Effective Control Strategy against Bacterial Blight on Carrot (당근 세균잎마름병에 대한 효과적 방제 수단)

  • Bacterial blight of carrot caused by Xanthomonas hortorum pv. carotae (Xhc) is one of the serious diseases of carrot, of which control measures has not been still established in the domestic farm. In this study, in order to select effective sterilizer for bacterial blight of carrots, three antibiotics such as streptomycin, oxolinic acid, kasugamycin, two copper compounds like copper hydroxide and copper sulfate basic and three rhizobacteria Burkholderia gladioli MRL408-3, Pseudomonas fluorescens TRH415-2 and Bacillus cereus KRY505-3 were selected to investigate their direct antibacterial effects using artificial media, aiming to identify effective pesticides against Xhc. Among them, treated medium with antibiotics such as streptomycin, oxolinic acid, and the antagonistic rhizobacteria MRL408-3 were formed inhibition zone. The agrochemicals and the rhizobacteria MRL408-3, which showed antibacterial effects on carrot leaves, pre-treated on the carrot leaves and then inoculated with Xhc. High control effects were shown on the carrot leaves pre-treated with both streptomycin and oxolinic acid. Scanning electron microscopy images of the carrot leaf surfaces showed that the population of bacteria decreased significantly on leaves pre-treated with streptomycin and oxolinic acid. From these results, it can be inferred that antibiotics like streptomycin and oxolinic acid exhibit superior control effects compared to other agents. This study provides valuable insights towards establishing an effective control system for bacterial blight of carrot.

Virulence Factors and Antimicrobial Resistance of Vibrio parahaemolyticus Isolated from Commercial Fisheries Products (시판수산물에서 분리한 Vibrio parahaemolyticus의 병원성 인자와 항균제 내성 현황)

  • Vibrio parahaemolyticus causes food poisoning, mainly via marine fisheries products. We investigated the virulence factors and drug resistance of V. parahaemolyticus isolated from fisheries products purchased from the Yeosu Fisheries Market. The isolates were identified using a variety of biochemical tests and the detection of toxR and hns gene. The presence of the virulence factor-encoding genes tdh and trh in the isolates was also investigated by PCR. The resistance of the isolates to 13 antibacterial agents was tested using the disc-diffusion method and carriage of β-lactamase genes and class 1 integrons by ampicillin-resistant isolates was investigated by PCR. Four of seventeen isolates identified as V. parahaemolyticus by biochemical tests produced a species-specific PCR band. Those isolates showed >98% 16S rRNA gene sequence homology with V. parahaemolyticus and only one isolate harbored the tdh gene. All of the V. parahaemolyticus isolates were resistant to ampicillin and amoxicillin; moreover, VPA0477, a class A β-lactamase gene, and class 1 integrons were detected. Therefore, V. parahaemolyticus from fisheries products represents a low risk to human health. Also, V. parahaemolyticus is likely to develop multidrug resistance because it has class 1 integrons.

Identification of a de novo mutation (H435Y) in the THRB gene in a Korean patient with resistance to thyroid hormone (갑상선호르몬 수용체 베타 유전자 돌연변이(H435Y)가 확인된 갑상선호르몬 저항성 증후군 1례)

  • The syndrome of resistance to thyroid hormone (RTH) is characterized by reduced tissue sensitivity to thyroid hormone (TH). In the majority of subjects, RTH is caused by mutations in the thyroid hormone receptor beta ($TR{\beta}$) gene, located on the chromosome locus 3p24.3. RTH is inherited in an autosomal dominant manner. The clinical presentation of RTH is variable, but common features include elevated serum levels of thyroid hormone (TH), a normal or slightly increased thyrotropin (thyroid stimulating hormone, TSH) level that responds to thyrotropin releasing hormone (TRH), and goiter. We report a 4 year-old girl, who was clinically euthyroid in spite of high total and free $T_4$, and $T_3$ concentrations, while TSH was slightly increased. Sequence analysis of the thyroid hormone receptor beta gene (THRB) confirmed a heterozygous C to T change at nucleotide number 1303, resulting in a substitution of histidine by tyrosine at codon 435 (H435Y). Further analysis of her parents revealed that the H435Y variation was a de novo mutation since neither parents had the variation. Her parents' TH and TSH levels were within normal range.

  • Several recent advances in our knowledge of thyroid physiology have broad application to the diagnosis and management of thyroid disorders. For in the thyroid, more than other endocrine organs, pathophysiology can be translated directly into the diagnosis and management of thyroid disease. Graves' disease is a syndrome including goiter with hyperthyroidism, exophthalmos and dermopathy. The pathogenesis of Graves' disease is not yet clearly identified, but various autoantibodies to the thyroid gland and immunopathologic studies indicate that autoimmune processes are involved in the pathogenesis of the disease. The diagnosis and management of Graves' disease are largely dependent on radionuclide techniques as radioimmunoassay, radioactive iodine therapy and so on. Several laboratory tests are also developed to determine the remission of this disase including TRH stimulation test, $T_3$ suppression test and detection of thyroid stimulating immunoglobulins. Autoimmune thyroiditis is almost certainly a primary immunologic disease and the incidence tends to increase recently, mainly due to the application of biopsy technique in thyroid diseases. Thyroid nodules have been a great challenge to physicians because of the possibility of malignancy. But recently, cytologic examination of thyroid aspirate provides a very simple and also reliable diagnostic method in patients with thyroid nodules. In 163 patients with thyroid nodules, only 19.3% was revealed to be malignant. Therefore cytologic examination of thyroid aspirate and thyroid biopsy should be included in the diagnosis of nodular patients prior to surgical intervention. In this paper, a comprehensive review is presented on the pathogenesis, clinical features, laboratory findings and therapeutic modalities of various thyroid diseases on the basis of over 80 researches performed during the past 20 years at radioisotope clinic, Seoul National University Hospital.

Isolation, Molecular Characterization and Antibiotic Susceptibility Pattern of Vibrio parahaemolyticus from Aquatic Products in the Southern Fujian Coast, China

  • Vibrio parahaemolyticus is a major gastroenteritis-causing pathogen in many Asian countries. Antimicrobial resistance in V. parahaemolyticus has been recognized as a critical threat to food safety. In this study, we determined the prevalence and incidence of antimicrobial resistance in V. parahaemolyticus in the southern Fujian coast, China. A total of 62 isolates were confirmed in retail aquatic products from June to October of 2018. The serotype O3:K6 strains, the virulence genes tdh and trh, antibiotic susceptibility and molecular typing were investigated. Then plasmid profiling analysis and curing experiment were performed for multidrug-resistant strains. The results showed that the total occurrence of V. parahaemolyticus was 31% out of 200 samples. Five strains (8.1%) out of 62 isolates were identified as the V. parahaemolyticus O3:K6 pandemic clone. A large majority of isolates exhibited higher resistance to penicillin (77.4%), oxacillin (71%), ampicillin (66.1%) and vancomycin (59.7%). Seventy-one percent (44/62) of the isolates exhibited multiple antimicrobial resistance. All 62 isolates were grouped into 7 clusters by randomly amplified polymorphic DNA, and most of the isolates (80.6%) were distributed within cluster A. Plasmids were detected in approximately 75% of the isolates, and seven different profiles were observed. Seventy-six percent (25/33) of the isolates carrying the plasmids were eliminated by 0.006% SDS incubated at 42℃, a sublethal condition. The occurrence of multidrug-resistant strains could be an indication of the excessive use of antibiotics in aquaculture farming. The rational use of antimicrobial agents and the surveillance of antibiotic administration may reduce the acquisition of resistance by microorganisms in aquatic ecosystems.

Virulence Factors and Antimicrobial Susceptibility of Vibrio parahaemolyticus Isolated from the Oyster Crassostrea gigas (양식 굴(Crassostrea gigas)에서 분리된 장염비브리오균의 독소 유전자 보유 및 항균제 감수성)

  • This study investigated the prevalence of Vibrio parahaemolyticus in the oyster Crassostrea gigas, which is commonly consumed raw. The presence of virulence factors and the antimicrobial susceptibility of isolates were also investigated. The overall prevalence rate of V. parahaemolyticus in oysters was 37.5% (36/96) and the range of concentrations was 30-11,000 MPN/100 g. PCR-based assays indicated that 9.6% (11/115) of the isolates were positive for the thermostable direct hemolysin-related hemolysin gene (trh), while none of the isolates were positive for the thermostable direct hemolysin gene (tdh). The Multiple Antibiotics Resistance (MAR) index was measured for 16 common antimicrobial agents and 46.1% (53/115) of the isolates had a MAR index > 0.2. The MAR index ranged from 0.07 to 0.73. The highest MAR index was observed with strain s150608, isolated in June 2015, which exhibited resistance to 11 antimicrobial agents. Our results demonstrate that oysters are high-risk sources of V. parahaemolyticus, although no antimicrobial agent was being used to promote growth or to treat bacterial infections in the sampled oyster-growing areas.

Temperature-Dependency Urease Activity in Vibrio parahaemolyticus is Related to Transcriptional Activator UreR

  • Vibrio parahaemolyticus possessing urease-positive property is relatively rare, but such strains consistently exhibit the TDH-related hemolysin (TRH) gene. In this study, we examined the effects of incubation temperature on urease activity expression, using the TH3996 and AQ4673 strains where the enzyme activity is known to be temperature-dependent and -independent, respectively. In the TH3996 strain, $\beta$-galactosidase activity was 4.4-fold lower after $30^{\circ}C$ cultivation than after $37^{\circ}C$ in a ureR-lacZ fusion strain, but temperature dependency was not found in ureD- or nikA-lacZ fusion strains. However, ureR-, ureD-, and nikA-lacZ fusions of the AQ4673 strain was not influenced by incubation temperature. We compared the promoter sequences of ureR between the above two strains. Intriguingly, we detected mismatches of two nucleotides between the two strains located at positions -66 and -108 upstream of the methionine initiation codon for UreR. Additionally, urease activity was not affected by culture temperature at either $30^{\circ}C$ or $37^{\circ}C$ by allelic introduction of the AQ4673 ureR gene into the TH3996 ureR deletion mutant. Taken together, our study demonstrates that the transcriptional factor UreR is involved in the temperature dependency of urease activity, and two nucleotides within the ureR promoter region are of particular importance for the urease activity dependency of V. parahaemolyticus.

Antimicrobial Resistance and Minimum Inhibitory Concentrations of Vibrio parahaemolyticus Strains Isolated from Seawater and Commercial Fisheries (해수 및 시판 수산물에서 분리한 장염비브리오균(Vibrio parahaemolyticus)의 항균제 내성 및 최소발육억제농도의 규명)

  • Eighty-three Vibrio parahaemolyticus isolates from surface seawater in Gomso Bay on the west coast of Korea, and commercial fisheries from Gunsan fisheries center were analyzed for the presence of virulence genes and susceptibility to 30 different antimicrobials. All 83 isolates were examined for the presence of two virulence genes (tdh or trh) using polymerase chain reaction; however, neither gene was found in any of the isolates. A disk diffusion susceptibility test, showed that all of the strains studied were resistant to clindamycin, oxacillin, ticarcillin, and vancomycin, and also revealed varying levels of resistance to ampicillin (98.8%), penicillin G (95.2%), streptomycin (20.5%), cefoxitin (14.5%), amikacin (6.0%), cephalothin (4.8%), and erythromycin (3.6%). However, all of the strains were susceptible to 19 other antimicrobial agents, including cefepime, cefotaxime, chloramphenicol, gentamycin, nalidixic acid, sulfamethoxazole/trimethoprim, and trimethoprim. All 83 isolates (100%) were resistant to five or more classes of antimicrobials, and two strains exhibited resistance to ten antimicrobial agents. The average minimum inhibitory concentrations against V. parahaemolyticus of clindamycin, oxacillin, ticarcillin, and vancomycin were 55.9, 98.3, 499.3, and 44.3 ㎍/mL, respectively. These results provide new insight into the necessity for seawater sanitation in Gomso Bay and commercial fisheries, and provide evidence to help reduce the risk of contamination by antimicrobial-resistant bacteria.

The Role of $K^+$ Channels on Spontaneous Action Potential in Rat Clonal Pituitary $GH_3$ Cell Line

  • The types of $K^+$ channel which determine the pattern of spontaneous action potential (SAP) were investigated using whole-cell variation of patch clamp techniques under current- and voltage-clamp recording conditions in rat clonal pituitary $GH_3$ cells. Heterogeneous pattern of SAP activities was changed into more regular mode with elongation of activity duration and afterhyperpolarization by treatment of TEA (10 mM). Under this condition, exposure of the class III antiarrhythmic agent E-4031 $(5\;{\mu}M)$ to $GH_3$ cells hardly affected SAP activities. On the other hand, the main $GH_3$ stimulator thyrotropin-releasing hormone (TRH) still produced its dual effects (transient hyperpolarization and later increase in SAP frequency) in the presence of TEA. However, addition of $BaCl_2$ (2 mM) in the presence of TEA completely blocked SAP repolarization process and produced membrane depolarization in all tested cells. This effect was observed even in TEA-untreated cells and was not mimicked by higher concentration of TEA (30 mM). Also this barium-induced membrane depolarization effect was still observed after L-type $Ca^{2+}$ channel was blocked by nicardipine $(10\;{\mu}M).$ These results suggest that barium-sensitive current is important in SAP repolarization process and barium itself may have some depolarizing effect in $GH_3$ cells.

Rat Gonadotropes and Somatotropes Express Growth Hormone Releasing Hormone Gene in the Pituitary (흰쥐 뇌하수체 Gonadotropes와 Somatotropes에서의 Growth Hormone Releasing Hormone 유전자 발현)

  • Several lines of evidence indicate that some neuropeptides classically associated hypothalamus have been found in pituitary gland, suggesting the existence of local regulation of pituitary function. Among the hypothalamic releasing hormones, genes for TRH and GnRH are expressed in the rat anterior pituitary gland. The present study was carried out to investigate the expression of the GHRH gene in rat anterior pituitary and the pituitary-derived cell lines. The presence of GHRH transcripts in pituitary tissue was shown by 3'rapid amplification of cDNA end (3'-RACE) analysis. In reverse transcription-polymerase chain reaction (RT-PCR) study, GHRH cDNA fragments were amplified from two pituitary-derived cell lines, $\alpha$T3 cells originated from mouse gonadotrope and GH3 cells from rat somatolactotrope. Immunoreactive GHRH was detected in large and medium-sized pituitary cells by immunocytochemistry. Significant amounts of GHRH-like molecules were found in the GH3 cell extracts. In RNase protection assay, the level of pituitary GHRH mRNA was augmented by ovariectomy. These results demonstrate that GHRH gene is expressed in the rat gonadotropes and somatotropes, and suggest that the pituitary GHRH could be participated in the paracrine and/or autocrine regulation of cell proliferation, as well as promoting growth hormone secretion.
