circ}C$ (P < 0.05). The barbels of the Chinese longsnout catfish was the tender and flexible type, and our experimental findings provide evidence of temperature-dependent regeneration. Additional investigation of the behavior and physiology of the Chinese longsnout catfish is needed, particularly histological studies of regenerated barbels and the measurements of the numbers of taste buds per barbel under various environmental conditions.

Development of Ice Cream Prepared from Lactic Fermented Egg White Food Added with Cream (크림 첨가 난백젖산균발효식품으로 만든 아이스크림의 개발)

  • Ice cream was prepared from cream added egg white mix(EWM) food fermented with Lactobacillus acidophilus(KCTC 2182). Change of viable cells during fermentation of cream added EWM food and sensory properties and hardness of ice cream prepared from lactic fermented EWM food added with cream were investigated. (1) As the ratio of cream to EWM was higher, stationary phase of growth curve of L. acidophilus was longer and number of viable cells during fermentation was higher. (2) Sensory properties of ice cream prepared from Formula 4 (EWM 60 : cream 40) and Formula 5 (EWM 50 : cream 50) were better than those of other formulas. Optimum condition of mix formula and fermentation time was Formula 4 fermented for 15 hr. (3) As the ratio of cream to EWM was higher, the hardness value of ice cream was lower. Therefore, the hardness value of ice cream prepared from Formula 5 was the lowest.
