Development of Production Techniques for Korean Native Cattle Calves from Embryos by In Vitro Technology 3. Effects of Culture Medium on Blastocyst Development and Effects of Sucrose and Trehalose on the Survival Rate of In Vitro Developed Embryos after Thawing (체외배양기술로 생산된 초기배에 의한 한우 송아지 생산기술 III. 배반포와 발생에 미치는 발생배지의 효과와 동결융해후의 생존율에 미치는 Sucrose와 Trehalose의 효과)

  • The objective of this study was to evaluate the embryonic development ability and the appearance of blastocysts of bovine in vitro fertilized oocytes cultured in different culture media, and also to evaluate survival rate after thawing of frozen embryos by using 1.5 or 1.8M ethylene glycol(EG) with sucrose or trehalose. Fertilized oocytes were divided into three groups; i ) monolayer of cumulus /granulosa cell prepared by TGM 199+5% calf serum(TGM199), ii)GRlaa+5% CS, iii)SOF+5% CS, and they were cultured after insemination for 9 days, at 39˚C, under 5% $CO_2$ in air, but SOF+5% CS was cultured at 39˚C, under 5% 02, 5% GO2, 99% N2. Blastocysts derived from GRlaa + 5% CS on day 7~8 after insemination were frozen by using 1.5M EG or 1.8M EG with/without 0.2M sucrose or O.1M trehalose. The development rate of blastocysts on day 7 after insemination in SOF+5% CS was significant higher than in TCM199 or CR1aa(P<0.05). The appearance rate of blastocysts on day 7-8 after insemination was higher than in TCM199, when fertilized oocytes were cultured in GRlas or SOF. The survival rate of frozen blastocysts after thawing tended to increase, when blastocysts were frozen by using 1.8M EG with 0.2M sucrose or O.1M trehalose. These results indicated that SOF or CRlaa media with amino acids was superior to TCM199 with monolayer in terms of blastocyst development in culturing of in vitro fertilized bovine nocytes, and sucrose or trehalose was supposed to prevent embryos from the freezing shock.

HaCaT Keratinocytes and Primary Epidermal Keratinocytes Have Different Transcriptional Profiles of Cornified Envelope-Associated Genes to T Helper Cell Cytokines

  • HaCaT cells are the immortalized human keratinocytes and have been extensively used to study the epidermal homeostasis and its pathophysiology. T helper cells play a role in various chronic dermatological conditions and they can affect skin barrier homeostasis. To evaluate whether HaCaT cells can be used as a model cell system to study abnormal skin barrier development in various dermatologic diseases, we analyzed the gene expression profile of epidermal differentiation markers of HaCaT cells in response to major T helper (Th) cell cytokines, such as $IFN{\gamma}$, IL-4, IL-17A and IL-22. The gene transcriptional profile of cornified envelope-associated proteins, such as filaggrin, loricrin, involucrin and keratin 10 (KRT10), in HaCaT cells was generally different from that in normal human keratinocytes (NHKs). This suggests that HaCaT cells have a limitation as a model system to study the pathophysiological mechanism associated with the Th cell cytokine-dependent changes in cornified envelope-associated proteins which are essential for normal skin barrier development. In contrast, the gene transcription profile change of human ${\beta}2$-defensin (HBD2) in response to $IFN{\gamma}$, IL-4 or IL-17A in HaCaT cells was consistent with the expression pattern of NHKs. $IFN{\gamma}$ also up-regulated transglutaminase 2 (TGM2) gene transcription in both HaCaT cells and NHKs. As an alternative cell culture system for NHKs, HaCaT cells can be used to study molecular mechanisms associated with abnormal HBD2 and TGM2 expression in response to $IFN{\gamma}$, IL-4 or IL-17A.

Enhancement of skin barrier and hydration-related molecules by protopanaxatriol in human keratinocytes

  • Background: Protopanaxatriol (PPT) is a secondary intestinal metabolite of ginsenoside in ginseng. Although the effects of PPT have been reported in various diseases including cancer, diabetes and inflammatory diseases, the skin protective effects of PPT are poorly understood. Methods: HaCaT cells were treated with PPT in a dose-dependent manner. mRNA and protein levels which related to skin barrier and hydration were detected compared with retinol. Luciferase assay was performed to explore the relative signaling pathway. Western blot was conducted to confirm these pathways and excavated further signals. Results: PPT enhanced the expression of filaggrin (FLG), transglutaminase (TGM)-1, claudin, occludin and hyaluronic acid synthase (HAS) -1, -2 and -3. The mRNA expression levels of FLG, TGM-1, HAS-1 and HAS-2 were suppressed under NF-κB inhibition. PPT significantly augmented NF-κB-luc activity and upregulated Src/AKT/NF-κB signaling. In addition, PPT also increased phosphorylation of the mitogen-activated protein kinases (MAPKs) ERK, JNK and p38 and upstream MAPK activators (MEK and MKK). Furthermore, transcriptional activity of AP-1 and CREB, which are downstream signaling targets of MAPK, was enhanced by PPT. Conclusion: PPT improves skin barrier function and hydration through Src/AKT/NF-κB and MAPK signaling. Therefore, PPT may be a valuable component for cosmetics or treating skin disorders.

  • Mechanical structures of military equipment have been mainly applied with black oxide coating due to the limitation of surface treatment thickness. However, the mechanical structures applied by the black oxide coating treatment is constantly being corroded by calcium chloride and humidity. Since this can cause serious problems in the operation of equipment, a review to improve surface treatment and corrosion resistance is required. Therefore, in this study, surface treatment methods that can enhance corrosion resistance were selected and corrosion resistance performance was verified through experiments describing harsh field conditions. Thus, applying a proven surface treatment method to future military equipment will prevent corrosion.

Isolation and identification of antifungal compounds from Spatholobus suberectus Dunn (계혈등(Spatholobus suberectus Dunn)으로부터 항균활성 물질의 분리 및 구조결정)

  • In the continued research on natural fungicides for control of plant diseases by using plant-derived products, we found that Spatholobus suberectus Dunn had a strong fungicidal activity against several plant pathogens. S. suberectus (1 kg) was extracted with 80% aqueous MeOH and then the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH and $H_2O$ successively. The four layers were tested their disease contron efficacies against 6 plant diseases such as rice blast (RCB), rice sheath blight (RSB), tomato grey mold (TGM), tomato late blight (TLB), wheat leaf rust (WLR), and barley powdery mildew (BPM). The EtOAc fraction was highly active showing over 80% control against RCB, TGM, TLB, and BPM. By using silica gel chromatography, preparative TLC and HPLC, six compounds that were expected to have antifungal activity were separated. Their chemical structures were identified as ethanone, hydroxytyrosol, epicatechin, procyanidin B2, dimethoxy daizein and formononetin by ESI-MS, $^1H$-NMR, $^{13}C$-NMR, and 2D-NMR spectroscopic analyses. The chemicals except epicatechin were first reported in S. suberectus. Study on in vitro and in vivo antifungal activities of the isolated compounds is in progress.

Skin Hydration Effect of Brasenia schreberi Mucilage Polysaccharide Extract (순채 점액질 다당체 추출물의 피부 보습 효과 연구)

  • In this study, we evaluated the skin moisturizing effect of Brasenia schreberi (B. schreberi) mucilage polysaccharide on human skin and in vitro and the potent cosmetic ingredient for skin. To protect skin from various environmental stresses and aging, we should increase moisture content of skin and prevent water loss. We have found that polysaccharides extracted from mucilage of B. schreberi improved the roughness of skin with its lubricating behavior. In vitro, the expression of transglutaminase 1 (TGM1) gene, which plays a role in cross-linking the skin barrier, was increased when the keratinocytes were treated with B. schreberi polysaccharides. In addition, the expression of hyaluronan synthase 3 (HAS3) gene, an enzyme that synthesizes water-binding matrix hyaluronic acid, aquaporin 3 (AQP3), which regulates the movement of water and glycerol were also increased. In addition, an experiment to evaluate its potential as a cosmetic ingredient has shown anti-inflammatory and collagen synthesis-promoting effects. As a result, the mucilaginous polysaccharide from natural products which has not existed before, showed moisturizing effect, anti-inflammation and collagen synthesis-promoting effects for skin protection and hydration.

Effect of Halophyte (Spartina anglica and Calystegia soldanella) Extracts on Skin Moisturizing and Barrier Function in HaCaT Cells (염생식물인 갯끈풀과 갯메꽃 추출물의 HaCaT 세포에서 피부 보습 및 피부 장벽 기능에 미치는 영향)

  • As aging progresses, reactive oxygen species (ROS) reduces skin moisturization and collapses skin barrier function. In this study, we evaluated the efficacy of skin moisturizing and skin barrier function enhancement by extracts from halophytes using HaCaT cells. Spartina anglica (S. anglica; SAE) and Calystegia soldanella (C. soldanella; CSE), a kind of halophytes, were collected from Dongmak beach in Incheon, and extracted with 70% ethanol. At the first, we evaluated the cytotoxicity of extracts in HaCaT cell using WST-8 Kit. As a result, the other experiment was conducted by setting the concentration at which the cell viability was 90% or more. SAE and CSE showed high radical scavenging activity through ABTS assay. Expression levels of genes related to skin moisturizing and skin barrier functions, were analyzed by real-time qPCR. As a result, it showed that the expression of aquaporin 3, hyaluronan synthase 2, and transglutaminase 1 was increased by SAE treatment but not changed by CSE. Activation of extracellular signal regulated kinase (ERK), c-Jun N-terminal kinase (JNK), and p38 mitogen activated protein kinase was induced by SAE. These results suggest that SAE can be used as functional materials for cosmetics for skin moisturizing and barrier function enhancement.

A Review on Total Gaseous Mercury Concentration Levels in the East Asia

  • In this work, the present concentration levels of total gaseous mercury (TGM) are compared among three East Asian countries, Korea, China, and Japan. Comparison of Hg distribution patterns was made by selecting representative figures for each type of land use among those three countries. The results of the analysis indicate that Hg concentrations within a country can fall into a wide range due probably to the diversity of source processes. It is seen that the urban areas of China and Japan generally exhibit large spatial variability with notably high Hg levels (above 10 ng m$^{-3}$), compared to their Korean counterparts. Although the presence of high Hg levels in Chinese locations can be accounted for by the major man-made source processes (e.g., the use of coal), the causes of unexpectedly high Hg data in Japanese sites appear to be rather complicated. The Hg concentration levels in relatively remote locations however show much reduced values, above 3 ng m$^{-3}$, which is still higher than the typical background concentrations of 1 to 2 ng m$^{-3}$ in Europe or America. Considering that the presence of unusually high Hg levels in urban areas of Asia is the consequence of man-made activities, the prevalence of excessively high Hg values in certain regions of the Asian continent needs further research to accurately assess the fundamental picture of Hg geochemistry in the East Asia.

Effect of Co-culture with Porcine Endometrial Cell Monolayers on the Development of In Vitro Produced Porcine Zygotes (자궁내막세포막의 공배양이 돼지 체외수정란의 초기발달에 미치는 영향)

  • This study was conducted to investigate the effects of co-culture for the development rate to morula /blastocyst stages of early porcine embryos, derived from oocytes matured and fertilized in vitro, with porcine endometrial cell monolayers(PEM) in the two different media, respectively. The rates of embryos developed to 2-, 4-, 8~16-cell and morula /blastocyst stage were 49.6, 40.5, 28.2 and 15.3% in Ham's F-10 with PEM, and 55.3, 45.9, 32.7, and 17.6% in TCM-HEPES with PEM, respectively. The above development rates to morula /blastocyst stages were significantly higher than those of the embryos cultured in the Ham's F-10 and TGM-HEPES without PEM(P<0.05). The in vitro development rates to the morula /blastocyst stage of 1-cell embryos cultured in Ham's F-10 and TCM-HEPES without PEM were 0~1.2%. Especially, most of embryos were observed to arrest the development beyond 4-cell stages. As shown in the above results, the co-culture of in vitro produced porcine embryos with PEM in the two different media enhanced the development of fertilized eggs to morula /blastocyst stages in vitro. However, we didn't find out any differences for the in vitro development to morula /blastocyst stages between Ham's F-10 and TcM-HEPES media.

Isolation and identification of antifungal compounds from Reynoutria elliptica (호장근(Reynoutria elliptica)으로부터 항균활성 물질의 분리 및 구조결정)

The four layers were tested their disease control efficacies against 4 plant disease such as rise blast (RCB), tomato grey mold (TGM), tomato late blight (TLB), and barly powdery mildew (BPM). The n-hexane fraction was highly active showing over 95% control against TLB and BPM. and the EtOAc fraction was highly active showing over 95% control against RCB, TLB, and BPM. By using silica gel chromatography, MPLC, and HPLC, three compounds that were expected to have antifungal activity were isolated. Their chemical structures were elucidated as physcion, emodin, and emodie-8-O-glucoside by EI-MS and NMR spectroscopic analyses.
