Effect of Taraxacum herba Extract on the Hepatic Xanthine Oxidase Activity in Rats (포공영 추출물이 흰쥐간 Xanthine Oxidase 활성에 미치는 영향)

  • This study was undertaken to investigate the effect of Taraxacum herba extract on the hepatic xanthine oxidase activity as a oxygen free radical generating enzyme in vitro and in vivo. It was observed that partial purified hepatic xanthine oxidase (type O) activity was strongly inhibited by the addition of Taraxacum herba n-butanol extract in vitro. The Km value of xanthine oxidase without affecting the Vmax value for xanthine was significantly increased by the addition of ta-dase (type O) activity was significantly inhibited by the treatment of Taraxacum gerba n-butanol ex-tract for 5days(over 40mg/kg, i.p), whereas, xanthine oxidase (type D) activity was not changed by the injection of Taracacum herba n-butanol extract. Meanwhile, liver weight / body weight(%), serum alanine aminotransferase activity and hepatic lipid peroxide content in Taraxacum herba n-buta-nol extract-treated rat were not changed. These findings led us to conclude that Taraxacum herba n-butanol extract may regulate the hepatic xanthine oxidase type O activity to prevent toxic effect of oxidative stress by the oxygen free radicals.

  • A study on the antitoxic effects of Herba Taraxaci extracts against Cadmium Chloride Toxicity in Rats. This study was performed to find out the effect of taraxacum platy carpum against Cadmium toxicity. The experimental rats were divided into 5 groups such as control group, Cadmium alone treatment group, three simultaneous treatment groups of taraxacum platy carpum and cadmium. Rat were given pellets administration with three dosage of taraxacum platy carpum such as 4 mg/kg body weight for four weeks. The results were summarized as follows on: 1. The simultaneously administration of taraxacum platy carpum and cadmium significantly more decreased cadmium concentration in liver and kidney tissues compared to the administration of cadmium alone. (p<0.05) 2. The simultaneously administration of taraxacum platy carpum and cadmium significantly more increased metallothionein concentration in liver and kidney tissues compared to administration alone. (p<0.05) 3. When liver and kidney tissues were observed with optical microscope obvious changes were visible in those tissue.

  • Objectives : The purpose of this study was to characterize the effect of the fraction of Taraxacum officinale and T.coreanum on the learning and memory impairments induced by scopolamine. Methods : The cognition-enhancing effect of Taraxacum officinale and T.coreanum was investigated using a passive avoidance test, the Morris water maze test and Y-maze test in mice. Drug-induced amnesia was induced by treating animals with scopolamine (1 mg/kg, i.p.). Results : The results showed that the Aug harvested T.offiicinale extract-treated group (200 mg/kg, p.o.) and the tacrine-treated group (10 mg/kg, p.o.) significantly ameliorated scopolamine-induced amnesia based on the Passive avoidance Y-maze test and Watermaze test. And these results are same manner in DPPH radical scavenger effect and Acetylcholineseterase inhibition effect. Conclusions : These results suggest that Taraxacum officinale extract maybe a useful cognitive impairment treatment, and its beneficial effects are depending on the collecting time and origin plants. As a result, Taraxacum officinale harvested in August improve memory most.

  • Glutamate-induced oxidative injury contributes to neuronal degeneration in many central nervous system (CNS) diseases, such as epilepsy and ischemia. Inducible heme oxygenase (HO)-1 acts against oxidants that are thought to play a role in the pathogenesis of these diseases. In the present study, we investigated the neuroprotective effects of the standard extracts of Taraxacum officinale Weber, one of the original plants of Taraxaci Herba, on glutamate-induced oxidative injury in mouse hippocampal HT22 cells. The standard EtOH extract of the aerial parts of T. officinale (NNMBS270) showed significant cytoprotective effects on glutamate-induced neurotoxicity and induced the expression of heme oxygenase (HO)-1 in the mouse hippocampal HT22 cells, while the roots' extract (NNMBS271) did not show neuroprotective effect. These results suggest that the extract of the aerial parts of T. officinale could be an effective candidate for the treatment of ROS-related neurological diseases.

Genetic Diversity of the Original Plant for Taraxaci Herba, Taraxacum spp. by the Analysis of AFLP (AFLP 분석을 통한 포공영 기원식물 민들레의 유전 다양성 분석)

  • Collected germplasms of five representative dandelion species (Taraxacum ohwianum, T. platycarpum, T. platypecidum, T. officinale, and T. coreanum) were 104 lines from different habitates in Korea and China. Their genetic diversity was analyzed by genomic fingerprinting method using amplified fragment length polymorphism (AFLP). AFLP results of 6 primer combinations were revealed 1,176 total DNA fragments and 523 polymorphic bands with a 44.4% ratio of polymorphism. On the basis of similarity coefficient analysis by unweight pair group method with arithmetic averages (UPGMA), 104 dandelion germplasm lines were ranged from 0.64 to 0.99 and clustered distinct five group depending on the species. Furthermore, a principal coordinate analysis (PCA) by the application of multi-variate analysis indicated significantly greater differences among species than geographical origins.

  • The polysaccharide fraction from Taraxii Herba showed potent immunopotentiating activities with antitumor activities. The fraction having small amount of protein inhibited the growth of solid tumor and increased peritoneal exudate cells and immunoorgan weights in normal mice, and also increased hypersensitivities in tumor bearing mice.

Mouse Single Oral Dose Toxicity Test of Taraxaci Herba Aqueous Extracts (포공영(蒲公英) 추출물의 마우스 단회 경구투여 독성 실험)

  • The object of this study was to obtain acute information (single oral dose toxicity) of Taraxaci Herba (Dried total parts of Taraxacum platycarpum. H.Dahlstedt (Compositae)), has been traditionally used in Korean medicine for treating various inflammatory diseases. In order to observe the 50% lethal dose (LD 50), approximate lethal dosage (ALD) and target organs, test articles were once orally administered to female and male ICR mice at dose levels of 2,000, 1,000, 500 mg/kg according to the recommendation of Korea Food and Drug Administration Guidelines. The mortality and changes on body weight, clinical signs and gross observation were monitored during 14 days after single oral treatment of Taraxaci Herba aqueous extracts according to KFDA Guidelines with organ weights and histopathological observations of 12 types of principle organs. After single oral treatment of Taraxaci Herba aqueous extracts, we could not find any mortality and toxicological evidences up to 2,000 mg/kg treated group, the limited dosages in rodents at body and organ weights, clinical signs, gross and histopathological observations. Except for slight soft feces, which were detected in male mice treated with 2,000 mg/kg of Taraxaci Herba aqueous extracts at 1 day after end of treatment. The results obtained in this study suggest that the LD 50 and ALD of Taraxaci Herba aqueous extracts in both female and male mice after single oral treatment were considered as over 2,000 mg/kg because no mortalities were detected up to 2000 mg/kg that was the highest dose recommended by KFDA and OECD. However, it also observed that the possibility of digestive disorders, like diarrhea when administered over 2,000 mg/kg of Taraxaci Herba aqueous extracts in the present study, but these possibilities of digestive disorders can be disregard in clinical use because they ate transient in the highest dosages male only.

Biological and Antifungal Activity of Herbal Plant Extracts against Candida Species (수종의 한약재 추출물의 항산화 활성 및 항진균 활성)

  • Anticandidial activity of seven herbal extracts, Taraxacum Platycarpum, Houttuyniae Herba, Lonicerae Flos, Anemarrhena Rhizome, Forsythia Fruit, Paeoniae Ratix, and Coptidis Rhizoma, were determined against five different Candida sp. by agar diffusion assay. The concentration of total phenolic compounds of seven herbal extracts ranged from 0.6 to $2.5{\mu}g/mg$. The total antioxidant activities showed that Taraxacum Platycarpum and Houttuyniae Herba were 60% in 80% ethanol extract and Lonicera Flower and Paeoniae Ratix were 70, 75%, respectively, in 100% ethanol extract. Coptidis Rhizoma extract showed antifungal activity against non-Candida albicans, C. tropicalis and C. glabrata. The MIC values of a compound separated in TLC from Coptidis Rhizoma extract were 24, and $48{\mu}g/mL$ against C. tropicalis and C. glabrata. The above compound showed the same retention time with berberin in HPLC analysis.

Anti-inflammatory effect and contents from the aerial part and root of the various Taraxacum spp. distributed in Korea (국산 5종 포공영(蒲公英)의 항염 효과 및 성분 함량 비교 연구)

  • Objectives : Taraxaci Herba et Radix (THR) is widely used as a food and medicinal herb in Korea. It has been used for treatment of virus inflammatory disease, liver diseases and gastritis. So far, anti-inflammatory effects and constituents of various species in THR has not been studied for comparison. The aim of this study is to compare the anti-inflammatory effects of the aerial part and root from various THR. Also, we have compared the contents of its known constituents with each. Methods : In this study, we estimated anti-inflammatory effect and compared their constituent by HPLC. For the determination of anti-inflammatory effects, we investigated NO and $PGE_2$ production by ELISA. The expressions of iNOS was determined by western blotting in LPS-induced RAW 264.7 macrophage cells. And, standard compounds which are methyl gallate, gallic acid, syringic acid and esculetin of THR were analyzed by HPLC using a $C_{18}$ column. Results : Methanol extracts of THR decreased NO and $PGE_2$ production. The expressions of iNOS protein were also decreased in methanol extracts of THR. As a result, HPLC analysis showed that they showed similar patterns. Methyl gallate and esculetin showed the highest content. Methyl gallate was included over 10% content in each aerial part and root of THR. Conclusions : These results indicate that most of THR distributed in Korea might represent therapeutic agent for treatment of inflammatory diseases.

Functional Biological Activity of Hot Water and Ethanol Extracts from Taraxaci Herba (포공영의 열수 및 에탄올 추출물의 기능적 생리활성)

  • This study was conducted to investigate the functional biological effects of hot water and ethanol extracts from Taraxacum mongolicum (TM). Then, the hot water and ethanol extracts of TM were measured for total flavonoids content, total phenolics content, electron donating ability, nitrite-scavenging ability, SOD-like activity, tyrosinase inhibitory effect, and elastase inhibitory effect. Total flavonoids contents of hot water and ethanol extracts from TM were 7.80$\pm$0.97 mg/g and 9.12$\pm$0.51 mg/g, respectively, and total phenolics contents were estimated as 54.20$\pm$1.95 mg/g for water extract and 79.43$\pm$4.44 mg/g for ethanol extract. The $RC_{50}$ values for electron donating ability of hot water and ethanol extracts were 943.98 $\mu$g/mL and 309.41 $\mu$g/mL. SOD-like activity and nitrite-scavenging ability were dependent on concentration of hot water and ethanol extracts, and the activity of ethanol extract was higher than that of hot water extract. However, hot water and ethanol extracts from TM showed no inhibitory activities on the elastase and tyrosinase inhibitory activities. Based on the above results, the ethanol extract of TM seems to be the most pertinent for use as functional food and cosmetic.
