Occurrence of Sweet-potato Whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae) and Its Response to Insecticide in Gyeonggi Area (경기지역에서 담배가루이의 발생 및 약제반응)

  • This study investigated the occurrence of sweet potato whitefly, Bemisia tabaci affecting cucumber, eggplant and red pepper, as well as sweet potato species, and its response to insecticides in Gyeonggi province from 2010 to 2011. Sweet potato whitefly is widespread throughout the southern part of Gyeonggi province. Most regional populations of B. tabaci belong to biotype Q having been reported in the south Korea since 2005, but in Goyang mixed populations of two biotypes (B and Q) were found. Survey results of Tomato Yellow Leaf Curl Virus (TYLCV) disease that was vectored by B. tabaci indicated that this virus disease was not spread throughout the Gyeonggi province. Biotype Q of B. tabaci was found to be resistant to neonicotinoid insecticides, whereas biotype B was highly susceptible to them.

Incidence of Viral Diseases on Solanaceae Crops in Gangwon Province (강원지역 가지과 작물 바이러스 발생현황)

  • This study researched the current occurrence status of viral diseases of solanaceous crops in the main production areas of Gangwon region, for four years from 2014 to 2018. From 2014 to 2018, in eight cities and counties including Chuncheon, CMV, BBWV2, PMMoV, TSWV, and PVY occurred in chilli peppers. In case of tomato, the research was performed in 2017 and 2018 while TSWV and TYLCV were verified in Chuncheon, Hoengseong, and Cheorwon. Especially, TYLCV occurred in Chuncheon region for the first time in 2017, which was verified in 2018. Lastly, the current occurrence status of viral diseases of bell peppers was researched in seven cities and counties like Gangneung, Hoengseong, and Cheorwon from 2015 to 2018. In the results, CMV, BBWV2, PMMoV, PepMoV, and TSWV were verified in bell peppers of Gangwon region. During the research period, there were no crops suspected or infected by BWYV, so that it is judged that there is no villiferous or infected aphid by this virus in Gangwon-do. Such results of this study could be used as the basic data for the stable production of crops through the early prevention of plant diseases occurring in the solanaceous crops of Gangwon region.

Occurrence of Virus Diseases on Major Crops in 2008 (2008년 우리나라 주요 작물 바이러스병 발생 상황)

  • Viruses diagnosed on crops including rice plants from farmers or agricultural extension agencies cover the country were 11 species including Broad bean wilt virus 2 (BBWV2) in 2008. Tomato spotted wilt virus (TSWV) was the most important virus having the detection rate of 22.9%. Two viruses of Tomato yellow leaf curl virus (TYLCV) and Tobacco leaf curl virus (TLCV) inducing leaf yellow and curl diseases on tomatoes were occurred newly with the detection rate of 12.2% and 4.0%, respectively, in 2008. Rice stripe virus (RSV) was occurred on 869.5 ha mainly at Jindo and Haenam areas in Jeollanamdo province. At Jindo area, 12 plots were damaged severely with the infected hill rate of 83.8%. At the main production area of oriental melon at Seongju, almost all fruits from whole sale market at Seongju were infected with Cucumber green mottle mosaic virus (CGMMV) as the detection rate of 87%. The areas occurred TSWV in Korea were 25 totally from 2003 including 7 areas newly reported in 2008 including Naju in Jeoallanamdo. TSWV could be reduced as 0.1 % from 5.3% by covering insect proof net in vinyl house after chemical soil sterilization. Tomato yellow leaf curl disease was occurred on April in 2008 at Tongyoung area in Kyeongsangnamdo, and detected continuously at 13 areas, 7 in Kyeongsangnamdo, 4 in Jeollabukdo and 2 in Jejudo. Potato spindle tuber viroid (PSTVd) was occurred abruptly in a confined space of a civil breeding greenhouse and a cultivar evaluation field followed by disuse 17.4 M/T of potato tubers. No PSTVd was detected at 17 fields cultivated the related potatoes to the bred company by RT-PCR.

  • Bemisia tabaci is one of polyphagous insect pests that transmits Tomato Yellow Leaf Curl Virus (TYLCV) and Cassava Brown Streak Disease (CBSD). Insecticides are primarily applied to control B. tabaci, but it has limits due to the development of resistance. As a result, a fixed precision sampling plan was developed for its integrated pest management (IPM). The tomato plants were divided into top (more than 130cm from the ground), middle (70 cm to 100 cm above the ground), and bottom (50 cm or less above the ground) strata, before visual sampling of the larvae of B. tabaci. The spatial distribution analysis was conducted using Taylor's power law coefficients with pooled data of top, middle, bottom strata. Fixed precision sampling plan and control decision-making were developed with precision levels and action threshold recommended from published scientific papers. To assess the validation of the developed sampling plans, independent data not used in the analysis were evaluated using the Resampling Validation for Sampling Plan (RVSP) program.

Construction of cDNA Library for Using Virus-induced Gene Silencing (VIGS) Vector with the Sweetpotato Whitefly, Bemisia tabaci(Hemiptera: Aleyrodidae) (담배가루이(Bemisia tabaci, Aleyrodidae, Hemiptera)에서 Virus-induced Gene Silencing (VIGS) Vector를 이용하기 위한 cDNA Library 제작)

  • The sweetpotato whitefly, Bemisia tabaci, is the major insect pest that transmitted over 100 plant viruses including tomato yellow leaf curl virus (TYLCV) of tomato plant as virus vector in the world. In this study, cDNA library of whitefly was constructed using Gateway system for selecting target gene in order to control of B. tabaci using virus-induced gene silencing (VIGS) vector with RNAi. First of all, when using oligo d(T) rimer, the calculated titer of cDNA library was confirmed with $1.4{\times}10^4$ clones and average insert sizes was confirmed with 1 kb. However, insert size was very big for construction of cDNA. Otherwise, when using attB-N25 random primer and sonication for 6 sec, the calculated titer of cDNA library was confirmed with $1.04{\times}10^5$ clones. But mostly insert band wasn't identified on the electrophoresis, because it seemed that insert size is too small (${\leq}100bp$), also the size of identified insert was somewhat big. Finally, when using oligo d(T) primer and sonication for 1 sec, cDNA insert of whitefly was appropriated for VIGS with 300-600 bp. However, cDNA sequence included a poly A and titer was very low to $5.2{\times}10^2$ clones. It was supposed that heat shock transformation was used instead of electro-transformation. It is considered that when constructing cDNA library for using VIGS vector, (1) random primer should be used for First strand cDNA synthesis in order to remove poly A and (2) sonication for 1 sec should be performed in order to get appropriated insert size and (3) electro-transformation should be performed in order to improve transformation efficiency.

Occurrence of Virus Diseases on Major Crops in 2010 (2010년 우리나라 주요 작물 바이러스병 발생 상황)

  • The kinds of crop requested from agricultural actual places of famers, Agricultural extension services and so forth was 8 including red pepper for vegetables, 4 including apple for fruit trees and 6 including chrysanthemum for flowers in 2010. The important vegetables in clinical diagnosis of viral diseases were tomato, watermelon and red pepper having the requested rate of 31.8%, 21.4% and 19.5%, respectively. On fruit trees, grape and apple were most common with the requested rate of 63.6% and 33.0%, orderly. On floral crops, tulip and cactus were damaged by viral diseases with the requested rate of 60.0% and 20.0%, orderly. On peppers and tomatoes, six viruses including Cucumber mosaic virus (CMV) and Broad bean wilt virus 2 (BBWV2) infected. Five viruses including Melon necrotic spot virus (MNSV) and CMV were identified from watermelons. On grapes, six viruses including Grapevine fleck virus (GFkV) infected. CMV was identified from six vegetables including pepper out of 8 kinds of vegetables and tulip plant. Total agents of virus and viroid species were 32 and 4 species, respectively, in 2010. Tomato yellow leaf curl disease by Tomato yellow leaf curl virus and Tobacco yellow leaf curl virus was occurred newly at 18 Si/Gun areas including Buan, Jeonbuk province in 2010 and the total areas were increased up to 58 Si/Gun from the first incidence in 2008. Tomato spotted wilt virus (TSWV) occurred newly at two areas of Jinan, Jeonbuk and Jeju in Jeju province in 2010, and the incidence areas were expanded to 25 Si/Gun areas from severe occurrence at Anyang area in 2004. No incidence of TSWV was recorded only in Gyeongbuk and Chungbuk province. Tomato bushy stunt virus occurred newly at Jinju, Gyeongnam, and it had the total incidence areas of 5 Si/Gun after first observation at Sacheon, Gyeongnam in 2004.

Colors and Sizes of Insect Screen Net Influence Physical Control of Bemisia tabaci and Frankliniella occidentalis under Controlled Environments (환경제어 조건에서 방충망 색과 크기가 담배가루이 및 꽃노랑총채벌레의 물리적 방제에 미치는 영향)

  • BACKGROUND: The tobacco whitefly(Bemisia tabaci Gennadius) and western flower thrips(Frankliniella occidentalis Pergande) seriously damaged to several greenhouse crops and transmitted plant viruses such as the Tomato Yellow Leaf Curl Virus(TYLCV) and Tomato Spotted Wilt Virus(TSWV). Objective of this study was to elucidate exclusion effects of insect screen nets by various hole sizes and colors for control of the two insect pests in controlled environments such as a closed plant production system.METHODS AND RESULTS: The exclusion effects to various hole sizes of three other colors with 30 individuals of two insect pests was evaluated. B. tabaci was not showed not difference to different colors and sizes. F. occidentalis showed that 0.2 mm black screen was the most effective exclusion than other colors of 0.6 and 0.8 mm.CONCLUSION: The two insects were different reponses to various hole sizes of white and other color screen nets. It was proved that the 0.4 mm white screen net used in this experimental condition was suitable for exclusion of B. tabaci and 0.2 mm black forF. occidentalis.
