Botulinum toxin type A was intramuscularly administered to Sprague-Dawley rats in both single and 28-day repeated dose toxicity studies. In the single dose toxicity study performed at 25, 50, 100, and 200 ng/kg, LD50 was estimated to be 70.71 ng/kg for males and 97.63 ng/kg for females.(omitted)
Tan spot, caused by the fungus Pyrenophora tritici-repentis, is economically important foliar disease in Latvia, Lithuania, and Romania; however, race structure from Baltic States and Romania is not known. In this study, we performed genotypic and phenotypic race characterization of a large collection of P. tritici-repentis isolates from these countries to determine race structure and utilize this information for better disease management and breeding wheat for tan spot resistance. We characterized 231 single spore isolates from Latvia (n = 15), Lithuania (n = 107), and Romania (n = 109) for Ptr ToxA and Ptr ToxB genes using two genes specific primers. A subset (139) of 231 isolates were further characterized for their race structure by inoculating them individually on tan spot wheat differentials set. Majority (83%) of the 231 isolates amplified Ptr ToxA gene suggesting prevalence of race 1 and 2. Further, phenotypic characterization of 139 isolates also showed wide prevalence of races 1 (68%), 2 (8%), 3 (11%), and 4 (5%) were also identified from Baltic States as well as Romania. Eighteen of the isolates (13%) did not seem to be of any of the eight known races as they lacked Ptr ToxA gene but they behaved like either race 1 or race 2, suggesting possibility of novel toxins in these isolates as their virulence tools.
Purpose: Acute gastroenteritis (AGE) is a major cause of morbidity and remains a major cause of hospitalization. Following the Syrian refugee crisis and insufficient clean water in the region, this study reviews the etiological and epidemiological data in Lebanon. Methods: We prospectively analyzed demographic, clinical and routine laboratory data of 198 children from the age of 1 month to 10 years old who were admitted with the diagnosis of AGE to a private tertiary care hospital located in the district of Nabatieh in south Lebanon. Results: Males had a higher incidence of AGE (57.1%). Pathogens were detected in 57.6% (n=114) of admitted patients, among them single pathogens were found in 51.0% (n=101) of cases that consisted of: Entamoeba histolytica 26.3% (n=52), rotavirus 18.7% (n=37), adenovirus 6.1% (n=12) and mixed co-pathogens found in 6.6% (n=13). Breast-fed children were significantly less prone to rotavirus (p=0.041). Moreover, children who had received the rotavirus vaccine were significantly less prone to rotavirus (p=0.032). Conclusion: Our findings highlight the high prevalence of E. histolytica infection as the major cause of pediatric gastroenteritis in hospitalized children, during the summer period likely reflecting the insanitary water supplies and lack of hygiene. Moreover the 42.4% of unidentified causative pathogens should prompt us to widen our diagnostic laboratory arsenal by adopting new diagnostic technologies.
Background: The number of synthesized chemicals has rapidly increased over the past decade. For many chemicals, there is a lack of information on toxicity. With the current movement toward reducing animal testing, the use of toxicity big data and deep learning could be a promising tool to screen potential toxicants. Objectives: This study identified potential chemicals related to reproductive and estrogen receptor (ER)-mediated toxicities for 1135 cleaning products and 886 laundry products. Methods: We listed chemicals contained in cleaning and laundry products from a publicly available database. Then, chemicals that potentially exhibited reproductive and ER-mediated toxicities were identified using the European Union Classification, Labeling and Packaging classification and ToxCast database, respectively. For chemicals absent from the ToxCast database, ER activity was predicted using deep learning models. Results: Among the 783 listed chemicals, there were 53 with potential reproductive toxicity and 310 with potential ER-mediated toxicity. Among the 473 chemicals not tested with ToxCast assays, deep learning models indicated that 42 chemicals exhibited ER-mediated toxicity. A total of 13 chemicals were identified as causing reproductive toxicity by reacting with the ER. Conclusions: We demonstrated a screening method to identify potential chemicals related to reproductive and ER-mediated toxicities utilizing chemical toxicity big data and deep learning. Integrating toxicity data from in vivo, in vitro, and deep learning models may contribute to screening chemicals in consumer products.
Purpose: Campylobacter species are currently the most common cause of bacterial gastroenteritis. In Lebanon, Campylobacter infection occurrence is underdiagnosed owing to the lack of specific culture and rapid test kits, particularly among children. This study aimed to evaluate the prevalence, laboratory findings, and clinical characteristics of Campylobacter infection in hospitalized children with acute gastroenteritis in South Lebanon. Methods: We conducted a 6-month retrospective cohort study between January and June 2018, including 291 children aged between 1 month and 12 years, who were admitted to a tertiary healthcare center in South Lebanon. The medical files of the patients were reviewed to retrieve the required clinical information, including clinical and laboratory data. Results: The prevalence of campylobacteriosis agents in pediatric patients with acute gastroenteritis is 12.02%. Patients infected with Campylobacter had more severe acute gastroenteritis than Campylobacter-negative patients and often presented with high-grade fever, diarrhea episodes more than six times per day, diarrhea lasting for more than five days, and dehydration. Indeed, children with high-grade fever (≥38.5℃) were five times more likely to test positive for Campylobacter than those with low-grade fever. In addition, the results showed a higher Vesikari score for the majority of Campylobacter-positive patients with severe acute gastroenteritis compared to a moderate profile for Campylobacter-negative patients. Conclusion: The present study findings highlight that Campylobacter infection is frequent among children with acute gastroenteritis. Therefore, the detection of Campylobacter should be carried out for the diagnosis of human gastroenteritis in Lebanon, along with the detection of routine enteropathogens.
Toxicogenomics combines transcriptome, proteome and metabolome profiling with conventional toxicology to investigate the interaction between biological molecules and toxicant or environmental stress in disease caution. Toxicogenomics faces the problems of comparison and integration across different sources of data. Cause of unusual characteristics of toxicogenomic data, researcher should be assisted by data analysis and annotation for getting meaningful information. There are already existing repositories which claim to stand for toxicogenomics database. However, those just contain limited abilities for toxicogenomic research. For supporting toxicologist who comes up against toxicogenomic data flood, now we propose novel toxicogenomics knowledgebase system, XPERANTO-TOX. XPERANTO-TOX is an integrated system for toxicogenomic data management and analysis. It is composed of three distinct but closely connected parts. Firstly, Data Storage System is for reposit many kinds of '-omics' data and conventional toxicology data. Secondly, Data Analysis System consists of analytical modules for integrated toxicogenomics data. At last, Data Annotation System is for giving extensive insight of data to researcher.
Objectives: The purpose of this study was to perform an aggregate human risk assessment for benzene in an industrial complex using the CalTOX model and to improve the reliability and predictability of the model by analyzing the uncertainty and sensitivity of the predicted assessment results. Methods: The CalTOXTM 4.0 beta model was used to evaluate a selected region, and @Risk 7.6 software was used to analyze uncertainty and sensitivity. Results: As a result of performing the aggregate risk assessment on the assumption that 6.45E+04 g/d of benzene would be emitted into the atmosphere over two decades, 3% of the daily source term to air remained in the selected region, and 97% (6.26E+04 g/d) moved out of the region. As for exposure by breathing, the predicted LADDinhalation was 2.14E-04 mg/kg-d, and that was assessed as making a 99.99% contribution to the LADDtotal. Regarding human Riskcancer assessment, the predicted human cancer risk was 5.19E-06 (95% CI; 4.07E-06-6.81E-06) (in the 95th percentile corresponding to the highest exposure level, a confidence interval of 90%). As a result of analyzing sensitivity, 'source term to air' was identified as the most influential variable, followed by 'exposure time, active indoors (h/day)', and 'exposure duration (years)'. Conclusions: As for the results of the human cancer risk assessment for the selected region, the predicted human cancer risk was 5.19E-06 (95% CI; 4.07E-06-6.81E-06) (in the 95th percentile, corresponding to the highest exposure level, a confidence interval of 90%). As a result of analyzing sensitivity, 'source term to air' was found to be most influential.
Objectives: The aim of this study was to secure the reliability of using the CalTOX model when evaluating LADD (or ADD) and Risk (or HQ) among local residents for the emission of BTEX (Benzene, Toluene, Ethylbenzene, Xylene) and by closely examining the difference in the confidence interval of the assessment outcomes according to the difference in the probability density function of input variables. Methods: The assessment was made by dividing it according to the method ($I^{\dagger}$) of inputting the probability density function in meteorological variables of the model with log-normal distribution and the method of inputting ($II^{\ddagger}$) after grasping the optimal probability density function using @Risk. A T-test was carried out in order to analyze the difference in confidence interval of the two assessment results. Results: It was evaluated to be 1.46E-03 mg/kg-d in LADD of Benzene, 1.96E-04 mg/kg-d in ADD of Toluene, 8.15E-05 mg/kg-d in ADD of Ethylbenzene, and 2.30E-04 mg/kg-d in ADD of Xylene. As for the predicted confidence interval in LADD and ADD, there was a significant difference between the $I^{\dagger}$ and $II^{\ddagger}$ methods in $LADD_{Inhalation}$ for Benzene, and in $ADD_{Inhalation}$ and ADD for Toluene and Xylene. It appeared to be 3.58E-05 for risk in Benzene, 3.78E-03 for HQ in Toluene, 1.48E-03 for HQ in Ethylbenzene, and 3.77E-03 for HQ in Xylene. As a result of the HQ in Toluene and Xylene, the difference in confidence interval between the $I^{\dagger}$ and $II^{\ddagger}$ methods was shown to be significant. Conclusions: The human risk assessment for BTEX was made by dividing it into the method ($I^{\dagger}$) of inputting the probability density function of meteorological variables for the CalTOX model with log-normal distribution, and the method of inputting ($II^{\ddagger}$) after grasping the optimal probability density function using @Risk. As a result, it was identified that Risk (or HQ) is the same, but that there is a significant difference in the confidence interval of Risk (or HQ) between the $I^{\dagger}$ and $II^{\ddagger}$ methods.

Single and 28-day repeated dose toxicity studies of botulinum toxin type A(BTA) were carried out in ICR mice and SD rats. respectively. In the single dose toxicity study. BTA was injected intraperitoneally to male and female mice at a single dose of 40, 59, 89, 133 and 200 ng/kg. All animals died from 59 ng/kg. Some clinical signs were observed in most of both sexes from 59 ng/kg, but no signs were seen in all animals at 40 ng/kg. (omitted)
