활성탄에 의한 Tartrazine의 흡착 특성 (Adsorption Charateristics of Tartrazine by Activated Carbon)

  • The adsorption characteristics of tartrazine by granular activated carbon were experimentaly investigated in the batch adsorber and the packed column. The adsorptivity of activated carbon for tartrazine was largely improved by pH control, and 98 percent of initial concentration could be removed at pH 3. It was estabilished that the adsorption equilibrium of tartrazine on granular activated carbon was sucessfully fitted by Freundlich isotherm equation in the concentration range from 50 mg/L to 1,000 mg/L. The characteristics of breakthrough curve of activated carbon packed column, which depend on the design variables such as initial concentration, bed height, and flow rate, were studied.

식용 Azo 색소가 Hepatic Azo Reductase에 미치는 영향과 Flavin의 효과 (Influence of Food Dye on the Activity of Hepatic Azo Reductase and the Effect of Flavin)

  • 식품의 상품화에 따라 식품 첨가물의 사용이 증가되고 있다. 식품 첨가물로서 색소는 고유의 색소를 보존하기 어려운 식품에 따라 사용되며 tar 색소, 식물성 색소 및 광물성 색소로 나눌 수 있는데 tar 색소는 독성이 강한 것이 많으므로 현재 15종만이 법적으로 허용되어 있다. 이중 azo 화합물인 amaranth, tartrazine, sunset yellow, allura red에 대하여 흰 주위 hepatic azo reductase의 활성을 측정 비교하였고 이에 대한 flavin의 효과를 보았다. 1. Amaranth를 기질로 사용하여 kinetic constants $Km=645\;\mu\textrm{M}$, Vmax=50 n mol/min/mg protein의 값을 얻었다. 2. Amaranth의 농도를 일정하게 하고 FAD의 농도를 증가시켰을 때 hepatic azo reductase의 활성은 현저히 증가하였으나 NADPH-generating system에 의한 환원은 작은 증가를 나타냈다. 3. 기타 식용 azo 색소 tartrazine, sunset yellow, allura red를 기지로 azo reductase activity를 측정한 결과 sunset yellow는 amaranth와 비슷한 환원활성은 나타냈고, tartrazine은 보다 낮은 환원활성을 allura red는 보다 높은 환원활성을 나타냈으며, $300\;\mu\textrm{M}-FAD$에 의해 기질 모두에서 환원활성이 증가함을 알 수 있었다.

  • 암모니아수와 zinc acetate로부터 액상 수열합성법에 의한 ZnO의 제조에 있어 반응온도, 반응물의 농도와 혼합방법, 용액의 pH 등 반응조건에 따른 ZnO 입자의 형상과 입자분포 등 제조특성을 고찰하고, UV 조사하에 tartrazine의 광분해를 측정하여 합성된 ZnO의 광촉매로서의 성능을 확인하였다. 반응용액의 pH가 높을수록 ZnO 입자의 평균 크기는 증가하였고, zinc acetate의 농도가 증가할수록 그리고 반응온도가 증가할수록 입자의 크기는 감소하였다. 반응용액의 혼합시 암모니아수 주입 후에 zinc acetate를 첨가하였을 경우 더 작은 입자를 얻을 수 있었다. 최소 크기의 ZnO 입자의 생성을 위한 최적 조건은 용액의 pH 11.2, zinc acetate의 농도 0.6 M, 반응온도 $90^{\circ}C$였으며, 입자 평균크기는 3.133 ${\mu}$m이었다. 합성온도 $80^{\circ}C$, zinc acetate 농도 1.0M 및 반응용액의 pH 11.2의 조건에서 합성된 ZnO에 의한 tartrazine의 광촉매 분해는 분해시간 60분에서 약 97%의 분해율을 보였다.

The Binding of Food Dyes with Human Serum Albumin

  • The binding interactions between human serum albumin (HSA) and the edible food dyes amaranth, tartrazine and sunset yellow have been studied. Intrinsic association constants and the free energy changes associated with dye-protein binding at physiological pH for amaranth and tartrazine, and at two different pH values for sunset yellow have been calculated from ultrafiltration data. The temperature dependence $(20-40^{\circ}C)$ of the intrinsic association constants at pH 7.4 for amaranth-HSA and tartrazine-HSA mixtures have been measured, from which a plot of the van't Hoff isochore exhibits a marked change in slope around $30^{\circ}C$ indicating a possible change in protein conformation. The number of dye binding sites on HSA is reported for all the above conditions. HSA-ligand binding enthalpies have been used in conjunction with the N-B transitional binding enthalpy for HSA, to calculate the enthalpy for the N-B transition when ligands are bound with the protein.

Simultaneous Detection of Tartrazine-Sunset Yellow in Food Samples Using Bioxide/Carbon Paste Microcomposite with Lanthanum and Titanium

  • This report describes the simultaneous detection of the two dyes most commonly used in food, tartrazine (TZ) and sunset yellow (SY), based on a microcomposite of carbon paste decorated with La2O3 and TiO2. Anodic currents for SY-TZ were observed at 0.89-1.21 V by cyclic voltammetry (CV) separated with a ΔV of 0.32 V. The increased anodic peak currents compared to that of the unmodified carbon paste electrode were almost 50 and 41% for SY-TZ, respectively. The detection limits with the optimal amount of La2O3-TiO2 were 0.02 and 0.03 µmol/L, respectively. The relative standard deviation (RSD) based on fifty measurements was less than 3%. The versatility and novelty of the sensor were tested with food samples containing these substances and other substances.

  • 식용색소(食用色素) Tartrazine을 토끼의 경우 $7.5{\sim}50mg/kg$, 몰모트의 경우 $1,000{\sim}2,000mg/$수(首)를 경구(經口) 또는 복막주사(腹膜注射), 정맥주사(靜脈注射)로 투여(投與)하여 그 동물(動物)에 있어서의 색소(色素)의 독성(毒性)을 관찰조사(觀察調査)하였다. 혈청(血淸)은 화학분석(化學分析)하였고 각各 장기(臟器)는 현미경적(的) 관찰(觀察)을 통(通)하여 조사(調査)하였다. 그 결과(結果) 사용(使用)한 양(量)의 색소(色素)로서 대부분(大部分)의 장기(臟器)가 충혈(充血) 또는 출혈(出血)되였고 특(特)히 간장기능장해(肝臟機能障害)를 인정(認定)할 수 있었다.

산화아연 현탁액에 의한 타르색소의 광분해 (Photodecomposition of Tar Colorant With Zinc Oxide Suspension)

  • The characteristics of photocatalytic degradation of tar colorants such as brilliant blue FCF(BBF) and tartrazine(TTZ) with zinc oxide suspension was studied in a batch reactor under irradiation of ultra-violet ray. Photocatalytic degradation of TTZ with ZnO was more higher than that of BBF, and was Increased with dosage of ZnO below 5g, but was nearly affected with initial pH of two tar colorants aqueous solution. Ammonium persulfate was more effective oxidant than potassium bromate which slightly increased the degradation of BBF, but not increased the degradation of TTZ. The photocatalytic degradation rates of BBF and TTZ were pseudo-first order with rate constants of 0.0066, 0.0092 and $0.015min^{-1}$ for BBF, 0.042, 0.017 and $0.110min^{-1}$ for TTZ at the dosage of 1, 2 and 5g ZnO, respectively.

  • The binding of bovine serum albumin (BSA)-edible dyes was studied by spectrophotometric method. The edible dyes used in this study were amaranth, erythrosine, tartrazine and sunset yellow. The binding free energies and binding sites were determined at pH 7.4. The ranges of edible dye concentration were from 0.3 to $7{\times}10^{-5}$M, and those of BSA were from 0.15 to $3{\times}10^{-5}$M. The binding free energies of BSA-edible dyes were from -6, 300 to -8, 100 cal/mole.

실리카졸의 표면 전하 밀도와 염료 흡착과의 상관성 (Relationship between the Adsorption of Dye and the Surface Charge Density of Silica Sol)

  • In this study, we investigated the relationship between the adsorption property of Methyl violet or Tartrazine dye onto silica sol surface and surface charge density of the sol. The adsorption ratio of Methyl violet dye on silica sol decreased to 74% and 92% for the 68nm and 94nm silica sol, respectively, at dye concentration of $175{\mu}g/m^2$. However, the adsorption ratio of Tartrazine dye on 68nm and 94nm silica sol was about 0% for both of them. The surface charge density is $-0.40C/m^2$, $-0.26C/m^2$ for 68nm and 94nm silica sol, respectively. The amount of Methyl violet dye adsorbed on silica sol increased with an increase of surface charge density of particle at the same concentration of the dye. The adsorbed amount of the silica having high surface charge density is larger at high pH domain. But adsorbed amount of the silica having low surface charge density is larger at low pH domain.

Embryotoxic and Teratogenic Effects of Tartrazine in Rats

  • Tartrazine (TAZ) is one of the most commonly used artificial dyes for foods and drugs. We determined the effect of TAZ on fetal development by examining morphological, visceral, and skeletal malformations in rat fetuses following daily oral administration of TAZ to pregnant Wistar rats at the 6th-15th day of gestation. TAZ at 0.45 and 4.5 mg/kg induced 6.0 and 7.1% fetal resorptions, as well as 10.0 and 10.5% fetal mortality, respectively. Fetal body weight and length were significantly lower in the groups treated with TAZ at 0.45 ($3.97{\pm}0.21g$ and $27.3{\pm}0.54mm$, respectively) and 4.5 mg/kg ($3.48{\pm}0.15g$ and $23.22{\pm}1.02mm$, respectively) than in the control group ($4.0{\pm}0.15g$ and $30.01{\pm}0.42mm$, respectively). TAZ at 0.45 and 4.5 mg/kg induced hepatic damage (20 and 33.3%, respectively), dark brown pigmentation due to hemosiderin in the splenic parenchyma (16.7 and 21.7%, respectively), as well as destructed and necrotic renal tubules (16.7 and 26.7%, respectively) in the fetuses. Moreover, TAZ at 0.45 and 4.5 mg/kg caused one or more missing coccygeal vertebrae (20 and 40%, respectively), missing sternebrae (6 and 10%, respectively), missing hind limbs (24 and 4%, respectively), and irregular ribs (16 and 20, respectively) in the fetuses. We concluded that TAZ has embryotoxic and teratogenic potentials in rats.
