Clinical Application of Serum CEA, SCC, Cyfra21-1, and TPA in Lung Cancer (폐암환자에서 혈청 CEA, SCC, Cyfra21-1, TPA-M 측정의 의의)

  • Background : Tumor markers have been used in diagnosis, predicting the extent of disease, monitoring recurrence after therapy and prediction of prognosis. But the utility of markers in lung cancer has been limited by low sensitivity and specificity. TPA-M is recently developed marker using combined monoclonal antibody of Cytokeratin 8, 18, and 19. This study was conducted to evaluate the efficacy of new tumor marker, TPA-M by comparing the estabilished markers SCC, CEA, Cyfra21-1 in lung cancer. Method : An immunoradiometric assay of serum CEA, sec, Cyfra21-1, and TPA-M was performed in 49 pathologically confirmed lung cancer patients who visited Keimyung University Hospital from April 1996 to August 1996, and 29 benign lung diseases. Commercially available kits, Ab bead CEA (Eiken) to CEA, SCC RIA BEAD (DAINABOT) to SCC, CA2H (TFB) to Cyfra2H. and TPA-M (DAIICHI) to TPA-M were used for this study. Results : The mean serum values of lung cancer group and control group were $10.05{\pm}38.39{\mu}/L$, $1.59{\pm}0.94{\mu}/L$ in CEA, $3.04{\pm}5.79{\mu}/L$, $1.58{\pm}2.85{\mu}/L$ in SCC, $8.27{\pm}11.96{\mu}/L$, $1.77{\pm}2.72{\mu}/L$ in Cyfra21-1, and $132.02{\pm}209.35\;U/L$, $45.86{\pm}75.86\;U/L$ in TPA-M respectively. Serum values of Cyfra21-1 and TPA-M in lung cancer group were higher than control group (p<0.05). Using cutoff value recommended by the manufactures, that is $2.5{\mu}/L$ in CEA, $3.0{\mu}/L$ in Cyfra21-1, 70.0 U/L in TPA-M, and $2.0{\mu}/L$ in SCC, sensitivity and specificity of lung cancer were 33.3%, 78.6% in CEA, 50.0%, 89.7% in Cyfra21-1, 52.3%, 89.7% in TPA-M, 23.8%, 89.3% in SCC. Sensitivity and specificity of nonsmall cell lung cancer were 36.1%, 78.1% in CEA, 50.1%, 89.7% in Cyfra21-1, 53.1%, 89.7% in TPA-M, 33.8%, 89.3% in SCC. Sensitivity and specificity of small cell lung cancer were 25.0%, 78.5% in CEA, 50.0%, 89.6% in Cyfra21-1, 50.0%, 89.6% in TPA-M, 0%, 89.2% in SCC. Cutoff value according to ROC(Receiver operating characteristics) curve was $1.25{\mu}/L$ in CEA, $1.5{\mu}/L$ in Cyfra2-1, 35 U/L in TPA-M, $0.6{\mu}/L$ in SCC. With this cutoff value, sensitivity, specificity, accuracy and kappa index of Cyfra21-1 and TPA-M were better than CEA and SCC. SCC only was related with statistic significance to TNM stages, dividing to operable stages(TNM stage I to IIIA) and inoperable stages (IIIB and IV) (p<0.05). But no tumor markers showed any correlation with significance with tumor size(p>0.05). Conclusion : Serum TPA-M and Cyfra21-1 shows higher sensitivity and specificity than CEA and SCC in overall lung cancer and nonsmall cell lung cancer those were confirmed pathologically. SCC has higher specificity in nonsmall cell lung cancer. And the level of serum sec are signiticantly related with TNM staging.

Evidence for Regulation of Interaction of Endogenous Protein Kinase C(Pkc) Substrates with Plasma Membrane by PKC Down-Regulation in K562 Cells

  • In the particulate fraction obtained from PKC-down regulated K562 cells by treatment for 24 h with 200nM TPA, phosphorylation of two proteins with molecular weight, 100 kDa and 23 kDa (designated p100 and p23, respectvely) was depleted and addition of exogenous purified PKC to this fraction failed to testore their phosphorylation. However, in the soluble fraction, all of phosphoproteins abolished by long-term treatment with TPA were restored by exogenously added PKC. Phosphorylation of two proteins was increased by short-term tretment (20 min), and diminished with the persistant exposure to TPA as well as at a concentration as low as 1nM. When K562 cells were treated with 1 nM and 200 nM TPA for 24 h, phosphorylation of p100 was restored with or without exogenous PKC on 2-3day and 6day after removal of treated TPA, respectively. Two-dimensional electrophoresis of phosphoproteins revealed that phosphorylated p100 (pl=5.9) and p66 species were completely absent from the particulate fraction of K562 cells treated with 200nM TPA for 24 h. These results suggest that the interaction of sensitive endogenous substrates, p100 and p23 with the plasma membrane might be regulated by PKC-down regulation without displacement to the cytosol and the interaction of p100 with the membrane might be reveersible.

Effects of TPA and IAA on Corn Coleoptile Elongation (옥수수 자엽초의 신장에 미치는 TPA와 IAA의 효과)

  • In view of the well-established role of protein kinase C effector element in signal transduction of animal systems, the possibility of diacylglycerol (DAG) and its analog 12-O-tetradecanoylphorbol13-acetate (TPA) having an effect on auxin-induced growih of com coleoptiles was explored. Both DAG and TP A were found to promote cell elongation in the coleoptile tissue_ Treatment of tissue with these protein kinase C-activating agents resulted in increase in the growth rate over the control by about 300%. When 1M was applied to TPA-pretreated coleoptiles. auxin effect appeared synergistic. Morever. coleoptile growth was found to be inhibited by staurosporine and methylated TPA, both of which are known to specifically inhibit protein kinase C. Electrophoretic and autoradiographic patterns of soluble proteins from the coeoptiles indicated that either 1M or TPA tereatment resulted in increased phosphorylation of certain proteins of 205 Kd. 66 Kd and 32 Kd in size. The results obtained from the present work suggest that protein kinase C may be associated with auxin action on cell elongation in the corn coleoptile segments.gments.

A Study on Performance Improvement of Glucose Sensor Adopting a Catalyst Using New Cross Liker (새로운 가교제를 적용한 촉매를 이용한 글루코스 센서의 성능향상 연구)

  • In this study, we synthesized a new biocatalyst consisting of glucose oxidase (GOx), polyethyleneimine (PEI) and carbon nanotube (CNT) with addition of terephthalaldehyde (TPA) (TPA/GOx/PEI/CNT) for fabrication of glucose sensor that shows improved sensing ability and stability compared with that using other biocatalysts. Main bonding of the new TPA/GOx/PEI/CNT catalyst is formed by Aldol condensation reaction of functional end groups between GOx/PEI and TPA. Such formed bonding structure promotes oxidation reaction of glucose. Catalytic activity of TPA/GOx/PEI/CNT is evaluated quantitatively by electrochemical measurements. As a result of that, large sensitivity value of $41{\mu}Acm^{-2}mM^{-1}$ is gained. Regarding biosensor stability of TPA/GOx/PEI/CNT catalyst, covalent bonding formed between GOx/PEI and TPA prevents GOx molecules from becoming leaching-out and contributes improvement in biosensor stability. With estimation of the biosensor stability, it is found that the TPA/GOx/PEI/CNT catalyst keeps 94.6% of its initial activity even after three weeks.

Determination of Eu(III) by Fluorescence Spectrometry (형광분광법에 의한 Eu(Ⅲ)의 정량)

  • Methods to determine EU(Ⅲ) ion in aqueous solution by fluorescence spectrometry based upon the ligand sensitized fluorescence of Eu(Ⅲ)-terephthalic acid (TPA) complex ion have been studied. The effects of excitation wavelength, pH, concentration of TPA and emission wavelength on the fluorescence intensity were investigated. The fluorescence intensity of the Eu(Ⅲ) complex ion was further increased with addition of trioctylphosphine oxide (TOPO). In this case Triton X-100 was used to dissolve TOPO in aqueous solution. The calibration curve for Eu(Ⅲ) was linear over the range from $1.0{\times}10^{-6}M\;to\;4.0{\times}10^{-4}M$ and the detection limit was $1.0{\times}10^{-6}M$ under the experimental conditions of 256 nm, 5.6, $3.5{\times}10^{-4}$M$ and 615 nm for excitation wavelength, pH, concentration of TPA and emission wavelength, respectively. When TOPO was added to the Eu(Ⅲ)-TPA system, the concentration range of linear response and the detection limit were $1.0 {\times}10^{-9}M\;to\;1.0{\times}10^{-4}M,\;1.0{\times}10^{-7}M,$ respectively under the experimental conditions of 284 nm, 4.4 and $1.0{\times} 10^{-4}M$ for excitation wavelength, pH and concentration of TOPO, respectively. Effects of interferences from various cations for the determination of Eu(Ⅲ) ion were also investigated.

Phorbol Ester TPA Modulates Chemoresistance in the Drug Sensitive Breast Cancer Cell Line MCF-7 by Inducing Expression of Drug Efflux Transporter ABCG2

  • Recent studies have indicated a link between levels of cyclooxygenase-2 (COX-2) and development of the multidrug resistance (MDR) phenotype. The ATP-binding cassette sub-family G member 2 (ABCG2) is a major MDR-related transporter protein that is frequently overexpressed in cancer patients. In this study, we aimed to evaluate any positive correlation between COX-2 and ABCG2 gene expression using the COX-2 inducer 12-O-tetradecanoylphorbol-13-acetate (TPA) in human breast cancer cell lines. ABCG2 mRNA and protein expression was studied using real-time RT-PCR and flow cytometry, respectively. A significant increase of COX-2 mRNA expression (up to 11-fold by 4 h) was induced by TPA in MDA-MB-231 cells, this induction effect being lower in MCF-7 cells. TPA caused a considerable increase up to 9-fold in ABCG2 mRNA expression in parental MCF-7 cells, while it caused a small enhancement in ABCG2 expression up to 67 % by 4 h followed by a time-dependent decrease in ABCG2 mRNA expression in MDA-MB-231 cells. TPA treatment resulted in a slight increase of ABCG2 protein expression in MCF-7 cells, while a time-dependent decrease in ABCG2 protein expression was occurred in MDA-MB-231 cells. In conclusion, based on the observed effects of TPA in MDA-Mb-231 cells, it is proposed that TPA up-regulates ABCG2 expression in the drug sensitive MCF-7 breast cancer cell line through COX-2 unrelated pathways.

  • In this study, we fabricated the catalysts for enzymatic biofuel cell anode with carbon nanotube (CNT), glucose oxidase (GOx) and various molecular weights branched poly(ethyleneimine)(bPEI) and terephthalaldehyde (TPA) as cross-linker. In case of GOx/bPEI/CNT using only physical entrapments for immobilization, the molecular weights of bPEI didn't affect to electrochemical performances and long term stability. but that of the catalysts cross linked via TPA (TPA[GOx/bPEI/CNT]) improved and the mass transfer of glucose to FAD was interrupted as increasing of the bPEI's molecular weights. Furthermore, it was confirmed that the optimum molecular weight of PEI for TPA [GOx/bPEI/CNT]) structure is 750k that showed marvelous high performance (maximum power density of $0.995mW{\cdot}cm^{-2}$).

The Inhibitory Effect of Chlorophyllin is Influenced by Different Promotion Stages in DMBA-TPA-induced Mouse Skin Carcinogenesis

  • To develop a chemopreventive strategy based on the different stages of premalignant lesions, we hypothesized that the inhibitory effect of chemopreventive agents is influenced by different promotion stages during carcinogenesis. DMBA-TPA-induced skin carcinogenesis was used with ICR mice and chlorophyllin (CHL) was applied as a chemopreventive agent. In vitro assay was performed with Salmonella typhi. TA100 to observe any anti-mutagenic activity of CHL against DMBA. Pre-initiation and pre-promotion effects of CHL were observed by CHL treatment before initiation and before promotion. To evaluate the inhibitory effect at different promotion stages, each group was divided into three subgroups after TPA promotion for 6, 18 and 24 weeks, respectively ; the first subgroup was immediately sacrificed after termination of TPA, the second subgroup was treated with CHL, and the third subgroup was sacrificed 8 weeks after termination of TPA without CHL treatment. The degrees of epithelial dysplasia, papilloma formation, and invasive carcinoma were observed histologically, and GST-Pi expression was observed immunohistochemically. ODC mRNA level was analyzed by reverse transcriptase-polymerase chain reaction. Results showed : CHL dose-dependently inhibited the mutation of Salmonella typhi. TA100; the incidence of epithelial dysplasia and papilloma formation was lower in pre-initiation and pre-promotion CHL-treated mice than DMBA-TPA-treated mice; no invasive carcinoma developed in pre-initiation CHL-treated groups, while 67% of DMBA-TPA treated mice had carcinomas; GST-Pi expression decreased when CHL was treated before initiation and before promotion; and when CHL was treated after termination of TPA application at 18 and 24- week-TPA promotion stages, respectively, the incidence of epithelial dysplasia and papilloma was markedly reduced compared to non-treated groups. When comparing the incidence of epithelial dysplasia and papilloma between the immediately-sacrificed subgroup and the non-treated group with a waiting period, we speculated that the 18-week-TPA promotion stage might belong to the promoter-independent progression stage. At the 18-week-TPA promotion stage, the level of ODC mRNA in the CHL-treated group was clearly reduced to the level of normal tissue. Taking these results together, CHL showed both anti-initiation and anti-promotion effects, while the inhibitory effect of CHL was prominent in the 18-week-TPA promotion stage. However, CHL seems to be incapable of completely blocking the progression in the 24-week-TPA promotion stage.

Effect of Terephthalaldehyde to Facilitate Electron Transfer in Heme-mimic Catalyst and Its Use in Membraneless Hydrogen Peroxide Fuel Cell (테레프탈알데하이드의 전자전달 강화효과에 따른 헴 단백질 모방 촉매의 성능 향상 및 이를 이용한 비분리막형 과산화수소 연료전지)

  • Terephthalaldehyde (TPA) is introduced as a cross liker to enhance electron transfer of hemin-based cathodic catalyst consisting of polyethyleneimine (PEI), carbon nanotube (CNT) for hydrogen peroxide reduction reaction (HPRR). In the cyclic voltammetry (CV) test with 10 mM H2O2 in phosphate buffer solution (pH 7.4), the current density for HPRR of the suggested catalyst (CNT/PEI/hemin/PEI/TPA) shows 0.2813 mA cm-2 (at 0.2 V vs. Ag/AgCl), which is 2.43 and 1.87 times of non-cross-linked (CNT/PEI/hemin/PEI) and conventional cross liker (glutaraldehyde, GA) used catalyst (CNT/PEI/hemin/PEI/GA), respectively. In the case of onset potential for HPRR, that of CNT/PEI/hemin/PEI/TPA is observed at 0.544 V, while those of CNT/PEI/hemin/PEI and CNT/PEI/hemin/PEI/GA are 0.511 and 0.471 V, respectively. These results indicate that TPA plays a role in facilitating electron transfer between the electrodes and substrates due to the π-conjugated cross-linking bonds, whereas conventional GA cross-linker increases the overpotential by interrupting electron and mass transfer. Electrochemical impedance spectroscopy (EIS) results also display the same tendency. The charge transfer resistance (Rct) of CNT/PEI/hemin/PEI/TPA decreases about 6.2% from that of CNT/PEI/hemin/PEI, while CNT/PEI/hemin/PEI/GA shows the highest Rct. The polarization curve using each catalyst also supports the superiority of TPA cross liker. The maximum power density of CNT/PEI/hemin/PEI/TPA (36.34±1.41 μWcm-2) is significantly higher than those of CNT/PEI/hemin/PEI (27.87±0.95 μWcm-2) and CNT/PEI/hemin/PEI/GA (25.57±1.32 μWcm-2), demonstrating again that the cathode using TPA has the best performance in HPRR.

Preparation Nanosized TPA-Silicalite-1 with Different Silica Sources and Promoters (다양한 실리카 원과 결정화 촉진제를 이용한 나노크기의 TPA-Silicalite-1 제조)

  • In this study, nanosized TPA-silicalite-1 was synthesized with a suitable molar composition of TPAOH: $SiO_2$: $H_2O$ for the development of zeolite ceramic membranes to utilize as gas separation. As silica sources, TEOS, LUDOX AS-40 and CAB-O-SIL were used with the starting material of TPAOH. $NaH_2PO_4$, and a variety of acids and bases were used as promoters after TPAOH, $SiO_2$, $H_2O$ gel synthesis. To decrease synthesis time, a two step temperature change method was applied to the synthesis of TPA-silicalite-1 at a low temperature. TPA-silicalite-1 synthesized was analyzed with XRD, SEM, BET and TGA. As a result, TPA-silicalite-1 powders with a particle size of 100 nm and a specific surface area of $416m^2/g$ were obtained as optimum synthesis conditions when the two stage temperature change method was used with $NaH_2PO_4$ as promoter.
