The impact of different diets and genders on fecal microbiota in Hanwoo cattle

  • Bovine fecal microbiota is important for host health and its composition can be affected by various factors, such as diet, age, species, breed, regions, and environments. The objective of this study was to evaluate the impact of diet and gender on fecal microbiota in Korean native Hanwoo cattle. The 16S rRNA gene amplicon sequencing of fecal microbiota was conducted from 44 Hanwoo cattle divided into four groups: (1) 11 heifers fed an oat hay plus total mixed ration (TMR) diet for breeding (HOTB), (2) 11 heifers fed an early fattening TMR diet (HEFT), (3) 11 steers fed the early fattening TMR diet (SEFT), and (4) 11 steers fed the late fattening TMR diet (SLFT). Firmicutes and Bacteroidota were the first and second most dominant phyla in all the samples, respectively. The Firmicutes/Bacteroidota (F/B) ratio associated with feed efficiency was significantly greater in the SLFT group than in the other groups. At the genus level, Romboutsia, Paeniclostridium, and Turicibacter were the most abundant in the SLFT while Akkermansia, Bacteroides, and Monoglobus were the most abundant in the HOTB group. Although the same early fattening TMR diet was fed to Hanwoo heifers and steers, Marvinbryantia and Coprococcus were the most abundant in the HEFT group while Alistipes and Ruminococcus were the most abundant in the SEFT group. Shannon and Simpson diversity indices were significantly lower in the SLFT group than in the other groups. Distribution of fecal microbiota and functional genetic profiles were significantly different among the four treatment groups. The present study demonstrates that different diets and genders can affect fecal microbiota and the F/B ratio may be associated with feed efficiency in Hanwoo cattle. Our results may help develop strategies to improve gut health and productivity through manipulation of fecal microbiota using the appropriate diet considering Hanwoo cattle gender.

Effect of Broussonetia papyrifera L. (paper mulberry) silage on dry matter intake, milk composition, antioxidant capacity and milk fatty acid profile in dairy cows

  • Objective: This study was carried out to investigate the possible application of Broussonetia papyrifera (B. papyrifera) silage as a functional feeding stuff in dairy cattle. Methods: Seventy-two Holstein cows were divided into four groups randomly and allocated to 6 pens with 3 individuals in each group and fed the original total mixed ratio (TMR) in the dairy farm or the new TMR with 5%, 10%, and 15% B. papyrifera silage, separately. Feed intake were recorded, milk and blood samples were collected, and milk composition, blood metabolites and milk fatty acids composition were measure at the end of the experiment. Results: Dry matter intake of cows decreased when they fed on diet with B. papyrifera, but no differences were observed in body condition score, milk yield, milk protein and lactose, feed efficiency and serum metabolites between groups. Both 10% or 15% of B. papyrifera silage in the diet significantly increased the immunoglobulin A (IgA) and IgG in serum, 15% of B. papyrifera silage increased the content of serum catalase, superoxide dismutase, total antioxidant capacity, and decreased the content of 8-hydroxy-2'-deoxyguanosine. Furthermore, 10% or 15% of B. papyrifera silage resulted in a significant decrease in the milk somatic cell count, and increased the polyunsaturated fatty acids content in the milk. Conclusion: The diets with 10% to 15% of B. papyrifera silage might enhance the immune and antioxidant function of dairy cows and increase the polyunstaturated fatty acid concentration in the milk.

Fruit and vegetable discards preserved with sodium metabisulfite as a high-moisture ingredient in total mixed ration for ruminants: effect on in vitro ruminal fermentation and in vivo metabolism

  • Objective: Our recent series of laboratory- and large-scale experiments confirmed that under aerobic and anaerobic conditions, sodium metabisulfite (SMB) was effective in preserving nutrients and antioxidant capacity of highly perishable fruit and vegetable discards (FVD). Hence, the purpose of this study was to examine how partial inclusion of SMB-treated FVD in total mixed ration (TMR) influences in vitro ruminal fermentation, whole-tract digestibility, nitrogen metabolism, blood metabolites, and voluntary feed intake of sheep. Methods: The FVD were mixed thoroughly with 6 g SMB/kg wet biomass and kept outdoors under aerobic conditions for 7 days. Four TMRs including four levels of SMB-treated FVD (as-fed basis) at 0%, 10%, 20%, and 30% (equaling to 0%, 1.9%, 3.8%, and 5.7% on dry matter basis, respectively), were prepared as replacement for corn grain. The ruminal fermentation metabolites were studied using an in vitro gas production test. Four mature male Corriedale sheep were assigned at random to the 4 diets for two separate sub-experiments; i) digestibility trial with four 21-d periods, and ii) voluntary feed intake trial with four 28-d periods. Results: Inclusion of SMB-treated FVD in the TMR tended to quadratically increase partitioning factor. No effect was seen on total-tract digestibility of organic matter, ether extract, crude protein, and acid detergent fiber, except for neutral detergent fiber digestibility that tended to linearly increase with increasing SMB-treated FVD in the TMR. The progressive increase of FVD preserved with SMB in the diet had no effect on nitrogen metabolism. Treatment had no effect on serum antioxidant capacity and blood metabolites assayed. Voluntary feed intake was not impaired by inclusion of SMB-treated FVD in the TMR. Conclusion: It appears that FVD preserved with SMB can be safely incorporated into TMR as replacement of corn grain without impairment of nutrient metabolism and feed intake.

Analysis of the influence of sex, slaughter season, and feeding system on carcass traits in Hanwoo

  • This study was conducted to investigate the effects of sex, environmental factors, and feeding system on the carcass traits of Hanwoo (Korean Native Cattle). Data were collected from 7,866 heads slaughtered in Chungnam province during one-year period. Using the collected carcass traits data which greatly influence a Hanwoo's carcass grade, the effects of sex class, slaughter season, and feeding system were estimated. Backfat thickness of steers was significantly higher than that of bulls (p < 0.05). Marbling score was also the highest in steers when compared with cows and bulls (p < 0.05). Live weight and carcass weight were significantly higher in winter than in summer (p < 0.05). However, backfat thickness was significantly lower in summer than in other seasons (p < 0.05). Marbling score was higher in spring and fall than in summer and winter (p < 0.05). In terms of feeding systems, TMR (Total mixed ration) and TMF (Total mixed fermentation feed) fed groups showed the highest carcass grade (p < 0.05). However, the group fed TMR and formula feed at the final fattening period showed the lowest performance (p < 0.05) and it is assumed that some stress was associated to the feed change. The results reconfirm that castration may be recommended in order to improve meat quality and marbling scores in bulls. There was no consistent trend of seasonal effects of slaughter on carcass traits although some traits were significantly affected. Regarding the feeding system, either TMR or TMF can be supplied to achieve high feed efficiency and good carcass characteristics in Hanwoo.

섬유질배합사료 급여가 비육후기 거세한우의 반추위 발효성상, 소화율 및 산육성에 미치는 영향 (Effects of Total Mixed Rations on Ruminal Characteristics, Digestibility and Beef Production of Hanwoo Steers)

  • 본 연구는 시판 비육후기용 사료와 볏짚을 급여한 관행적 비육후기 사료급여형태(대조구)와 특정 비육농가에서 급여하고 있는 섬유질배합사료의 단독급여 효과를 비교하기 위해 증체량, 사료효율, 도체등급 등을 조사하였고(시험 1), 결과 해석을 위해 시험 2에서 반추위 발효성상에 미치는 영향과 소화율을 조사하였다. 비육후기의 섬유질배합사료 사료의 급여효과 시험에서는 섬유질배합사료의 건물 사료섭취량이 1일 7.4kg으로 대조구와 차이가 없었고, 일당 증체량에서는 718g으로 대조구 775g보다 낮았으나 유의성은 없었다. 1kg 증체에 필요한 사료요구량에서는 섬유질배합사료구가 10.5kg으로 대조구보다 약 0.8kg 더 높았다. 대조구의 A등급 출현율은 22%였으나, 섬유질배합사료구는 33%로 섬유질배합사료구의 A등급 출현율이 높았고, 육질 등급에서도 1등급 출현율이 대조구와 섬유질배합사료구에서 각각 56%와 75%로 섬유질배합사료구가 현저히 높았다. 섬유질배합사료 사료의 건물을 비롯한 조단백, 조지방, 그리고 총에너지의 전장소화율은 대조구와 비교하여 약 1.2-1.6배 더 유의적으로 높았던 것으로 나타났다. 반추위 내의 pH 변화는 섬유질배합사료구가 배합사료를 급여한 관행구보다 높은 pH 6.0이상에서 비교적 안정적으로 유지되었다. 반추위내 NH3-N 농도는 섬유질배합사료구가 사료급여 후 8시간 후까지 높은 수준에서 유지되었고, 특히 사료급여 2시간 후 까지 대조구의 2배에 이르는 28.2mg/$d\ell$까지 급격히 증가하였다(P<0.05). 대조구와 섬유질배합사료구의 총 VFA 함량은 사료급여 후 2시간까지 증가하였다가 그 후 감소하는 경향이 비슷하였으며, 유의적인 차이도 없었다. 그러나 branch chained fatty acid (BCFA)인 iso-butyric acid와 iso-valeric acid의 비율에서는 섬유질배합사료와 대조구간에 차이가 있었고, 특히 사료급여 후 3-5시간 내에 섬유질배합사료구가 유의성있게(P<0.01) 높았다. 본 시험의 결과, 섬유질배합사료 급여는 한우의 반추위 발효를 안정화시키고 비육후기 산육성 및 육질등급을 높이는 효과가 있었다.

Effects of Italian ryegrass silage-based total mixed ration on rumen fermentation, growth performance, blood metabolites, and bacterial communities of growing Hanwoo heifers

  • This study utilized Italian ryegrass silage (IRGS) - based total mixed ration (TMR) as feedstuff and evaluated its effects on rumen fermentation, growth performance, blood parameters, and bacterial community in growing Hanwoo heifers. Twenty-seven Hanwoo heifers (body weight [BW], 225.11 ± 10.57 kg) were randomly allocated to three experimental diets. Heifers were fed 1 of 3 treatments as follows: TMR with oat, timothy, and alfalfa hay (CON), TMR with 19% of IRGS (L-IRGS), and TMR with 36% of IRGS (H-IRGS). Feeding high levels of IRGS (H-IRGS) and CON TMR to heifers resulted in a greater molar proportion of propionate in the rumen. The impact of different TMR diets on the BW, average daily gain, dry matter intake, and feed conversion ratio of Hanwoo heifers during the growing period did not differ (p > 0.05). Furthermore, the blood metabolites, total protein, albumin, aspartate aminotransferase, glucose, and total cholesterol of the heifers were not affected by the different TMR diets (p > 0.05). In terms of rumen bacterial community composition, 264 operational taxonomic units (OTUs) were observed across the three TMR diets with 240, 239, and 220 OTUs in CON, L-IRGS, and H-IRGS, respectively. IRGS-based diets increased the relative abundances of genera belonging to phylum Bacteroidetes but decreased the abundances of genus belonging to phylum Firmicutes compared with the control. Data showed that Bacteroidetes was the most dominant phylum, while Prevotella ruminicola was the dominant species across the three TMR groups. The relative abundance of Ruminococcus bromii in the rumen increased in heifers fed with high inclusion of IRGS in the TMR (H-IRGS TMR). The relative abundance of R. bromii in the rumen significantly increased when heifers were fed H-IRGS TMR while P. ruminicola increased in both L-IRGS and H-IRGS TMR groups. Results from the current study demonstrate that the inclusion of IRGS in the TMR is comparable with the TMR containing high-quality forage (CON). Thus, a high level of IRGS can be used as a replacement forage ingredient in TMR feeding and had a beneficial effect of possibly modulating the rumen bacterial community toward mainly propionate-producing microorganisms.

Effects of Lactic Acid Bacteria Inoculant on Fermentation Quality and in vitro Rumen Fermentation of Total Mixed Ration

  • Fermented total mixed ration (TMR) is a novel feed for ruminants in South Korea. The purpose of this study was to evaluate the effects of lactic acid bacteria (LAB) on the quality of TMR and in vitro ruminal fermentation. Strains of three LAB spp. (Lactobacillus plantarum, L. brevis, L. mucosae) were used in fermentation of TMR. Inoculations with the three LAB spp. lowered pH and increased concentrations of lactic acid, acetic acid, and total organic acid compared to non-LAB inoculated control (only addition of an equivalent amount of water) (p<0.05). Bacterial composition indicated that aerobic bacteria and LAB were higher. However, E. coli were lower in the fermented TMR than those in the control treatment (p<0.05). Among the treatments, L. brevis treatment had the highest concentration of total organic acid without fungus detection. Gas production, pH, and ammonia-nitrogen during ruminal in vitro incubation did not differ throughout incubation. However, ruminal total VFA concentration was higher (p<0.05) in the LAB spp. treatments than the control treatment at 48 hours. Overall, the use of L. brevis as an inoculant for fermentation of high moisture. TMR could inhibit fungi growth and promote lactic fermentation, and enhance digestion in the rumen.

사료급여체계가 재래종 및 교잡종 흑염소의 영양소 이용율, 질소축적 및 혈액성상에 미치는 영향 (Effects of Different Feeding Systems on Nutrient Availability, Nitrogen Retention and Blood Characteristics in Native or Crossbred Korean Black Goats)

  • 본 시험은 흑염소의 효율적인 사양관리체계의 확립에 기여하기 위하여, 재래흑염소 10두(male)와 교잡흑염소(재래흑염소$\times$호주야생염소 교잡종; 50:50%) 10두(male)를 공시하여, 시판 배합사료와 조사료를 급여하는 관행사료와 TMR 사료를 각각 급여하였을 때, 흑염소의 사료섭취량, 영양소소화율, 질소축적율 및 혈액성상에 미치는 영향을 조사하였으며, 실험기간은 사료 적응기간 14일 예비기간 10일을 거친 후, 14일간의 본 실험 기간을 두어 총 38일간 실시하였다. 결과를 살펴보면 1일 두당 건물의 섭취량은 TMR을 급여한 교잡종이 가장 높았고(p<0.05), 품종간에서는 재래종 보다 교잡종지, 사료급여체계간에는 관행구보다 TMR구가 높았다(p<0.05). 일당증체량은 재래종 관행구, 재래종 TMR구, 교잡종 관행구, 교잡종 TMR구의 순으로 유의하게 높아졌다(p<0.05). 건물 소화율은 사료급여체계간에서는 TMR구가 관행구보다 유의하게 높았으며, 품종간에도 교잡종이 재래종 보다 높았다(p<0.05). 질소축적량 및 질소축적율은 교잡종 TMR구가 가장 높았고, 재래종 관행구가 가장 낮았다(p<0.05). 혈중 glucose의 함량과 PUN(plasma urea nitrogen)은 교잡종이 재래종 보다 유의하게 높았다(p<0.05). 이상의 결과를 종합하면, TMR 형태의 사료급여가 재래종 보다는 교잡종에 흑염소의 생산성과 사료섭취량, 소화율, 질소축적을 및 혈액성상에 양호한 영향을 나타내어, 효율적인 사양관리체계의 확립과 함께 흑염소 생산농가에 경제적 이득을 줄 수 있을 것으로 판단된다.

맥주박을 첨가한 섬유질 배합사료가 번식흑염소의 영양소 이용율에 미치는 영향 (Effects of Total Mixed Ration with Wet Brewer's Grain on Nutrient Utilization in Breeding Korean Native Goats)

  • 본 연구는 섬유질 배합사료에 맥주박 수준을 달리 첨가하여 번식 흑염소의 사료 및 영양소 섭취량, 영양소 소화율 및 질소 축적율에 미치는 영향을 조사하여 번식 흑염소의 사료로서 맥주박의 사료가치를 평가하고자 실시하였다. 공시축은 번식 흑염소 12두를 4처리구(관행사료구, 맥주박 20, 30 및 40% 첨가구)로 나누어 처리구당 3두씩 대사케이지에 완전임의 배치법으로 배치하여 시험을 실시하였으며 그 결과는 다음과 같다. 맥주박 첨가 수준별 섬유질 배합사료의 일반 조성분은 건물 함량이 $61.46{\sim}65.81%$, 조단백질 함량은 $14.42{\sim}15.59%$의 수준이었으며, ADF 와 NDF 함량은 $28.32{\sim}28.52$$53.46{\sim}54.16%$로 나타나 맥주박 첨가 비율에 따른 섬유소 함량 의 차이는 없었으나, NFC 함량은 맥주박 첨가 비율이 높을수록 NFC 함량이 낮아졌다. 건물 섭취량은 맥주박을 20% 첨가하였을 때 대조구보다 다소 높은 경향이었다. 조단백질과 가소화 조단백질 섭취량은 맥주박 첨가구가 대조구 보다 높은 경향을 나타내었고, NFC 섭취량과 가소화 NFC 섭취량은 대조구가 맥주박첨가구보다 높은 경향이었고 맥주박첨가수준이 높을수록 낮아지는 경향이었다. 건물, ADF, NDF 및 NFC 소화율은 대조구가 맥주박 첨가구 보다 높았으며 (p<0.05), 조단백질 소화율은 맥주박 첨가구가 대조구보다 높은 경향이었고 질소 축적율은 맥주박첨가구가 대조구보다 다소 높은 경향이었다.

Effect of Alcohol Fermented Feed on Lactating Performance, Blood Metabolites, Milk Fatty Acid Profile and Cholesterol Content in Holstein Lactating Cows

  • A feeding experiment with 40 lactating Holstein cows and 4 dietary treatments was conducted to investigate supplementation with different levels of alcohol fermented feed to the TMR on lactating performance, blood metabolites, milk fatty acid profile and cholesterol concentration of blood and milk. Forty Holstein lactating cows ($106{\pm}24$ d post-partum; mean${\pm}$SD) were distributed into four groups and randomly assigned to one of four treatments with each containing 10 cows per treatment. The treatment supplemented with TMR (DM basis) as the control (CON), and CON mixed with alcohol-fermented feeds (AFF) at a level of 5%, 10% and 15% of the TMR as T1, T2 and T3, respectively. Dry matter intake and milk yield were not affected by supplementation of AFF. An increased 4% FCM in the milk occurred in cows fed T3 diet compared with CON, while T1 and T2 diets decreased 4% FCM in a dose dependent manner. Supplementation of AFF increased the concentration of albumin, total protein (TP), ammonia, and high density lipoprotein-cholesterol in serum compared with CON. In contrast, supplementation with AFF clearly decreased concentration of blood urea nitrogen (BUN) and total cholesterol (TC) compare with CON. AFF supplementation increased the proportion of C18:1n9 and C18:2n6 compared to CON. A decrease in the concentration of saturated fatty acid (SFA) for T1, T2 and T3 resulted in an increased unsaturated fatty acid (USFA) to SFA ratio compared to CON. Concentration of cholesterol in milk fat was reduced in proportion to the supplemental level of AFF. Feeding a diet supplemented with a moderate level AFF to lactating cows could be a way to alter the feed efficiency and fatty acid profile of milk by increasing potentially human consumer healthy fatty acid without detrimental effects on feed intake and milk production. A substantially decreased cholesterol proportion in milk induced by supplementation AFF suggests that alcohol fermented feed may improve milk cholesterol levels without any negative effects in lactating cows.
