Bacterial Identification and Detection of Equol in Korean Soybean Paste (한국 된장에서 Equol의 검출 및 미생물 동정)

  • Equol has beneficial effects on human health. Fermented soy products contain equol, and many microbes participate in the equol production process. This study investigated fermented Korean soybean paste, doenjang. Thirty seven doenjang samples collected from different manufacturers were examined. Equol was detected in 3 samples (D2, D13, and D19) at the maximum content of 507 ng/100 g by liquid chromatography-tandem mass spectrometry (LC-MS/MS). Fifteen microbial species were isolated and identified by 16S rRNA gene sequence analysis and by matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS). Bacillus spp, Paenibacillus spp, Tetragenococcus spp, Stapylococcus spp, and Clostridium species were the predominant bacteria in equol containing doenjang samples.

  • 닭의 간은 해독작용, 당의 저장, 혈장 단백질의 합성 등 주요 기능을 하는 것으로 알려져 있다. 본 연구는 간에서 성장 단계별로 발현량에 차이를 보이는 단백질들을 비교해 보았다. 2차원적 전기 영동에 의해 분리된 300개 이상의 단백질들이 확인되었으며 이 중 성장 단계별 특이적인 13개의 단백질은 MALDI-TOF MS에 의하여 분석이 되어졌다. 본 연구를 통하여 밝혀진 단백질들은 생화학적인 연구에 중요한 자료를 제공할 것으로 사료된다.

Multidrug-resistant Staphylococci Found on Book Surfaces in East London Libraries

  • There is an increase in the presence of drug-resistant staphylococci outside of the nosocomial and healthcare setting. Although the presence of staphylococci has been studied in several public spaces, nothing is known on the presence of staphylococci in public libraries. Book surfaces from public libraries in the East London area, United Kingdom were swabbed and cultured and identity of the isolates determined by matrix-assisted laser desorption/ionization time of flight (MALDI-TOF) mass spectrometry (MS). Seven different staphylococcal species were identified by MALDI-TOF-MS analysis. This short study provides evidence of the presence of multidrug-resistant staphylococci in public libraries in the East London area.

Characterization of Polysaccharides from Artemisia capillaris and Artemisia sylvatica (사철쑥과 그늘쑥의 다당류 연구)

  • Two polysaccharides, ACP-UMP and ACP-ULF, were purified from the aerial part of Artemisia capillaris by anion-exchange chromatography, ultrafiltration, and gel filtration chromatography. The polysaccharides appeared to be homogenious from the results of HPLC. The molecular weights of ACP-UMF and ACP-ULF were estimated to be 16305.92 D and 3292.26 D, respectively, by MALDI-TOF MS. The sugar compositions were determined by GC to be arabinose 10.05%, xylose 1.67%, mannose 5.45G, galactose 39.06%, glucose 15.43% for ACP-UMF and arabinose 11.60%, xylose 11.15%, mannose 6.37% galactose 32.47%, glucose 18.35% for ACP-ULF. A polysaccharide, SP-M was determined to be 2462.52 D by MALDI-TOF MS. SP-M consisted mainly of rhamnose 36.49%, arabinose 29.00%, and glucose 19.38%. Incubation of CCl4-intoxicated hepatocytes with ACP-UMF reduced the levels of glutamic pyruvic transaminase (GPT) and cellular malondialdehyde (MDA) to 62.8% and 23.8%. ACP-ULF also reduced the levels of GPT and MDA to 46.1% and 38.1% and 26.3%, respectively.

Analysis of Folate by Matrix-Assisted Laser Desorption/Ionization Time-of-Flight Mass Spectrometry

  • Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) was used to observe folic acid and its derivatives such as tetrahydrofolate and 5-methyltetrahydrofolate in a vitamin tablet and in foods. Folic acid in a vitamin tablet was determined using angiotensin I as an internal reference. Tetrahydrofolic acid, 5-methyltetrahydrofolic acid, and an oxygenated folate were observed from a human blood sample using graphite plate. The results show that these mass spectrometric methods are useful for quickly obtaining a profile of folates.

  • A three-layered poly(benzyl ether) dendrimer having a bis-ethynylbenzene core was synthesized and characterized with $^{1}H$ NMR and MALDI-TOF-MS spectroscopy. The dendrimer was reacted with platinum complexes to obtain platinum-acetylide based organometallic polymers. When the dendrimer was reacted with trans-[$PtCl_{2}(PEt_{3})_{2}$], a high molecular weight polymeric compound was formed, whereas, with cis-[$PtCl_{2}dppp$], a uniform molecular weight compound was formed, which was found to be a dimeric metallacycle by $^{1}H\;NMR,\;^{31}P\;NMR$ and ESI-TOF-MS spectroscopy. Both these complexes exhibited relatively a strong emission around 440 nm, indicating that they could be potential candidates for blue emitting polymer LEDs.

  • The cleavage of fullerene oxides with several aromatic amines such as 4-nitroaniline, 3-nitroaniline, 4-isopropylaniline in the present of $FeCl_3$ occurred under ultrasonic irradiation in air at $25{\sim}43^{\circ}C$. The aminated fullerenes were confirmed by MALDI-TOF-MS and UV-vis spectra.

  • Inclusion compounds using cyclic poly(oxyethylene)s as host molecules have been used to polymerize pyrrole chemically in aqueous medium. This general synthetic strategy makes it possible to grow rigid aromatic polymers in aqueous medium by chemical oxidation method. It is an easy method to obtain rigid polymers in a very mild manner. Some threaded and water-soluble polypyrroles are obtained, and their characterization is performed by NMR, IR, UV, and MALDI-TOF MS measurements.
