Polarization of THP-1-Derived Macrophage by Magnesium and MAGT1 Inhibition in Wound Healing

  • Background Macrophages play a major role in wound healing and prevent infection from the outside. Polarization conversion of macrophages regulates aspects of inflammation, and two macrophages, M1 (classically activated) and M2 (alternatively activated), exist at both ends of broad-spectrum macrophage polarization. Thus, we aimed to investigate whether macrophage polarization can be artificially regulated. To this end, MgSO4 and small-interfering RNA (siRNA) targeting magnesium transport 1 (MAGT1) were used to investigate the effects of intracellular magnesium (Mg2+) concentrations on the differentiation of macrophages in vitro. Methods THP-1 derived macrophages maintained in a culture medium containing 5 mM MgSO4 and siRNA to inhibit the expression of MAGT1. As comparative groups, THP-1 derived macrophages polarized into M1 and M2 macrophages by treatment with M1, M2 inducer cytokine. The polarization status of each group of cells was confirmed by cell surface antigen expression and cytokine secretion. Results We found that MgSO4 treatment increased CD163 and CD206, similar to the effect noted in the M2 group. The expression of CD80 and HLA-DR was increased in the group treated with MAGT1 siRNA, similar to the effect noted in the M1 group. Functional assays demonstrated that the group treated with MgSO4 secreted higher levels of IL-10, whereas the MAGT1 siRNA-treated group secreted higher levels of IL-6 cytokines. Additionally, the conditional medium of the Mg2+ treated group showed enhanced migration of keratinocytes and fibroblasts. Conclusion Mg2+ can help to end the delay in wound healing caused by persistent inflammation in the early stages.

Lycopene의 새로운 혈관내피세포 생리활성 (Novel Function of Lycopene in Vascular Endothelial Cell)

  • 항암 효과와 항산화 기능을 가지는 것으로 알려진 lycopene은 심혈관에서의 기능이 현재까지 잘 알려져 있지 않다. 본 연구에서는 lycopene의 심혈관 기능을 알기 위해 혈관내피세포를 이용해 다양한 세포실험을 수행하였다. 그 결과, lycopene은 내피세포의 성장 및 이동을 촉진하였으나 세포사멸에는 영향이 없었다. 또한, 백혈구의 혈관내피세포 부착을 억제하였고 내피세포 내 신호전달물질인 MAPK들의 활성을 촉발하였다. MAPK의 활성 억제제를 이용한 신호전달기전 연구실험 결과, ERK와 p38 MAPK의 활성은 세포성장에 관여하고, JNK의 활성은 세포이동에 관여함을 확인하였다. 종합하면, lycopene은 혈관내피세포의 신호전달 물질인 MAPK들를 통해 세포성장 및 이동을 촉진시키며, LPS에 의한 THP-1의 내피세포 부착을 억제 하는 등 혈관내피세포의 다양한 생리활성을 조절한다. Lycopene의 이러한 혈관내피세포 기능들은 lycopene이 혈관질환 치료제로 응용될 가능성이 있음을 의미한다.

Rapamycin-resistant and torin-sensitive mTOR signaling promotes the survival and proliferation of leukemic cells

  • The serine/threonine kinase mTOR is essential for the phosphoinositide 3-kinases (PI3K) signaling pathway, and regulates the development and function of immune cells. Aberrant activation of mTOR signaling pathway is associated with many cancers including leukemia. Here, we report the contributions of mTOR signaling to growth of human leukemic cell lines and mouse T-cell acute leukemia (T-ALL) cells. Torin, an ATP-competitive mTOR inhibitor, was found to have both cytotoxic and cytostatic effects on U-937, THP-1, and RPMI-8226 cells, but not on Jurkat or K-562 cells. All cells were relatively resistant to rapamycin even with suppressed activity of mTOR complex 1. Growth of T-ALL cells induced by Notch1 was profoundly affected by torin partially due to increased expression of Bcl2l11 and Bbc3. Of note, activation of Akt or knockdown of FoxO1 mitigated the effect of mTOR inhibition on T-ALL cells. Our data provide insight on the effect of mTOR inhibitors on the survival and proliferation of leukemic cells, thus further improving our understanding on cell-context-dependent impacts of mTOR signaling.   • There is the urgent need of new human health care's technology against cancers or tumors based on plasma electronics, medicine and biology. Main target of our study is to enhance efficacy and selectivity of plasma on cancer cells with metabolic modifiers and by inducing immune-modulations. We have evaluated the combination effect of plasma with metabolic modifiers (2-DG) on various solid and liquid cancers. Our findings suggest that 2-DG enhances the efficacy and selectivity of plasma and induces apoptosis in blood cancer cells through glucose deprivation. Finally, we conclude that 2-DG with non-thermal plasma may be used as a combination treatment against cancer cells. Our work also comprises plasma induced activation of immune cells; which find applications for curing various kinds of resistant tumors and other dreadful diseases. Plasma significantly activates immune cells which increases cell death in solid tumors in co-culture conditions.

Chunghyul-dan acts as an anti-inflammatory agent in endothelial cells by regulating gene expression

  • Chunghyul-dan (CHD) is a combinatorial drug known to exert anti-inflammatory effects in endothelial cells. In this study, we employed global transcriptional profiling using cDNA microarrays to identify molecular mechanisms responsible for the anti-inflammatory activity of CHD in endothelial cells. An analysis of the microarray data revealed that transcript levels of monocyte chemotactic protein-1 (MCP-1), vascular cell-adhesion molecule-1 (VCAM-1) and activated leukocyte cell-adhesion molecule were dramatically altered in CHD-treated endothelial cells. These changes in gene expression were confirmed by RT-PCR, Western blotting and ELISA. Chronic CHD treatment also appeared to decrease MCP-1 secretion, probably as a result of decreased MCP-1 expression. In addition, we determined that chronic CHD treatment inhibited lipopolysaccharide-stimulated adhesion of THP-1 leukocytes to endothelial cells. The inhibitory effect of CHD on LPS-stimulated adhesion resulted from downregulation of VCAM-1 expression. Transmigration of THP-1 leukocytes through endothelial cells was also inhibited by chronic CHD treatment. In conclusion, CHD controls a variety of inflammatory activities by regulating MCP-1 and VCAM-1 gene expression.

  • Background: Mycobacterial antigens released as PIM, LM, LAM, lipoproteins and other cellular factors may contribute to macrophage and dendritic cell activation through pattern recognition receptors such as TLRs. In this study, we assessed cytokine production and ERK activation with stimulation of several major mycobacterial antigens. Methods: Purified mycobacterial antigens (10, 22, 30, 38kDa) and recombinant antigens (6, 16, 19, 38kDa, Ag85A antigen) were studied. The production of cytokines (TNF-${\alpha}$, IL-12, IL-6) was measured by ELISA. The ERK activation was detected by western blotting. The expression of TLR2 or TLR4 was measured by flow cytometry. Results: Among purified antigens only 30kDa antigen induced production of IL-6 or TNF-${\alpha}$ in THP-1 macrophage cells. When THP-1 macrophage cells were treated with 30kDa antigen, phosphorylation of ERK was detected. ERK activation also occurred in TLR2 transfectant HEK293 cells with 30kDa antigen stimulation. Conclusion: 30kDa antigen is one of the major mycobacterial antigens inducing cytokine production and MAP kinases phosphorylation in macrophages.

Anti-inflammatory Effect of LFR on LPS-stimulated THP-1 Cells

  • Background and Objective: Luffae Fructus Retinervus (LFR) is used for investigating symptoms of inflammation. We have evaluated the anti-inflammatory effect of LFR by analyzing the expression of pro-inflammatory cytokines. Materials and Methods : We differentiated THP-l cells into macrophage-like cells by treatment with PMA. Inflammation was induced by treatment with LPS and PMA. We determined the safe concentration of LFR by using the MTS and MTT assays and using PD 98059 as a negative control for comparison of the anti-inflammatory effect of LFR. Results : The MTS and MTT analysis showed that the cell survival rate was >80% within the LFR concentration range of 10-100 ng/ml and began to decrease to >80% at 1 ${\mu}g/ml$. By RT-PCR analysis, the gene expression of TNF-${\alpha}$, IL-8, TGF-${\beta}$, IL-6, IL-${\beta}$1, and IL-10 levels were down-regulated when monocyte-derived macrophages were treated with concentrations of LFR between 10 ng/mL and 100 ng/mL. Conclusion : We conclude that LFR exerts an anti-inflammatory effect by inhibiting the expression of pro-inflammatory activity. The results suggest a promising way to treat general inflammatory diseases.

Interaction of Der p 2 with Toll-like Receptor 4 and its Effect on Cytokine Secretion

  • Der p 2, which is a major allergen of house dust mite, plays an important role in the pathogenesis of allergic disease. There is controversy regarding whether Der p 2 binds to Toll-like receptor 4 (TLR4), and its inflammatory effect has not yet been elucidated. In the current study, we examined the interaction of Der p 2 with TLR4 and the effect of Der p 2 on cytokine release in THP-1 cells and lymphocytes. Among house dust mite extracts, recombinant TLR4 protein interacted with Der p 2. The overall structure of Der p 2 is characteristic of the immunoglobulin superfamily and contains ten ${\beta}-strands$, forming a ${\beta}-cup$ fold with two anti-parallel ${\beta}-sheets$, and a short 310 helix. The two sheets can be separated, further allowing the formation of a large internal pocket, which is narrow and suitable for binding large flat molecules such as lipid-like molecules. Der p 2 caused increased secretion of IL-6, IL-8, and MCP-1, which are neutrophil survival factors, in human monocytic THP-1 cells in a time-dependent manner. Der p 2 also induces the release of cytokines in normal and allergic lymphocytes. Supernatant after treatment with Der p 2 inhibited neutrophil apoptosis. In coculture of lymphocytes with neutrophils, Der p 2 inhibited spontaneous apoptosis of allergic neutrophils. In summary, Der p 2 binds to TLR4 and induces an inflammatory response such as cytokine secretion in immune cells. These findings may enable elucidation of allergy pathogenesis by specific allergen of house dust mite.

결핵균이 감염된 대식세포의 면역반응을 통한 유향(Frankincense)의 항결핵효과 (Anti-tuberculosis effects of frankincense through immune responses of Mycobacterium tuberculosis-infected macrophages)

  • 현재 시판되는 항생제 및 항결핵약제 대부분은 균의 성장을 저해하는 기전에 맞추어 개발되었고, 지금도 새로운 약제들이 개발되고 있다. 이러한 약제들은 사용빈도가 증가함에 따라 자연발생적으로 약제내성이 출현하게 되고, 약제 내성 결핵의 경우 치료가 더 어려울 뿐만 아니라 치료에 소요되는 시간과 비용이 기하급수적으로 증가한다. 이런 관점에서 볼 때, 인체 면역을 활성화시켜 결핵을 치료하는 방법이 결핵균을 직접적으로 사멸시키는 방법보다 더 안전하고 효과적인 방법이 될 수 있을 것이다. 본 연구에서는 오랜 시간 동안 민간요법으로 사용되어지고 있으면서 탐식구 활성화를 촉진시키는 것으로 알려져 있지만, 항결핵 효과는 검증되어 있지 않은 FRM에 의한 항결핵효과를 검증하였다. 유향의 세포독성효과를 확인하기 위해 MTT reduction assay를 실시하여 31.3, 62.5 ㎍/mL의 농도에서 독성효과를 나타내지 않는 것을 확인하였다. 또한 형태학적 관찰을 통해서도 세포독성 효과를 나타내지 않은 것을 확인하였다. 대식세포의 면역 작용기전을 측정하는 중요한 지표인 cytokine TNF-α, IL-1β의 발현양의 변화를 통해서 면역반응을 확인하였다. 결핵균이 감염된 대조군에서는 이 cytokine들의 발현이 증가한 반면, FRM을 처리한 세포에서는 발현이 감소하였고, 결핵균 또한 감소하였다. 이러한 현상은 FRM이 necrosis를 감소시키면서 apoptosis로 면역반응을 유도하여 발생하는 것으로 가정할 수 있으며, 결과적으로 결핵균 제거 효과를 보여주었다. 현재 치료가 힘든 다제내성결핵, 광범위내성결핵에서 유향이 대식세포의 면역력 활성화를 통한 항결핵효과를 나타내는 치료제로서의 잠재적인 가능성을 제시하였다. 다만, 추가적인 항결핵효과의 검증 및 면역기전에 대한 연구가 이루어져야 할 것으로 사료된다.

양혈윤부탕(養血潤膚湯)의 면역(免疫) 조절작용(調節作用)을 통한 항알러지 효능(效能) (Inhibitory Effects of Yanghyelyoonbutang (YHYBT) on Allergic Reaction and Pro-Inflammatory Cytokines in Various Cell Lines)

  • This study saw the anti-allergy effect by the immunity regulation action of Yanghyelyoonbotang (YHYBT) consists 12 kinds of herbal medicine agents. Consequently, YHYBT controlled the amount of secretion of various infla- mmatory cytokines, chemokine, monocyte chemotactic protein and histamine from cells (HMC-1, THP-1, EoL-1) stimulated by PMA, A23187 or HDM. 1. YHYBT did not show cytotoxicity on cultured human fibroblast cells under 250 ${\mu}g/m\ell$ concentration. 2. YHYBT suppressed IL-8, TNF-$\alpha$, IL-6 mRNA expression in the HMC-1 cell stimulated with PMA and A23187. 3. YHYBT significantly suppressed IL-6 release in the THP-1 and EoL-1 cell stimulated with HDM. 4. YHYBT significantly suppressed histamine release in the HMC-1 cell stimulated with PMA and A23187 in a dose-dependent. 5. YHYBT significantly suppressed $\beta$-Hexosaminidase release in the HMC-1 cell stimulated with A23187 in a dose-dependent. 6. YHYBT suppressed NF-$\kappa$B gene expression in the RBL-2H3 cell stimulated with PMA in a dose-dependent. These results suggested that YHYBT has suppressive effects on allergic reaction and pro-inflammatory cytokines in various cell lines through the regulation of immune system. YHYBT has potential to use as an antiallergic agents.
