Synthesis of Reconstituted High Density Lipoprotein (rHDL) Containing apoA-I and apoC-III: the Functional Role of apoC-III in rHDL

  • Apolipoprotein (apo) C-III is a marker protein of triacylglycerol (TG)-rich lipoproteins and high-density lipoproteins (HDL), and has been proposed as a risk factor of coronary heart disease. To compare the physiologic role of reconstituted HDL (rHDL) with or without apoC-III, we synthesized rHDL with molar ratios of apoA-I:apoC-III of 1:0, 1:0.5, 1:1, and 1:2. Increasing the apoC-III content in rHDL produced smaller rHDL particles with a lower number of apoA-I molecules. Furthermore, increasing the molar ratio of apoC-III in rHDL enhanced the surfactant-like properties and the ability to lyse dimyristoyl phosphatidylcholine. Furthermore, rHDL containing apoC-III was found to be more resistant to particle rearrangement in the presence of low-density lipoprotein (LDL) than rHDL that contained apoA-I alone. In addition, the lecithin:cholesterol acyltransferase (LCAT) activation ability was reduced as the apoC-III content of the rHDL increased; however, the CE transfer ability was not decreased by the increase of apoC-III. Finally, rHDL containing apoC-III aggravated the production of MDA in cell culture media, which led to increased cellular uptake of LDL. Thus, the addition of apoC-III to rHDL induced changes in the structural and functional properties of the rHDL, especially in particle size and rearrangement and LCAT activation. These alterations may lead to beneficial functions of HDL, which is involved in anti-atherogenic properties in the circulation.

The Effects of Weight Training by Intensity for 8 Weeks of Metabolic Syndrome Factor Improvement in Overweight High School Students (8주간의 강도별 웨이트트레이닝이 비만고교생의 대사증후군 인자 개선에 미치는 영향)

  • The purpose of this study was to examine the most effective weight training (WT) intensity for the improvement of metabolic syndrome factors in overweight high school students. Sixty overweight high school students were randomly divided into 4 groups (15 subjects/group) according to WT intensity: A group - control group, B group - 40-55% repetition maximum (RM) group, C group - 60-75% RM group, and D group - 80-90% RM group. They exercised between 60-70 minutes a day, three times a week, for eight weeks. In order to determine the most effective WT intensity, five metabolic syndrome factors--waist size, fasting glucose, triglyceride (TG), high-density lipoprotein-cholesterol (HDL-C) and blood pressure--were measured for each subject prior to and after the experiment. Waist size decreased in all exercise groups, but the results were more prominent in the 40-55% RM and 60-75% RM groups. TG levels also decreased in all groups but results were more prominent in the 40-55% RM group. HDL-C levels in the 80-90% RM group were significantly higher than the control group. Groups did not show any significant difference in the diastolic blood pressure, but the 40-55% RM group's systolic blood pressure significantly decreased compared to other groups. Groups did not show significant difference in the fasting glucose level. These results indicate that lower-intensity weight training is generally more effective than the higher-intensity weight training in improving metabolic syndrome factors in overweight high school students and that the most effective WT intensity is 40-55% RM.

The Effect of Music Rope-jumping with Weight Loading on Middle School Girls` Blood Lipids and Growth Hormone (중량부하 음악줄넘기 운동이 여자중학생의 혈중지질과 성장호르몬에 미치는 영향)

  • The purpose of this study is to identify the effect of rope-jumping to music with weight loading on middle school girls' blood lipid and growth hormone. Toward this end, the study targeted 24 middle school girls at Y Girls' Middle School in Yangsan, Gyeongnam, who have no health problems or exercise experience, after sufficiently explaining to them the study's purpose. The selected students were categorized into a music rope-jumping group (8), a music rope-jumping group with weight loading (8), and a control group (8). In a comparison of blood lipid within the exercise groups showed that the music rope-jumping groups' measures changed significantly by demonstrating increases or decreases in RBC, T-C, TG, HDL-C and LDL-C (p<0.05). The music rope-jumping group with weight loading showed significant differences in WBC, RBC, glucose, T-C, TG, HDL-C and LDL-C by demonstrating increases of decreases (p<0.05). A comparison between the exercise groups revealed that the music rope-jumping group showed significant increases in growth hormone (p<0.05). The music rope-jumping group with weight loading also showed significant increases in growth hormone (p<0.05). The study's results identified music jump-roping with weight loading to effect students' blood lipids and growth hormone.

Effect of Genetic Predisposition on Blood Lipid Traits Using Cumulative Risk Assessment in the Korean Population

  • Dyslipidemia, mainly characterized by high triglyceride (TG) and low high-density lipoprotein cholesterol (HDL-C) levels, is an important etiological factor in the development of cardiovascular disease (CVD). Considering the relationship between childhood obesity and CVD risk, it would be worthwhile to evaluate whether previously identified lipid-related variants in adult subjects are associated with lipid variations in a childhood obesity study (n = 482). In an association analysis for 16 genome-wide association study (GWAS)-based candidate loci, we confirmed significant associations of a genetic predisposition to lipoprotein concentrations in a childhood obesity study. Having two loci (rs10503669 at LPL and rs16940212 at LIPC) that showed the strongest association with blood levels of TG and HDL-C, we calculated a genetic risk score (GRS), representing the sum of the risk alleles. It has been observed that increasing GRS is significantly associated with decreased HDL-C (effect size, $-1.13{\pm}0.07$) compared to single nucleotide polymorphism combinations without two risk variants. In addition, a positive correlation was observed between allelic dosage score and risk allele (rs10503669 at LPL) on high TG levels (effect size, $10.89{\pm}0.84$). These two loci yielded consistent associations in our previous meta-analysis. Taken together, our findings demonstrate that the genetic architecture of circulating lipid levels (TG and HDL-C) overlap to a large extent in childhood as well as in adulthood. Post-GWAS functional characterization of these variants is further required to elucidate their pathophysiological roles and biological mechanisms.

Relationships Between Alcohol Intake and Blood Pressure, Serum Lipids, Liver Function Tests and Obesity Indices in Middle-Aged Male Workers (중년 남성 근로자의 알코올 섭취와 혈압, 혈청 지질, 간기능검사치 및 비만지표와의 관련성)

  • The purpose of this study was to investigated the effects of alcohol intake on blood pressure, serum lipids, liver function tests and obesity indices in middle-aged male workers. The 2,175 male workers aged 30-59 years who examined health checkup were recruited as the study subjects during the period between January to December, 2013. We observed the relationships between the amount of alcohol intake and blood pressure, serum lipids, liver function tests and obesity indices. As a result, the mean score of SBP, DBP, LDL-C and TG were significantly higher increasing with amount of alcohol increased, but HDL-C were significantly lower increasing with amount of alcohol increased. AST, ALT, ALP and ${\gamma}$-GTP were significantly higher increasing with amount of alcohol increased, but obesity indices were not significant. Age adjusted odds ratios(ORs) increasing with abnormal levels of SBP, DBP, HDL-C, LDL-C, TG, AST and ALT were significantly increased in heavy drinking group than light drinking group. Above results suggested that the heavy drink related with blood, serum lipid levels and liver function test levels, but not find out with obesity indices.

Effects of 12-week aquarobics exercise on the blood lipids, atherogenic index and liver functions of elderly women (12주간의 아쿠아로빅 운동이 여성 노인의 혈중지질, 동맥경화지수 및 간 기능에 미치는 영향)

  • The purpose of this study was to investigate the effects of 12-week aquarobics exercise on blood lipids, atherogenic index, and liver functions in elderly women. The subjects for the study were 25 healthy elderly women volunteers, aged 65 to 80 years, composed of the aquarobics exercise group (n=13) and control group (n=12). Twice a week for 60 minutes for 12 weeks. The exercise intensity was set to RPE 11-12 for 1 week to 4 weeks, RPE 13-14 for 5 week to 8 weeks and RPE 15-16 for 9 week to 12 weeks respectively. The test data were analyzed by two-way repeated measures ANOVA, paired t-test, and independent t-test. The alpha level of .05 was set for all tests of significance. The results of the study in the aquarobics exercise group were as follows; LDL-C (p<.05), AST (p<.05) had significantly decreased, with TC (p<.01), TG (p<.05), LDL-C (p<.05), LDL-C/HDL-C (p<.05), TG/HDL-C (p<.05), AST (p<.001) significantly lower than control group. Consequently, these findings suggest that regular aquarobics exercise will be effective for preventiy the metabolic syndrome and cardiovascular health in elderly women.

Relationship between Lipoprotein(a) and Dyslipidemia in the Elderly over 60 Years (60세 이상 노인에서 혈중 지단백(a)와 이상지질혈증의 관련성)

  • The relationship between lipoprotein(a) and dyslipidemia is not clear. This study was therefore undertaken to investigate the relationship between lipoprotein(a) and dyslipidemia in elderly patients over 60 years. From 2014 to 2020, a total of 2,580 adults aged 60 years or older (73.31±7.24 years, 1,954 males) were enrolled in the study. The patients had checked into a general hospital, and data were obtained for lipoprotein(a), LDL-C, TG, HDL-C, hs-CRP, HbA1c, sex, age, BMI, dyslipidemia diagnosis, and use of lipid-lowering agents. BMI and HbA1c showed no correlation with lipoprotein(a), but hs-CRP (r=0.138), LDL-C (r=0.097), HDL-C (r=-0.089), TG (r=-0.073), and age (r=0.072) were significantly correlated to lipoprotein(a). The partial correlation between lipoprotein(a) and LDL-C, which was adjusted for variables, was significant only in the male gender (r=0.158, P<0.001). As the odds ratio of the 4th quartile of lipoprotein(a) (OR=1.376, 95% CI=1.038~1.822) for dyslipidemia was found to be significant in this study when the level of LDL-C, the primary target, could not be reduced even by taking lipid-lowering drugs, we propose that lipoprotein(a) should also be included among the several factors considered as secondary targets. Our results indicate that studies on various lipid factors considering the sex, age, types and use of lipid-lowering agents, are warranted.

Effects of Kimchi on Tissue and Fecal Lipid Composition and Apolipoprotein and Thyroxine Levels in Rats (흰쥐에서 김치식이가 조직과 분변의 지질조성과 Apo단백 및 Thyroxine 농도에 미치는 영향)

  • This study was carried out to examine whether kimchi has hypolipidemic effect and to know how it exert lipid-lowering effect in rats. Male Sprague-Dawley rats were fed with kimchi-fee diet, or 3%, or 5%, or 10% kimchi diets for 6 weeks. Plasma cholesterol level was lowered in rats fed all concentrations of kimchi diets, and plasma triglyceride(TG) level was lowered in 10% kimchi diet group compared with that of control significantly(p<0.05). Th intake of kimchi lowered VLDL-cholesterol and VLDL-TG levels, whereas increased HDL-cholesterol level significantly(p<0.05). LDL-cholesterol level was lowered only in 5% kimchi diet group and LDL-TG level was lowered in all kimchi diet groups compared with those of control significantly (p<0.05). the intake of 5% and 10% kimchi diets also lowered the levels of hepatic cholesterol, TG, total lipid, and apolipoprotein B, whereas increased the levels of fecal total fat, cholesterol, TG, and apolipoprotein A-1 significantly(p<0.05). Triiodothyronine(T$_3$) level was elevated in rats fed kimchi diet, whereas thyroxine(T$_4$) level was not affected by kimchi treatment. These observations support that the intake of kimchin in rats loweres plasma and hepatic lipid levels by increasing the excretion of TG and cholesterol through feces, by the elevation of T$_3$ level, and by the altered lipoprotein metabolism.

Effect of Pyroligneous Liquor on Lipid Metabolism in Serum of CD Rats (혈청의 지질대사에 미치는 목초액 (Pyroligneous Liquor)의 영향)

  • This study was designed to investigate the effects of pyroligneous liquor on lipid metaboilsm in serum of SPF/VAF Cri/Bgi CD rats. Male CD rats were fed basic diet prepared in our Lab., and PL-0 (control), PL-1, PL-25, PL-50, PL-75 groups were prepared with pyroligneous liquor (35% of Choa Co. Ltd.) of 0%, 1%, 25%, 50% and 75% in purified water per day for 8 weeks. Body weights and food intakes almost did not change in these five groups, and triglyceride (TG) levels were significantly decreased (20.5% and 19.5%, respectively) in these PL-25 and PL-50 groups compared with control group. Total and LDL-cholesterol levels were markedly decreased (20.7%, 16.3% and 20.3%, 116.9%, respectively) in these PL-25 and PL-50% groups, but HDL-cholesterol levels were significantly increased about 36.4% and 25.5% in serum of PL-25 and PL-50 groups compared with control group. Atherogenic indices were also remarkedly decreased (63.0%, and 50%, respectively) in these PL-25 and PL-50 groups compared with control group. These results suggest that long term administration orally of pyroligneous liquor in 25 to 50% may inhibit chronic degenerative disease through improving of lipid metabolism of lower TG & LDL-cholesterol and higher HDL-cholesterol, and lower AJ.

Effects of Music Rope-Skipping exercise on health fitness, blood lipids and growth-related factors in male middle school Boys (음악줄넘기가 남자중학생의 건강체력, 혈중지질, 성장관련인자에 미치는 영향)

  • The purpose of this study was to find out the effects of music rope-skipping on health fitness, blood lipids and growth-related factors in male middle school students. For this purpose, twenty subjects were participated in this study and the participants were 'After School Class' boys in B middle school located in Gimhae City. They divided into exercise group(n=10) and control group(n=10). Exercise group conducted 45minutes per a day, four times a week for 12 weeks. And the control group continued their normal life. The results were as follows; First, in exercise group, lean body mass, muscular strength, muscular endurance, cardiovascular endurance and flexibility were all significantly increased after program execution. But %body fat was significantly decreased. In control group, muscular strength, muscular endurance and cardiovascular endurance were significantly increased. There were significant differences in lean body mass, %body fat, muscular strength and flexibility between two groups after program execution. Second, in exercise group, HDL-C was significantly increased after program execution. In control group, TG was significantly decreased. But HDL-C was significantly increased. There were no significant differences in all blood lipids(TC, TG, HDL-C and LDL-C) between two groups after program execution. Third, in exercise group, GH and IGF-I were significantly increased after program execution. In control group, IGF-I was significantly decreased. There were significant differences in IGF-I between two groups after program execution. These results indicate that music rope-skipping is effective to improve lean body mass, muscular strength, flexibility, GH and IGF-I in normal middle school male students.
