Dietary Effect of Puer Tea Extract on the Body Weight in Rats

  • Puer tea is a traditional beverage originating from Yunnam area of China. It supplies basic nutrients such as vitamin C. It has been well reported that daily drinking of Puer tea can help the digestion and ease the stomachache after food intake. Puer tea also contains various polyphenols which may exert antibacterial activity against Staphylococcus aureus and Listeria monocytogenes. Because of these functional effects on digestive system we suspected if Puer tea can display any dietary effect or decrease the obesity after long-term drinking. We employed 6-week old SD rats as experimental animal and treated them with extract of Puer tea in relation to the body weights. Rats were divided into 5 groups (NC, PC, E, E+P, E+P5). NC group was experimental control and rest of them are as follows: water only (PC), water with exercise (E), water with exercise and Puer tea extract (E+P), water with exercise and 5X extract of Puer tea (E+P5). Feeding was carried out every day for 5 weeks by oral administration. Reduction rate of body weights was highest in E group. Relative ratio of losing weight was as follows: PC group (100.78%), E group (95.57%), E+P group (94.53%) and E+P5 group (74.22%), respectively. Exercise was more helpful to control the body weight. The result strongly suggests that Puer tea is highly effective to control the body weight and could be used for pharmaceutical purpose to treat obesity without side effects.

Effects of Green Tea [Camellia sinensis (L.) O. Kuntze] Extract on Lipid Metabolism in F1B Golden Syrian Hamsters Fed with the Atherogenic Diet (녹차[Camellia sinensis (L.) O. Kuntze] 추출물의 투여가 동맥경화 유발식이를 급여한 F1B Golden Syrian hamster의 지질대사에 미치는 영향)

  • This study examined the effects of green tea extract supplementation (500 or 1,000 mg/kg b.w. per day) in conjunction with an atherogenic diet (10% coconut oil (w/w), 0.1% cholesterol) on plasma lipid composition, regression of pre-existing foam cells, and on the mRNA levels of hepatic HMG-CoA reductase and LDL receptor. Compared to groups fed only with the atherogenic diet, the addition of green tea extract to atherogenic diet-fed groups significantly down-regulated plasma triglyceride and total cholesterol levels, dose-dependently. Supplementation of 1,000 mg/kg b.w. of green tea extract with the atherogenic diet induced significant up-regulation of both HMG-CoA reductase and LDL receptor messenger RNA levels in liver as compared to the group receiving green tea extract supplementation at 500 mg/kg b.w. The F1B hamsters fed the atherogenic diet had greater foam cell accumulation compared to those fed a normal diet, or the atherogenic diet supplemented with green tea extract. Regression of fatty streak lesions was achieved by atherosclerosis in fat- and cholesterol-fed hamsters and this effect was associated with down-regulation of plasma cholesterol and up-regulation of hepatic LDL receptor expression.

Effects of Addition Levels of Coffee and Green Tea By-products Extract including Polyphenols on in vitro Rumen Fermentation and Methane Emission (폴리페놀을 다량 함유한 커피박 및 녹차박 추출물의 수준별 첨가가 반추위 발효 및 메탄 발생량에 미치는 영향)

  • This study was conducted to investigate the effect of addition levels of coffee and green tea by products extract including polyphenols through hot water extraction on rumen fermentation. The treatment groups consisted of coffee extract (CO), green tea extract (GR) and mixed extract (MIX), and the addition level was 10 µL, 20 µL and 30 µL of three levels. The experiment consisted of a total of 10 experimental groups including the control group, and a full factorial design was used. The effect of polyphenol addition in coffee and green tea by-products was analyzed through main and interaction effect of statistical analysis. The total polyphenol content of the extracts was 106.15, 79.10 and 185.25 ㎍ GAE/g DM for coffee by-product, green tea by-product and mixture, respectively. Total gas production was significantly lower in the treatment groups than in the control (114.00 mL/gDM) (p<0.05). Methane emission tended to decrease as the polyphenol addition level increased. Moreover, the MIX showed the lowest methane emission when 30 µL was added (p<0.05). Volatile fatty acids showed a significant difference compared to the treatment group as a control (98.06 mM) (p<0.05), but there was no change according to the level of polyphenols. As a result of the main effect and interaction, it is thought that the effect on methane reduction and improvement of rumen fermentation in MIX20 can be expected. In a series of studies, the addition of 20 µL of a blended extract of coffee and green tea by-products is thought to reduce methane to levels that do not inhibit rumen fermentation.

Bioavailability and Efficiency of Ten Catechins as an Antioxidant

  • Tea is a pleasant, popular and safe beverage in the world. During the past decade, epidemiological studies have shown that tea catechins intake is associated with lower risk of cardiovascular disease. Tea provides a dietary source of health-promoting components to help humans reduce a wide variety of cancer risks and chronic diseases. The antioxidative activity of tea-derived catchins has been extensively studied. The antioxidant effect is a synergistic action between catechins e.g. EGCG, EGC, ECG, EC, pheophytins a and b, and other components in tea leaves, which aye more bioavailable for human body. Green tea has a Higher content of catechins than other kinds of tea. Green tea extract with hot water has high potential and more efficiency to reduce cancer risk than any other tea products or pure EGCG. Protein, iyon, and other food components may interfere with the bioavailability of ten catechins. Interaction of catechins with drug affects the cancer-preventive activity of some cancer-fighting medication. Further studies are required to determine the bioavailability of tea catechins and cancer-preventive functionality.

Management of Earthworm Casts using Pesticides and Tea Tree (Camellia oliefera) Extracts in Turfgrass of Golf Courses (골프장에서 차나무류 추출물과 화학 방제제를 이용한 지렁이 분변토 발생 관리)

  • Earthworm cast is a harmful factor which reduces the uniformity of management of turfgrass and play quality in golf courses and playground. This research was conducted to carry out the efficacy of some pesticide (bifenthrin GR + clothianidin GR, ethoprophos GR, imidacloprid SC, picoxystrobin WG, thiophanate-methyl WP) and tee tree (Camellia oliefera) extracts against reducing earthworm cast in golf courses. All tested materials were acted reducing earthworm cast, thiophanate-methyl among them showed the highest effect reducing earthworm casts in golf courses. Tea tree extracts was occurred similar efficacy among tested chemical pesticides. However, pellet type byproduct of tea seed extract and liquid formulation of tea tree extract were better than granule formulation of them. Byproduct of tea seed and tea tree extract can be used for alternative biological management material for reducing earthworm cast in heavily casting occurrence area.

Effect of Clerodendron trichotomum Thunberg tea on anti-hypertension (누리장나무차가 항고혈압에 미치는 영향)

  • Objectives : Clerodendron trichotomumThunberg grows wild fields and mountains in South Korea and China. They have been used for anti-hypertension disease. This study was performed in order to observe the effects ofClerodendron trichotomumThunberg tea on anti-hypertension.Methods : For the study ofClerodendron trichotomumThunberg tea, we had divided male SHR (Spontaneously Hypertensive Rat) to ten groups. Control group is SHR. The treatment group is SHR which is administered tea extract at dose of 200 mg/kg/day and 500 mg/kg/day orally for 8 weeks. We measured systolic blood pressure, diastolic blood pressure, number of pulse, body weight, total cholesterol and triglyceride from blood serum.Results : The level of systolic and diastolic blood pressure were significantly decreased by 12.5% and 44.9% inClerodendron trichotomumThunberg tea groups than control group. Heart rate ofClerodendron trichotomumThunberg tea groups, 24.5%, was significantly lower than control group. We found that treatment with the water extract fromClerodendron trichotomumThunberg leave and flower significantly decreased weight of body, total cholesterol and triglyceride to 20.4% and 27.7% compared to untreated control group, significantly (P< 0.05).Conclusions : These results suggest that leaf and flower tea ofClerodendron trichotomumThunberg tea is effective in prevention and treatment of hypertension and decreasing cholesterol. Long term consumption ofClerodendron trichotomumThunberg leaf and flower tea can be help in lowering high systolic and blood diastolic pressure, heart rate and blood serum in SHR.

Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats (추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. Nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice (갈근 및 녹차추출물의 아질산염 소거작용 및 간독성 보호효과)

  • This study was designed to evaluated the nitrite scavenging activity and protective effect of the Puerariae Radix and green tea extract on lead acetate and cadmium-induced liver damage in mice. The quantitative analytical method for major antioxidants, isoflavones, puerarin, catechine and caffeine in galgun (Puerariae Radix) and green tera extract were established by HPLC. Contents of isoflavones, such as daidzin, genistin, daidzein and genistein were 4.23g/100g, 0.13g/100g, 0.07g/100g, and 0.03g/100g, and puerarin contents was 8.99g/100g, respectively. The total catechins and caffeine contents of green tea extract were 49.24g/100g and 6.53g/100g. The nitrite scavenging ability of galgun extract (pH 1.2, 100mg/ml) was 98.07% and it was higher than those of other extracts. It was the highest at the pH 1.2 and more than 64% in 25~100mg green tea extract, and was dependents on pH and concentration of the samples. The hepatoprotective effects of an aqueous extract from the root of gal gun and standard puerarin were evaluated against lead acetate and cadmium-induced liver damage in mice. Galgun extract and standard at a dose of 100mg/kg and 10mg/kg, 50mg/kg were administered orally once daily for successive 5 days and then a lead acetate and cadmium were orally at 3 hrs after the every day administration of galgun. The substantially elevated serum enzymatic activities of alanine and aspartate aminotransferase were due to lead acetate and cadmium treatment was dose dependently restored to the near normal level. In addition, galgun extract also significantly prevented the elevation of hepatic malon-dialdehyde formation in the liver of lead acetate and cadmium intoxicated mice in a dose-dependent manner. The results of this study clearly indicated that green tea and galgun extracts had nitrite scavenging ability and galgun extract had potent hepato-protective effects against lead acetate and cadmium-induced hepatic damage in mice and standard puerarin was also showed similar to the results of the galgun extracts.

Evaluation of Toxicity of Green Tea Extract in Chilled Boar Spermatozoa

  • The cold shock of spermatozoa is associated with oxidative stress induced by reactive oxygen species. This study was conducted to evaluate the toxicity of natural antioxidant green tea extract (GTE) in lactose-egg yolk (LEY) extender during boar sperm cooling prior to freezing. Spermatozoa were cooled to $5^{\circ}C$ for 3 h in LEY extender containing 0 (control), 1, 10, 100 or 1,000 mg/l of GTE, re-suspended with LEY-glycerol-Equex extender and cooled at $5^{\circ}C$ for 30 min. Sperm progressive motility, viability and phosphatidylserine (PS) translocation were evaluated. PS translocation was assayed by flow cytometry using Annexin V-FITC apoptosis detection kit. The sperm function including progressive motility, viability and PS translocation was not significantly different regardless of GTE concentrations (P>0.05). In conclusion, this study demonstrated non-toxicity of GTE supplement in LEY extender during sperm cooling.

Antimutagenic Effects of Persimmon Leaf Tea Extract (PLTE) in Mice Using Micronucleus Induction (MN) Test (마우스 소핵(Micronuclei, MN) 시험방법을 이용한 감잎차 추출물의 돌연변이 억제효과)

  • The antimutagenic effects of persimmon leaf tea extract (PLTE) at concentration levels consumed by human were examined in mice using micronuleus induction with MMC(mitomycin C) or 4-NQO (4-nitroquinoline-1-oxide). When mice received oral gavage of 10 equivalent to PLTE 24 hr and 6 hr before, and 5 equivalent to PLTE 6 hr before and 3 hr after intraperitoneal injection of MMC, a significant decrease in the frequency of micronuclei were observed. The induction of micronuclei by 4-NQO was suppressed by oral dosage of PLTE at 5 equivalent to PLTE 6 hr before and 3 hr after, 10 equivalent to PLTE 3 hr before and 3 hr after intraperitoneal injection of MMC. Though the components of PLTE have not been analyzed so far, our present results suggest the existence of several bio-antimutagens and/or desmutagens in PLTE, beside catechin, well-known antimutagen.
