Effect of Cinnamomum Cassia on Cartilage Protection in Rabbit and Human Articular Cartilage

  • Background & Objective: Articular cartilage is a potential target for drugs designed to inhibit the activity of matrix metalloproteinases (MMPs) to stop or slow the destruction of the proteoglycanand collagen in the cartilage extracellular matrix. The purpose of this study was to investigate the effects of Cinnamomum cassia in inhibiting the release of glycosaminoglycan (GAG), the degradation of collagen, and MMP activity in rabbit and human articular cartilage explants. Methods: The cartilage-protective effects of Cinnamomum cassia were evaluated by using glycosaminoglycan degradation assay, collagen degradation assay, colorimetric analysis of MMP activity, measurement of lactate dehydrogenase activity and histological analysis in rabbit cartilage explants culture. Results: Interleukin-1a (IL-1a) rapidly induced GAG, but collagen was much less readily released from cartilage explants. Cinnamomum cassia significantly inhibited GAG and collagen release in a concentration-dependent manner. Cinnamomum cassia dose-dependently inhibited MMP-1, MMP-3 and MMP-13 activities from IL-1a-treated cartilage explants culture when tested at concentrations ranging from 0.02 to 1 mg/ml. Conclusion : These results indicate that Cinnamomum cassia inhibits the degradation of proteoglycan and collagen through the down regulation of MMP-1, MMP-3 and MMP-13 activities of IL-1a-stimulated rabbit and human articular cartilage explants.

A Study on the Time Delay Compensate Algorithm in Uniform Linear Array Antenna on Radar System (레이더시스템의 등 간격 선형 배열 안테나에서 시간 지연 보상 알고리즘 연구)

  • This paper proposed a control algorithm to compensate the delay time to improve the signal to noise, and the proposed control algorithm estimate the target information to apply the continuous wave radar equation. The proposed control algorithm improves the output signal of each array element bv multiplying the weight of the receive signal to the signal to noise ratio. Radar radiate a signal in spatial and the target information is estimated by the echoed signal from the target. But the signal in the wireless communication environment occurs the delay time due to the multipath which appear human and natural structures. It is difficult to accurately estimate the desired information because of the degradation for the system performance due to the interference signal and the signal distortion. The target information can be improved by compensating the delay signal to apply the weight to the received signal by using the uniform linear array antenna. As a simulation result, we show that the performance of the proposed control algorithm and the non-compensated delay time are compared. The proposed control algorithm proved that the target distance estimation information is improved.

Molecular Analysis of the Interaction between Human PTPN21 and the Oncoprotein E7 from Human Papillomavirus Genotype 18

  • Human papillomaviruses (HPVs) cause cellular hyperproliferation-associated abnormalities including cervical cancer. The HPV genome encodes two major viral oncoproteins, E6 and E7, which recruit various host proteins by direct interaction for proteasomal degradation. Recently, we reported the structure of HPV18 E7 conserved region 3 (CR3) bound to the protein tyrosine phosphatase (PTP) domain of PTPN14, a well-defined tumor suppressor, and found that this intermolecular interaction plays a key role in E7-driven transformation and tumorigenesis. In this study, we carried out a molecular analysis of the interaction between CR3 of HPV18 E7 and the PTP domain of PTPN21, a PTP protein that shares high sequence homology with PTPN14 but is putatively oncogenic rather than tumor-suppressive. Through the combined use of biochemical tools, we verified that HPV18 E7 and PTPN21 form a 2:2 complex, with a dissociation constant of 5 nM and a nearly identical binding manner with the HPV18 E7 and PTPN14 complex. Nevertheless, despite the structural similarities, the biological consequences of the E7 interaction were found to differ between the two PTP proteins. Unlike PTPN14, PTPN21 did not appear to be subjected to proteasomal degradation in HPV18-positive HeLa cervical cancer cells. Moreover, knockdown of PTPN21 led to retardation of the migration/invasion of HeLa cells and HPV18 E7-expressing HaCaT keratinocytes, which reflects its protumor activity. In conclusion, the associations of the viral oncoprotein E7 with PTPN14 and PTPN21 are similar at the molecular level but play different physiological roles.

Study on Optimization of Operating Conditions for High Temperature PEM Fuel Cells Using Design of Experiments (실험계획법을 이용한 고온 고분자 전해질 막 연료전지의 운전조건 최적화 연구)

  • High temperature proton exchange membrane fuel cells (PEMFCs) using phosphoric acid (PA) doped polybenzimidazole (PBI) membranes have been concentrated as one of solutions to the limits with traditional low temperature PEMFCs. However, the amount of reported experimental data is not enough to catch the operational characteristics correlated with cell performance and durability. In this study, design of experiments (DOE) based operational optimization method for high temperature PEMFCs has been proposed. Response surface method (RSM) is very useful to effectively analyze target system's characteristics and to optimize operating conditions for a short time. Thus RSM using central composite design (CCD) as one of methodologies for design of experiments (DOE) was adopted. For this work, the statistic models which predict the performance and degradation rate with respect to the operating conditions have been developed. The developed performance and degradation models exhibit a good agreement with experimental data. Compared to the existing arbitrary operation, the expected cell lifetime and average cell performance during whole operation could be improved by optimizing operating conditions. Furthermore, the proposed optimization method could find different new optimal solutions for operating conditions if the target lifetime of the fuel cell system is changed. It is expected that the proposed method is very useful to find optimal operating conditions and enhance performance and durability for many other types of fuel cell systems.

  • Handover of WiBro is based on 802.16e hard handover scheme. When PSS is handover, it is handover that confirm neighbor's cell condition and RAS ID in neighbor advertisement message. Serving RAS transmits HO-notification message to neighbor RAS. Transmiting HO-notification message to neighbor RAS, it occurs many signaling traffics. Also, When WiBro is handover, It occurs many packet loss. Therefore, user suffer service degradation. LPM handover is supporting seamless handover because it buffers data packets during handover. So It is proposed scheme that predicts is LPM handover and reserves target RAS with pre-authentication. These schemes occur many signaling traffics. In this paper, we propose PSS Movement Prediction to solve signaling traffic. Target RAS is decided by old data in history cache. When serving RAS receives HO-notification-RSP message to target RAS, target RAS inform to crossover node. And crossover node bicast data packet. If handover is over, target RAS forward data packet. Therefore, It reduces signaling traffics but increase handover success rate. When history cache success, It decrease about 48% total traffic. But When history cache fails, It increase about 6% total traffic

Efficient Verification of X-ray Target Replacement for the C-series High Energy Linear Accelerator

  • The manufacturer of a linear accelerator (LINAC) has reported that the target melting phenomenon could be caused by a non-recommended output setting and the excessive use of monitor unit (MU) with intensity-modulated radiation therapy (IMRT). Due to these reasons, we observed an unexpected beam interruption during the treatment of a patient in our institution. The target status was inspected and a replacement of the target was determined. After the target replacement, the beam profile was adjusted to the machine commissioning beam data, and the absolute doses-to-water for 6 MV and 10 MV photon beams were calibrated according to American Association of Physicists in Medicine (AAPM) Task Group (TG)-51 protocol. To verify the beam data after target replacement, the beam flatness, symmetry, output factor, and percent depth dose (PDD) were measured and compared with the commissioning data. The difference between the referenced and measured data for flatness and symmetry exhibited a coincidence within 0.3% for both 6 MV and 10 MV, and the difference of the PDD at 10 cm depth ($PDD_{10}$) was also within 0.3% for both photon energies. Also, patient-specific quality assurances (QAs) were performed with gamma analysis using a 2-D diode and ion chamber array detector for eight patients. The average gamma passing rates for all patients for the relative dose distribution was $99.1%{\pm}1.0%$, and those for absolute dose distribution was $97.2%{\pm}2.7%$, which means the gamma analysis results were all clinically acceptable. In this study, we recommend that the beam characteristics, such as beam profile, depth dose, and output factors, should be examined. Further, patient-specific QAs should be performed to verify the changes in the overall beam delivery system when a target replacement is inevitable; although it is more important to check the beam output in a daily routine.

A Study on Persulfate Oxidation to Remove Chlorinated Solvents (TCE/PCE) (과황산(persulfate) 산화반응을 이용한 염소계 화합물(TCE, PCE) 분해에 관한 연구)

  • In situ chemical oxidations (ISCO) are technologies for destruction of many contaminants in soil and groundwater, and persulfate has been recently studied as an alternative ISCO oxidant. Trichloroethylene (TCE) and tetrachloroethylene (PCE) were chosen for target organic compounds. The objective of this study is to demonstrate the influence of initial pH (3, 6, 9, 12), oxidant concentrations (0.01, 0.05, 0.1, 0.3, 0.5 M), and contaminants concentrations (10, 30, 50, 70, 100 mg/L) on TCE/PCE degradation by persulfate oxidation. The maximum TCE/PCE degradation occurred at pH 3, and the removal efficiencies with this pH condition were 93.2 and 89.3%, respectively. The minimum TCE/PCE degradation occurred at pH 12, and the removal efficiencies were 55.0 and 31.2%, respectively. This indicated that degradation of TCE/PCE decreased with increasing the initial pH of solution. Degradation of TCE/PCE increased with increasing the concentration of persulfate and with decreasing the concentration of contaminants (TCE/PCE). The optimum conditions for TCE/PCE degradation were pH 3, 0.5 M of persulfate solution, and 10 mg/L of contaminant concentration. At these conditions, the first-order rate constants ($k_{obs}$) for TCE and PCE were 1.04 and 1.31 $h^{-1}$, respectively.

Characteristics of the TCE removal in FeO/Fe(II) System (FeO/Fe(II) 시스템에서 TCE의 제거 특성)

  • The reaction between iron oxide and ferrous iron is known to be the adsorption of ferrous iron onto the oxide surfaces that produces Fe(II)-Fe(III) (hydr)oxides and ferrous oxide oxidized to ferric ion which is the reducing agent of the target compounds. In our investigations on DS/S using ferrous modified steel slag, the results did not follow the trends. FeO and Fe(II), the major component of steel slag, were used to investigate the degradation of TCE. Degradation did not take place for the first and suddenly degraded after awhile. Degradation of TCE in this system was unexpected because Fe(II)-Fe(III) (hydr)oxides could not be produced in absence of ferric oxide. In this study, the characteristics of FeO/Fe(II) system as a reducing agent were observed through the degradation of TCE, measuring byproducts of TCE and the concentration of Fe(II) and Fe(III). Adsorption of ferrous ion on FeO was observed and the generation of byproducts of TCE showed the degradation of TCE by reduction in the system is obvious. However it did not correspond with the typical reducing mechanisms. Future research on this system needs to be continued to find out whether new species are generated or any unknown mineral oxides are produced in the system that acted in the degradation of TCE.

In situ Recovery of hCTLA4Ig from Suspension Cell Cultures of Oryza sativa (형질전환 벼 현탁세포 배양에서 hCTLA4Ig의 in situ 회수)

  • In this research, recombinant human cytotoxic T-lymphocyte antigen 4-immunoglobulin (hCTLA4Ig) was produced by transgenic rice cells. RAmy3D promoter was used for overcome the limitation of low expression level in transgenic plant cells, and the secretion of target protein was accomplished by signal peptide. However, the RAmy3D promoter system which can be induced only by sugar starvation causes the decrease of cell viability. As a result, cell death promotes the release of protease which degrades the target proteins. The protein stability and productivity can be significantly influenced by proteolysis activity. Therefore, development of new strategies are necessary for the in situ recovery of target proteins from cell culture media. In this study, in situ recovery was performed by various strategies. Direct addition of Protein A resin with nylon bag leads to cell death by increased shear stress and decrease in production of hCTLA4Ig by protease. Medium exchange through modified flask could recover hCTLA4Ig with high cell viability and low protease activity, on the other hand, the productivity was lower than that of control. When in situ recovery was conducted at day 7 after induction in air-lift bioreactor, 1.94-fold of hCTLA4Ig could be recovered compared to control culture without in situ recovery. Consequently, in situ recovery of hCTLA4Ig from transgenic rice cell culture could enhance productivity significantly and prevent degradation of target proteins effectively.

Evaluation of Stream Water Quality to Select Target Indicators for the Management of Total Maximum Daily Loads (수질오염총량관리 대상물질 선정을 위한 하천수질 평가)

  • It is one of the most critical steps identifying impaired waterbodies exactly in the selection of target water quality indicators for the management of Total Maximum Daily Loads (TMDLs). Excess ratio and excess level were applied and analyzed by the stream zone basis in order to evaluate water impairment for Nakdong, Geum, Youngsan and Seomjin rivers. Each river basin was divided into stream zones in the light of its watershed and waterbody characteristics. Selected water quality parameters discussed in this study were pH, DO, BOD, COD, SS, T-P, T-Coli and F-Coli. The excess ratios of the water quality parameters were used to discriminate water bodies that did not meet water quality standards. The excess levels were used to classify the degradation of water quality. The excess ratios and the excess levels to the water quality criteria of the medium influence areas were used for each stream zone. The results indicate that the excess ratios and the excess levels are varied on the stream zone in each river basin. Three parameters, pH, DO and SS, met water quality standards in all stream zones. The other five parameters indicated very high excess ratios in most waterbodies, and especially T-P and T-Coli revealed to be very high excess levels in some waterbodies. These parameters could be considered as major target indicators for the management of TMDLs.
