Chemical Analysis and Isolation of Antibacterial Compound from Ulmus Species (II) : Isolation and Chemical Structure of Antibacterial Compound (느릅나무 근피의 화학조성분 및 항균성 물질(II) : 항균성 물질의 단리 및 화학구조)

  • The methanol extractives from root bark of Ulmus davidiana var japonica nakai were fractionated with n-hexane, ethyl ether, ethyl acetate and waster, the water soluble fraction was also fractionated with silicagel column chromatograhy. The chemical structure of purifided compounds were identified with UV, IR, 1H-NMR and 13C-NMR spectra and the antibacterial activities also were investigated. Two different antibacterial compounds (compound A and B) were fractionated with silicagel chromatography and TLC. Compounds B was identified as a catechin rahmnoside. The both of compounds had antibacterial activity on Staphylococcus aureus and Salmonella typhimurium.

Comparison of Shear Bond Strength of Different Restorative Materials to Tricalcium Silicate-Based Pulp Capping Materials (Tricalcium Silicate-Based 치수복조재에 대한 수 종 수복재의 전단결합강도 비교)

  • The aim of this study was to evaluate the shear bond strength (SBS) of three typical restorative materials - glass ionomer cement (GIC), resin-modified glass ionomer (RMGIC) and composite resin (CR) - to different pulp capping materials, i.e., Theracal $LC^{TM}$ (TLC), $Biodentine^{TM}$ (BD), and $ProRoot^{TM}$ white MTA (WMTA). 90 acrylic blocks with a center hole were prepared. The holes were completely filled with three pulp capping materials (TLC, BD, and WMTA), with 30 specimens per capping material. The samples were then randomly divided into 3 subgroups of 10 specimens each and were overlaid with GIC, RMGIC, or CR. A total 9 specimen groups were prepared. The SBS was assessed using a universal testing machine. Kruskal-Wallis test and Mann-Whitney's test were performed to compare the SBS among the subgroups (p < 0.05). After the SBS test, the fractured surfaces were examined under a stereomicroscope at a magnification of $25{\times}$. The highest and lowest SBS values were recorded for TLC-CR and TLC-GIC, respectively. With regard to the SBS to the three pulp capping materials, CR was found to be superior to RMGIC and GIC. BD showed a higher SBS compared to TLC and WMTA when used with GIC.

Antifungal activity of essential oil from Cryptomeria japonica against dermatophytic fungi (삼나무 정유의 표재성 진균에 대한 항진균 활성)

  • In this study, the antifungal activity of Cryptomeria japonica essential oil against superficial and allergic fungi, Trichophyton schoenleinii, Trichophyton tonsurans, Trichophyton mentagrophytes, Trichophyton rubrum, Epidermophyton floccosum, and Aspergillus fumigatus, was evaluated for determining the potential compound as dermatitis treatment. Minimum Inhibitory Concentration (MIC) measurement, TLC bioassay and agar dilution methods were used for determining the antifungal activity of crude essential oil and its fractions from C. japonica. Also, their major constituents were analyzed by GC/MS. The MICs were below 500 ppm at all superficial fungi, and spot 1 of C. japonica essential oil showed highly effective antifungal activity by TLC bioassay. In antifungal activity by agar dilution methods, crude oil showed high antifungal effect at more than 500 ppm and fraction D was significantly effective at even 100 ppm except for A. fumigatus. The major compounds of spot 1 and fraction D of C. japonica oil determined by GC/MS were elemol, ${\gamma}$-eudesmol, and ${\beta}$-eudesmol, which could be used as atopic dermatitis treatment material.

Quality inspections of active components in oxidative hair coloring products by UPLC (UPLC를 이용한 염모제품에 함유된 유효성분에 대한 품질검사)

  • To identify oxidative hair dyes in hair-coloring products, the thin-layer chromatography (TLC) screening method was used in accordance with Korean Quasi-drug Codex. However, the TLC method is not reliable when there are very small amount of materials to be tested or when $R_f$ values of several components are similar. In this study, Ultra Performance Liquid Chromatography (UPLC) with a rapid sample preparation method was developed for the reliable and sensitive identification of active components contained in oxidative hair-coloring products. Hexane-distilled water was used for the extraction of active components contained in the products prior to UPLC analysis. The limit of detection of active components was 6.7-77.9 ${\mu}g/L$, and the limit of quantitation was 22.3-259.7 ${\mu}g/L$. Except for ${\alpha}$-naphthol, the range of recovery ratio was 96.2-101.5%. From this study, we demonstrated that oxidative active hair-coloring components can easily be analyzed by rapid extraction method followed by UPLC analysis.

Component Analysis and Study on Anti-elastase Activity of Equisetum arvense Extracts(II) (쇠뜨기 추출물의 성분 분석과 엘라스타제 활성 저해 효과 연구(II))

  • In the previous study, we reported the antioxidative activity of Equisetum arvense extracts. In this study, its inhibitory effect on elastase and components were investigated. Aglycone fractions obtained from the deglycosylation reaction of ethylacetate fraction among the Equisetum arvense extracts, showed 4 bands and 4 peaks in TLC and HPLC experiments, respectively. Four components were identified as luteolin(composition ratio, 19.12%), quercetin(12.87), apigenin(15.81) and kaempferol(52.20). TLC chromatogram of ethylacetate fraction of Equisetum arvense extract revealed 7 bands and HPLC chromatogram showed 8 peaks, which were identified as kaempferol-3,7-O-diglucoside(composition ratio, 15.74%), luteolin-5-O-glucoside(galuteolin, 11.91), apigenin-5-O-glucoside(12.91), kaempferol-3-O-glucoside(astragalin, 27.94), quercetin-glycoside(10.81, structure was not determined), kaempferol-glycoside (12.33, structure was not determined), luteolin(3.72) and apigenin(4.62) in the order of elution time. The inhibitory effect of aglycone fraction on elastase($IC_{50}$, $9.8{\mu}g/mL$) was very high. But ethylacetate fraction(flavonoid glycosides) rarely exhibited the inhibitory activity on elastase. Combined with the previous results of the antioxidative activity of Equisetum arvense extracts, it is concluded that the inhibitory activity on elastase of the aglycone fraction could be applicable to new functional cosmetics for smoothing wrinkles.

  • The ALE specification of EPCglobal is leading the development of RFID standards, includes the Event Cycle Specification (ECSpec) describing how long a cycle is, how to filter RFID tag data and which reader is interested in. The ECSpec is a specification for filtering and collecting RFID tag data. It is registered to a middleware for long time and is evaluated to return results satisfying the requirements included in it. Thus, it is quite similar to the continuous query. It can be transformed into a continuous query as its predicate in WHERE clause is characterized by the long interval. Long intervals cause problems deteriorating insertion and search performance of existing query indices. In this paper, we propose a TLC-index as a new query index structure for long interval data. The TLC-index has hybrid structure that uses the cell construct of CQI-index with the virtual construct of VCR-index for partitioning long intervals. The TLC-index can reduce the storage cost and improve the insertion performance through decomposing long intervals into one or more cell constructs that have long size. It can also improve the search performance through decomposing short intervals into one or more virtual constructs that have short size enough to fit into those intervals.

Studies on antimicrobial agent produced by lactobacillus acidophilus (Lactobacillus acidophilus가 생산한 항균물질(抗菌物質)에 관한 연구(硏究))

  • The research was conducted(1) to confirm the agent(s) responsible for the antimicrobial activity contained in the fermented tomato juice with L. acidophilus(2) to extract and purify the antimicrobial agent(s)(3) to find the biological, physical and chemical properties of the agent(s). The following results were obtained and summarized as followings; 1. The agent responsible for the inhibitory activity was confirmed by both well assay method using fermented tomato juice with L. acidophilus and turbidimetric technique using the cell-free filtrate or neutralized filtrate of tomato acidohilus culture and found exerted antimicrobial agent other than lactic acid. 2. The procedures of purification : The isolation and purification of antimicrobial agent from the lyophilized acidophilus tomato culture were carried out by (1) methanol extraction (2) acetone extraction, (3) Sephadex G-50 gel filtration (4) paper chromatography and (5) thin layer chromatography. 3. The biological, physical and chemical properties of antimicrobial agent: The biological, physical, chemical properties of the purified antimicrobial agent were: (1) The antimicrobial activity was strong against test organisms; Bacillus subtilis(ATCC 6633), Escheichia coli(ATCC 25922), Staphylococcus aureus(ATCC 167), Pseudomonas fluorescens(KFCC 32394), Proteus vulgaris and Shigella dysenteriae. (2) The pH value of the agent was 2.0 and the inhibitory activity was lost when it was neutralized at 7.0 of pH and the agent was heat stable at $121^{\circ}C$ for 60 minutes. (3) The ultraviolet light absorption spectra of methanol-acetone extract and TLC fraction exhibited a maximum absorption at 260nm and 224nm respectively. (4) The most purified agent from TLC plate increased about 130-fold in activity. (5) The agent isolated from TLC plate was free from $H_2O_2$ or lactic acid. 4. Bioautographic assy: By means of bioautography of the agent on silica gel of TLC plate a strong inhibitory activity against B. subtilis was demonstrated. 5. Mass spectrometry: The agent obtained from TLC plate was analyzed by mass spectrometry which show the parent peak at m/e 264 suggesting the molecular weight of the compound and molecular group such as [$C_2H{^+}_4$], [CO], [CH=NH], [$C_3{H^}4_7$], [$\begin{array}{rcl}O\\{\parallel}\\CH_3-C\\\end{array}$], [$C_6-H{^+}_{11}$], [$C_5H{^+}_{11}$], [$\begin{array}{rcl}O\\{\parallel}\\C_5H_7-C^+\\\end{array}$] were suggested.

Isolation and Purification of Tyrosinase Inhibitors from the Seeds of Thuja orientalis L. (백자인(Thuja orientallis L.)으로부터 tyrosinase 저해제의 분리 및 정제)

  • Previously, the methanolic extracts of thirty Korean medicinal plant seeds were screened for tyrosinase inhibitors using a rapid and simple TLC method, which was superior to a conventional spectrophotometrical in vitro assay. As a result, the methanolic extracts of Thuja orientalis seeds was found to have strong tyrosinase inhibitory activity. To isolate active tyrosinase inhibitors, the seeds were defatted with n-hexane under reflux, and then extracted twice with methanol under reflux at 90$^{\circ}C$. The methanolic extract was evaporated to a small volumn in vacuo, and then successively fractionated with ether, ethyl acetate and n-butanol. The ether extract showing significant tyrosinase inhibitory activity was solubilized with 5% NaHCO$_3$and then acidified with 6N HCI. The ether souble acidic fraction was successively ohromatographed on silica gel, Sephadex LH-20 and preparative TLC. Among four compounds isolated, two of them showed stronger tyrosinase inhibitory activity, comparable to that of L-ascorbic acid (IC$\sub$50/=28$\mu\textrm{g}$/$m\ell$). These results suggest that Thuja orientalis seeds may be useful as potential sources of antibrowning agents in fruits and vegetables, and anti-melanoma agents in cosmetics and phamaceuticals.

Degradation and Metabolism of Phorate in Soil (토양중(토양중) Phorate의 분해(분해)와 대사(대사))

  • To get further information on the behavior of phorate(0,0-diethyl S-ethylthiomethyl phosphorodithioate) in soil under the subtropical conditions, a field experiment has been conducted. Phorate granule (10%) was applied to silt loam soil at the rate of 40kg a.i./ha and incorporated to 10cm soil depth. Residues of phorate and its metabolites in soil were determined with GLC and confirmed qualitatively with TLC. Phorate was rapidly oxidized to its sulfoxide and sulfone. Therefore, main metabolic pathway of phorate in soil was the oxidation of phorate to phorate sulfoxide and sulfone. Phorate sulfoxide and sulfone were relatively more persistent than phorate itself. Phoratoxon was detected at low level only up to 30 days after treatment and its sulfoxide and sulfone were not detected during the whole experimental period. Toluene-acetonitrile-nitromethane(40 : 30 : 30, v/v/v) solvent system separated satisfactorily phorate and its five metabolites. Most of the residues was found in the initial incorporation depth $(0{\sim}10cm)$. Consequently, insecticides showed a little downward movement.

TLC, HPTLC FINGERPRINTING AND ACUTE ORAL TOXICITY EVALUATION OF HABB-E-AZARAQI: A NUX-VOMICA-BASED TRADITIONAL UNANI FORMULATION

  • Background and Objective: Nux-vomica based traditional Unani formulation, Habb-e-Azaraqi (HAZ) is an important drug used by Unani physicians since several decades. It possesses Muqawwi-i-A'sab (nervine tonic), Muharrik-i-A'sab (nervine stimulant) properties and is an effective treatment option for diseases like Laqwa (facial palsy), Falij (paralysis), Niqris (gout) and Waja'al-Mafasil (arthritis) etc. The aim of the study is to access and provide information of HAZ for its TLC, HPTLC Fingerprinting defining its clear qualitative perspective and acute oral toxicity evaluation for its safety assessment which was not done earlier, thus contributing in the field of research. Materials and Methods: The chief ingredient, nux-vomica was detoxified as per method mentioned in Unani Pharmacopeia before its use in formulation. TLC and HPTLC was developed under four detection system i.e., UV 366nm, UV 254nm, exposure to iodine vapours and after derivatization with anisaldehyde sulphuric acid. Acute toxicity studies were performed as per OECD Guidelines 425 at a limit dose of 2000 mg/kg. Observations were done for signs of toxicity, body weight, and feed consumption at regular intervals followed by haematological and biochemistry evaluation. Results: The generated data proved the authenticity and established the TLC and HPTLC profile of the formulation. Acute toxicity revealed no significant differences in HAZ-treated animals with respect to body weight gain, feed consumption, haematology, clinical biochemistry evaluation. No significant gross pathological observation was noticed in necropsy. Conclusion: Data of the present study is substantial and scientific proof of HAZ in terms of standardization and toxicity study that can be utilize in future research activities.
