Differential Effects of Tautomycetin and Its Derivatives on Protein Phosphatase Inhibition, Immunosuppressive Function and Antitumor Activity

  • In the present work, we studied the structure-activity relationship (SAR) of tautomycetin (TMC) and its derivatives. Further, we demonstrated the correlation between the immunosuppressive fuction, anticancer activity and protein phosphatase type 1 (PP1) inhibition of TMC and its derivatives. We have prepared some TMC derivatives via combinatorial biosynthesis, isolation from fermentation broth or chemical degradation of TMC. We found that the immunosuppressive activity was correlated with anticancer activity for TMC and its analog compounds, indicating that TMC may home at the same targets for its immunosuppressive and anticancer activities. Interestingly, TMC-F1, TMC-D1 and TMC-D2 all retained significant, albeit reduced PP1 inhibitory activity compared to TMC. However, only TMC-D2 showed immunosuppressive and anticancer activities in studies carried out in cell lines. Moreover, TMC-Chain did not show any significant inhibitory activity towards PP1 but showed strong growth inhibitory effect. This observation implicates that the maleic anhydride moiety of TMC is critical for its phosphatase inhibitory activity whereas the C1-C18 moiety of TMC is essential for the inhibition of tumor cell proliferation. Furthermore, we measured $in$ $vivo$ phosphatase activities of PP1 in MCF-7 cell extracts treated with TMC and its related compounds, and the results indicate that the cytotoxicity of TMC doesn't correlate with its $in$ $vivo$ PP1 inhibition activity. Taken together, our study suggests that the immunosuppressive and anticancer activities of TMC are not due to the inhibition of PP1. Our results provide a novel insight for the elucidation of the underlying molecular mechanisms of TMC's important biological functions.

Formulation of Cyclic Plasticity Model and FE Analysis for SM490 TMC (SM490 TMC 강재의 반복소성모델의 정식화 및 유한요소해석)

  • In this paper, cyclic plasticity model of SM490 TMC was formulated by basing on monotonic loading test and cyclic loading test. For exact description of cyclic performance and plastic deformation capacity of steel member using SM490 TMC, formulated cyclic plasticity model and finite deformation theory were applied to 3-dimensional elastic-plastic FE analysis. Cyclic plastic behavior of pipe-section steel column using SM490 TMC was clarified by carrying out numerical analysis. Also, in order to clarifying seismic performance of pipe-section steel column using SM490 TMC, analysis results were compared with analysis results of pipe-section steel column using SM490. A comparison of analysis results shows that SM490 TMC pipe-section steel column has a better cyclic performance for strength and energy dissipation than SM490 pipe-section steel column under cyclic loading

Evaluation of Forming Performance of TMC Steel Pipes & Tubes for Building Structure (건축구조용 TMC 강관의 가공성능 평가)

  • With building structures becoming higher and having longer spans, new structural steel with better strength, thicker plate, and performance may be required rather than conventional structural steel. TMC steel is widely used in building structures largely due to its excellent seismic performance, superior weldability, and design strength that is not affected by plate thickness. To make use of TMC steel in pipe structures with large diameter and heavy wall, however, the this study, the degradation of material properties in submerged are welded SM520TMC steel pipes and tubes was evaluated using variable fabrication process and material change. Degradation test results showed that the yield and ultimate strength increased and elongation decreased regardless of the mode of fabrication, i.e., through roll bending or press forming, or steel used, i.e., domestic SM520TMC steel or SM520TMC steel from Japan.

A Study for Structural Stabilities of Beams Built with TMC Fire Resistant Steels by Analytical Method at High Temperatures (해석적 방법에 의한 TMC 건축용 내화강재 적용 보부재의 고온 내력평가 연구)

  • Steel framed building can be destroyed due to deteriation of structural stabilities in a fire. This leads a TMC Fire Resistant Steels and this study analyzed the structural stabilities such as a deflection and a reduction of maximum load capacity for the structural beams built with a TMC Fire Resistant Steel. In this study the structural stabilities were evaluated using a mechanical properties in high temperatures not only a heat transfer theory but a heat stress anlaysis with a statistically determinated beam and a statistially indeternated one. The results showed that a TMC Fire Resistant Steels demonstrated a little lower those of Fire Resistant Steels.

  • Taemyeongcheong (TMC) is Korean traditional extracted drink with various health ingredients. TMC has been used to treat hepatic damage, obesity, gastritis, and colitis. However, the role of TMC on allergic reaction has not been studied yet. In this study, we investigated the anti-allergic effects of TMC against a compound 48/80-induced systemic anaphylactic reaction and IgE-mediated passive cutaneous anaphylaxis (PCA). TMC significantly inhibited the compound 48/80-induced systemic anaphylactic reaction and IgE-mediated PCA reaction. Furthermore, TMC reduced the levels of interleukin (IL)-1β, IL-4, IL-13, and vascular endothelial growth factor in the serum of mice under PCA reaction. Taken together, these results suggest that TMC can play a useful role as an anti-allergic agent.

A Study on the Materials Characteristics of SM570TMC Plates (SM570TMC 강재의 재료특성에 관한 연구)

  • There is increasing need for high-strength steel especially for the high-rise steel building structure. High- strength steels, however, may have mechanical properties that are significantly different from those of the conventional steels.The application of high-strength steels to building structures should be reviewed as to whether inelastic behavior equivalent to that of conventional steels can be attained or not. In this study, SM570TMC steel, which was developed recently in Korea, was tested to evaluate mechanical properties and welding characteristics for use as a structural steel. Yield phenomenon of SM570TMC steel, which is continuous yielding, is quite different from that of conventional steel. The distributions of yield strength of SM570TMC steel were uniform regardless of thickness, while the minimum yield strength was 440MPa. Also, the flat distributions of hardness in z-direction were found to comprise one of the important properties. Results of the charpy impact tests at -5oC revealed toughness values of SM570TMC steel, which were higher than those of equivalent as rolled steel. Carbon equivalent(Ceq) values of SM570TMC steel, which were related to welding performance, were lower than those of equivalent as rolled steel.

Short-term Supplementation with a Trace Mineral-fortified Microbial Culture May Increase Trace Minerals in Longissimus dorsi Muscle and Prevent Incidence of Urolithiasis in Finishing Hanwoo Steers

  • This study evaluated the effects of TMC (trace mineral-fortified microbial culture) supplementation on growth performance, carcass characteristics, and meat quality parameters of Hanwoo steers during the last 4 months of finishing period. The TMC was a combination of 0.4% trace minerals, 20.0% Na-bentonite, and 79.6% feedstuffs, which was inoculated with a mixed microbial culture (Enterobacter ludwigii, Bacillus cereus, B. subtilis, Lactobacillus plantarum, and Saccharomyces cerevisiae). Twenty-four steers were blocked by initial BW ($634{\pm}16kg$) and randomly allocated to one of two treatments (control vs. 3.3% TMC). The effect of TMC supplementation on the growth performance was not significant. There was no incidence of urolithiasis in TMC-fed steers. However 3 out 12 steers (25%) fed the control diet were observed to have urinary calculi. The carcass yield and meat quality parameters were not affected by TMC supplementation, however marbling score was increased in TMC-fed steers (P = 0.08). There was no effect of TMC treatment on the chemical composition of longissimus dorsi muscle (LM). The TMC supplementation increased concentrations of manganese (P < 0.01), cobalt (P = 0.02), iron, and copper (P = 0.06) in LM. In conclusion, TMC treatment did not negatively affect growth performance and meat quality parameters, and positively affected the trace minerals profile of LM.

Hysteretic Behavior Characteristics of SM490-TMC Steel Column (SM490-TMC 강재를 적용한 기둥부재 이력거동의 특성)

  • In design of steel column member using TMCP steels, hysteretic behavior characteristics of steel column must be clarified. To predict hysteretic behavior of steel column using TMCP steels, a cyclic plasticity model is necessary which can consider the mechanical characteristics and stress-strain relationship of TMCP steels. In this paper, a cyclic plasticity model of SM490-TMC was formulated based on monotonic and cyclic loading tests. The formulated cyclic plasticity model was applied to 3-dimensional finite element analysis. Hysteretic behavior characteristics of steel circular column and H-section column using SM490-TMC was presented by carrying out numerical analysis. Also, influence of SM490-TMC on hysteretic behavior of steel column was presented by comparing analysis results both SM490 and SM490-TMC steel column.

  • Various kinds of water soluble N,N,N-trimethylchitosan ammonium chloride(TMC) with different molecular weights were synthesized to examine the antimicrobial activity against Methicillin Sensitive Staphylococcus aureus(MSSA) and Methicillin Resistant Staphylococcus aureus(MRSA), which causes serious hospital infection, and to apply them to antimicrobial finishing agents for textiles. Chitosan samples were highly deacetylated with sodium hydroxide solution and degraded with hydrogen peroxide to control the molecular weight. TMC has the antimicrobial activities against MRSA and MSSA. TMC showed an excellent antimicrobial activity below the molecular weight of 70,000, especially at 40,000. The minimum inhibitory concentration (MIC) of TMC with optimum molecular weight against MRSA and MSSA was 250ppm. Because MRSA did not resist TMC in the subculture test of bouillon medium, it was expected that the successive use of TMC against MRSA was possible.

  • The objective of this study is to evaluate the yield stress of SM570TMC CFT column subject to axial force. These columns were evaluated and compared by statistical tests, during which the displacements and axial loads of column specimens were measured. Test results showed that the yield stress of CFT columns under axial load could be predicted using the previously proposed the yield stress of steel columns.
