Streptococcal infection is one of the most serious disease of cultured olive flounder, Paralychthys olivaceus in Korea and caused by more than one species. However, there has been considerable confusions about the taxonomic position of the fish pathogenic streptococci. In this study, We performed the randomly amplified polymorphic DNA(RAPD) pattern analysis to evaluate the possible classification in 8 streptococci isolated from diseased olive flounder and reference strains based on their DNA structure. RAPD PCR with DNA solution prepared by simple boiling and 10-mer random primer was appeared to be a good tool for discrimination of different streptococcal strains. Phenotypical characters by simple biological test and API 20 Strep corresponded well to the specific profiles of RAPD in streptococcal isolates of this study. Therefore, the RAPD profile was considered as one of differential characters to discriminate the streptococcal isolates from diseased olive flounder.
Taxonomic review of Cobitis laterimaculata was carried out based on 17 specimens collected from the Caozejiang River, Huangzezhen, Shengxian county, Zhejiang province, China. The species shares several notable characters with Niwaella spp., including the posterior position of the dorsal fin, small head, color pattern without pigmentary zones of Gambetta, short and curved suborbital spine, short and subequal barbels, strong carina on caudal peduncle, and lack of a lamina circularis on the pectoral fin base in males. We conclude that Cobitis laterimaculata should be transferred from Cobitis to Niwaella.

Two different molecular markers, the plastid rbcL and mitochondrial cox1 genes, were used to define the taxonomic position of the northwest Pacific Ocean species currently named Gracilaria chorda. We analyzed both genes (1,222 bp for rbcL and 1,245 bp for cox1) from 18 specimens collected in Korea, Japan, and China. Phylogenetic reconstruction revealed that this organism should be classified in the genus Gracilariopsis, rather than in the Gracilaria. Thus, Gracilariopsis chorda (Holmes) Ohmi is the legitimate name for Gracilaria chorda Holmes. Within the species, the sequences differed by 8 bp (0.7%) in rbcL and 5 bp (0.4%) in cox1. Six haplotypes of cox1 tended to be geographically organized. Gp. chorda is characterized by coarse, elongate terete axes, short filiform branchlets usually at irregular intervals, an abrupt transition in cell size from medulla to cortex, cystocarps without tubular nutritive cells connecting the gonimoblast to the upper pericarp, and relatively large gonimoblast cells of the cystocarp in the specimens collected from Wando in southern Korea.
Ganoderma lucidum has a long history of use as a traditional medicine in Asian countries. However, the taxonomy of Ganoderma species remains controversial, since they were initially classified on the basis of their morphological characteristics. Recently, it was proposed that G. lucidum from China be renamed as G. sichuanense or G. lingzhi. In the present study, phylogenetic analysis using the internal transcribed spacer region rDNA sequences of the Ganoderma species indicated that all strains of the Korean 'G. lucidum' clustered into one group together with G. sichuanense and G. lingzhi from China. However, strains from Europe and North American, which were regarded as true G. lucidum, were positioned in a clearly different group. In addition, the average size of the basidiospores from the Korean cultivated Yeongji strains was similar to that of G. lingzhi. Based on these results, we propose that the Korean cultivated Yeongji strains of 'G. lucidum' should be renamed as G. lingzhi.
This paper is a report of studies to determine the amino acid composition of Plant Pathogenic Corynebacteria and to assess whether these characteristics can be correlated with the taxonomic position of these organisms. The results indicated that plant pathogenic Corynebacteria contained on average less cystein, trypthophane, histidine, phenylalanine, isoleucin and total protein than did the other genera of bacteria. However, in general, the quantities of both cell protein and amino acids contained in bacterial cells were characteristics of the species or an individual strain of the organism and were not related to its classification.
Two species of Moroco oxycephalus and M. lagowskii were studied to examine patterns of genic variation and morphological difference within and between populations of two species, and to clarify their taxonomic status and geographic distribution. The number of scales above lateral line(SAL) was the key character to classify these species. On the average, M.lagowskii had 22 or more SAL whereas M.oxycephalus had 20 or less. Previously known character of the position of dorsal fin was found to be not appropriate to distinguish them. Five loci, Gp, st-1, Est-2, Est-3 and Got-1 , showed fixed difference electrophoretically between two species and these could be used as genetic markers to identify them. The degree of genic variation of M. oxycephalus was four fold higher(H=0.032) than that of M. lagowskii(H=0.008) but both species were far less than the average genic variation of freshwater fish in general. Rogers' genetic similarity coefficients between two species were S=0.692 and their presumed divergent time was estimated to be sbout 1.8million years ageo. Detailed survey of the geographic distribution of thses revealed that M.lagowskii was distributed in northeastern part of South Korea(Ganseong , Gangreung, Wangsan, and Oggye) and M. oxycephalus was occupied rest of the peninsula. The distrance between Oggye, the southern limit of M. lagowskii distribution , and Samwha (near Samcheog), the northern limit of M.oxycephalus, was aobut 15 Km apart and no symparty was found in between.
This study was conducted to clarify the phylogenetic position and relationships of Korean Poaceae taxa. A total of 438 taxa including 155 accessions of Korean Poaceae (representing 92% and 72% of Korean Poaceous genera and species, respectively) were employed for phylogeny reconstruction. Sequence data of eight chloroplast DNA markers were used for molecular phylogenetic analyses. The resulted phylogeny was mostly concordant with previous phylogenetic hypotheses, especially in terms of subfamilial and tribal relationships. Several taxa-specific indels were detected in the molecular phylogeny, including a 45 bp deletion in rps3 (PACMAD [Panicoideae, Arundinoideae, Chloridoideae, Micrairoideae, Aristidoideae, Danthonioideae] clade), a 15 bp deletion in ndhF (Oryzeae + Phyllorachideae), a 6 bp deletion in trnLF (Poeae s.l.), and two (17 bp and 378 bp) deletions in atpF-H (Pooideae). The Korean Poaceae members were classified into 23 tribes, representing eight subfamilies. The subfamilial and tribal classifications of the Korean taxa were generally congruent with a recently published system, whereas some subtribes and genera were found to be non-monophyletic. The taxa included in the PACMAD clade (especially Andropogoneae) showed very weak and uncertain phylogenetic relationships, presumably to be due to evolutionary radiation and polyploidization. The reconstructed phylogeny can be utilized to update the taxonomic positions of the newly examined grass accessions.
In order to solve the taxonomic problems of the genus Juniperus growing in South Korea, an identification key of the genus and species was developed bayed un flower structure, cane and seed shape, branching habit, tree form, leaf characteristics etc. of the 7 native species and the a exotic cultivars. The typical pattern of karyotype found by chromosome analysis of the species was used for the identification among morphologically similar species. The length of chromosome were ranged $9{\sim}15{\mu}m$ in all studied specie. J. chinensis, var. procumbens, and var. kaizuka sere tetraploid, 4n=44, var. globosa, var. procumbens, var. horizontalis, J. virginiada, J. rigida, J. rigida var. longicarpa, and J. coreana were diploid, 2n=22. The species in the Sabina section showed large variation in the length of chromosome and kinetochore position. The species in the Oxycedrus section showed the cytological characteristics that the 11th chromosome t-type(acrocentric), and the m-type abundant chromosome set was relatively uniform as compared to those of the Sabina section. The species in the Sabina section, which are planted in the large city area, show great morphological variation because many different ecotypes were mixed and often crossed among them. In summary, this study was able to make clear identification and to find out similarity among Juniperus, species by the morphological and cytological analysis.
The internal transcribed spacer (ITS) regions of nuclear ribosomal DNA from genus Chrysosplenium were sequenced to address phylogenetic relationship. ITS including 5.8S sequence varied in length from 647 bp to 653 bp. Among them, 219 sites were variable sites with parsimony-informative. The aligned sequences were analyzed by maximum parsimony (MP) and neighbor-joining (NJ) methods. In the strict consensus trees of parsimony analysis, the monophyly of Chrysosplenium was supported by 100% bootstrap value. The first clade, C. pseudofauriei was at the basal position of the genus, and others formed two clades with high bootstrap support. The second clade included Ser. Pilosa and Ser. Oppositifolia and third clade included Ser. Alternifolia and Ser. Flagellifera. The NJ trees showed essentially the same topology. Finally, DNA sequences of ITS regions were useful phylogenetic marker in this genus. Based on the ITS and ridge seed morphological results, C. sphaerospermum Maxim. and C. valdepilosum (Ohwi) S.H. Kang & J.W. Han were discussed their scientific names and taxonomic positions.
