Taxonomic study of Korean Scirpus L. s.l. (Cyperaceae) I. Morphology of Bolboschoenus (Asch.) Palla, Schoenoplectus (Rchb.) Palla, Schoenoplectiella Lye, Scirpus L., and Trichophorum Pers. (한국산 광의의 고랭이속(사초과)의 분류학적 연구 I. 매자기속, 큰고랭이속, 올챙이골속, 고랭이속, 애기황새풀속의 형태적 특성)

  • Scirpus L. s.l. of Cyperaceae has been regarded as a monophyletic group, but the taxa of Scirpus s.l. were rearranged into the five independent genera in recent molecular phylogenetic research: Bolboschoenus (Asch.) Palla, Schoenoplectus (Rchb.) Palla, Schoenoplectiella Lye, Scirpus L., and Trichophorum Pers. In this study, we examined the morphological characteristics and presented the delineation of the five genera of Scirpus s.l., including 21 taxa from South Korea. As a result of our investigation and analysis of domestic collections and those from abroad, the each taxon could be identified using key characteristics of the rhizome features; a cross-section of culm; the degree of development and the width of leaves; the shapes of the membranous region of leaf sheath; and the shapes and size of inflorescence, bract, floral scale, and achene. These major characteristics are illustrated and artificial keys are suggested in this study. In addition, we suggest a new combination for an unrecorded taxon of Schoenoplectiella that was discovered in Gangwon-do, Goseong-gun and give it the Korean name, as 'a-do-song-i-go-raeng-i'.

Morphological Development of Eggs, Larvae and Juveniles of the Sterlet Sturgeon, Acipenser ruthenus (Pisces: Acipenseridae) (스텔렛 철갑상어 (Acipenser ruthenus)의 난 발생과 자치어 형태발달)

  • To develop cultivation technique for Acipenser ruthenus and secure basic taxonomic materials, this study examined egg development and larva morphological development. This study used a couple of male and female broodstork matured in a PVC circle water tank (Ø 5 m) with the water temperature of $16.0{\pm}1.0^{\circ}C$ in June 2005. They were oval opaque and sinking cohesive eggs and the sizes of them were between $3.83{\sim}3.85(3.84{\pm}0.01)mm$ (n=10). As for the egg development, the dry process was adopted with artificial insemination in this study, and an hour after the insemination, the embryo disks were developed, 31 hours after the insemination, they reached the end of gastrula stage, and 82 hours (50%) after the insemination, they were hatched. Right after the hatch, the total lengths of larvae were $10.1{\sim}10.3(10.2{\pm}0.11)mm$ (n=10) with big yolks in the venter. 17 days after the hatch, the total lengths of them were $26.2{\sim}32.4(29.3{\pm}4.39)mm$ (n=10) and in the front of a dorsal fin there were 11 scute scales that started to make tracing development. As 50 days after the hatch, the total lengths were $86.4{\sim}93.1(89.8{\pm}4.71)mm$ (n=10), and fin rays all reached integer, they were moved to the larva apparatus.

Phylogenetic and Morphological Comparison between Thamnaconus septentrionalis and T. modestus Collected in Southwest Seashore (서남해에서 채집된 말쥐치 (Thamnaconus modestus)와 유사종 (T. septentrionalis)의 형태 및 계통유전학적 비교)

  • Thamnaconus modestus, distributed in the Northwest Pacific, has high economic value and is used in various seafood. In this study, the morphological and genetic characteristics of T. modestus and T. septentrionalis were compared and analyzed. We observed the external and internal morphology of T. modestus, sketched skeletal elements, and analyzed phylogenetic evolutionary relationships using the cytochrome c oxidase subunit I (COI) gene on mitochondrial DNA compared to T. septentrionalis. The T. modestus observed in this study had blackish-brown patterns irregularly scattered on the gray-brown body, and the fins were blue-green. Genetic analysis results based on the COI sequences of T. modestus showed seven types of base sequence variation; however, the homology was more than 98.8%. In addition, as a result of comparison of the COI nucleotide sequences and phylogenetic analysis in Tetraodontiformes, two T. septentrionalis sequences (JN813099, MW485059) were similar to T. modestus with 99% homology, and the other two T. septentrionalis sequences (EF607583, KP267619) were similar to those of species belonging to another genus Thamnaconus with 95% homology with T. modestus. It was not easy to classify the species based on morphological characteristics, and phylogenetic analysis between T. modestus and T. septentrionalis confirmed the difference in classification. These results provide the external and internal morphology of T. modestus and will be used as important information for the taxonomic study of T. modestus and T. septentrionalis.

Re-examination of the vascular plants on Hongdo Island, Korea (홍도 관속식물상 재검토)

  • This study was carried out to survey the flora of Hongdo Island in Sinan-gun, Jeollanam-do, South Korea. Specimens collected from previous Hongdo flora studies were reexamined using a relevant biodiversity database, and field surveys were carried out 22 times from April of 2003 to October of 2020. Based on the specimens collected during both previous studies and this study, the identified vascular plants of Hongdo consisted of 472 taxa comprising 102 families, 296 genera, 425 species, 6 subspecies, and 41 varieties. Among them, 111 taxa are newly recorded in this study, and 6 taxa are described in detail in terms of their morphological characteristics and habitat. Also, 29 taxa were reviewed or re-identified with corresponding taxonomic annotations. In Korea, Hongdo represents the northern distributional limit of the 4 taxa of Goodyera biflora, Damnacanthus major, Calanthe aristulifera, and Hemerocallis hongdoensis. Moreover, Hosta yingeri and Saussurea polylepis are endemic to Hongdo and nearby islands in Korea. Distribution maps of these species were prepared. Protected species designated by the Ministry of Environment were 7 taxa consisting of 2 taxa of level I, specifically Sedirea japonica and Neofinetia falcata, and 5 taxa of level II, which were Cymbidium macrorhizon, Woodwardia japonica, Dendrobium moniliforme, Calanthe aristulifera, and Bulbophyllum inconspicuum. Red list plants as designated by the National Institute of Biological Resources numbered 11 taxa. Naturalized plants numbered 40 taxa.

A Study on Microorganism Dominant Species in Bench-scale Shipboard STP Using Combined SBR and MBR Process (SBR 및 MBR 복합공정을 적용한 Bench-scale Shipboard STP에서의 미생물 우점종에 관한 연구)

  • International Maritime Organization (IMO) is one of the most effective organizations in evolving international law for the protection and conservation of the marine environment. The IMO, MARPOL(Marine Pollution) 73/78 contains six Annexes that provide an overarching framework for the objectives of the international marine pollution. Annex IV was regulated by 64 th resolution in 2012 to control sea pollution from sewage. In 2014 large-scale wastewater treatment and nutrient removal device was developed with a grant from the Ministry of Oceans and Fisheries. A combined new process of Sequence Batch Reactor (SBR) and Membrane Bioreactor(MBR) was developed to overcome the pollution caused by shipboard sewage. In the present study, shipboard sewage wastewater was treated by mixing and aeration cycle in the newly developed SBR process. Furthermore, during analysis by NGS technique(Macrogen Co., Ltd.), dominant species of bacteria were found in the aeration tank of the Bench-scale wastewater treatment facility. Bacteroidetes and Gammaproteobacteria accounted for 27.1 % of the aerobicbacteria and 16.8 % of the anaerobicbacteria, respectively. Microorganisms play a vital role in shipboard wastewater treatment. A further detailed study is required to understand the precise role of the microorganisms in the wastewater treatment.

Analysis of gut bacterial diversity and exploration of cellulose-degrading bacteria in xylophagous insects (목질섭식곤충의 장내 세균 다양성 분석 및 섬유소 분해균 탐색)

  • In this study, gut bacterial communities in xylophagous insects were analyzed using the pyrosequencing of 16S rRNA genes for their potential biotechnological applications in lignocelluloses degradation. The result showed that operational taxonomic units (OTUs), species richness and diversity index were higher in the hindgut than in the midgut of all insect samples analyzed. The dominant phyla or classes were Firmicutes (54.0%), Bacteroidetes (14.5%), ${\gamma}-Proteobacteria$ (12.3%) in all xylophagous insects except for Rhinotermitidae. The principal coordinates analysis (PCoA) showed that the bacterial community structure mostly clustered according to phylogeny of hosts rather than their habitats. In our study, the two carboxymethyl cellulose (CMC)-degrading isolates which showed the highest enzyme activity were most closely related to Bacillus toyonensis $BCT-7112^T$ and Lactococcus lactis subsp. hordniae $NCDO\;2181^T$, respectively. Cellulolytic enzyme activity analysis showed that ${\beta}-1,4-glucosidase$, ${\beta}-1,4-endoglucanase$ and ${\beta}-1,4-xylanase$ were higher in the hindgut of Cerambycidae. The results demonstrate that xylophagous insect guts harbor diverse gut bacteria, including valuable cellulolytic bacteria, which could be used for various biotechnological applications.

Change in Taxonomic Composition of Phytoplankton and Environmental Factors after Construction of Dike in Yeongsan River Estuary (하구언 건설 전.후의 영산강 하구 식물플랑크톤 군집 및 환경 변화)

  • The Yeongsan River estuary, located in the south west coast, was a typical estuary before a dike was constructed in December 1981. After the construction, the water quality and plankton communities are expected to change. We investigated the change of phytoplankton community and environmental properties in the Yeongsan River estuary by comparing the data collected recently with the results reported before and/or shortly after the construction. Concentrations of $NH_4$-N were significantly increased in the freshwater zone. Concentrations of DO at the surface, $NO_2+NO_3$ and $NH_4$-N were increased while the concentrations of $PO_4$ at the surface were decreased in the seawater zone. The number of green algae species was decreased in the freshwater zone. The number of diatom species was decreased while the numbers of green algae and dinoflagellates were increased in the seawater zone. This study can provide information for better management of the Yeongsan River estuary since no comparison study has been documented between the phytoplankton community before and after the construction of the dike in the estuary.

Enhancement of the germination efficiency of Gastrodia elata seeds using a new Mycena species (신규 애주름버섯 NIFOS101을 이용한 천마 종자의 발아효율 증진)

  • This study investigated the in vitro symbiotic seed germination of the achlorophyllous orchid, Gastrodia elata, using a new Mycena species. A leaf-disc ($2{\times}2cm$) of Quercus accutissima was inoculated with either of the two fungal species, NIFOS101 (NCBI accession number KY449288) or KFRI1212 (HQ662845), following which it was placed on water agar medium, prior to sowing seeds of G. elata. After 20 days of symbiotic culture, NIFOS101 and KFRI1212 germinated 94% and 70% of G. elata seeds, respectively, although the initiation of protocorm development was induced much earlier by KFRI 1212 than by NIFOS101. Furthermore, the NIFOS101 mycelia grew much faster than KFRI 1212 at all temperatures tested. A phylogenetic analysis using the internal transcribed spacer (ITS) sequences showed that NIFOS101 belonged to a clade with M. purpureofusca, which completely differed from the clade with KFRI1212. This study not only identified a new fungal species, NIFOS101, which improved the rate of symbiotic seed germination up to 94% as compared to KFRI1212 (70%), but also revealed that G. elata required a broad taxonomic range of fungi for its symbiotic germination.

Phylogenetic Analysis of Ji-Mo (Anemarrhena asphodeloides) on the Basis of Chloroplast DNA Sequences (엽록체 DNA 염기서열을 이용한 한약재 지모의 기원 확인 및 유연관계 분석)

  • Anemarrhena asphodeloides (Korean name "Ji-Mo") has been used for oriental medicinal purposes in Korea, China and Japan. In this study, 29 A. asphodeloides samples were collected including 3 certified A. asphodeloides plants and many commercially marketed A. asphodeloides products. Chloroplast trnL-F regions of the "Ji-Mo" samples were sequenced and used to identify whether the samples were genuine A. asphodeloides or not. As the result, the trnL-F sequences of all the "Ji-Mo" samples were shown to be identical and it was proven that commercially available medicinal products "Ji-Mo" are genuine A. asphodeloides. Phylogenetic tree of. A. asphodeloides using the trnL-F sequences was constructed and compared with phylogenetic tree using rubisco large subunit (rbcL) gene sequences. In these tree, A. asphodeloides was affiliated in the family Agavaceae in the order Asparagales. It is proven that trnL-F phylogenetic tree is useful to study taxonomic position of A. asphodeloides.

Biomass and Species Composition of Phytoplankton and Zooplankton along the Salinity Gradients in the Seomjin River Estuary (섬진강 하구역에서 염분경사에 따른 동${\cdot}$식물플랑크톤의 현존량 및 종조성)

  • Biomass and species composition of phytoplankton and zooplankton along the salinity gradients in the Seomjin River estuary were investigated in March, July, September and November, 1999. The locations of sampling sites were set based on the surface salinity during each cruise rather than geographic locations. A total of 96 phytoplankton species were identified with 60 diatom species in the study area. The species number of green algae was relatively high in low salinity region while the number of dinoflagellates increased in high salinity areas. Monthly mean of phytoplankton biomass ranged from 183 cells/ml to 833 cells/ml. There was a diatom bloom of Skeletonema costatum in November at the 5-15 psu salinity region and sharp decrease of biomass in very low salinity region occurred in March. During the rainy season in July with the salinity less than 8.1 psu, the green algae dominated in most of the study areas. Chroomonas spp. were highly dominant in March and September and the biomass increased towards the high salinity regions. A total of 83 zooplankton taxa were identified with 72 marine and 11 freshwater taxa. Major group of marine form was copepods with34 taxonomic groups. Most freshwater taxa were aquatic insects. Threshold salinities of freshwater zooplankton appearance were 8.1 psu in July, 4.7 psu in September, and 0.2 psu in November. The number of taxa appeared and abundances of zooplanktons were minimal in the middle of salinity gradients in this estuary.
